Growth and synthesis of rubratoxin by Penicillium rubrum in a chemically defined medium fortified with organic acids and intermediates of the tricarboxylic acid cycle.

A sterile glucose-mineral salts broth was fortified with equimolar concentrations (10--3 M) of various organic acids and intermediates in the tricarboxylic acid cycle. Appropriate media were neutralized with 2 N NaOH, inoculated with spore suspensions or mycelial pellets of Penicillium rubrum and incubated quiescently for 14 days or with shaking for 5 days. Rubratoxins were recovered from culture filtrates by ether extraction and resolved by thin-layer chromatography. Toxin formation in quiescent cultures was enhanced by malonate but was not markedly affected by ethyl malonate, shikimate, and

Fisetinidin chloride ameliorates carbon tetrachloride-induced hepatotoxicity in HepaRG cells.

Liver diseases poses a significant global health burden. This study investigated the hepatoprotective effects of fisetinidin chloride (FC), a natural flavonoid, against carbon tetrachloride (CCl₄)-induced hepatotoxicity in HepaRG cells, a model mimicking human hepatocyte responses. Co-treatment with FC restored cell viability and reduced cellular steatosis, and minimized lactate dehydrogenase leakage, demonstrating membrane stabilization. FC mitigated oxidative stress by reducing mitochondrial reactive oxygen species (ROS) and lipid peroxidation, while enhancing antioxidant defenses through up

Uncovering metabolic shifts: an ¹H-NMR study on the dual influence of HIV/TB coinfection and ART.

BACKGROUND: Human immunodeficiency virus (HIV) and tuberculosis (TB) coinfection pose a substantial global health burden, with each infection amplifying the immune and metabolic impacts of the other. Investigating the compounded metabolic disruptions in HIV/TB coinfection and how these are modulated by antiretroviral therapy (ART) offers information into disease mechanisms and therapeutic strategies. METHODS: This study used untargeted proton nuclear magnetic resonance metabolomics to characterize the urine samples from three participant groups: healthy controls, treatment-naive HIV/TB coinfe

Two cycles of chemotherapy modulate itaconic acid levels via ATF3/TET2/NF-κB pathways in ovarian cancer among Han Chinese women.

BACKGROUND: The metabolic rationale chemotherapy cycles after debulking surgery in ovarian cancer (OC) remain unclear. This prospective cohort was aimed to define metabolic changes induced by chemotherapy and assess longitudinal effects. METHODS: Metabolites between treatment-naïve OC patients (n = 26) and age-matched healthy controls (n = 30) were identified by gas chromatography-mass spectrometry (GC-MS). Chemotherapy-related signaling molecules were quantified via enzyme-linked immunosorbent assay. RESULTS: Our analysis identified 38 differentially expressed metabolites, including critical

Inflammation-Responsive Polyion Complex Vesicles for Autoimmune Disease Therapy via Cell-Free DNA Scavenging and Inflammatory Microenvironment Modulation.

Cell-free DNA (cfDNA) scavenging represents a promising anti-inflammatory modality for autoimmune disease (AID) treatment. However, it remains challenging for existing systems to achieve inflammation-targeted cfDNA scavenging and the management of cfDNA-unrelated inflammatory pathways. Herein, inflammation-responsive polyion complex vesicles (PICsomes) are developed, bridging inflammation-instructed cfDNA scavenging, and methotrexate (MTX) delivery for AID management. A positively charged, PEGylated polypeptide with guanidine side chains (PEG-PG) is developed, which self-assembles with a negat

Method based on GC-MS to study the influence of tricarboxylic acid cycle metabolites on cardiovascular risk factors.

Metabolites involved in the tricarboxylic acid (TCA) cycle have previously been proposed as cardiovascular biomarkers. This cycle plays a key role in cell metabolism and the levels of the involved metabolites can also be affected by other physiological factors. The influence of three cardiovascular risk factors such as obesity, hypercholesterolemia, and smoking habit on serum levels of TCA-cycle metabolites has been studied in patients diagnosed with significant coronary lesion. For this purpose, a method based on GC-MS for determination of the target metabolites (viz. citric/isocitric, pyruvi

Testicular ACE regulates sperm metabolism and fertilization through the transcription factor PPARγ.

Testis angiotensin-converting enzyme (tACE) plays a critical role in male fertility, but the mechanism is unknown. By using ACE C-domain KO (CKO) mice which lack tACE activity, we found that ATP in CKO sperm was 9.4-fold lower than WT sperm. Similarly, an ACE inhibitor (ACEi) reduced ATP production in mouse sperm by 72%. Metabolic profiling showed that tACE inactivation severely affects oxidative metabolism with decreases in several Krebs cycle intermediates including citric acid, cis-aconitic acid, NAD, α-ketoglutaric acid, succinate, and L-malic acid. We found that sperms lacking tACE activi

Metabolite therapy guided by liquid biopsy proteomics delays retinal neurodegeneration.

BACKGROUND: Neurodegenerative diseases are incurable disorders caused by progressive neuronal cell death. Retinitis pigmentosa (RP) is a blinding neurodegenerative disease that results in photoreceptor death and progresses to the loss of the entire retinal network. We previously found that proteomic analysis of the adjacent vitreous served as way to indirectly biopsy the retina and identify changes in the retinal proteome. METHODS: We analyzed protein expression in liquid vitreous biopsies from autosomal recessive (ar)RP patients with PDE6A mutations and arRP mice with Pde6ɑ mutations. Proteo

The FAD-dependent tricarballylate dehydrogenase (TcuA) enzyme of Salmonella enterica converts tricarballylate into cis-aconitate.

Tricarballylate is the causative agent of grass tetany, a ruminant disease characterized by acute magnesium deficiency. Tricarballylate toxicity has been attributed to its ability to chelate magnesium and to inhibit aconitase, a Krebs cycle enzyme. Neither the ruminant nor the normal rumen flora can catabolize tricarballylate to ameliorate its toxic effects. However, the gram-negative enterobacterium Salmonella enterica can use tricarballylate as a carbon and energy source, providing an opportunity to study the genes and enzymes required for tricarballylate catabolism. The tricarballylate util

Functional metabolite signatures reveal biochemical recovery trajectories in oil-polluted soils undergoing bioremediation.

Reliable candidate indicators of recovery following petroleum contamination remain limited, as remediation success is commonly assessed using hydrocarbon removal alone rather than restoration of soil characteristics. This study integrated physicochemical profiling with untargeted liquid chromatography-mass spectrometry (LC-MS) metabolomics to resolve biochemical recovery trajectories across polluted soil (PS), soil undergoing bioremediation (UB), bioremediated soil (BS), and uncontaminated reference soil (CNTR) in Ogoniland, Nigeria. Total petroleum hydrocarbon (TPH) concentrations decreased p

Independent pathways can transduce the life-cycle differentiation signal in Trypanosoma brucei.

African trypanosomes cause disease in humans and livestock, generating significant health and welfare problems throughout sub-Saharan Africa. When ingested in a tsetse fly bloodmeal, trypanosomes must detect their new environment and initiate the developmental responses that ensure transmission. The best-established environmental signal is citrate/cis aconitate (CCA), this being transmitted through a protein phosphorylation cascade involving two phosphatases: one that inhibits differentiation (TbPTP1) and one that activates differentiation (TbPIP39). Other cues have been also proposed (mild ac

Metabolomic and immunological signatures of asthma severity in children.

BACKGROUND: Multiple risk factors for asthma severity have been identified by epidemiological studies. Yet, the pathophysiological mechanisms driving the severity of clinical asthma manifestations remain incompletely understood. In asthmatic children, Notch4 expression on circulating Treg cells and levels of circulating GDF15 have been shown to be increased as a function of disease severity, suggesting a contribution of Treg dysfunction to disease phenotype. METHODS: 126 children with asthma (intermittent = 40; mild persistent = 43; moderate persistent = 29, and severe persistent = 14) and 83

Integrative metagenomics and metabolomics reveal age-associated gut microbiota and metabolite alterations in a hamster model of COVID-19.

Aging is a key contributor of morbidity and mortality during acute viral pneumonia. The potential role of age-associated dysbiosis on disease outcomes is still elusive. In the current study, we used high-resolution shotgun metagenomics and targeted metabolomics to characterize SARS-CoV-2-associated changes in the gut microbiota from young (2-month-old) and aged (22-month-old) hamsters, a valuable model of COVID-19. We show that age-related dysfunctions in the gut microbiota are linked to disease severity and long-term sequelae in older hamsters. Our data also reveal age-specific changes in the

Akkermansia muciniphila reduces neuroinflammation and Aβ deposition via tryptophan metabolism in the APP/PS1 mouse model of Alzheimer's disease.

Akkermansia muciniphila (A. muciniphila), a beneficial gut bacterium, has increasingly attracted interests in Alzheimer's disease (AD) research, its specific role in the microbiota-gut-brain axis still remains unclear. In this study, we demonstrated that A. muciniphila administration improve cognitive deficits and reduce amyloid-beta (Aβ) deposition in APP/PS1 mice, a transgenic model of AD. Subsequently, it is revealed that A. muciniphila administration significantly alters gut microbiota diversity and composition. Mechanically, our metabolomics analysis of cecal contents indicates A. mucinip

Suppression of HIV-TAT and cocaine-induced neurotoxicity and inflammation by cell penetrable itaconate esters.

HIV-associated neurological disorder (HAND) is a serious complication of HIV infection marked by neurotoxicity induced by viral proteins like Tat. Substance abuse exacerbates neurocognitive impairment in people living with HIV. There is an urgent need for therapeutic strategies to combat HAND comorbid with Cocaine Use Disorder (CUD). Our analysis of HIV and cocaine-induced transcriptomes in primary cortical cultures revealed significant overexpression of the macrophage-specific gene aconitate decarboxylase 1 (Acod1). The ACOD1 protein converts the tricarboxylic acid intermediate cis-aconitate

Assessment of taste profile dynamics during crabapple (Malus prunifolia (Willd.) Borkh.) ripening by metabolomics and electronic tongue analysis.

Crabapple (Malus prunifolia (Willd.) Borkh) is valued for its ornamental appearance, effectiveness as a rootstock, and rich nutritional profile. However, the flavor profile and metabolite composition are poorly characterized, which hinders a comprehensive assessment of its overall value. This study combined broad-spectrum untargeted metabolomics with electronic tongue analysis to systematically characterize the flavor and metabolite profiles of M. prunifolia across four distinct ripening stages, thereby clarifying the relationship between flavor and metabolites and uncovering the metabolic bas

Lysine biosynthesis in microbes: relevance as drug target and prospects for β-lactam antibiotics production.

Plants as well as pro- and eukaryotic microorganisms are able to synthesise lysine via de novo synthesis. While plants and bacteria, with some exceptions, rely on variations of the meso-diaminopimelate pathway for lysine biosynthesis, fungi exclusively use the α-aminoadipate pathway. Although bacteria and fungi are, in principle, both suitable as lysine producers, current industrial fermentations rely on the use of bacteria. In contrast, fungi are important producers of β-lactam antibiotics such as penicillins or cephalosporins. The synthesis of these antibiotics strictly depends on α-aminoadi
