Taurine-magnesium coordination compound, a potential anti-arrhythmic complex, improves aconitine-induced arrhythmias through regulation of multiple ion channels.

Taurine-magnesium coordination compound (TMCC) exhibits antiarrhythmic effects in cesium-chloride-and ouabain-induced arrhythmias; however, the mechanism underlying these effects on arrhythmia remains poorly understood. Here, we investigated the effects of TMCC on aconitine-induced arrhythmia in vivo and the electrophysiological effects of this compound in rat ventricular myocytes in vitro. Aconitine was used to induce arrhythmias in rats, and the dosages required to produce ventricular premature contraction (VPC), ventricular tachycardia (VT), ventricular fibrillation (VF), and cardiac arrest

Increased concentration of plasma cyclic GMP from aconitine-induced atrial fibrillation in dogs and patients with paroxysmal atrial fibrillation.

The concentrations of plasma cyclic nucleotides in patients with atrial fibrillation and in dogs with aconitine-induced atrial fibrillation were measured. Radioimmunoassay was used for the determination of cyclic nucleotides. The concentration of plasma cGMP in dogs with aconitine-induced atrial fibrillation was significantly elevated at 30 and 60 min after the onset of atrial fibrillation, whereas that of plasma cAMP showed no significant changes. After the injection of atropine in dogs with atrial fibrillation and with increased plasma cGMP, the plasma levels decreased to the normal level at

Integrating proteomic, lipidomic and metabolomic data to construct a global metabolic network of lethal ventricular tachyarrhythmias (LVTA) induced by aconitine.

Lethal ventricular tachyarrhythmias (LVTA)-related sudden cardiac death (SCD) is one of the major causes of death worldwide. However, the mechanisms underlying LVTA induced by myocardial ion channel diseases (MICDs) are not yet fully understood. Here, we produced an LVTA rat model induced by aconitine, to mimic MICDs-elicited LVTA, and constructed a global pathway network via integrating proteomic and lipidomic data, and our previously published metabolomic data. Results showed that both proteome and lipidome were disturbed during the LVTA process. Most of the differentially expressed proteins

Aconitine induces mitochondrial energy metabolism dysfunction through inhibition of AMPK signaling and interference with mitochondrial dynamics in SH-SY5Y cells.

Aconitine, a highly toxic alkaloid derived from Aconitum L., affects the central nervous system and peripheral nervous system. However, the underlying mechanism of aconitine-induced neurotoxicity remains unclear. This study investigates the effects and mechanism of aconitine on mitochondrial energy metabolism in SH-SY5Y cells. Results demonstrated that aconitine exposure suppressed cell proliferation and led to an increase in reactive oxygen species (ROS) and excessive lactate dehydrogenase (LDH) release. Aconitine (400 μmol/L) induced abnormal mitochondrial energy metabolism that quantified b

Assessment of potential disease-drug interactions when prescribed Fuzi for rheumatoid arthritis treatment, with focus on the pharmacokinetics of aconitine, mesaconitine, and hypaconitine.

ETHNOPHARMACOLOGICAL RELEVANCE: Fuzi is a toxic traditional Chinese medicine that is commonly prescribed for rheumatoid arthritis (RA), while CYP3A4 and P-gp are primarily responsible for the metabolism and efflux of its toxic alkaloids: aconitine (AC), mesaconitine (MA), and hypaconitine (HA). However, the RA condition may induce disease-drug interactions due to the elevated IL-6 and TNF-α levels, which significantly inhibit the function of CYPs and P-gp. AIM OF THE STUDY: To investigate the existence of disease-drug interactions during Fuzi treatment for RA, focusing on the pharmacokinetics

Aconitine outperforms mesaconitine and hypaconitine in triggering excessive mitophagy via lysosomal two-pore channels disruption in SH-SY5Y cells and zebrafish.

The genus Aconitum L. is widely used in the treatment of rheumatoid arthritis, tumors, and cardiovascular diseases due to its prominent pharmacological properties. However, increasing scientific attention has been directed toward its neurotoxicity. Diester-diterpenoid alkaloids (DDAs), such as aconitine (AC), mesaconitine (MA), and hypaconitine (HA), have been identified as the principal toxic constituents of Aconitum. Although disruption of calcium homeostasis has been demonstrated to mediate DDAs-induced neurotoxicity, the key neurotoxic components and their underlying molecular mechanisms r

Microcirculatory Changes in a Venous-Arterial Extracorporeal Membrane Oxygenation-Supported Patient with Cardiac Arrest Due to Aconitine Poisoning: A Case Report.

BACKGROUND: Aconitine, a highly toxic diterpenoid alkaloid derived from the Aconitum genus, continues to be utilized in traditional Chinese medicine. Aconitine poisoning is common in China and often causes malignant arrhythmias and cardiac arrest, with a mortality rate of approximately 15.1%. Using venous-arterial extracorporeal membrane oxygenation (VA-ECMO) in this context has been rarely reported, and associated microcirculatory changes have not been described. CASE REPORT: We present a case of a 59-year-old man who ingested a topical medicinal wine prepared from Aconitum brachypodum Diels

An interesting two-phase solvent system and its use in preparative isolation of aconitines from aconite roots by counter-current chromatography.

Two-phase solvent system plays crucial role in successful separation of organic compounds using counter-current chromatography (CCC). An interesting two-phase solvent system, composed of chloroform/ethyl acetate/methanol/water, is reported here, in which both phases contain sufficient organic solvents to balance their dissolving capacities. Adjusting the solvent system to get satisfactory partition coefficients (K values) for target compounds becomes relatively simple. This solvent system succeeded in sample preparation of aconitine (8.07 mg, 93.69%), hypaconitine (7.74 mg, 93.17%), mesaconiti

Combined blood purification and antiarrhythmic therapy for acute aconitine poisoning with refractory arrhythmias: a case-based mechanistic evaluation and treatment strategy optimization.

INTRODUCTION: Aconitine poisoning from traditional Chinese medicine is life-threatening, associated with arrhythmias and shock. Early diagnosis and multidisciplinary treatment are essential due to the lack of specific antidotes. This study aimed to present a severe case of aconitine poisoning and to evaluate the effectiveness of combined blood purification and antiarrhythmic therapy, thereby providing practical insights for clinical management. A 70-year-old man presented with coma, recurrent ventricular arrhythmias, and severe hypotension (nadir: 44/24 mmHg) after ingesting Fuzhi (Aconitum ta

The role of efflux transporters on the transport of highly toxic aconitine, mesaconitine, hypaconitine, and their hydrolysates, as determined in cultured Caco-2 and transfected MDCKII cells.

Aconitum alkaloids including aconitine (AC), mesaconitine (MA), hypaconitine (HA), are highly toxic. Their hydrolysates, such as benzoylaconine (BAC), benzoylmesaconine (BMA), benzoylhypaconine (BHA), aconine, and mesaconine, are considerably less toxic. Efflux transporters, including P-glycoprotein (P-gp), breast cancer resistance protein (BCRP), and multidrug resistance-associated protein isoform 2 (MRP2), act as a first line of defence and play key roles in toxicity prevention. The aim of the present study was to determine the role of efflux transporters in the transport of Aconitum alkaloi

Metabolic Behaviors of Aconitum Alkaloids in Different Concentrations of Aconiti Lateralis Radix Praeparata and Effects of Aconitine in Healthy Human and Long QT Syndrome Cardiomyocytes.

Aconiti Lateralis Radix Praeparata (Fu Zi) is the processed lateral root of Aconitum carmichaelii Debx, which is widely used in emergency clinics. Poisoning incidents and adverse reactions occur with the improper intake of Fu Zi. Metabolic characteristics of aconitum alkaloids of Fu Zi may vary, and the effects of Fu Zi in healthy and Long QT syndrome (LQTS) patients is unknown. In this experiment, 24 Sprague Dawley rats were randomly divided into three groups: 2.0, 1.0, and 0.5 g/kg dose groups, and blood samples were collected after the oral administration of Fu Zi extract. We used an ultra-

Effects of sodium-calcium exchange inhibitors, KB-R7943 and SEA0400, on aconitine-induced arrhythmias in guinea pigs in vivo, in vitro, and in computer simulation studies.

The sodium-calcium exchange (NCX) plays a pivotal role in regulating contractility and electrical activity in the heart. However, the effects of NCX blockers on ventricular arrhythmias are still controversial. We examined the effects of KB-R7943 (KBR) and SEA0400 (SEA), two NCX blockers, on aconitine-induced arrhythmias in guinea pigs using the ECG recordings and the current-clamp method. Using Luo's and Rudy's computer model (1991 Circ Res 68:1501-1526) for ventricular myocytes, we simulated abnormal membrane activity produced by NCX inhibition. In the whole-animal model, KBR in a dose range

Comparative analysis of six aconitine-type alkaloids in decocting extracts between the different processed Fuzi (Aconiti Lateralis Radix praeparata) and its pharmacokinetic behavior in rats by HPLC-MS/MS.

Aconiti Lateralis Radix praeparata (also known as Fuzi in Chinese) has been extensively used in clinic. However, the toxicity issues limit the therapeutic range of Fuzi. Thus, a rapid and sensitive HPLC-MS/MS method was developed to simultaneously quantify and compare six active/toxic constituents in decocting extracts from four different processed Fuzi products and in rat plasma after oral administration of its decocting extracts. The selectivity, linearity, sensitivity, precisions, accuracy, matrix effects, extraction recoveries, and stability were validated. The comparative analysis of six

Forensic and toxicological insights into aconite poisoning: A retrospective analysis of clinical and postmortem findings.

Aconitine, a highly toxic alkaloid derived from the aconite plant, presents significant clinical and forensic challenges due to its cardiotoxic and neurotoxic effects. It may induce fatal ventricular arrhythmias and severe central nervous system disturbances when improperly used. The diagnosis of aconitine poisoning remains difficult, particularly in postmortem contexts, owing to its rapid metabolism and the non-specific nature of autopsy findings. In this retrospective study, we analyzed 35 reported cases of aconitine poisoning published between 1958 and 2024. Each case was evaluated for demo
