Actinomyces species associated with intrauterine contraceptive devices and pelvic inflammatory disease.

Swabs from 112 removed intrauterine contraceptive devices (IUCD), endocervical swabs from these women and from 65 women with pelvic inflammatory disease (PID) were studied for actinomyces, using direct fluorescent antibody test and culture. Endocervical swabs from 50 control subjects were also studied. Actinomyces species could be detected in 23 (20.5 percent) of IUCD wearers and 8 (12.3 percent) of patients with PID. In control cases, no actinomyces were detected. The isolation rate using a selective medium (Actino Blood Agar) was 71.4 percent. Actinomyces israelii, Actinomyces naeslundii and

Characterization of unusual tetracycline-resistant gram-positive bacteria.

Tetracycline-resistant Tet M-negative isolates of Actinomyces viscosus, Eubacterium lentum, Mobiluncus curtisii, and Mobiluncus mulieris were screened with the Tet K, Tet L, and Tet O DNA probes. Ten (71%) of the resistant Mobiluncus strains hybridized with the Tet O probe, two of the three E. lentum strains hybridized with the Tet K probe, and the A. viscosus isolate hybridized with the Tet L probe.

Emended description of Actinomyces naeslundii and descriptions of Actinomyces oris sp. nov. and Actinomyces johnsonii sp. nov., previously identified as Actinomyces naeslundii genospecies 1, 2 and WVA 963.

Actinomyces naeslundii is an important early colonizer in the oral biofilm and consists of three genospecies (1, 2 and WVA 963) which cannot be readily differentiated using conventional phenotypic testing or on the basis of 16S rRNA gene sequencing. We have investigated a representative collection of type and reference strains and clinical and oral isolates (n=115) and determined the partial gene sequences of six housekeeping genes (atpA, rpoB, pgi, metG, gltA and gyrA). These sequences identified the three genospecies and differentiated them from Actinomyces viscosus isolated from rodents. Th

In vitro activity of amoxicillin, clindamycin, doxycycline, metronidazole, and moxifloxacin against oral Actinomyces.

Actinomyces spp have been increasingly associated with endodontic infections. However, the antimicrobial susceptibility of this genus has not been studied extensively. The objective of this study was to determine the susceptibility of oral isolates of Actinomyces naeslundii, Actinomyces gerencseriae, Actinomyces israelii, Actinomyces viscosus, and Actinomyces odontolyticus to amoxicillin, clindamycin, doxycycline, metronidazole, and moxifloxacin using in vitro assays. The minimum inhibitory concentration (MIC) of each bacterial isolate was determined by using E-test strips (AB Biodisk, Solna,

A bacteriocin of Actinobacillus actinomycetemcomitans.

An inhibitory factor from Actinobacillus actinomycetemcomitans Y4 was isolated, and its properties indicated that it was a bacteriocin (actinobacillicin). The bacteriocin was active against Streptococcus sanguis strains, Streptococcus uberis (FDC1), and Actinomyces viscosus T14 as well as other strains of A. actinomycetemcomitans, but not against other crevicular bacteria, including other streptococci and actinomycetes. The activity of this bacteriocin was inhibited by pronase, trypsin, and heat (45 min at 56 degrees C) but not by DNase, RNase, phospholipase, exposure to UV light, or low pH (1

Effect of co-aggregation on the pathogenicity of oral bacteria.

The pathogenicity of oral bacteria was studied by measuring the development of subcutaneous abscesses in mice after infection with Actinomyces viscosus and Streptococcus mitis either singly or as co-aggregated pairs. Heat-treated cells were also tested. The pathogenicity of the co-aggregates was examined in various viable and heat-treated combinations of the two bacterial species. More abscesses were formed by A. viscosus than S. mitis at all the bacterial concentrations tested. Also, abscess formation by co-aggregates of the two strains produced a higher percentage of abscess formation than t

Actinomyces of infected dental root canals.

The aim of the present study was to identity Actinomyces occurring in infected dental root canals. Out of twenty-five isolated Actinomyces strains, four were A. israelii, four were A. naeslundii, four were A. odontolyticus, and six were A. viscosus. Seven strains could be identified only as Actinomyces spp. The relative frequency of various species of Actinomyces isolated from root canals was similar to that of carious dentin. The Actinomyces strains were usually part of polymicrobial infections. These infections could usually be eliminated by one or two conventional endodontic treatments. In

Occurrence of Actinomyces in infections of endodontic origin.

Species of Actinomyces have been associated with endodontic treatment that failed to heal. In this study polymerase chain reaction was used with a pair of universal primers for Actinomyces and species-specific primers to evaluate the contents of infected root canals and aspirates from abscesses or cellulitis for the presence of Actinomyces israelii, A. naeslundii, and A. viscosus. DNA was extracted from 131 clinical samples. DNA from 2 of the original 131 samples was not available for polymerase chain reaction with the universal primer for Actinomyces and A. naeslundii. Immunocytochemical demonstration of Actinomyces species and Arachnia propionica in periapical infections.

Immunocytochemical methods were used for the demonstration of Actinomyces israelii, Actinomyces naeslundii and Arachnia propionica in bacterial colonies found in 7 routine biopsies from periapical inflammatory lesions. All 3 species were found in one specimen, both A. israelii and A. propionica in 3 specimens, and one of each species in the remaining 3 biopsies. Specificity controls by enzyme immunoassay showed that antiserum to A. israelii reacted also with A. odontolyticus, and that to A. naeslundii with A. viscosus, while antiserum to A. propionica did not show any cross-reactions with Acti

Detection of Actinomyces spp. in cervical exudates from women with cervical intraepithelial neoplasia or cervical cancer.

PURPOSE: Under certain circumstances, Actinomyces behaves as an opportunistic microorganism and can cause actinomycosis, a chronic and inflammatory granulomatous infection. The purpose of this project was to detect the presence of Actinomyces in cervical exudates from women with cervical intraepithelial neoplasia (CIN) and women with cervical cancer. METHODOLOGY: Cervical samples from 92 women were divided into three groups: CIN, cervical cancer and healthy women. Metagenomic DNA extraction was performed following the Qiagen QIAamp Mini Kit protocol. A specific fragment (675 bp) was amplified

Partial 16S rRNA primary structure of five Actinomyces species: phylogenetic implications and development of an Actinomyces israelii-specific oligonucleotide probe.

The intra- and intergeneric relationships of the genus Actinomyces were determined by comparing long 16S rRNA sequences, generated by reverse transcriptase. All species formed a phylogenetically coherent cluster in which Actinomyces bovis, A. viscosus, A. naeslundii, A. odontolyticus and A. israelii constituted genetically well defined species. A. israelii DSM 43322 (serotype 2) was not closely related to three other strains of this species (serotype 1) and, as judged from phylogenetic distances, could be accommodated within A. naeslundii, or represent a new species. In contrast to previous fi

Tetracycline and its derivatives strongly bind to and are released from the tooth surface in active form.

Several antibiotics were found to adsorb to saliva-coated enamel and to inhibit in vitro plaque formation by pure cultures of oral bacteria: Actinomyces viscosus, Actinomyces naeslundii and Streptococcus mutans. Tetracycline, minocycline and oxytetracycline adsorbed to the greatest degree, showing 100-fold higher adsorption than spiramycin, the test antibiotics with least adsorption. Inhibition of in vitro plaque formation was found to require both drug substantivity (capacity for adsorption) and antimicrobial activity. Inhibition of plaque formation in the in vitro assay employed correlated w

An unusual case of actinomycosis.

We describe an unusual case of actinomycosis that appeared as a nasal mass. The diagnosis was established when Actinomyces viscosus was isolated from pus obtained from the right maxillary antrum by intranasal antrostomy. The patient was treated with intravenous crystalline penicillin and made an uneventful recovery within 10 days.

Host genotype, pathogenic challenge and periodontal bone loss in the mouse.

Periodontal bone loss was induced in mice of different genotypes by oral inoculation with Actinomyces viscosus. Relatively high bone loss occurred in the mutant X-linked immune deficiency, implicating the humoral immune system. Maximum bone loss was always associated with inoculations of 1.6 x 10(7) colony forming units (c.f.u.), while inoculations of 1.6 x 10(6) or 1.6 x 10(9) c.f.u. resulted in bone loss little different from that in uninoculated controls. This 1000-fold range in inoculum size was not reflected in the number of A. viscosus organisms recovered from the molar teeth at the end

Pulmonary actinomycosis of periodontal origin.

A case of pulmonary actinomycosis of periodontal origin is presented. Microbiologic testing including culture and typing of multiple systemic and oral lesions revealed the presence of Actinomyces naeslundii and A viscosus in all sites. In addition, biopsies demonstrating branching filamentous forms and "sulfur granules" were found in exudates from the lungs, submandibular region and left quadrants of the oral cavity. Seeding of tooth-associated materials containing Actinomyces sp into the pulmonary field may have resulted in this case of pulmonary actinomycosis. Therapy of the pulmonary and pe

Microbiological and toxicity analyses of the synthetic polymer polyhexamethylene guanidine hydrochloride against endodontic microorganisms.

Failures in endodontic treatments are common due to microbial resistance in the pulp canal. The study evaluated the in vitro activity of polyhexamethylene guanidine hydrochloride (PHMGH) against endodontic strains, as well as in vivo toxicity. Using minimum inhibitory concentration and minimum bactericidal concentration techniques, PHMGH was effective against all microorganisms, even at low concentrations. At 50.0 µg/mL, it inhibited Enterococcus faecalis; furthermore, when compared to chlorhexidine (CLX), it demonstrated values 19 times lower against Candida albicans. The polymer's activity w

[Effect of systemic oral administration of tetracycline on experimental gingivitis in golden hamsters].

Local and systemic administrations of tetracycline have been used in human periodontal treatment for conditions including juvenile periodontitis and rapidly progressive periodontitis, although microbiological effects of the treatment have not been clear. The effect of systemic oral administration of tetracycline on subgingival bacteria in experimental periodontal disease in hamsters as an animal model has not yet been reported. The aim of this study was to investigate changes in subgingival bacteria and bone resorption at the lower left first molar, and supragingival plaque formation on the lo

The in vitro inhibition of microbial growth and plaque formation by surfactant drugs.

Several cationic, mixed and amphoteric surfactants were tested for their antimicrobial activity and ability to inhibit the formation of in vitro plaque by oral microorganisms. All had antimicrobial activity against Actinomyces viscosus. Actinomyces naeslundii and Streptococcus mutans. Cationic surfactants were comparable to chlorhexidine in antimicrobial activity but were less effective in inhibiting plaque formation. Amphoteric surfactants were less effective than other detergents in antibacterial activity and had very limited capacity for the inhibition of plaque formation. Comparison of dru
