Lipidomics of brown and white adipose tissue: Implications for energy metabolism.

Adipose tissue exerts multiple vital functions that critically maintain energy balance, including storing and expending energy, as well as secreting factors that systemically modulate nutrient metabolism. Since lipids are the major constituents of the adipocytes, it is unsurprising that the lipid composition of these cells plays a critical role in maintaining their functions and communicating with other organs and cells. In both positive and negative energy balance conditions, lipids and free fatty acids secreted from adipocytes exert either beneficial or detrimental effects in other tissues,

Brown adipose tissue ameliorates autoimmune arthritis via inhibition of Th17 cells.

The functions of adipose tissue are associated with autoimmune diseases, such as rheumatoid arthritis (RA). Some studies have shown that the three compositions of adipose tissue (white, brown, and beige) have different functions. Brown adipose tissue (BAT) is known to secrete several factors that differ from those in white adipose tissue. This suggests that BAT might have potential positive advantages in the physiology of autoimmune diseases. We compared the functions of collagen-induced arthritis mice-derived BAT (CIA BAT) with normal mice-derived BAT. DBA/1J mice (6-7 weeks of age) were immu

Brown adipose tissue activation in a rat model of Parkinson's disease.

Loss of body weight and fat mass is one of the nonmotor symptoms of Parkinson's disease (PD). Weight loss is due primarily to reduced energy intake and increased energy expenditure. Whereas inadequate energy intake in PD patients is caused mainly by appetite loss and impaired gastrointestinal absorption, the underlying mechanisms for increased energy expenditure remain largely unknown. Brown adipose tissue (BAT), a key thermogenic tissue in humans and other mammals, plays an important role in thermoregulation and energy metabolism; however, it has not been tested whether BAT is involved in the

Variable Cold-Induced Brown Adipose Tissue Response to Thyroid Hormone Status.

BACKGROUND: In addition to its role in adaptive thermogenesis, brown adipose tissue (BAT) may protect from weight gain, insulin resistance/diabetes, and metabolic syndrome. Prior studies have shown contradictory results regarding the influence of thyroid hormone (TH) levels on BAT volume and activity. The aim of this pilot study was to gain further insights regarding the effect of TH treatment on BAT function in adult humans by evaluating the BAT mass and activity prospectively in six patients, first in the hypothyroid and then in the thyrotoxic phase. METHODS: The study subjects underwent 18

Hypothalamic melanocortin system regulates sympathetic nerve activity in brown adipose tissue.

To clarify the neuronal mechanism of the hypothalamic melanocortin system in regulating energy metabolism, we investigated the effects of centrally administered alpha-melanocyte-stimulating hormone (alpha-MSH) and agouti-related protein (AGRP), an agonist and an antagonist for the melanocortin 4 receptor (MC4-R), respectively, on the activity of sympathetic nerves innervating brown adipose tissue (BAT) and on BAT temperature. A bolus infusion of alpha-MSH (1 nmol) into the third cerebral ventricle (i3vt) significantly increased sympathetic nerve activity and elevated BAT temperature (P<0.05).

Accumulation of brown adipose tissue in patients with Chagas heart disease.

Brown adipose tissue (BAT) is an important source of non-shivering thermogenesis. Increased BAT amounts have been reported to occur in association with several diseases, including congestive heart failure. The objective of the present study was to determine whether BAT accumulation occurs in patients with Chagas disease. Histological sections of peri-adrenal tissue obtained at autopsy from 259 patients were examined. Of these patients, 58 had the digestive form of Chagas disease, 50 had the cardiac form without heart failure and 201 had the cardiac form with heart failure. All cases were inves

Control of norepinephrine turnover in brown adipose tissue of Syrian hamsters.

Sympathetic nervous system activity in brown adipose tissue (BAT) of normal and myopathic Syrian hamsters was assessed by measuring norepinephrine turnover rate (NETR) using [3H]norepinephrine. Acute exposure of normal hamsters to cold (4 degrees C) for 4 or 24 h increased norepinephrine secretion but not resynthesis. By 3 days and at 2 wk in the cold, NETR increased but returned to a normal level by 6 wk. Hamsters were initially hypothermic (to 3 days) then normothermic (2 and 6 wk). Adaptation of normal or myopathic hamsters to high-fat diet or short photoperiod (4 h light, 20 h dark) for up

Brown adipose tissue in ruminants: development, thermogenic function, and nutritional regulation.

Brown adipose tissue (BAT) is essential for neonatal ruminants to maintain body temperature and survive postnatal cold stress through non-shivering thermogenesis. At birth, BAT is abundant and mainly distributed in the neck, shoulder, perirenal region, and around the heart, but declines with age ("whitening"). This review summarizes BAT developmental dynamics, physiological functions, and the nutritional regulation of BAT, focusing on how fatty acids, carnitine, vitamins, and minerals modulate thermogenic activity through effects on mitochondrial function, lipid metabolism, and key signaling p

Fueling or Fighting Cancer? The Thermogenic Paradox of Brown Adipose Tissue.

PURPOSE OF REVIEW: The global rise in obesity and metabolic syndrome has intensified interest in brown adipose tissue (BAT) as a regulator of energy metabolism and potential modulator of cancer risk. BAT-mediated thermogenesis and the browning of white adipose tissue (WAT) confer metabolic benefits that may reduce oncogenic susceptibility. However, emerging evidence reveals a paradoxical role for BAT in cancer progression, where tumor-induced thermogenic activation contributes to cancer-associated cachexia (CAC). This review article examines the cellular, molecular, and translational dimension

ETS-2 deficiency impairs brown adipose tissue function through Prdm16 repression.

BACKGROUND: Brown adipose tissue (BAT) and beige adipocytes regulate adaptive thermogenesis and systemic energy balance, whereas BAT dysfunction contributes to obesity and metabolic disease. Although ETS transcription factors participate in adipocyte biology, the role of ETS-2 in BAT remains unclear. METHODS: Adipocyte-specific Ets-2 knockout mice were generated using the Adipoq-Cre system. BAT morphology, thermogenic responses, energy expenditure, and mitochondrial structure and function were assessed under basal conditions, cold exposure, β3-adrenergic stimulation, and mTOR activation. Deciphering the Presence of Active Interscapular Brown Adipose Tissue in Humans.

Brown adipose tissue (BAT) is increasingly recognized as a metabolically active tissue in humans, although its physiological relevance remains incompletely understood. In rodents, BAT is well characterized, with interscapular BAT (iBAT) representing the main thermogenic depot. In contrast, the existence and persistence of iBAT in adult humans have long been overlooked. In this review, we synthesize anatomical, histological, imaging, and molecular evidence supporting the presence of a potentially active iBAT depot within the dorsocervical subcutaneous adipose tissue in humans. Gene expression a

Brown adipose tissue transplantation ameliorates hindlimb ischemic damage in diabetic mice.

Peripheral arterial disease (PAD) is a common complication associated with diabetes, which can lead to foot ischemia. The condition is often accompanied by infection and necrosis, ultimately leading to diabetic foot ulcers and the risk of amputation. Brown adipose tissue (BAT) and its secreted cytokines play an essential role in the regulation of glucose homeostasis, the modulation of inflammatory responses, and vascular endothelial cell proliferation. The transplantation of BAT into ischemic regions may offer therapeutic benefits in alleviating the symptoms associated with PAD. A diabetic mou

Metabolically active brown adipose tissue in PPGL: an observational cohort study.

Brown adipose tissue (BAT) activity, identifiable through fluorodeoxyglucose positron emission tomography (FDG-PET), has gained interest due to its potential link with metabolic disorders and tumour pathophysiology. This study aims to explore the activation of BAT in patients with phaeochromocytoma/paraganglioma (PPGL) and its clinical relevance. This retrospective observational study, conducted in a large academic centre in London, reviewed FDG-PET images of 62 confirmed PPGL patients, collected between 2013 and 2021. We assessed patient demographics, biochemistry, radiological features, muta

Senescent immune cells accumulation promotes brown adipose tissue dysfunction during aging.

Brown adipose tissue (BAT)-mediated thermogenesis declines with age. However, the underlying mechanism remains unclear. Here we reveal that bone marrow-derived pro-inflammatory and senescent S100A8+ immune cells, mainly T cells and neutrophils, invade the BAT of male rats and mice during aging. These S100A8+ immune cells, coupled with adipocytes and sympathetic nerves, compromise axonal networks. Mechanistically, these senescent immune cells secrete abundant S100A8 to inhibit adipose RNA-binding motif protein 3 expression. This downregulation results in the dysregulation of axon guidance-relat

White-brown adipose tissue interplay in polycystic ovary syndrome: Therapeutic avenues.

This study highlights the therapeutic potential of activating brown adipose tissue (BAT) for managing polycystic ovary syndrome (PCOS), a prevalent endocrine disorder associated with metabolic and reproductive abnormalities. BAT plays a crucial role in regulating energy expenditure and systemic insulin sensitivity, making it an attractive target for the treatment of obesity and metabolic diseases. Recent research suggests that impaired BAT function and mass may contribute to the link between metabolic disturbances and reproductive issues in PCOS. Additionally, abnormal white adipose tissue (WA

Brown adipose tissue remodelling induced by corticosterone in male Wistar rats.

NEW FINDINGS: What is the central question of this study? Does glucocorticoid excess disrupt brown adipose tissue (BAT) phenotype and function? What is the main finding and its importance? Glucocorticoid excess induced an extensive remodelling of interscapular BAT, resulting in a white-like phenotype in association with metabolic disturbances. Glucocorticoids might be an important modulator of BAT physiology and BAT may have a role in pathophysiology of metabolic disturbances induced by glucocorticoid excess. ABSTRACT: In mammals, brown adipose tissue (BAT) is centrally involved in energy met

Modulation of musculoskeletal hyperalgesia by brown adipose tissue activity in mice.

Cold exposure and a variety of types of mild stress increase pain in patients with painful disorders such as fibromyalgia syndrome. Acutely, stress induces thermogenesis by increasing sympathetic activation of beta-3 (β3) adrenergic receptors in brown adipose tissue. Chronic stress leads to the hypertrophy of brown adipose, a phenomenon termed adaptive thermogenesis. Based on the innervation of skeletal muscle by collaterals of nerves projecting to brown adipose, we theorized an association between brown adipose tissue activity and musculoskeletal hyperalgesia and tested this hypothesis in mic

Hypothalamic overexpression of mutant huntingtin causes dysregulation of brown adipose tissue.

Expression of mutant huntingtin (htt) protein has been shown to cause metabolic imbalance in animal models of Huntington disease (HD). The pathways involved are not fully understood but dysfunction of both the hypothalamus and brown adipose tissue (BAT) has been implicated. Here we show that targeted expression of mutant HTT in the hypothalamus leads to loss of the A13 dopaminergic cell group located in the zona incerta and reduced mRNA expression of neuropeptide Y1 receptor in the hypothalamus. Furthermore, this is accompanied by downregulation of uncoupling protein 1 expression and PPARγ coa
