BACKGROUND: Acute myeloid leukemia (AML) remains associated with high relapse rates and poor long-term survival, particularly in refractory patients or those ineligible for hematopoietic stem cell transplantation (HSCT). Adoptive transfer of natural killer (NK) cells has emerged as a promising immunotherapeutic strategy due to intrinsic cytotoxicity, low risk of graft-versus-host disease (GVHD), immune restoration capacity, and feasibility of allogeneic off-the-shelf manufacturing.
METHODS: A systematic review of phase 1-2 clinical trials evaluating non-genetically modified NK-cell adoptive t

Adoptive transfer therapy has great potential to treat diseases such as cancer as well as autoimmune and infectious diseases. Identification of chain-centric T cell receptors (TCRs) with the dominant-active antigen-specific α-chains (TCRα) can significantly improve the efficacy of adoptive cell therapy while reducing time, labor, and costs of generation of TCR-modified antigen-specific T cells. This protocol describes how to generate salmonella-specific TCRα-modified mouse T cells by retroviral transduction and evaluate their functional activity in vivo in the mouse model of salmonellosis. Tumor cells from NPC patients are regularly and latently infected with EBV. To examine whether the virus serves as target for immune intervention of the cancer, we determined levels of EBV-specific CTLp in peripheral blood from NPC patients, long-term survivors of the cancer and healthy subjects. CTLp levels of test subjects varied between 3- 3,000/10(6) PBMCs. The plasma EBV burden increased when the CTLp level fell below 150, whereas the EBV burden of PBMCs was not correlated with CTLp level. Compared with healthy carriers, CTLp levels of patients were lower and varied over a wider range, be

The effector mechanism of the protection against re-infection with eggs of Taenia taeniaeformis was evaluated in rats by analysing protection dynamics against an ongoing primary infection. Immune spleen cells were prepared from F344 donor rats 10 days after oral inoculation with 200 eggs and used for adoptive transfer experiments. When F344 recipient rats were injected intravenously with immune spleen cells (2 x 10(8)) from 2 to 48 h after a primary egg inoculation, there was a statistically significant reduction in the number of metacestodes recovered at day 30 of the infection. The highest p

INTRODUCTION: Glioblastoma (GBM) is an aggressive brain tumor with limited treatment options and poor immune cell infiltration. Although cellular immunotherapies have transformed cancer treatment, they remain largely ineffective against GBM due to the restrictive blood-brain barrier (BBB) and a profoundly immunosuppressive tumor microenvironment. Innate lymphoid cells type 2 (ILC2s) have recently emerged as potential candidates for immune-based approaches because of their regenerative and immunomodulatory functions.
METHODS: Bone marrow-derived ILC2s from C57BL/6 mice were fluorescently label

The density of NK cells in tumors correlates positively with prognosis in many types of cancers. The average number of infiltrating NK cells is, however, quite modest (approximately 30 NK cells/sq.mm), even in tumors deemed to have a "high" density of infiltrating NK cells. It is unclear how such low numbers of tumor-infiltrating NK cells can influence outcome. Here, we used ovalbumin-expressing tumor cell lines and TCR transgenic, OVA-specific cytotoxic T lymphocytes (OT-I-CTLs) to determine whether the simultaneous attack by anti-tumor CTLs and IL-2-activated NK (A-NK) cells synergistically

Immunotherapeutic approaches to the treatment of advanced melanoma have relied on strategies that augment the responsiveness of endogenous tumor-specific T-cell populations [eg, cytotoxic T lymphocyte-associated antigen 4 (CTLA-4) blockade-mediated checkpoint inhibition] or introduce exogenously prepared tumor-specific T-cell populations [eg, adoptive cell transfer (ACT)]. Although both approaches have shown considerable promise, response rates to these therapies remain suboptimal. We hypothesized that a combinatorial approach to immunotherapy using both CTLA-4 blockade and nonlymphodepletiona

Acute infection with Toxoplasma gondii (T. gondii) during pregnancy is associated with adverse outcomes. The mechanisms that cause this phenomenon are not clear. Regulatory T cells (Tregs) are involved in maternal tolerance, and here we observed a decrease in the absolute numbers of CTLA-4(+) Tregs and PD-1(+) Tregs in spleen and at the fetal-maternal interface in T. gondii-infected mice. Our results suggest that T. gondii induces apoptosis of Tregs. Additionally, we found that the expression of CTLA-4 and PD-1 on Tregs at fetal-maternal interface were higher than on spleen cells from normal p

Adjuvant arthritis (AA) can be induced in Lewis rats by a single injection of either heat-killed Mycobacterium tuberculosis or the lipoidal amine CP20961. Concanavalin A (Con A)-stimulated T cells isolated from AA rats are able to adoptively transfer the disease to naive syngeneic recipients. It is unclear, however, whether these transferred cells traffic directly to the joint and initiate arthritis, or whether secondary host cells are responsible for activation of the disease. In the current investigation, T cells labelled with the vital fluorescent dyes Hoechst H33342 and Zynaxis PKH26-G wer

BACKGROUND: To elucidate the mechanisms of immune-related myocardial injuries, we examined whether autoimmune myocarditis was passively transferable by use of humoral or cellular factors.
METHODS AND RESULTS: Active myocarditis was elicited in Lewis rats by immunization with human cardiac myosin fraction in complete Freund's adjuvant. This experimental myocarditis was characterized by macroscopic features such as pericardial effusion, enlargement of the heart, and gray discoloration of the cardiac surface. Histologically, extensive myocardial necrosis and numerous inflammatory cell infiltrati

Spleen and lymph node cells taken from strain 2 and strain 13 guinea pigs at the peak of their primary immune response to cutaneous syphilitic infection could transfer partial protection to symptomatic disease to normal syngeneic recipients challenged with the Nichols strain of Treponema pallidum. These recipients of immune cells had significantly fewer treponemes disseminating to the regional lymph nodes and developed fewer and less severe cutaneous lesions that resolved faster than those in guinea pigs that had been infused with normal lymphoid cells. Immune donor cells also had the capacity

The relationship between the gut and mesenteric lymph node in the kinetics of mucosal mast cells (MMC) was further analysed. A Trichinella-induced synchronous proliferation of MMC occurred in the intestine of thymus-bearing and not in athymic mice, both at sites of parasite invasion (jejunum) and elsewhere (caecum) and in the mesenteric lymph node. The mastocytopoietic ability could be adoptively transferred with immune mesenteric lymph node cells to the jejunum and caecum of infected thymus-bearing recipients. The enhanced mastocytopoiesis which was both antigen- and thymus-dependent, was onl

Adoptive transfer of immunity against the enteral phase of Strongyloides ratti was monitored using four parameters; namely, number of worms recovered, position occupied in the host's small intestine, worm length and fecundity. When immune mesenteric lymph node cells (IMLNC) were transferred from donors infected for 20, 26 or 32 days into recipients infected for 3 days, a marked acceleration of worm expulsion was evident by day 16 post-infection (p.i.). IMLNC from day-16 p.i. donors did not transfer expulsion. In an experiment in which recipients were given 2 x 10(8) or 1 x 10(8) IMLNC from don

Lymphoid cells from New Zealand Black (NZB) and (C57BL × NZB)F1 mice with Coombs positive haemolytic anaemia and the other disease features of these strains, and also cells from young NZB mice without established disease, were transferred to BALB/c and C57BL recipients treated with anti-lymphocyte globulin. Donor strain skin grafts and anti-allotype antibody to (C57BL × NZB)F1 IgG were used to assess the fate of the transferred lymphoid cells. Haematological, serological, biochemical and histological criteria were used to indicate successful transfer of NZB disease to the normal recipients. Sp

OBJECTIVE: Progressive multifocal leukoencephalopathy (PML) is still burdened by high mortality in a subset of patients, such as those affected by hematological malignancies. The aim of this study was to analyze the safety and carry out preliminary evaluation of the efficacy of polyomavirus JC (JCPyV)-specific T cell therapy in a cohort of hematological patients with PML.
METHODS: Between 2014 and 2019, 9 patients with a diagnosis of "definite PML" according to the 2013 consensus who were showing progressive clinical deterioration received JCPyV-specific T cells. Cell lines were expanded from

Sepsis, a condition caused by severe infections, affects more than 30 million people worldwide every year and remains the leading cause of death in hospitals1,2. Moreover, antimicrobial resistance has become an additional challenge in the treatment of sepsis3, and thus, alternative therapeutic approaches are urgently needed2,3. Here, we show that adoptive transfer of macrophages containing antimicrobial peptides linked to cathepsin B in the lysosomes (MACs) can be applied for the treatment of multidrug-resistant bacteria-induced sepsis in mice with immunosuppression. The MACs are constructed b

BACKGROUND: Current FDA-approved therapeutic options for patients with metastatic melanoma include dacarbazine, interleukin 2, ipilimumab, vemurafenib, dabrafenib, and trametinib, but long-term tumor regression using available agents remains out of reach for most patients. Adoptive cell transfer (ACT) with autologous tumor-infiltrating lymphocytes (TILs) has shown encouraging results in clinical trials, with evidence of durable ongoing complete responses in patients with advanced melanoma. Emerging techniques to engineer T-cell receptors (TCRs) or chimeric antigen receptors (CARs) using lympho

BACKGROUND: Intestinal infections with the protozoan parasite Cryptosporidium parvum are prevalent in both immunocompetent and immunocompromised hosts. Although C parvum is an important cause of outbreaks and opportunistic infections worldwide, little is known about protective mucosal immune responses. This is in part because animal models of infection are limited to those with genetic or induced immunodeficiencies.
METHOD: In this report, we isolated immune (primed) or nonimmune (unprimed) intraepithelial lymphocytes (IEL) from BALB/cJ mouse intestines, adoptively transferred them into C par

Using lethal irradiation of recipients, adoptive transfer of experimental allergic encephalomyelitis (EAE) into Lewis (LEW) rats using (LEW x PVG/c)F1 (LPVGF1) spleen cells was successfully achieved. Recipient rats usually developed clinical signs of EAE 5 or 6 days after transfer. The EAE was characterized by the presence of a number of petechiae over the spinal cord. Immunohistochemical examination using OX27, a monoclonal antibody specific for RT1.Ac, revealed the localization of transferred F1 (RT1(1/c] cells in the LEW recipients (RT1(1]. Most of the inflammatory cells in the spinal cord

INTRODUCTION: Cancer immunotherapy holds promise for the treatment of malignancies by mobilizing the immune system; however, its efficacy remains limited in tumors that lack immune infiltration. Innovative approaches are therefore required to convert these immunologically "cold" tumors into "hot" tumors that are more susceptible to immune-mediated attack. Therapeutic vaccination represents one such strategy, capable of triggering robust immune activation and durable memory responses. Given that the vast majority of the global population has either been vaccinated against or exposed to SARS-CoV
