The adrenomedullary chromaffin cells' hormonal pathway has been related to the pathophysiology of diabetes mellitus. In mice, the deletion of insulin receptor substrate type 2 (Irs2) causes peripheral insulin resistance and reduction in β-cell mass, leading to overt diabetes, with gender differences on adrenergic signaling. To further unravel the relevance of Irs2 on glycemic control, we analyzed in adult Irs2 deficient (Irs2-/-) mice, of both sexes but still normoglycemic, dopamine effects on insulin secretion and glycerol release, as well as their adrenal medulla by an immunohistochemical an

We investigated the alteration of tyrosine hydroxylase (TH) mRNA expression and androgen receptor in the adrenal medulla of spontaneously hypertensive rats (SHR). The TH mRNA expression in the adrenal medulla of SHR was higher than that of Wistar Kyoto rats (WKY). The testosterone increased the TH mRNA expression of SHR, but not WKY. The affinity of androgen receptor in the adrenal medulla of SHR was higher than that of WKY; however, the number of androgen receptor was not different. These results suggested that the increased binding affinity of the androgen receptor of SHR may be related to T

25 of 85 (29.4%) consecutive patients operated on for pheochromocytoma had other neuroectodermal abnormalities. Medullary thyroid carcinoma was the most common associated neuroectodermal abnormality followed by von Recklinghausen's neurofibromatosis. Other abnormalities were intracranial tumors, parathyroid hyperplasia and midgut carcinoid. The adrenal medulla was studied to find out morphological characteristics in patients with associated neuroectodermal abnormalities. All patients with multiple pheochromocytomas (n = 7) and all patients with hyperplasia of the extratumoral adrenal medulla (

To assess adrenal medulla activity in states of hyperfunction, a single 0.3 mg oral dose of clonidine hydrochloride (Catapres) was given to twelve patients with varying evidence of familial adrenal medullary hyperplasia and pheochromocytomas from kindreds with Multiple Endocrine Neoplasia type 2 syndrome (MEN-2), seven patients with sporadic pheochromocytomas and six normal subjects. Mean arterial blood pressure and plasma norepinephrine (NE) levels were lower than baseline values 2 h after clonidine in the normal subjects. Plasma epinephrine (E) rose in one normal but fell in the remainder af

1. The release of catecholamines from the adrenal medulla, in response to carotid body hypoxia, may outlast the stimulus by more than 30 min. 2. After denervation of the adrenal gland the immediate release of catecholamines in response to carotid hypoxia is abolished, but the prolonged release remains. 3. The prolonged release of catecholamines is abolished by cycloheximide. 4. Both corticotrophin in vivo and hydrocortisone in the isolated perfused adrenal gland release adrenomedullary catecholamines. 5. It is concluded that a component of the response of the adrenal medulla to carotid body hy

The uptake of 22Na+ and secretion of catecholamines by primary cultures of adrenal medulla cells under the influence of a variety of agonists and antagonists were determined. Veratridine, batrachotoxin, scorpion venom, and nicotine caused a parallel increase in 22Na+ uptake and Ca2+-dependent catecholamine secretion. Ba2+, depolarizing concentrations of K+, and the Ca2+ ionophore Ionomycin stimulated secretion of catecholamines but did not increase the uptake of 22Na+. Tetrodotoxin inhibited both 22Na+ uptake and catecholamine secretion evoked by veratridine, batrachotoxin, and scorpion venom,

The left adrenal medullas of 20 (5 litters of 4) day-old dogs were studied with the electron microscope. The right adrenal medullas of the same animals had been examined by fluorescence microscopy and the results reported previously. In each litter one animal served as a coenothermic (37 degrees C) and one a hypothermic control (15 degrees C). The third animal (at 37 degrees C) was asphyxiated until twice its time of last gasp (about 32 min) and the fourth was cooled to 15 degrees C and asphyxiated for the same length of time as the third animal. The induction of hypothermia (to 15 degrees C)

The article presents a description of a clinical case of an ectopic focus of adrenocorticotropic hormone (ACTH) hypersecretion located in the adrenal medulla in a 64-year-old obese woman without other characteristic clinical signs of hypercorticism, with complaints of rapidly progressing lower limb edema, severe muscle weakness, loss of appetite, weight loss by 4 kg, as well as stage 2 arterial hypertension and type 2 diabetes mellitus. The patient's blood biochemistry showed severe hypokalemia, up to 2.2 mmol/l, despite infusion and oral therapy with potassium preparations. Taking into accoun

Intermittent hypoxia (IH) increases reactive oxygen species generation resulting in oxidative stress in the adrenal medulla (AM), a major end-organ of the sympathetic nervous system which facilitates catecholamine secretion by hypoxia. Here, we show that carotid body chemoreflex contributes to IH-induced oxidative stress in the AM. Carotid bodies were ablated by cryocoagulation of glomus cells, the putative O(2) sensing cells. Carotid body ablated (CBA) and control rats were exposed to IH and the redox state of the AM was assessed biochemically. We found that IH raised reactive oxygen species

The administration of nicotine stimulates the transcription rate of the tyrosine hydroxylase gene in rat adrenal medulla. This stimulation occurs very rapidly (within 10 min) after the subcutaneous injection of nicotine and persists for at least 1 hr after a single injection of the drug. Repeated injections of the drug (seven injections once every 30 min) are associated with a more persistent activation of the gene (for at least 3 hr) and elicit the induction of tyrosine hydroxylase mRNA and tyrosine hydroxylase protein. Quantitatively, the increases in tyrosine hydroxylase gene transcription

This study was performed to determine if epinephrine (EPI) present in hypophysial portal blood has a peripheral or a central origin. Pituitary stalk and femoral arterial EPI plasma levels were simultaneously measured in rats anesthetized with thiopental or urethane. EPI was measured by two different methods: radioenzymatic assay or HPLC followed by electrochemical detection. In thiopental-anesthetized rats, levels were higher in hypophysial portal than in femoral arterial plasma with both methods. In urethane-anesthetized rats, no difference in EPI concentrations was found between peripheral a

Dopamine-beta-hydroxylase (DBH), the enzyme that catalyzes the conversion of dopamine to norepinephrine, remains the topic of many unanswered questions. We isolated DBH cDNA clones from a bovine adrenal medulla cDNA library in the vector lambda gt10. The longest cDNA had an open reading frame encoding an entire mature DBH 578 amino acid (64,808 dalton) polypeptide chain, though lacking a portion of the signal peptide. Additional 5' clones, obtained by the polymerase chain reaction, established the sequence of a 19 amino acid signal peptide. The mature protein sequence was 84% homologous to tha

Parkinson's disease most consistently involves pathologic changes in the substantia nigra, which is the major source of dopamine to the striatum. It has been shown that either fetal substantia nigra or adrenal medulla tissue implanted into the rat brain survives, produces dopamine, and improves behavioral abnormalities induced by deprivation of the caudate nucleus of its dopaminergic innervation. Thus, catecholamine-containing grafts could be potential replacements for destroyed or damaged dopaminergic neurons in patients with Parkinson's disease. To explore the potential of this therapeutic a

Ultraviolet (UV) radiation has a strong biological effect on skin biology, and it switches on adaptive mechanisms to maintain homeostasis in organs such as the skin, adrenal glands, and brain. In this study, we examined the adaptation of the body to repeated bouts of UVB radiation, especially with respect to the catecholamine synthesis pathway of the adrenal glands. The effects of UVB on catecholamine-related enzymes were determined by neurochemical and histological analyses. To evaluate catecholamine changes after chronic excessive UVB irradiation of mouse skin, we examined dopamine and norep

The mammalian adrenal gland is composed of two main components; the catecholaminergic neural crest-derived medulla, found in the center of the gland, and the mesoderm-derived cortex producing steroidogenic hormones. The medulla is composed of neuroendocrine chromaffin cells with oxygen-sensing properties and is dependent on tissue interactions with the overlying cortex, both during development and in adulthood. Other relevant organs include the Zuckerkandl organ containing extra-adrenal chromaffin cells, and carotid oxygen-sensing bodies containing glomus cells. Chromaffin and glomus cells rev

The effects of hypothermia and hypoglycaemia on adrenal catecholamines and dopamine-beta-hydroxylase were compared in control and carbon disulphide (CS2) exposed rats 24 h after the last of ten daily 4 h inhalation exposures to CS2, 2 mg 1(-1) air. Animals were either kept in a cold room (0 degrees C) for 210 min with or without immobilization or were injected with insulin 100 u kg-1. Before these treatments CS2 exposed rats had more dopamine and less adrenaline in their adrenals than controls, and CS2 exposure also elevated the adrenal synthesis of catecholamines. Cold with immobilization or

The cardiovascular effects of intravenous and intracisternal administration of neurohypophysial peptides were compared in normal, diabetes insipidus and adrenal demedullated chloralose anaesthetized dogs. In normal dogs, intravenous lysine vasopressin (0.1 to 100 mU/kg) induced a dose-dependent increase in blood pressure with bradycardia whereas intracisternal injection (0.01 to 10 mU/kg) elicited a dose-related decrease in blood pressure but no change in heart rate. Intracisternally injected oxytocin (1 and 10 mU/kg) increased blood pressure. The central hypotensive effects of vasopressin wer
