Adenovirus-vectored novel African Swine Fever Virus antigens elicit robust immune responses in swine.

African Swine Fever Virus (ASFV) is a high-consequence transboundary animal pathogen that often causes hemorrhagic disease in swine with a case fatality rate close to 100%. Lack of treatment or vaccine for the disease makes it imperative that safe and efficacious vaccines are developed to safeguard the swine industry. In this study, we evaluated the immunogenicity of seven adenovirus-vectored novel ASFV antigens, namely A151R, B119L, B602L, EP402RΔPRR, B438L, K205R and A104R. Immunization of commercial swine with a cocktail of the recombinant adenoviruses formulated in adjuvant primed strong A

African Swine Fever Vaccinology: The Biological Challenges from Immunological Perspectives.

African swine fever virus (ASFV), a nucleocytoplasmic large DNA virus (NCLDV), causes African swine fever (ASF), an acute hemorrhagic disease with mortality rates up to 100% in domestic pigs. ASF is currently epidemic or endemic in many countries and threatening the global swine industry. Extensive ASF vaccine research has been conducted since the 1920s. Like inactivated viruses of other NCLDVs, such as vaccinia virus, inactivated ASFV vaccine candidates did not induce protective immunity. However, inactivated lumpy skin disease virus (poxvirus) vaccines are protective in cattle. Unlike some e

Analysis of acidified feed components containing African swine fever virus.

Mitigation of African swine fever (ASF) virus in contaminated feed materials would assist control activities. Various finely-ground pig feed ingredients (5 cereals, 4 plant proteins, 2 animal proteins, 1 oil, 1 compound) were sprayed and mixed thoroughly with a buffered formic acid formulation (0, 1 or 2% vol/vol) to produce a consistent and durable level of formate (1% or 2%) with consistent acidification of cereal ingredients to less than pH 4. No such acidification was noted in other ingredients. Selected representative feed ingredients were further mixed with infectious ASF virus (106 TCID

Cyproheptadine hydrochloride inhibits African swine fever viral replication in vitro.

African swine fever (ASF) is an infectious disease caused by the African swine fever virus (ASFV), and has a high mortality rate. It has caused serious socioeconomic consequences worldwide. Currently, there are no available commercial vaccines or antiviral drug interventions. D1133L is one of the key genes for ASFV replication and antiviral drug screening. In this study, a virtual screening software program, PyRx, was used to screen libraries of compounds against the potential drug target D1133L. Twelve compounds with a high affinity for ASFV D1133L were screened, and cyproheptadine hydrochlor

Consumption coagulopathy associated with shock in acute African swine fever.

We studied the evolution of shock using a comprehensive array of haematological tests in pigs infected with the highly virulent strain Malawi '83 (Lilongwe 20/1). A sudden onset of illness was observed between day 5 and 7 after inoculation with development of flush, episodes of epistaxis and melaena. Prior to these clinical signs, initiation of a consumption coagulopathy was demonstrated with loss of antithrombin III and plasminogen activity. Our findings indicate that during infection with this highly virulent strain the development of a consumption coagulopathy precedes and possibly contribu

A synthetic genomics-based African swine fever virus engineering platform.

African swine fever (ASF) is a deadly viral disease in domestic pigs that has a large global economic impact for the swine industry. It is present in Africa, Europe, Asia, and in the Caribbean island of Hispaniola. There are no effective treatments or broadly licensed vaccines to prevent disease. Efforts to counteract ASF have been hampered because of the lack of convenient tools to engineer its etiological agent, ASF virus (ASFV), largely due to its large noninfectious genome. Here, we report the use of synthetic genomics methodology to develop a reverse genetics system for ASFV using a CRISP

Current state of knowledge about African swine fever: a review.

African swine fever (ASF) is a highly contagious animal disease caused by African swine fever virus (ASFV). It is listed by the World Organization for Animal Health (WOAH) as an animal disease subject to statutory reporting. ASFV, a large, enveloped double-stranded DNA virus with high genomic complexity, exhibits a case fatality rate of up to 100%, posing a significant threat to the global pig industry and food safety. To date, the absence of a safe commercial ASFV vaccine primarily stems from challenges in identifying immunogenic viral antigens, insufficient characterization of ASFV pathogene

Azvudine potently inhibits African swine fever virus replication in vitro.

African swine fever (ASF), caused by the African swine fever virus (ASFV), is a highly contagious and often fatal disease affecting domestic pigs and wild boars, resulting in substantial economic losses globally. The lack of commercially available vaccines or effective antiviral drugs highlights the urgent need for novel therapeutic strategies. In this study, we conducted a cell-based screening of a targeted anti-DNA virus compound library to identify inhibitors of ASFV replication. Among the candidates, Azvudine-a clinically approved nucleoside analog-demonstrated potent dose-dependent suppre

African Swine Fever Virus Interaction with Host Innate Immune Factors.

African swine fever virus (ASFV) adversely affects pig farming owing to its 100% mortality rate. The condition is marked by elevated body temperature, bleeding, and ataxia in domestic pigs, whereas warthogs and ticks remain asymptomatic despite being natural reservoirs for the virus. Breeding ASFV-resistant pigs is a promising solution for eradicating this disease. ASFV employs several mechanisms to deplete the host antiviral response. This review explores the interaction of ASFV proteins with innate host immunity and the various types of machinery encompassed by viral proteins that inhibit an

Inhibition of African swine fever virus replication by β-glucan.

BACKGROUND: African swine fever (ASF) is one of the most important diseases in pigs because of its effects on all ages and breeds. To date, commercial vaccines and drugs for the prevention of ASF are lacking in the market and the survival of African swine fever virus (ASFV) in various environmental, farm, and or feed matrices has allowed the virus to remain, causing new outbreaks in the pig population. Besides biosecurity and animal husbandry management practices, the improvement of the host immune responses is critical to control, managing, and preventing ASF. AIM: In this study, we investig

Berbamine Hydrochloride Inhibits African Swine Fever Virus Infection In Vitro.

African swine fever virus (ASFV) causes a viral disease in swine with a mortality rate of approximately 100%, threatening the global pig industry's economic development. However, vaccines are not yet commercially available, and other antiviral therapeutics, such as antiviral drugs, are urgently needed. In this study, berbamine hydrochloride, a natural bis-benzylisoquinoline alkaloid isolated from the traditional Chinese herb Berberis amurensis, showed significant antiviral activity against ASFV. The 50% cytotoxic concentration (CC50) of berbamine hydrochloride in porcine alveolar macrophages (

Evaluation of Cryopreserved Primary Swine Macrophages as a Substrate for African Swine Fever Virus Replication.

African swine fever (ASF) is a highly contagious and often fatal viral disease of pigs that poses serious economic consequences to the swine industry due to its high mortality rate and rapid spread. Currently, the identification of infectious ASF virus (ASFV) is the confirmatory test when clinical samples are positive for ASFV by any other diagnostic methods. Detection of infectious ASFV requires the availability of primary swine macrophage cultures as a cell substrate. We demonstrate here that cryopreserved swine primary macrophages are a suitable cell substrate for the detection, isolation a

Risk factors associated with the occurrence of African swine fever in Benin.

African swine fever (ASF) is a highly contagious viral disease affecting pigs that cause high mortality and threatens the swine industry. In Benin where the virus was first identified in 1997, epidemiological data remain limited. This study assessed potential factors associated with reported ASF occurrence in pig farms in southern Benin using 42 questions survey administered to 215 pig farms. Data collected followed descriptive and logistic regression analyses. Of the surveyed farms, 100 reported at least one ASF outbreak between 1998 and 2020. Univariate analysis identified several risk facto

Determinants of variation in epidemiological parameters for African Swine Fever virus.

African Swine Fever (ASF) is an infectious reportable disease of swine that can have devastating consequences for pork producers and consumers. ASF virus can persist in either domestic or wild swine through several epidemiological cycles. This has led to a wide variety of epidemiological traits, making it challenging to plan effective surveillance and mitigation strategies. Here, we compile and analyze data from experimental infections of ASF virus variants from genotype II that have been reported in peer-reviewed publications. We provide descriptive summaries of the distributions of epidemiol

Thoughts on the research of African swine fever live-attenuated vaccines.

African swine fever (ASF) is a contagious and fatal disease caused by the African swine fever virus (ASFV), which can infect pigs of all breeds and ages. Most infected pigs have poor prognosis, leading to substantial economic losses for the global pig industry. Therefore, it is imperative to develop a safe and efficient commercial vaccine against ASF. The development of ASF vaccine can be traced back to 1960. However, because of its large genome, numerous encoded proteins, and complex virus particle structure, currently, no effective commercial vaccine is available. Several strategies have bee

[Characteristics of African swine fever virus and difficulties in vaccine development].

African swine fever (ASF) is a devastating disease of pigs caused by African swine fever virus (ASFV), which is considered to be the No. 1 killer to the global pig industry. Highly virulent strains are usually responsible for the peracute and acute forms that provoke high mortality rates that may reach 100%. Since ASF was first introduced in August 2018 into China, 137 outbreaks in domestic and wild pigs had been reported from 32 provinces by June 06, 2019, causing severe socioeconomic consequences. Efforts to develop an ASFV vaccine began in the 1960s, but all failed, the major reason is the
