The African swine fever virus (ASFV) is a major global threat affecting pork production and strengthening biosecurity practices is an urgent priority, especially given the paucity of effective vaccines and antiviral drugs. Mitigation strategies focused on virus inactivation play an important role in controlling ASFV and there is growing recognition that multipronged mitigation strategies can not only achieve rapid decontamination of ASFV-exposed materials in different environmental settings but also support pig health by minimizing disease symptoms and preventing lateral transmission. Herein,

African swine fever virus (ASFV), a devastating pathogen of swine, poses a great threat to the global pork industry and food supply due to its high lethality and lack of effective countermeasures. In this review, we aim to elucidate the intricate interplay between ASFV and host autophagy-a cellular process with dual roles in viral pathogenesis. By dissecting the molecular mechanisms through which ASFV interacts with autophagy, this review resolves key controversies surrounding autophagy's context-dependent effects on viral replication, immune evasion, and tissue damage. The significance of thi

African Swine Fever Virus (ASFV) is of high concern in porcine livestock across the world due to both the high mortality rates and the trade restrictions imposed on affected regions. The viral genome is large and complex, and genomic analysis is essential for tracing its origin and evolution. Although several bioinformatics tools exist for genome assembly and analysis, no single platform integrates all necessary steps in an accessible and systematic way. In this study the authors developed 'PePApipe', a custom-built, user-friendly pipeline that enables rapid, complete, and efficient ASFV genom

African swine fever is a fatal, febrile, infectious disease that affects pigs and wild boars and is caused by infection with African swine fever virus (ASFV). The current pandemic strain, belonging to genotype II, first emerged in Georgia in 2007 and subsequently spread across Russia, Europe, Asia, and the Caribbean. Developing live attenuated viruses represents the most promising vaccine strategy; however, biosafety and biosecurity concerns due to long-term persistence of attenuated viruses in immunized animals may lead to secondary transmission or reversion to virulence. To address this conc

African swine fever (ASF) is a viral haemorrhagic disease found in domestic and wild boars caused by the African swine fever virus (ASFV). A highly virulent strain was used to evaluate the efficacy of newly developed vaccine candidates. The ASFV strain SY18 was isolated from the first ASF case in China and is virulent in pigs of all ages. To evaluate the pathogenesis of ASFV SY18 following intraoral (IO) and intranasal (IN) infections, a challenge trial was conducted in landrace pigs, with intramuscular (IM) injection as a control. The results showed that the incubation period of IN administra

BACKGROUND: African swine fever virus (ASFV) is one of the most fatal swine etiological agents and has a huge economic impact on the global pork industry. Given that no effective vaccines or anti-ASFV drugs are available, there remains a pressing need for novel anti-ASFV drugs. This study aimed to investigate the anti-African swine fever virus (ASFV) activity of brequinar, a DHODH inhibitor.
METHODS: The anti-ASFV activity of brequinar was investigated using IFA, HAD, HAD50, qRT-PCR, and western blotting assays. The western blotting assay was used to investigate whether brequinar inhibits ASF

African swine fever (ASF) is an important transboundary animal disease with a high mortality rate. The high African swine fever virus (ASFV) titers in the excretion of infected wild boar possibly contaminate the feed ingredient. Once contaminated, it could support persistent residual titer of the ASFV. The chemical inactivation of imported feed ingredients is a precautionary risk management measure to restrict the import risk of ASFV through international trade. The log ASFV titers were linearly reduced as a function of the inactivation time after exposure to 0.03%, 0.05%, 0.1%, and 0.2% forma

African swine fever virus (ASFV) is a double-stranded DNA virus that causes an acute and hemorrhagic disease in domestic swine, resulting in significant economic losses to the global porcine industry. The lack of vaccines and antiviral drugs highlights the urgent need for antiviral studies against ASFV. Here, we report that brequinar (BQR), which is a specific inhibitor of dihydroorotate dehydrogenase, robustly inhibits ASFV replication in Vero cells, as well as in porcine macrophages. We demonstrate that BQR exerts its antiviral activity in a dose-dependent manner through the depletion of pyr

Animal experiments on African swine fever virus (ASFV) are vital to the study of ASFV; however, ASFV can only infect pigs, and animal experiments need to be performed in animal biosafety level 3 (ABSL-3) laboratories, meaning that many small ABSL-3 laboratories are unable to carry out in vivo ASFV experiments. Therefore, miniaturized experimental animals for ASFV infection are urgently needed. Here, we successfully isolated genotype II of ASFV SY-1 from wild boars and evaluated ASFV-infected Bama minipigs in a negative-pressure isolator of a small ABSL-3 laboratory. The pathological changes of

African swine fever (ASF) is a highly contagious, often fatal viral disease caused by African swine fever virus (ASFV), leading to high fever, severe hemorrhages with high lethality in domestic pigs and wild boar. In 2007, ASF was reintroduced into Europe. Since then, ASF has spread to many European and Asian countries and now becomes a major concern to the swine industry worldwide. There have been various vaccine attempts, but no commercial ASF vaccines are available so far. A key hurdle in developing a safe and efficacious ASF vaccine is the limited understanding of innate and adaptive immun

The pathogenesis of African swine fever virus (ASFV) infection in Ornithodoros porcinus porcinus was examined in nymphal ticks infected with the ASFV isolate Chiredzi/83/1. At times postinfection (p.i.) ranging from 6 h to 290 days, ticks or dissected tick tissues were titrated for virus and examined ultrastructurally for evidence of virus replication. The ASFV infection rate in ticks was 100% in these experiments, and virus infection was not associated with a significant increase in tick mortality. Initial ASFV replication occurred in phagocytic digestive cells of the midgut epithelium. Subse

Thirteen sows between 38 and 92 days of pregnancy were exposed oronasally to African swine fever virus (1979 Dominican Republic isolate) to determine whether infection would affect their reproductive performance. Abortions occurred in sows in all stages of pregnancy. Of 11 sows, 7 aborted 5 to 8 (6.1 +/- 0.90) days after inoculation or 0 to 3 (1.4 +/- 0.98) days after fever development. Abortions occurred soon after clinical signs of infection first developed and viremia titers peaked. Swelling of the vulva and treading preceded abortion in some sows. Ecchymotic placental and petechial cutaneo

African swine fever virus (ASFV) relies on a virus-encoded transcription machinery to initiate gene expression within cytoplasmic viral factories. The viral early transcription factor (VETF), a heterodimer composed of pD1133L and pG1340L, is required for early transcription and viral replication, yet its structural organization and regulation remain poorly understood. Here, we present cryo-EM structures of ASFV VETF in five distinct conformations. The complex adopts an unusual chair-like architecture stabilized by an extensive buried interfacial area. The small subunit, pD1133L, contains a con

African swine fever virus (ASFV), a highly virulent double-stranded DNA virus, poses a significant threat to global pig farming, with mortality rates in domestic pigs reaching up to 100%. Originating in Kenya in 1921, ASFV has since proliferated to Western Europe, Latin America, Eastern Europe, and most recently China in 2018, resulting in substantial global agricultural losses. Antigenic epitopes, recognized by the immune system's T cells and B cells, are pivotal in antiviral immune responses. The identification and characterization of these antigenic epitopes can offer invaluable insights in

Due to the current unavailability of vaccines or treatments for African swine fever (ASF), which is caused by African swine fever virus (ASFV), rapid and reliable detection of the virus is essential for timely implementation of emergency control measures and differentiation of ASF from other swine diseases with similar clinical presentations. Here, an improved PCR assay was developed and evaluated for sensitive and universal detection of ASFV. Primers specific for ASFV were designed based on the highly conserved region of the vp72 gene sequences of all ASFV strains available in GenBank, and th

African swine fever virus (ASFV) devastates swine herds and lacks widely available antivirals. We show that the ASFV DNA polymerase beta (Pol β; gene G1211R) localizes to viral replication factories and is required for viral replication. Cryo-electron microscopy structures of Pol β in apo and double-stranded DNA (dsDNA)-bound states reveal pronounced conformational flexibility that enables transitions between functional states. Binding to dsDNA promotes higher-order oligomerization that is essential for catalytic activity, as demonstrated by structure-guided mutagenesis and biochemical assays.

Type II topoisomerases are ubiquitous enzymes that play a pivotal role in modulating the topological configuration of double-stranded DNA. These topoisomerases are required for DNA metabolism and have been extensively studied in both prokaryotic and eukaryotic organisms. However, our understanding of virus-encoded type II topoisomerases remains limited. One intriguing example is the African swine fever virus, which stands as the sole mammalian-infecting virus encoding a type II topoisomerase. In this work, we use several approaches including cryo-EM, X-ray crystallography, and biochemical assa

The rapid spread of the African swine fever virus (ASFV), causing severe disease with often high fatality rates in Eurasian suids, prevails as a threat for pig populations and dependent industries worldwide. Although advancing scientific progress continually enhances our understanding of ASFV pathogenesis, alternative transmission routes for ASFV have yet to be assessed. Here, we demonstrate that ASFV can efficiently be transferred from infected boars to naïve recipient gilts through artificial insemination (AI). In modern pig production, semen from boar studs often supplies many sow herds. Th
