Akkermansia muciniphila induces slow extramedullary hematopoiesis via cooperative IL-1R/TLR signals.

Bacterial infections can activate and mobilize hematopoietic stem and progenitor cells (HSPCs) from the bone marrow (BM) to the spleen, a process termed extramedullary hematopoiesis (EMH). Recent studies suggest that commensal bacteria regulate not only the host immune system but also hematopoietic homeostasis. However, the impact of gut microbes on hematopoietic pathology remains unclear. Here, we find that systemic single injections of Akkermansia muciniphila (A. m.), a mucin-degrading bacterium, rapidly activate BM myelopoiesis and slow but long-lasting hepato-splenomegaly, characterized by

Microbe on the move: Akkermansia in infectious diseases and emerging roles in gynecological health.

The genus Akkermansia was first described in 2004 following the identification of Akkermansia muciniphila, a Gram-negative, mucin-degrading bacterium of the intestine that constitutes 1-3% of the total adult fecal content. Since the interest in A. muciniphila in human health has increased over the past decade, an extensive amount of research examining the impact of A. muciniphila on metabolic disorders, non-communicable diseases, and during infection has been published. Furthermore, a rapidly evolving area of research is the role of A. muciniphila in gynecological health. Many studies have sho

Faecalibacterium prausnitzii and Akkermansia muciniphila: driving the multiple sclerosis-a systematic review.

The gut microbiota (GM) is a complex microbial ecosystem, and its alteration contributes to the development of several diseases including multiple sclerosis (MS). Progressive research concerning neurodegenerative diseases and GM reveals that GM dysbiosis has been linked to fostering the development and progression of MS. Among existing bacteria, F.prausnitzii and A.muciniphila are key species implicated in this disease. Thus, the present study systematically reviewed and synthesized the research on the involvement of F. prausnitzii and A. muciniphila in MS patients versus healthy individuals.

Potential role of Akkermansia massiliensis in multiple sclerosis protection by the FcRL3 gene.

The contribution of human genetics in shaping the abundance of gut microbiota is small, about 5%; however, it can inform about disease etiopathogenesis, including multiple sclerosis (MS). Searching for MS-gut microbiota cross-comparison of reported genome-wide association studies (GWAS), we identified a coincident association of the disease with a commensal - Akkermansia massiliensis-in the selective Fc receptor of secretory IgA gene (FcRL3). Those two signals colocalized, supporting that lower abundance of A.massiliensis may contribute to MS predisposition. In cis, the FcRL3-MS predisposing s

The role of Akkermansia muciniphila in inflammatory bowel disease: Current knowledge and perspectives.

Inflammatory bowel diseases, including Crohn's disease and ulcerative colitis, is a chronic relapsing gastrointestinal inflammatory disease mediated by dysregulated immune responses to resident intestinal microbiota. Current conventional approaches including aminosalicylates, corticosteroids, immunosuppressive agents, and biological therapies are focused on reducing intestinal inflammation besides inducing and maintaining disease remission, and managing complications. However, these therapies are not curative and are associated with various limitations, such as drug resistance, low responsiven

Characterization of antibiotic-resistance traits in Akkermansia muciniphila strains of human origin.

Akkermansia muciniphila, a commensal bacterium commonly found in healthy gut microbiota, is widely considered a next-generation beneficial bacterium candidate to improve metabolic and inflammatory disorders. Recently the EFSA's Panel on Nutrition, Novel food, and Food Allergens has declared that pasteurized A. muciniphila DSM 22959T (also MucT, ATCC BAA-835) can be considered safe as a novel food, opening the door to its commercialization as a food supplement. Despite its recognized health benefits, there is still little information regarding the antimicrobial susceptibility of this species an

Akkermansia muciniphila protects against endometritis through a tyrosol-mediated gut-uterus axis.

Endometritis is a common reproductive disorder in humans and domestic animals that causes infertility and livestock losses. However, few specific bacteria and related metabolites involved in its pathogenesis have been identified. Here, we find that endometritis cows (EC) exhibit gut dysbiosis, with depletion of Akkermansia muciniphila (AKK). Gut microbiota transplantation from EC (E-GMT) induces endometritis in mice, whereas gut microbiota transplantation from healthy cows (H-GMT) does not. Supplementation with live AKK or its culture supernatant, but not heat-inactivated AKK, alleviates Esche

Next-generation probiotics and microbiome-based therapeutics highlighting Akkermansia muciniphila and Amuc_1100.

The gut microbiota is pivotal in regulating host metabolism, immune signaling, and the integrity of the mucosal barrier, thus serving as a critical factor in various metabolic, inflammatory, and gastrointestinal disorders. Traditional probiotics, primarily comprising Lacticaseibacillus and Bifidobacterium species, have been utilized to enhance gut health; however, the strain-dependent effects and variable clinical outcomes associated with these probiotics have prompted increased interest in next-generation probiotics (NGPs). Sleep Deprivation Exacerbates Ischemic Stroke Outcomes via Akkermansia Depletion and Metabolic Dysregulation.

AIMS: Sleep disturbances are a potential risk factor for stroke. Our clinical cohort shows that insomnia is associated with a higher 90-day stroke recurrence rate, prompting an investigation into the underlying mechanisms, particularly the role of the gut microbiota. METHODS: We utilized a rat MCAO model of sleep deprivation (SD) combined with antibiotic (ABX)-induced gut dysbiosis. Neurological deficits were assessed through behavioral tests, and cerebral infarct volume was measured using TTC staining. Structural changes in brain and intestinal tissues were observed by H&E staining, while th

Akkermansia muciniphila in Central Nervous System Disorders: Mechanisms, Controversies, and Therapeutic Potential.

Akkermansia muciniphila (A. muciniphila) is an intestinal mucus-dwelling mucindegrading bacterium that has recently attracted great interest due to its involvement in several metabolic diseases, including obesity, diabetes, and non-alcoholic fatty liver disease. Recent findings have indicated that A. muciniphila plays an important role in central nervous system (CNS) disorders via the microbiota-gut-brain axis (MGBA). This review highlights its dual roles in neuroprotection and pathogenesis, focusing on three key mechanisms, including immunomodulation, metabolic regulation, and barrier reinfor

Akkermansia muciniphila alleviates experimental colitis through FXR-mediated repression of unspliced XBP1.

UNLABELLED: Endoplasmic reticulum (ER) stress-related mucin depletion could be involved in the pathogenesis of ulcerative colitis (UC). Akkermansia muciniphila (A. muciniphila) uses mucin as its sole energy source and shows potential in the treatment of colitis. However, the effects and underlying mechanisms of A. muciniphila on colonic epithelial ER stress in colitis are largely unknown. Colitis was induced by adding 2.5% dextran sulfate sodium (DSS) in drinking water. Mice were orally administered A. muciniphila (3*10^7, 3*10^8 cfu/day) once daily for 10 days during DSS intervention. Ultra h

Mucin-derived sugars act as metabolic brakes controlling growth initiation in Akkermansia muciniphila.

Akkermansia muciniphila is a key member of the gut microbiota and plays important roles in host metabolism and health. In the colon, A. muciniphila extracts nutrients from oligosaccharide-rich mucin glycans that comprise the mucosa. However, this environment is complex and shaped by dietary inputs, microbiome metabolism, and mucin glycan composition varying across hosts, gastrointestinal regions, and physiological states. How strains of A. muciniphila integrate these nutrient signals into growth initiation and niche colonization remains unclear. Here, we compare physiological responses of a hu
