Transparent, Aldehyde-Free Chitosan Aerogel.

Aldehyde-free, transparent chitosan aerogel is reported. The aerogel was prepared by thermal decomposition of urea to induce gelation of a chitosan solution, followed by solvent exchange to ethanol, and supercritical drying. Low urea concentrations (≤ 25 g L-1) result in transparent and highly mesoporous aerogels, while higher urea concentrations (≥ 30 g L-1) produce opaque, more macroporous aerogels. The high surface areas of > 400 m2 g-1, large mesopore volumes up to 3.5 cm3 g-1, and optical transparency of the low-urea aerogels indicate a high structural homogeneity at the mesoscale, and th

Role of Aldehyde Dehydrogenases in Physiopathological Processes.

Many different diseases are associated with oxidative stress. One of the main consequences of oxidative stress at the cellular level is lipid peroxidation, from which toxic aldehydes may be generated. Below their toxicity thresholds, some aldehydes are involved in signaling processes, while others are intermediaries in the metabolism of lipids, amino acids, neurotransmitters, and carbohydrates. Some aldehydes ubiquitously distributed in the environment, such as acrolein or formaldehyde, are extremely toxic to the cell. On the other hand, aldehyde dehydrogenases (ALDHs) are able to detoxify a w

Carbethoxylating agents as inhibitors of aldehyde dehydrogenase.

N,O-Dicarbethoxy-4-chlorobenzenesulfohydroxamate (1c) and O-carbethoxy-N-hydroxysaccharin (6), both potential carbethoxylating agents, inhibited yeast aldehyde dehydrogenase (AlDH) with IC50's of 24 and 56 microM, respectively. The esterase activity of the enzyme was commensurably inhibited. AlDH activity was only partially restored on incubation with mercaptoethanol (20 mM) for 1 h. On incubation with rat plasma, 1c liberated nitroxyl, a potent inhibitor of AlDH. Under the same conditions, nitroxyl generation from 6 was minimal, a result compatible with a previous observation that nitroxyl ge

Prodrugs of nitroxyl as inhibitors of aldehyde dehydrogenase.

In the preceding paper, analogs of chlorpropamide with an OMe substituent on the sulfonamide nitrogen were shown to inhibit aldehyde dehydrogenase (AlDH), and it was postulated that these compounds were bioactivated by O-demethylation to release nitroxyl (HN = O, nitrosyl hydride), which is an inhibitor of AlDH. Further evidence for the production of nitroxyl from compounds with O-acyl instead of OMe on the sulfonamide nitrogen is now presented. Thus, nitrous oxide (N2O), the end product of nitroxyl dimerization and disproportionation, was found to be generated on alkaline or enzymatic hydroly

Staining properties of aldehyde fuchsin analogs.

This investigation was designed to clarify the role of the aldehyde component of aldehyde fuchsin in its staining reactions. Several aldehyde fuchsin analogs were prepared by using different aldehydes. The staining quality of these analogs and pararosaniline-HCl was compared with that of aldehyde fuchsin prepared with paraldehyde in the usual way. The major findings of this investigation include: 1) Aldehyde fuchsin staining of nonoxidized pancreatic B cells requires a stain prepared with either paraldehyde or acetaldehyde. 2) An aldehyde moiety is required for aldehyde fuchsin staining of str

Aldehyde dehydrogenase-2 as a therapeutic target.

Introduction: Aldehyde dehydrogenase-2 (ALDH2) is a main contributor of the alcohol elimination process. Functional polymorphism in the ALDH2 gene and its inactive form causes unpleasant flushing responses after alcohol consumption and prevents excessive alcohol intake. ALDH2 plays a key role in removing endogenous aldehydes like 4-hydroxy-2-nonenal (4-HNE) and malondialdehyde (MDA) produced by lipid peroxidation triggered by oxidative stress. Additionally, ALDH2 is involved in the elimination of metabolites of neurotransmitters like 3,4-dihydroxyphenylacetaldehyde (DOPAL) and 3,4-dihydroxyphe

Effects of benzodiazepines on aldehyde dehydrogenase activity.

The reported ability of benzodiazepines to increase human erythrocyte aldehyde dehydrogenase (ALDH) activity and reverse the disulfiram-induced inhibition of ALDH was reexamined. When ALDH activity assays were carried out spectrophotometrically on a hemoglobin-free lysate of human erythrocytes with propionaldehyde as substrate, addition of diazepam (10 mumol/l) did not affect the enzyme activity. When assays were carried out on intact or hemolysed erythrocytes using high performance liquid chromatographic technique with 3,4-dihydroxyphenylacetaldehyde as substrate, no significant increase in e

Alcohol and aldehyde dehydrogenase polymorphisms and alcoholism.

The alcohol-flush reaction occurs in Asians who inherit the mutant ALDH2*2 allele that produces an inactive aldehyde dehydrogenase enzyme. In these individuals, high blood acetaldehyde levels are believed to be the cause of the unpleasant symptoms that follow drinking. We measured the alcohol elimination rates and intensity of flushing in Chinese subjects in whom the alcohol dehydrogenase ADH2 and ALDH2 genotypes were determined. We also correlated ADH2, ADH3, and ALDH2 genotypes with drinking behavior in 100 Chinese men. We discovered that ADH2*2 and ADH3*1, alleles that encode the high activ

Contact urticaria from cinnamic aldehyde.

Lip swelling developed following the use of a cinnamic aldehyde-containing mouthwash in a woman with allergic rhinitis and asthma. Open application of the mouthwash on the antecubital fossae of the patient and control subjects produced immediate erythematous or urticarial responses in all tested individuals. Subsequent investigation established cinnamic aldehyde as the responsible ingredient. The incidence of positive reactions was shown to depend on site of application, vehicle, concentration, and time of reading. Of several related cinnamates, only cinnamic acid produced easily detectable re

Lessons learned from aldehyde dehydrogenases as non-P450 aldehyde-oxidizing enzymes: implications for the 'exposome' and human health.

INTRODUCTION: Aldehyde dehydrogenases (ALDHs) are critical enzymes that protect against cellular damage by metabolizing reactive aldehydes derived from both endogenous processes and environmental exposures. Although cytochrome P450 enzymes dominate metabolic toxicology discussions, the non-P450 ALDH superfamily plays a unique underrecognized role in mitigating the health impacts of the exposome - the totality of lifetime environmental exposures. AREAS COVERED: This Special Report highlights key insights from recent research on ALDHs, with a focus on their enzymatic diversity, disease-relevant

Body odour aldehyde reduction by acetic acid bacterial extract including enzymes: alcohol dehydrogenase and aldehyde dehydrogenase.

OBJECTIVE: The major causes of unpleasant human body odour are aldehydes produced by axillary-resident bacteria. There are many methods of body odour prevention; however, they all carry risks of destroying indigenous dermal bacteria that are necessary for the maintenance of the normal physical function of the skin. Furthermore, some methods cannot directly reduce the concentrations of substances that cause body odour. Therefore, a novel method of reducing body odour more safely and effectively is required. We focused on acetic acid bacterial enzymes, which can convert aldehydes into carboxylic

Hydralazine as a selective probe inactivator of aldehyde oxidase in human hepatocytes: estimation of the contribution of aldehyde oxidase to metabolic clearance.

Aldehyde oxidase (AO) metabolism could lead to significant underestimation of clearance in prediction of human pharmacokinetics as well as unanticipated exposure to AO-generated metabolites, if not accounted for early in drug research. We report a method using cryopreserved human hepatocytes and the time-dependent AO inhibitor hydralazine (K(I) = 83 ± 27 μM, k(inact) = 0.063 ± 0.007 min(-1)), which estimates the contribution of AO metabolism relative to total hepatic clearance. Using zaleplon as a probe substrate and simultaneously monitoring the AO-catalyzed formation of oxozaleplon and the C

Enantioselective Vinyl Radical Addition to Aldehydes via Dual Chromium Catalysis.

Achieving stereocontrol in radical-mediated transformations remains a fundamental challenge, with the enantioselective manipulation of highly reactive vinyl radicals being particularly elusive. Here, we report a dual catalytic system that synergistically merges chiral chromium catalysis with organophotoredox catalysis to address this challenge. This platform enables a three-component, enantioselective 1,2-dialkylation of alkynes, directly converting simple alkyl radical precursors, alkynes, and aldehydes into axially chiral allylic alcohols with high efficiency, diastereoselectivity (up to >20

Direct electrochemical oxidation of alkylarenes to aryl aldehydes and ketones.

A green and sustainable electrochemical protocol for the direct benzylic C(sp3)-H oxidation of alkylarenes to aryl aldehydes and ketones is developed. The protocol employs potassium persulfate as a co-oxidant under constant current electrolysis. It exhibits broad substrate scope, excellent functional group tolerance, and high site-selectivity without over-oxidation. Mechanistic studies, including 18O-labeling experiments, confirm that water serves as the exclusive oxygen atom donor. This operationally simple approach offers an economical alternative for aldehyde and ketone synthesis.

Deoxygenative Cyanofunctionalization of Aldehydes and Ketones Enabled by Electrochemical Reduction.

The deoxygenative difunctionalization of carbonyls converts readily available carbonyl compounds into value-added gem-difunctionalized building blocks. We report an electrochemical carbonyl umpolung approach for the deoxygenative cyanofunctionalization of aldehydes and ketones, affording various α-functionalized nitriles. Further derivatization of these products provides a versatile and modular platform for geminal disubstitution of carbonyl oxygens.

Benzyl Alcohol Functionalization of [1.1.1]Propellane with Alkanes and Aldehydes.

Bicyclo[1.1.1]pentanes (BCPs) play a crucial role in drug discovery research as C(sp3)-rich bioisosteres of benzene rings. However, the preparation of BCPs with strong alkane C(sp3)-H bonds has not been reported to date. In this study, we reported a method for light-induced benzyl alcohol functionalization of [1.1.1]propellane with aliphatic hydrocarbons (which have not previously been explored for this purpose) and aldehydes under metal- and photocatalyst-free conditions. The BCP products could be transformed into various useful derivatives, demonstrating the utility of the method. Notably, w

Chemodivergent Reactions of Aromatic Ring-Annulated Hexahydrocyclopentafurans with Various Aldehydes.

Hexahydro-2H-cyclopenta[b]furan fused to electron-rich aromatic rings reacts with various aromatic aldehydes in different modes to build diverse frameworks. The reaction of a dimethoxybenzene-fused cyclopentafuran generated diquinanes fused with a hydrofuran ring, indenopyrans, or diarylindanes depending upon the type of aromatic aldehydes, whereas an indole-annulated cyclopentafuran generated another type of diquinane fused with hydrofuran ring or benzylidenecyclopentafuran. The chemodivergence is due to the different properties between indole- and dimethoxybenzene-fused hydrocyclopentafurans

NHPI-Catalyzed Electro-Oxidation of Alcohols to Aldehydes and Ketones.

A practical and recyclable electro-oxidation of alcohols to aldehydes and ketones by using N-hydroxyphthalimide (NHPI) as the catalyst is presented. Through an undivided pool, under constant current conditions, various alcohols can be oxidized to the corresponding aldehydes or ketones in a high yield. Compared with previous methods, this system has the following characteristics: (1) the catalyst, electrode, electrolyte, and solvent (mainly water) are recyclable; (2) it has many advantages such as mild reaction conditions, easy operation, and good tolerance of functional groups; and (3) it can
