Modeling the kinetics of perfluorooctanoic and perfluorooctane sulfonic acid biodegradation by Acidimicrobium sp. Strain A6 during the feammox process.

Per- and polyfluoroalkyl substances (PFAS) are emerging contaminants of concern due to their health effects and persistence in the environment. Although perfluoroalkyl acids (PFAAs), such as perfluorooctanoic acid (PFOA) and perfluorooctanesulfonic acid (PFOS) are very difficult to biodegrade because they are completely saturated with fluorine, it has recently been shown that Acidimicrobium sp. A6 (A6), which oxidizes ammonium under iron reducing conditions (Feammox process), can defluorinate PFAAs. A kinetic model was developed and tested in this study using results from previously published

Distribution of per- and poly-fluoroalkyl substances and their precursors in human blood.

Many studies have examined per- and poly-fluoroalkyl substances (PFASs) in human blood. However, the distribution of PFASs in human blood remains not well known, especially for perfluorooctane sulfonate (PFOS) precursors. In this study, human blood samples (n = 162) were collected from general Chinese population, and then the isomer-specific partitioning of PFASs between human plasma and red blood cells (RBCs) were investigated. Perfluorooctanoate (PFOA) and PFOS were consistently the predominant PFASs in both human plasma and RBCs. In human blood, among C4-C7 perfluoroalkyl carboxylates (PFCA

A systematic evidence map of chronic inflammation and immunosuppression related to per- and polyfluoroalkyl substance (PFAS) exposure.

BACKGROUND: The ability to induce chronic inflammation and immunosuppression are two key characteristics of carcinogens and important forms of immunotoxicity. The National Toxicology Program (NTP) evaluated the immunotoxicity of two per- and polyfluoroalkyl substances (PFASs), PFOA (perfluorooctanoic acid) and PFOS (perfluorooctane sulfonate), in 2016. However, the potential pro-inflammatory and immunosuppressive effects of other PFASs remain largely uncharacterized. METHODS: We developed an expanded set of search terms pertaining to the chronic inflammatory and immunosuppressive effects of P

Potency Ranking of Per- and Polyfluoroalkyl Substances Using High-Throughput Transcriptomic Analysis of Human Liver Spheroids.

Per- and polyfluoroalkyl substances (PFAS) are some of the most prominent organic contaminants in human blood. Although the toxicological implications of human exposure to perfluorooctane sulfonate (PFOS) and perfluorooctanoate (PFOA) are well established, data on lesser-understood PFAS are limited. New approach methodologies (NAMs) that apply bioinformatic tools to high-throughput data are being increasingly considered to inform risk assessment for data-poor chemicals. The aim of this study was to compare the potencies (ie, benchmark concentrations: BMCs) of PFAS in primary human liver microt

Nonideal Transport and Extended Elution Tailing of PFOS in Soil.

The objective of this research was to examine the influence of nonideal sorption/desorption on the transport of polyfluorinated alkyl substances (PFASs) in soil, with a specific focus on characterizing and quantifying potential extended, mass-transfer-limited elution behavior. Perfluorooctane sulfonic acid (PFOS) was used as a representative PFAS, and miscible-displacement experiments were conducted with two soils comprising contrasting geochemical properties. The influence of nonlinear, rate-limited, hysteretic, and irreversible sorption/desorption on transport was investigated through experi

Radiosynthesis of perfluorooctanesulfonate (PFOS) and perfluorobutanesulfonate (PFBS), including solubility, partition and adhesion studies.

Here, we describe for the first time the synthesis of [(35)S] PFOS and [(35)S] PFBS with sulfur-35 enriched sulfur dioxide as the radiolabelled reagent, resulting in 2.5 and 2.3 mCi of product, respectively. Basic information concerning the physicochemical properties of perfluorooctanesulfonate (PFOS), perfluorobutanesulfonate (PFBS) and perfluorooctanoic acid (PFOA) are still limited. Hence, we utilized these radiolabelled perfluoroalkanesulfonates (PFSAs), as well as carbon-14 labelled perfluorooctanoic acid ([(14)C] PFOA) to determine some basic characteristics of physiological and experime

Equilibrium dialysis of barbituric acids up to high concentrations of aqueous sodium alkylsulfonate solutions.

The partition coefficient K of four barbituric acids has been determined by the equilibrium dialysis method in aqueous solutions of sodium alkylsulfonate at 25 degrees C. The results obtained from dilute solutions to 0.3 M surfactant concentration are compared with solubility data for the same systems. The K values as deduced from Langmuir or Freundlich isotherms decrease with increasing surfactant concentration. The detergent used was an impure commercial product of known composition, which could be considered as a mixed surfactant. The change of partition coefficient with surfactant concentr

Atomistic simulation study of linear alkylbenzene sulfonates at the water/air interface.

Molecular dynamics simulations with the CHARMM atomistic force field have been used to study monolayers of a series of linear alkylbenzene sulfonates (LAS) at the water/air interface. Both the numbers of carbon atoms in the LAS alkyl tail (1 to 11) and the position of attachment of the benzene ring on the alkyl chain have been varied. Totally 36 LAS homologues and isomers have been investigated. The surface tensions of the systems and the average tilt angles of the LAS molecules are found to be related to both the length and the degree of branching of the alkyl tails, whereas the solubility an

Non-Negligible Source of PFOS: Biochemical Fate and Metabolic Activation of Precursors.

The widespread detection of PFOS precursors in the environment and human matrices raises a new wave of health concerns. However, their absorption, distribution, metabolism, and excretion (ADME) fates and toxicity profiles in mammals remain largely unexplored. Here, we investigated the toxicokinetic, metabolic transformation, and toxicity of three precursors, N-MeFOSA, N-EtFOSA, and N-EtFOSAA, by integrating toxicokinetic modeling in mice, rat liver microsome metabolism, and hepatocellular toxicity assays. N-MeFOSA and N-EtFOSA were rapidly eliminated in vivo and exhibited high efficiencies of

Hydrothermal synthesis of SnO2 nanoparticles for electrocatalytic detection of perfluorononanoic acid and perfluorooctanesulfonic acid in environmental samples using voltammetric techniques.

Perfluorochemicals are synthetic compounds used in many applications; however, their slow degradation classifies them as emerging micropollutants that persist in the environment. Long-term exposure to perfluorononanoic acid (PFNA) and perfluorooctanesulfonic acid (PFOS) via soil, water, and food can cause human health problems. In this research, SnO2 nanoparticles were synthesized hydrothermally to study the electrocatalytic detection of PFNA and PFOS. The tin oxide (SnO2) nanoparticles were characterized by scanning electron microscopy and X-ray diffraction, which showed a tetragonal rutile s

Investigating the degradation potential of microbial consortia for perfluorooctane sulfonate through a functional "top-down" screening approach.

Perfluorooctane sulfonate (PFOS) is a prominent perfluorinated compound commonly found in the environment, known to pose various risks to human health. However, the removal of PFOS presents significant challenges, primarily due to the limited discovery of bacteria capable of effectively degrading PFOS. Moreover, single degradation bacteria often encounter obstacles in individual cultivation and the breakdown of complex pollutants. In contrast, microbial consortia have shown promise in pollutant degradation. This study employed a continuous enrichment method, combined with multiple co-metabolic

Biogas utilization without desulfurization pretreatment in a bioelectrochemical system.

Utilizing biogas as a fuel for heating and power generation usually requires desulfurization pretreatment. In this study, the biogas utilization without desulfurization pretreatment in a bioelectrochemical system (BES) was explored. The results showed that the biogas-fueled BES was successfully started up within 36 d and the presence of hydrogen sulfide promoted both methane consumption and electricity generation. The optimal performance (i.e., a methane consumption of 0.523 ± 0.004 mmol/d, a peak voltage of 577 ± 1 mV, a coulomb production of 37.86 ± 0.43C/d, a coulombic efficiency of 9.37 ±

PFOS-induced thyroid hormone system disrupted rats display organ-specific changes in their transcriptomes.

Perfluorooctanesulfonic acid (PFOS) is a persistent anthropogenic chemical that can affect the thyroid hormone system in humans and animals. In adults, thyroid hormones (THs) are regulated by the hypothalamic-pituitary-thyroid (HPT) axis, but also by organs such as the liver and potentially the gut microbiota. PFOS and other xenobiotics can therefore disrupt the TH system at various locations and through different mechanisms. To start addressing this, we exposed adult male rats to 3 mg PFOS/kg/day for 7 days and analysed effects on multiple organs and pathways simultaneously by transcriptomics

Efficacy of binders on sequestering per- and polyfluoroalkyl substances under in vitro ruminal conditions.

The efficacy of 6 binders in sequestering perfluorooctane sulfonic acid (PFOS) under in vitro ruminal conditions was evaluated. Binders, including bentonite, montmorillonite aluminum pillared clay, chitosan, activated charcoal Norit, and cholestyramine resin (CHM), were tested at a dose of 1.8% (wt/wt; fresh basis). The PFOS (%) binding affinity of binders was calculated in relation to the control. A randomized complete block design (n = 4) was used to analyze the data. Only CHM, an anion-exchange resin, demonstrated strong potential to sequester PFOS (x_=52.5%) The binders did not affect the

Novel PFOS alternative OBS inhibits body growth of developing zebrafish by triggering thyroid function disorder and osteoclast differentiation.

The extensive use of the perfluorooctane sulfonate (PFOS) alternative sodium p-perfluorous nonenoxybenzene sulfonate (OBS) has resulted in its widespread detection in the environment and enrichment in wildlife and humans. However, little is known about its potential toxicity, particularly in terms of body development. In this study, zebrafish embryos were acutely exposed to PFOS and OBS for a comparative developmental toxicity assessment. Both PFOS and OBS led to lower body weight and shorter body length, and the damaging effects of PFOS were more severe than those of OBS at the same exposure

Per- and polyfluoroalkyl substances (PFAS) in grocery store foods: method optimization, occurrence, and exposure assessment.

Dietary exposure to per- and polyfluoroalkyl substances (PFAS) is poorly understood. Evaluating PFAS in food is complicated by the need to evaluate varied matrices and a lack of a standard, matrix-specific sample extraction methods. Prior food studies implemented universal rather than matrix-specific extraction approaches, which may yield false negatives and an underestimation of PFAS dietary exposure if methods are not suitable to all matrices. Here the objectives were to screen and optimize PFAS extraction methods for plants, tissues, and dairy; apply optimized extraction methods to a grocer

Accumulation and glucocorticoid signaling suppression by four emerging perfluoroethercarboxylic acids based on animal exposure and cell testing.

Various perfluoroethercarboxylic acids (PFECA) have emerged as next-generation replacements of legacy per- and polyfluoroalkyl substances (PFAS). However, there is a paucity of information regarding their bioaccumulation ability and hazard characterization. Here, we explored the accumulation and hepatotoxicity of four PFECA compounds (HFPO-DA, HFPO-TA, PFO4DA, and PFO5DoDA) in comparison to perfluorooctanoic acid (PFOA) after chronic low-dose exposure in mice. Except for HFPO-DA, the levels of all tested PFAS in the liver exceeded that in serum. High molecular weight PFECA compounds (PFO5DoDA
