Theoretical and experimental NMR studies on muscimol from fly agaric mushroom (Amanita muscaria).

In this article we report results of combined theoretical and experimental NMR studies on muscimol, the bioactive alkaloid from fly agaric mushroom (Amanita muscaria). The assignment of (1)H and (13)C NMR spectra of muscimol in DMSO-d6 was supported by additional two-dimensional heteronuclear correlated spectra (2D NMR) and gauge independent atomic orbital (GIAO) NMR calculations using density functional theory (DFT). The effect of solvent in theoretical calculations was included via polarized continuum model (PCM) and the hybrid three-parameter B3LYP density functional in combination with 6-3

Acute liver failure caused by Amanita verna: a case series and review of the literature.

BACKGROUND: Amanita verna is one of the most harmful wild fungi in China. Amanita verna poisoning occurs every year, and the mortality is as high as 50%. However, its clinical manifestations are complex and diverse. CASE PRESENTATION: In March 2019, three patients took a large amount of Amanita, and one of them received liver transplantation in Zhongshan hospital, Sun Yat-sen University. All patients had vomiting and diarrhea 8-12 h after eating wild mushrooms (Amanita). The patients were initially diagnosed with Amanita poisoning. One case (case 3) was complicated and diagnosed as mushroom p

Amanita poisoning during the second trimester of pregnancy. A case report and a review of the literature.

Amanita phalloides-type mushroom poisoning is well recognized as causing acute liver injury and often death. Less is known, however, of whether maternal Amanita poisoning is associated with fetal damage or not. In August 1991 four members of a family were hospitalized with food intoxication caused by Amanita phalloides and Amanita verna. One of them died from hepatic and renal failure. The survivors included a 26-year-old woman in the 23rd week of pregnancy. Her clinical symptoms and blood chemistry data (lowest prothrombin activity 23%) indicated intoxication of medium severity. The managemen

Acute renal failure associated with suspected Amanita smithiana mushroom ingestions: a case series.

Mushroom poisoning leading to acute renal failure is extremely rare in North America. Cortinarius species and Amanita smithiana both can cause acute renal failure and inhabit the Pacific northwest. This article describes 4 patients who presented in acute renal failure and who ingested mushrooms described as resembling A. smithiana. Two patients (a 74-year-old Korean couple) described eating mushrooms with an approximately 6 x 0.5-inch stipe with a white cap, 1.25-1.5 inches in diameter. The other 2 patients (a 55-year-old male and a 30-year-old female) also described a white-capped mushroom. A

Coagulation disorders secondary to acute liver failure in Amanita phalloides poisoning: a case report.

Amanita mushroom poisoning is a serious occurrence. Physicians need to recognize the syndrome promptly and institute effective treatment as soon as possible in order to avoid the often fatal complications. We report a case of a young man with mushroom poisoning of the genus Amanita with a clinical spectrum that ranged from hepatic failure and coagulation disorders to an outcome with a partial recovery. The aim of this report was to emphasize the clinical outcomes and treatment regimens in this type of mushroom poisoning seen with increasing frequency in our region.

Tumor therapy with Amanita phalloides (death cap): stabilization of B-cell chronic lymphatic leukemia.

BACKGROUND: Molecular events that cause tumor formation upregulate a number of HOX genes, called switch genes, coding for RNA polymerase II transcription factors. Thus, in tumor cells, RNA polymerase II is more active than in other somatic cells. Amanita phalloides contains amanitin, inhibiting RNA polymerase II. Partial inhibition with amanitin influences tumor cell--but not normal cell--activity. OBJECTIVES: To widen the treatment spectrum, homeopathic dilutions of Amanita phalloides, containing amanitin, were given to a patient with leukemia. Monitoring the leukemic cell count, different d

Renal and hepatic injury with elevated cardiac enzymes in Amanita phalloides poisoning: a case report.

Amatoxins are one of the most potent toxins that cause hepatic and renal failure. However, this is the first report demonstrating an elevation of cardiac enzymes in a patient with Amanita phalloides poisoning. A 56-year-old male was admitted to the emergency department (ED) 42 h after an unknown type of mushroom ingestion. Hepatic, renal function tests, amylase and cardiac enzymes (troponin I, creatine kinase (CK), CK-MB isoenzyme and myoglobin) were found elevated in his blood chemistry. The electrocardiogram disclosed sinus tachycardia. Aggressive treatment with fluids, activated charcoal, p

Genome Mining and Total Synthesis of Viduamanide, A Non-canonical Prolineless Cycloamanide from Amanita vidua.

The MeOH extract of the newly described deadly mushroom Amanita vidua was dominated by an unknown peptide. Despite the limited fungal material available, the compound was isolated and characterized as a homodetic cyclic hexapeptide by NMR and MS/MS analyses, and its structure was confirmed by high-efficiency solid-phase peptide synthesis. Recently implemented genome mining further validated the structure and provided a broader insight into the cyclopeptide-producing capabilities of this rare mushroom. Named viduamanide, this compound is a non-canonical cycloamanide of the "prolineless" subtype

Extrapolating Qualitative Mycochemical Profiling and Therapeutic Potential of Two Mycorrhizal Amanita Species (Agaricomycetes) from Pakistan.

The present study was an attempt to evaluate the antimicrobial and anthelmintic potential of two Amanita species, i.e., A. orsonii and A. glarea, along with their qualitative mycochemical screening. Maceration technique was adopted to make crude extracts in non-polar (petroleum ether and chloroform) and polar (ethanol and distilled water) solvents. Qualitative mycochemical screening revealed the presence of a variety of secondary metabolites like terpenoids, flavonoids, tannins, alkaloids, saponins, and cardiac glycosides. Antimicrobial activities were carried out by using agar well diffusing

Ibotenic Acid and Muscimol in Amanita muscaria: Chemistry, Sources of Variability, Analytical Determination, and Toxicological Significance.

Amanita muscaria contains two structurally related neuroactive isoxazole metabolites, ibotenic acid (IA) and muscimol (MUS), which differ markedly in pharmacological activity. IA acts as an agonist at NMDA and metabotropic glutamate receptors, whereas MUS is a potent GABAergic agonist. This review critically evaluates their chemistry and biosynthesis, biological variability, post-harvest transformation, analytical determination, and toxicological significance. Direct evidence demonstrates interspecific variation within Amanita section Amanita. Developmental evidence for A. muscaria is limited

Effects of Amanita muscaria extract on different in vitro neurotoxicity models at sub-cellular and cellular levels.

Muscimol is the main compound found in Amanita muscaria. Several studies have proven that muscimol has suppressive effects on essential tremor, without impairing speech and coordination. The effects of muscimol in Parkinson-affected patients is also described in a number of studies. These studies describe the free radical scavenging and antioxidant activity of the mushroom extract. We have evaluated the possible neuroprotective effects of a standardized extract from A. muscaria, containing high amounts of muscimol, on different models of neurotoxicity in rat brain microsomes, mitochondria, syn

Acute Amanita muscaria Toxicity: A Literature Review and Two Case Reports in Elderly Spouses Following Home Preparation.

Amanita muscaria (L.) Lam., commonly known as fly agaric, remains an uncommon yet clinically important cause of acute mushroom intoxication. Although typically associated with mild to moderate neuropsychiatric disturbances, the mushroom's toxic profile is highly variable and continues to attract scientific, toxicological, and public health interest. This work provides an integrative review of the biochemical composition, toxicodynamics, and clinical manifestations associated with A. muscaria exposure, with particular emphasis on the pharmacological actions of its principal constituents, iboten

Therapeutic plasma exchange in amatoxin associated acute liver failure-results from the multi-center Amanita-PEX study.

BACKGROUND: Amatoxin-related acute liver failure (AT-ALF) carries high mortality without liver transplantation (LTX). While therapeutic plasma exchange (PEX) might improve LTX-free survival in other ALF cases, its role in AT-ALF is unclear. Clinical practice varies, and, given the rarity of this ALF entity, the feasibility of conducting a randomized controlled trial to investigate PEX in AT-ALF is more or less impossible. METHODS: The Amanita-PEX study is a multi-center, international, retrospective study analyzing patients with AT-ALF from 2013 to 2024. The primary outcome was 28-day LTX-fre

Cimetidine protection against alpha-amanitin hepatotoxicity in mice: a potential model for the treatment of Amanita phalloides poisoning.

The ingestion of the mushroom Amanita phalloides is associated with hepatic necrosis appearing clinically two to three days after ingestion. The mechanism of this toxicity is unknown, and no reliable antidote is available. Because of the similarity of Amanita poisoning to other toxins affecting cytochrome P450, we investigated the use of cimetidine (as a P450 cytochrome inhibitor) as an antidote against a primary toxin of the mushroom alpha-amanitin. Mice injected with alpha-amanitin and given cimetidine either prophylactically or within six hours showed histologic protection from the hepatic
