Ameloblastic fibrodentinoma (AFD) is classified as a rarely seen benign mixed odontogenic tumor. Ameloblastic fibrodentinoma is common in children and young adults with painless swelling and mostly seen at the mandibular posterior region and usually associated with unerupted tooth. In this clinical report, an 11-year-old-boy referred to Ankara University, Faculty of Dentistry, with the complaint of pain and swelling at the mandibular posterior region, diagnosed as AFD, is presented. Extraoral examination showed swelling at the affected side. In intraoral examination, a rough painful swelling w

Ameloblastic fibroma (AF) is an extremely rare true mixed benign tumor that can occur in either the mandible or the maxilla, but is most frequently found in the posterior region of the mandible. It usually occurs in the first two decades of life and is associated with tooth enclosure, causing a delay in eruption or altering the dental eruption sequence. AF is diagnosed on routine radiographic evaluation and is clinically and radiographically similar to ameloblastic fibrodontoma and odontoma, which makes an accurate diagnosis mandatory. There is controversy in the literature as to whether treat

Nine cases of ameloblastic fibroma are described and compared with published data on this odontogenic tumour. The median age of the 7 male and 2 female patients was 11 years (range 2-17 yrs.). The tumour was localized in the posterior segment of the mandible and less often in the maxilla. The neoplasia was frequently associated with impaction and agenesis of teeth. Although recurrences of ameloblastic fibromas are rare, long-term follow-up is recommended.

Intake of excess amounts of fluoride during tooth development cause enamel fluorosis, a developmental disturbance that makes enamel more porous. In mild fluorosis, there are white opaque striations across the enamel surface, whereas in more severe cases, the porous regions increase in size, with enamel pitting, and secondary discoloration of the enamel surface. The effects of fluoride on enamel formation suggest that fluoride affects the enamel-forming cells, the ameloblasts. Studies investigating the effects of fluoride on ameloblasts and the mechanisms of fluorosis are based on in vitro cult

Ameloblastic carcinoma (AC) represents a distinct challenge in the realm of odontogenic malignancies due to its rarity and aggressive nature. We present a unique case of AC in a 70-year-old male, retired dry cleaner, with symptoms initially suggestive of chronic allergic rhinitis and recurrent acute sinusitis with asymmetric facial edema and paresthesia. Detailed evaluation revealed a prominent mass in the right maxillary sinus with extensive cortical destruction. Pathological assessment post-right maxillectomy identified a high-grade AC with malignant spindle cell transformation. The patient

Ameloblastic fibroma is a rare mixed odontogenic tumour which involves mostly the posterior mandible. Its peripheral variant is very rare. Only eight cases have been reported worldwide. In this report, we described a case of peripheral ameloblastic fibroma occurring in the maxillary gingiva in a 10 year-old child. The lesion was excised with a conservative surgical approach and no recurrence has occurred. Peripheral ameloblastic fibroma should be considered in the differential diagnosis of a slow growing lesion involving the gingiva.

Ameloblastic fibroma (AF) is a rare benign odontogenic tumour first described by Kruse in 1891. Although reported in a wide age range, most of the cases are seen in the first two decades of life with majority of cases being diagnosed before the age of 20 years. There are reported variations in the histopathological presentation of ameloblastic fibroma. In this case report, we present a case of ameloblastic fibroma with extensive dentinoid formation involving the mandible in an 8-year-old male patient which clinically presented as an aggressive lesion.

Dental enamel formation is coordinated by ameloblast differentiation, production of enamel matrix proteins, and crystal growth. The factors regulating ameloblast differentiation are not fully understood. Here we show that the high mobility group N (HMGN) nucleosomal binding proteins modulate the rate of ameloblast differentiation and enamel formation. We found that HMGN1 and HMGN2 proteins are downregulated during mouse ameloblast differentiation. Genetically altered mice lacking HMGN1 and HMGN2 proteins show faster ameloblast differentiation and a higher rate of enamel deposition in mice mola

Ameloblastic fibro-odontoma (AFO) is a rare, benign, and mixed odontogenic tumor that consists of both ectodermal and mesenchymal elements. AFO is more prevalent in young children and adolescents than in adults and is usually found in the molar area associated with a failure of tooth eruption. The purpose of this report is to discuss the differential diagnosis and treatment of a three-year-old girl diagnosed with an AFO around a primary canine. The manifestations of the lesion resembled localized periodontal disease caused by an enamel pearl. Excision and curettage were done and the separated

A 24-year-old womanpresented with ameloblastic fibrosarcoma arising from ameloblastic fibroma. The delayed presentation accounted for the extensive destruction of the mandible and complete occlusion of her oral cavity. This resulted in an inability to eat and maintain oral hygiene. A multidisciplinary team management approach involved nutritional optimisation, segmental mandibulectomy, reconstruction with a reconstructive plate and a free anterolateral thigh flap to line the the floor of mouth. Functional and aesthetic outcome was acceptable, and the patient is planned for secondary free fibul

Ameloblastic carcinoma (AC) is a rare malignant odontogenic tumor. Surgical resection of the tumor is the mainstay of its treatment. To date, radiotherapy for this tumor remains controversial. This report describes a case of AC with intracranial extension and provides the first report of the efficacy of single-fraction helical tomotherapy for the treatment of residual AC after surgical resection.

Ghost cell differentiation within an ameloblastic fibroma is extremely rare. The ghost cells found in an ameloblastic fibroma in previously reported cases were all associated with a typical calcifying odontogenic cyst. Here, we report a case of an ameloblastic fibroma with focal ghost cells and calcifications in some neoplastic epithelial islands, but without other histologic manifestations consistent with a calcifying odontogenic cyst. The patient was a 13-year-old Chinese boy who presented with a bony-hard swelling in the posterior mandibular region over a 6-month period. Radiographs showed

Ameloblastic carcinoma is a rare malignant lesion with characteristic histologic features and behaviour that dictates a more aggressive surgical approach than that of a simple ameloblastoma. However, reliable evidence of its biologic activity is currently unavailable due to the scarcity of well-documented cases. It occurs primarily in the mandible in a wide range of age groups; no sex or race predilection has been noted. It may present as a cystic lesion with benign clinical features or as a large tissue mass with ulceration, significant bone resorption and tooth mobility. Because the lesion i

Introduction: Developmental defects of the enamel manifest before tooth eruption and include amelogenesis imperfecta, a rare disease of underlying gene mutations, and molar-incisor hypomineralization (MIH), a prevalent disease in children originating from environmental and epigenetic factors. MIH enamel presents as the abnormal enamel marked by loss of translucency, demarcation between the healthy and affected enamel, and reduced mineral content. The pathophysiology of opaque, demarcated enamel lesions is not understood; however, the retention of enamel proteins in the matrix has been suggeste

The Sp7/Osterix transcription factor is essential for bone development. Mutations of the Sp7 gene in humans are associated with craniofacial anomalies and osteogenesis imperfecta. However, the role of Sp7 in embryonic tooth development remains unknown. Here we identified the functional requirement of Sp7 for dentin synthesis and tooth development. Sp7-null mice exhibit craniofacial dysmorphogenesis and are completely void of alveolar bone. Surprisingly, initial tooth morphogenesis progressed normally in Sp7-null mice. Thus the formation of alveolar bone is not a prerequisite for tooth morphoge

After addition of ethanol to the ordinary fodder of pregnant mini-pigs, ultrastructural changes were found in secretory ameloblasts from tooth germs of their mid-term fetuses. Compared with controls, many mitochondria showed abnormal shape and size, and some exhibited deposition of paracrystalline material in the matrix. An abnormal deposition of stippled material intercellularly was also observed. These changes were interpreted as signs of an abnormal secretory function.

Apoptotic reactions were demonstrated with a commercial detection kit in the reduced ameloblasts around the tip of the cusp just before and after exposure to the mouth but, as tooth eruption advanced, they were no longer seen in the remainder of the reduced ameloblasts. Although some of the reduced ameloblasts appear to be expelled as a consequence of apoptosis, the remainder appear to constitute the junctional epithelium.

Ultrastructural changes of secreting rat-incisor ameloblasts after administration of vincristine and vinblastine were observed. Polarity of nuclei and cytoplasmic organellae was remarkably disturbed; consequently, the regular secreting orientation of cellular products was altered greatly, leading to heterotopic deposition of large amounts of enamel matrix.

PURPOSE: Ameloblastic fibro-odontoma (AFO) is a rare benign mixed odontogenic tumor that, after being classified for years as a distinct entity, was redefined as a "developing odontoma" in the 2017 World Health Organization classification. This article presents a unique case of an AFO with an FGFR1 mutation.
METHODS: We present a case of an 8-year-old child with a slowly progressive swelling in the lower left mandible. Next-generation sequencing (TSO500 panel) was performed.
RESULTS: Panoramic radiography revealed an odontogenic tumor; therefore, a transoral enucleation was performed. Pathol
