Post-translational amino acid isomerization: a functionally important D-amino acid in an excitatory peptide.

The post-translational modification of an L- to a D-amino acid has been documented in relatively few gene products, mostly in small peptides under 10 amino acids in length. In this report, we demonstrate that a 46-amino acid polypeptide toxin has one D-phenylalanine at position 44, and that the epimerization from an L-Phe to a D-Phe has a dramatic effect on the excitatory effects of the peptide. In one electrophysiological assay carried out, the D-Phe-containing peptide was extremely potent, whereas the unmodified polypeptide had no biological activity, demonstrating that the chirality of the

Posttranslational amino acid epimerization: enzyme-catalyzed isomerization of amino acid residues in peptide chains.

Since ribosomally mediated protein biosynthesis is confined to the L-amino acid pool, the presence of D-amino acids in peptides was considered for many years to be restricted to proteins of prokaryotic origin. Unicellular microorganisms have been responsible for the generation of a host of D-amino acid-containing peptide antibiotics (gramicidin, actinomycin, bacitracin, polymyxins). Recently, a series of mu and delta opioid receptor agonists [dermorphins and deltorphins] and neuroactive tetrapeptides containing a D-amino acid residue have been isolated from amphibian (frog) skin and mollusks.

Plasma amino acid concentrations during branched-chain amino acid infusions in stressed patients.

To determine the effect of infusing large quantities of BCAA on plasma amino acid concentrations, plasma amino acid profiles were measured in 18 stressed patients before and 48 to 96 hours after initiation of amino acid solutions enriched with or exclusively containing BCAA (15.6+, 50%, 100%). Plasma concentrations of BCAA were elevated in the 100% and 50% BCAA groups, but not in the 15.6% group. Methionine, glycine, and phenylalanine concentrations were increased in the 15.6% BCAA group: methionine and glycine were decreased in the 100% BCAA groups. In the 50% BCAA group, nonbranched-chain am

Transforming natural amino acids into alpha-alkyl-substituted amino acids with the help of the HOF.CH3CN complex.

Alpha-alkyl amino acids can be efficiently prepared in high yields from the respective amino acids themselves. The key step is the oxidation of the amine function to create the corresponding alpha-nitro acid in a fast and very high yield reaction followed by phase-transfer alkylation and finally reduction to the desired alpha-alkyl amino acid. Several such acids containing aromatic rings or additional carboxylic groups and acids with steric hindrance at the alpha-position are suitable substrates. Several alkyl halides were examined as alkylating agents.

Amino acids and memory consolidation in the cricket. II: Effect of injected amino acids and opioids on memory.

The effect of injections of selected amino acids on memory, given before a maze-learning, was investigated. Thirsty crickets (Pteronemobius sp.) were trained to turn only to one side of a symmetrical Y-shaped maze using reinforcements with water. The insects retained the learned task 24 hr later. N2 anoxia applied immediately after training produced retrograde amnesia. Injections of Ala, Arg, Gln or morphine prior to training blocked the amnesic action of anoxia, whereas those of Cys, Met, Pro, Orn, octopamine or naloxone did not. Naloxone blocked long-term memory formation, but not learning,

Effects of dietary amino acids on brain amino acids and transmitter amines in rats with a portacaval shunt.

The etiologic relationship between disturbances in metabolism of amino acids and amines and hepatic coma was investigated by examining the effects of diets containing various mixtures of amino acids on brain amine metabolism in rats with a portacaval shunt, using a method for simultaneous analysis of amino acids and amines. Rats with a portacaval shunt were fed on four different amino acid compositions with increased amounts of various amino acids suspected to be etiologically related to hepatic coma, such as methionine, phenylalanine, tyrosine, and tryptophan. The animals were killed 4 weeks

Amino acid derangements in patients with sepsis: treatment with branched chain amino acid rich infusions.

Sepsis is a major catabolic insult resulting in modifications in carbohydrate and fat energy metabolism, and leading to increased muscle breakdown and nitrogen loss. Insulin resistance, which develops in sepsis, decreases glucose utilization, but plasma insulin levels are sufficiently elevated to prevent lipolysis, resulting in a further energy deficit. The availability of fuels in sepsis is therefore limited, and the body resorts to muscle breakdown, gluconeogenesis, and amino acid oxidation for energy supply. Previous work has not defined, however, the exact alterations in amino acid metabol

Effect of essential amino acid supplementation on muscle and plasma free amino acids in chronic uremia.

In chronic uremia patients on an 18-g protein diet for 2 to 3 weeks, the intracellular free amino acid pattern in muscle was grossly abnormal, with low concentrations of threonine, valine, carnosine, and high concentrations of aspartate, glutamine, glycine, alanine, and taurine. When the protein-poor diet was supplemented with essential amino acids (2 to 3 times that for minimum requirements) and histidine, these abnormalities were partly corrected. Other abnormalities, however, were observed, such as low intracellular concentrations of phenylalanine and tyrosine, and high concentrations of ly

Transport of amino acids and nucleic acid precursors in malarial parasites.

In vitro studies have shown that exogenously supplied amino acids are transferred into the malaria-infected cell, where they are incorporated into proteins. Most amino acids appear to enter the cell by facilitated or simple diffusion; however, the high distribution ratios seen in Plasmodium knowlesi-infected cells are difficult to explain on this basis. The changes (leakiness) observed in amino acid transport in P. lophurae infected cells are probably the result of ATP depletion in the host cell as well as the elaboration of plasmodial substances. Depletion of isoleucine, methionine, and cyste

Sulfuric Acid Catalyzed Esterification of Amino Acids in Thin Film.

The esterification reaction of different amino acids with methanol catalyzed by H2SO4 was first studied in the small volume of thin films generated by ESI microdroplet deposition. The reaction is promoted by the pneumatic spray of the ESI source and reaches its maximum efficiency at a thin film temperature of 70 °C. Selective esterification of the COOH moiety was demonstrated. Microdroplet size and thin film volume and lifetime are critical parameters that influenced the reaction outcome. As expected, l-tyrosine and l-phenylalanine having aromatic side chain substituents were the most reactive

Characteristics and formation of amino acids and hydroxy acids of the Murchison meteorite.

Eight characteristics of the unique suite of amino acids and hydroxy acids found in the Murchison meteorite can be recognized on the basis of detailed molecular and isotopic analyses. The marked structural correspondence between the alpha-amino acids and alpha-hydroxy acids and the high deuterium/hydrogen ratio argue persuasively for their formation by aqueous phase Strecker reactions in the meteorite parent body from presolar, i.e., interstellar, aldehydes, ketones, ammonia, and hydrogen cyanide. The characteristics of the meteoritic suite of amino acids and hydroxy acids are briefly enumerat

Inhibition of urease activity by hydroxamic acid derivatives of amino acids.

Hydroxamic acids have been reported to be potent and specific inhibitors of urease (EC 3.5.1.5) activity of plant and bacterial origin. The present investigation was performed on the inhibitory effect of hydroxamic acid derivatives of naturally occurring amino acids on the urease activity of the Jack Bean and the alimentary tracts of rats. Methionine-hydroxamic acid was the most powerful inhibitor (I50=3.9 X 10(-6) M) among nineteen alpha-aminoacyl hydroxamic acids. Phenylalanine-, serine-, alanine-, glycine-, histidine-, threonine-, leucine-, and arginine-hydroxamic acids followed, in order o

Influence of pH on radical reactions between kynurenic acid and amino acids tryptophan and tyrosine. Part I. Amino acids in free state.

In the human eye lens the endogenous chromophores of UV-A light (315-400 nm) are able to sensitize radical reactions leading to protein modifications during normal aging and the cataract progression. Kynurenic acid (KNA-) is the most photochemically active dye of the human eye lens reported to date with pKa(KNAH2•) 5.5 for its radical form. Cataract is thought to develop under oxidative stress which could be accompanied by acidosis, an acidification of the intracellular environment. Protonation of kynurenyl radicals at mildly acidic conditions may change the outcome of radical reactions leadin

Dinuclear chromium(V) amino acid complexes from the reduction of chromium(VI) in the presence of amino acid ligands: XAFS characterization of a chromium(V) amino acid complex.

The first synthesis and characterization of Cr(V) complexes of non-sulfur-containing amino acids are reported. The reduction of Cr(VI) in methanol in the presence of amino acids glycine, alanine, and 2-amino-2-methylpropanoic acid (alpha-aminoisobutyric acid, Aib) yielded several Cr(V) EPR signals. For the reaction involving glycine, the only Cr(V) EPR signals detected were those of the Cr(V)-intermediate methanol complexes, which were also observed in the absence of amino acids. The reaction involving alanine yielded one Cr(V) signal with a g(iso) value of 1.9754 (a(iso) = 4.88 x 10(-4) cm(-1

The kinetics of amino acid disappearance in the small intestine is related to the extent of amino acids absorbed in growing pigs.

This study evaluated the importance of a correction for amino acids (AA) released into the hindgut on a measure of AA absorption kinetics and tested whether AA absorption kinetics are related to the extent of AA absorption using the growing pig as a model for humans. Thirty-six nine-week-old pigs (22·3 kg) received a diet containing whey protein as the sole protein source for 8 d. Pigs received their last meal containing the indigestible marker titanium dioxide before being euthanised at 1, 2, 3, 4, 6 and 12 h post-feeding. The entire content of each gastrointestinal tract (GIT) region was col

Amino acid disorders detected by quantitative amino acid HPLC analysis in Thailand: an eight-year experience.

BACKGROUND: Amino acid disorders are a major group of inborn errors of metabolism (IEM) with variable clinical presentations. This study was aimed to provide the data of amino acid disorders detected in high-risk Thai patients referred to our metabolic lab from all over the country. METHODS: From 2001 to 2009, we analyzed amino acids by HPLC in 1214 plasma and cerebrospinal fluid specimens. These specimens were obtained from patients with clinical suspicion of IEM or with positive newborn screening. The clinical data of the patients with confirmed diagnoses of amino acid disorders were also a
