Neural correlates of concern about falling in multiple sclerosis: resting-state functional connectivity in amygdala-hippocampal and amygdala-cerebellar circuits.

BACKGROUND: Concern about falling (CAF) is common in multiple sclerosis (MS), impacting motor function, cognition, and emotional well-being. However, the underlying neural correlates remain understudied. Given the multifactorial nature of CAF, we hypothesized that neural correlates may involve interactions between brain regions involved in emotional (e.g., amygdala), motor (e.g., cerebellum), and cognitive functions (e.g., hippocampus). This study explored associations between CAF and resting-state functional connectivity (FC) in amygdala-hippocampal and amygdala-cerebellar circuits in MS. ME

Astrocyte Reactivity by Alcohol Dependence in the Central Amygdala.

Astrocytes play essential roles in maintaining brain homeostasis and in contributing to synaptic functions, but, in response to injury, infection, or disease, astrocytes can downregulate their homeostatic and physiological functions while increasing neuroinflammatory responses. The central amygdala (CeA) is important for stress responsivity and the development of alcohol (ethanol) dependence. Using a multi-omics approach in Aldh1l1-EGFP/Rpl10a mice and the chronic intermittent ethanol two-bottle choice (CIE-2BC) model, we have characterized the translational response of CeA astrocytes, as well

Dopamine modulation of basolateral amygdala activity and function.

The basolateral amygdala (BLA) is central to emotional processing, fear learning, and memory. Dopamine (DA) significantly influences BLA function, yet its precise effects are not clear. We present a mathematical model exploring how DA modulation of BLA activity depends on the network's current state. Specifically, we model the firing rates of interconnected neural groups in the BLA and their responses to external stimuli and DA modulation. BLA projection neurons are separated into two groups according to their responses-fear and safety. These groups are connected by mutual inhibition though in

Alterations of the amygdala in post-COVID olfactory dysfunction.

Olfactory dysfunction (OD) as a symptom of COVID-19 has received significant attention in research due to its high prevalence. While it is transient in the majority of individuals, post-COVID OD persists in a notable subset of patients even months to years after the acute infection. A deeper understanding of the underlying factors driving this phenomenon is essential. There is increasing evidence for an involvement of the central nervous system in this deficit. The objective of this study was to investigate the structural connectivity and integrity of white matter pathways in brain regions ass

Unraveling the amygdala: A review of its anatomy and functions.

The amygdala is a complex cellular structure situated centrally in the brain, adjacent to the hippocampus. It is an integral part of the limbic system and is essential for the processing of emotional reactions. The amygdala possesses extensive connections to multiple brain regions, enabling it to acquire sensory information and affect responses. The amygdaloid complex has over ten nuclei located in the mid-temporal lobe. It is quite likely that the primary root of certain anxiety disorders in humans, such as posttraumatic stress disorder, is a dysfunction in the processing of information assoc

Neuroinflammation in the Amygdala Is Associated With Recent Depressive Symptoms.

BACKGROUND: Converging evidence suggests that elevated inflammation may contribute to depression. Yet, the link between peripheral inflammation and neuroinflammation in depression is unclear. Here, using data from the UK Biobank, we estimated associations among depression, C-reactive protein (CRP) as a measure of peripheral inflammation, and neuroinflammation as indexed by diffusion basis spectral imaging-based restricted fraction (DBSI-RF). METHODS: DBSI-RF was derived from diffusion-weighted imaging data (N = 11,512) for whole-brain gray matter (global-RF), and regions of interest in the bi

The amygdala is not necessary for the familiarity aspect of recognition memory.

Dual-process accounts of item recognition posit two memory processes: slow but detailed recollection, and quick but vague familiarity. It has been proposed, based on prior rodent work, that the amygdala is critical for the familiarity aspect of item recognition. Here, we evaluated this proposal in male rhesus monkeys (Macaca mulatta) with selective bilateral excitotoxic amygdala damage. We used four established visual memory tests designed to assess different aspects of familiarity, all administered on touchscreen computers. Specifically, we assessed monkeys' tendencies to make low-latency fal

Anatomical Subpial Resection of Tumors in the Amygdala and Hippocampus.

OBJECTIVE: Surgical techniques to achieve complete resection of mesial-basal temporal tumors should be pursued by neurosurgical oncologists. We describe the anatomical subpial amygdalohippocampectomy (SpAH) technique for tumor resection. METHODS: The key anatomical landmarks and critical steps of the SpAH technique were outlined and emphasized with medical illustrations and intraoperative photographs. The senior author's 90-day surgical outcomes with this approach were reviewed. RESULTS: Twenty-five patients (men, 17 [68%]; women, 8 [32%]; median [range] age, 59 [23-80] years) with temporal

Amygdala GABA Neurons Project To vlPAG And mPFC.

The amygdala regulates multiple behaviors and emotions by projecting to multiple brain regions. However, the topographical distribution of amygdala neurons projecting to specific brain areas is still unclear. In the present study, we focus on determining whether single amygdala neurons project to the brain stem ventrolateral periaqueductal grey (vlPAG) and to the medial prefrontal cortex (mPFC). The mPFC neurons are involved in detecting emotional content while the vlPAG neurons are involved in regulating muscle tone. In VGAT-Cre mice a cre-inducible retrograde AAV tracer expressing tdTomato w

Functional connectivity of the human amygdala in health and in depression.

To analyse the functioning of the amygdala in depression, we performed the first voxel-level resting state functional-connectivity neuroimaging analysis of depression of voxels in the amygdala with all other voxels in the brain, with 336 patients with major depressive disorder and 350 controls. Amygdala voxels had decreased functional connectivity (FC) with the orbitofrontal cortex, temporal lobe areas, including the temporal pole, inferior temporal gyrus and the parahippocampal gyrus. The reductions in the strengths of the FC of the amygdala voxels with the medial orbitofrontal cortex and tem

Synaptic dysfunction in amygdala in intellectual disorder models.

The amygdala is a part of the limbic circuit that has been extensively studied in terms of synaptic connectivity, plasticity and cellular organization since decades (Ehrlich et al., 2009; Ledoux, 2000; Maren, 2001). Amygdala sub-nuclei, including lateral, basolateral and central amygdala appear now as "hubs" providing in parallel and in series neuronal processing enabling the animal to elicit freezing or escaping behavior in response to external threats. In rodents, these behaviors are easily observed and quantified following associative fear conditioning. Thus, studies on amygdala circuit in

Receptor-driven, multimodal mapping of the human amygdala.

The human amygdala consists of subdivisions contributing to various functions. However, principles of structural organization at the cellular and molecular level are not well understood. Thus, we re-analyzed the cytoarchitecture of the amygdala and generated cytoarchitectonic probabilistic maps of ten subdivisions in stereotaxic space based on novel workflows and mapping tools. This parcellation was then used as a basis for analyzing the receptor expression for 15 receptor types. Receptor fingerprints, i.e., the characteristic balance between densities of all receptor types, were generated in

Amygdala volume is reduced in early course schizophrenia.

Subcortical structural alterations have been implicated in the neuropathology of schizophrenia. Yet, the extent of anatomical alterations for subcortical structures across illness phases remains unknown. To assess this, magnetic resonance imaging (MRI) was used to examine volume differences of major subcortical structures: thalamus, nucleus accumbens, caudate, putamen, globus pallidus, amygdala and hippocampus. These differences were examined across four groups: (i) healthy comparison subjects (HCS, n=96); (ii) individuals at high risk (HR, n=21) for schizophrenia; (iii) early-course schizophr

Polymodal activation of the endocannabinoid system in the extended amygdala.

The reason why neurons synthesize more than one endocannabinoid (eCB) and how this is involved in the regulation of synaptic plasticity in a single neuron is not known. We found that 2-arachidonoylglycerol (2-AG) and anandamide mediate different forms of plasticity in the extended amygdala of rats. Dendritic L-type Ca(2+) channels and the subsequent release of 2-AG acting on presynaptic CB1 receptors triggered retrograde short-term depression. Long-term depression was mediated by postsynaptic mGluR5-dependent release of anandamide acting on postsynaptic TRPV1 receptors. In contrast, 2-AG/CB1R-

Amygdala mechanisms of Pavlovian psychostimulant conditioning and relapse.

Psychostimulant addiction often consists of periods of sustained drug abstinence disrupted by periods of relapse and renewed heavy drug use. Prevention of relapse remains the greatest challenge to the successful treatment of drug addiction. Drug-associated cues are a primary trigger for relapse, as they can elicit intense craving for the drug. These cues become associated with the drug reward through Pavlovian learning processes that develop over multiple drug-cue pairings. The amygdala (AMY) is critical for such drug-related learning. Intrinsic and extrinsic circuitry position the AMY to inte

Amygdala dysfunction in men with the fragile X premutation.

Premutation alleles (55-200 CGG repeats) of the fragile X mental retardation 1 (FMR1) gene are associated with autism spectrum disorder in childhood, premature ovarian failure, and the neurodegenerative disorder, fragile X-associated tremor/ataxia syndrome (FXTAS). FXTAS, and perhaps the other clinical presentations among carriers, are thought to be due to toxic gain-of-function of elevated levels of the expanded-repeat FMR1 mRNA. Previous structural MRI studies have implicated the amygdala as a potential site of dysfunction underlying social deficits and/or risk for FXTAS. As a preliminary in

Neurons in the amygdala of the monkey with responses selective for faces.

To investigate the functions of the amygdala in visual information processing and in emotional and social responses, recordings were made from single neurons in the amygdala of the monkey. A population of neurons (40 of more than 1000 recorded in 4 monkeys) was investigated which responded primarily to faces. These neurons typically (1) responded to some human or monkey faces, which were presented to the monkey through a large aperture shutter so that response latencies could be measured, or were simply shown to the monkey, (2) responded to 2-dimensional representations of these faces, as well
