A study of barosensitivity in the European eel (Anguilla anguilla, L.): effects of arterial pressure changes on heart rate and ventilation.

A study of the effects of arterial pressure changes on heart rate and ventilation was carried out on the eel (Anguilla anguilla L.). The anatomical characteristics of this fish enabled extracorporeal circulation in both the ventral and dorsal aorta to be set up. The pressure was changed mechanically, either by clamping external circulations or, in some cases, by modifying the frequency of a pulsatory pump placed in series with the heart, thus ensuring constant blood flows in the ventral aorta. The maximal pressure decreases obtained by clamping the external ventral aorta circulation (70%-80%)

Milt androgen profile and evaluation of sperm morpho-functional characteristics of wild-caught and farmed European eels (Anguilla anguilla).

Evaluating milt samples before their preservation and/or reproductive use is critical to correctly develop assisted reproductive strategies for teleost species. This is especially true in case of "critically endangered" species, like the European eel (Anguilla anguilla), whose life cycle is not closed yet, leading to the necessity of capturing younglings from natural populations for both research and productive purposes. Despite androgens being involved in spermatogenesis and playing pivotal roles in the pre-migratory morphological and physiological changes, their quantification in different b

Cucullanus egyptae sp. nov. (Nematoda, Cucullanidae) infecting the European eel Anguilla anguilla in Egypt. Morphological and molecular phylogenetic studies.

A total of 80 specimens of the European eel Anguilla anguilla were collected during the period from February 2013 to March 2014 at the coast of the Gulf of Suez (Red Sea, Egypt). A new species of parasitic nematodes was recovered and described as Cucullanus egyptae. It was found in the eel's intestine with a prevalence of 68.7%. The morphology of the recovered parasite was studied by light and scanning electron microscopy. The adult worms had a wide cephalic extremity with a slit-like oral aperture being surrounded by a cuticular ring and delimited internally by a row of small teeth in additio

Behaviour of European eel Anguilla anguilla transitioning from the yellow resident to the silver migratory stage in marine waters.

Current knowledge on the continental shelf and transatlantic spawning migration of the European eel (Anguilla anguilla) is based on silver eels outmigrating from fresh water. However, an unknown proportion of eels grow in marine coastal waters during the continental part of their lifecycle and therefore start their spawning migration while they are already in marine waters. To study the onset of the European eel's spawning migration in marine waters, we tagged yellow eels in Sandnesfjord, Norway. One hundred yellow and pre-migratory eels (stage FII or FIII) were tagged with acoustic transmitte

Bioaccumulation and the effects of organochlorine pesticides, PAH and heavy metals in the Eel (Anguilla anguilla) at the Camargue Nature Reserve, France.

Pesticides (organochlorines-OC), polycyclic aromatic hydrocarbons (PAH) and heavy metals are toxic to fish and may be taken in through gills, skin and contaminated foods. Here we measure concentrations of OC, PAH and heavy metals, and their effects in the eel Anguilla anguilla from three locations in the Camargue Reserve in southern France. The Camargue Biosphere Reserve is the largest coastal wetland in Western Europe, and A. anguilla is a common predator at the top of the food chain. Livers and spleens were analyzed for histopathological, chemical and organo-somatic (HSI and SSI) effects. Gi

A psychiatric drug found in waste-water plant effluents alters the migratory behavior of critically endangered Anguilla anguilla juveniles.

Migratory fishes cross or settle in several environments potentially polluted. Psychiatric drugs, which represent one growing pollution and are found in discharges from waste-water treatment plants, may alter individual behaviors. Here, we assessed behavioral alterations in the upstream migratory behavior of Anguilla anguilla caused by diazepam, an anxiolytic. We monitored the swimming activity, swimming behavior, and boldness to assess whether diazepam impacts them or not. Our 7-day behavioral follow-up allowed us to test the kinetics of the potential effects of diazepam. We found diazepam re

Effect of photoperiod on endocrine profiles and vitellogenin expression in European eels Anguilla anguilla during artificially induced ovarian development.

The aim of this work was to determine the effects of dark and light conditions on the E2, testosterone and thyroid hormones levels and on the gene expression levels (vitellogenin 1, vitellogenin 2, and estradiol receptor one) in European eels (Anguilla anguilla) during ovarian development induced by increasing doses of carp pituitary extracts (CPEs). The subjects were divided into 2 groups: 14-hour light:10-hour dark (Light Group) and 24-hour darkness (Dark Group). All the eels received intramuscular injections with CPE at a dosage of 10 mg/kg body weight (BW) once a week for the first 3 weeks

European eel (Anguilla anguilla) genotoxic and pro-oxidant responses following short-term exposure to Roundup--a glyphosate-based herbicide.

The glyphosate-based herbicide, Roundup, is among the most used pesticides worldwide. Due to its extensive use, it has been widely detected in aquatic ecosystems representing a potential threat to non-target organisms, including fish. Despite the negative impact of this commercial formulation in fish, as described in literature, the scarcity of studies assessing its genotoxicity and underlying mechanisms is evident. Therefore, as a novel approach, this study evaluated the genotoxic potential of Roundup to blood cells of the European eel (Anguilla anguilla) following short-term (1 and 3 days) e

Effects of cold seawater pre-treatments on induction of early sexual maturation and sperm production in European eel (Anguilla anguilla).

To induce sexual maturation in captivity, eels rely on hormonal treatments, but this process is costly and time-consuming. As an alternative, different types of conditioning, also referred as pre-treatment, have been assessed to ease hormonal treatment response. Recent studies have shown that migrating eels experience a wide range of temperatures, varying from 12 °C at night to as low as to 8 °C during the day. Therefore, this study evaluates the effects of low-temperature (10 °C) seawater pre-treatments of different durations (2 and 4 weeks) on male eel reproduction. The eye, gonadosomatic an

Changes in ovarian gene expression profiles and plasma hormone levels in maturing European eel (Anguilla anguilla); Biomarkers for broodstock selection.

Complete sexual maturation of European eels (Anguilla anguilla) in captivity can only be achieved via injections with gonadotropins. For female eels this procedure takes 4-6months and the response ranges from "unresponsive" to final maturation and ovulation. Reproductive success could be significantly increased via early selection of responders based on predictive markers and minimally invasive sampling methods. To get a better understanding of the genetic background of ovarian maturation of the European eel we performed a pilot deep-sequencing transcriptome analysis of ovarian tissue derived

Lipid peroxidation and its control in Anguilla anguilla hepatocytes under silica-coated iron oxide nanoparticles (with or without mercury) exposure.

Having multidisciplinary applications, iron oxide nanoparticles can inevitably enter aquatic system and impact inhabitants such as fish. However, the studies in this context have ignored the significance of obvious interaction of iron oxide nanoparticles with other persistent co-contaminants such as mercury (Hg) in the modulation of the toxicity and underlying mechanisms of iron oxide nanoparticles and Hg alone, and concomitant exposures. This study aimed to evaluate lipid peroxidation (LPO) and its control with glutathione (GSH) and associated enzymes (such as glutathione reductase, GR; gluta

Branchial expression of an aquaporin 3 (AQP-3) homologue is downregulated in the European eel Anguilla anguilla following seawater acclimation.

A cDNA encoding the homologue of mammalian aquaporin 3 (AQP-3) was isolated by reverse transcription-polymerase chain reaction from the gill of the European eel. The derived amino acid sequence shares 67-70% homology with other vertebrate AQP-3 homologues. Northern blot analysis revealed two AQP-3-specific mRNA species of 2.4 kb and 7 kb. AQP-3 mRNA is expressed predominantly in the eye, oesophagus, intestine (as found in mammals) and the gill; no expression could be demonstrated in the stomach and only low and sporadic levels in the kidney. Quantitative studies demonstrated that, following th

Swimming activity and behaviour of European Anguilla anguilla glass eels in response to photoperiod and flow reversal and the role of energy status.

To better understand migratory divergences among Anguilla anguilla glass eels, the behaviour of individuals caught at the time of their estuary entrance was studied through their response to a light:dark cycle and then to both water current reversal and light:dark cycle. In a first experiment, fish moving with the flow in response to dusk (M(+) fish) and fish that had not exhibited any movement (M(-) fish) were distinguished. Anguilla anguilla from these two groups were then individually marked and their response to water current reversal compared. M(+) individuals mainly exhibited negative rh

Cutaneous and gill O2 uptake in the European eel (Anguilla anguilla L.) in relation to ambient PO2, 10-400 Torr.

The partition of O2 uptake between gills and skin was examined in the freshwater eel (Anguilla anguilla L.) at ambient PO2 ranging from hyperoxia (PO2 = 400 Torr) to severe hypoxia (PO2 = 10 Torr), using a technique of open-flow respirometry. All the expired water was collected, and the ventilatory flow and the mixed expired water PO2 were directly measured. The ventilatory water flow decreased moderately in hyperoxia, increased markedly between normoxia and 40 Torr, and below 40 Torr, hyperventilation was gradually reduced. Between PO2 400 and 70 Torr, the total O2 uptake was constant and the

Experimental infection with Anguillicola crassus alters immune gene expression in both spleen and head kidney of the European eel (Anguilla anguilla).

Invasive parasites have been implicated in the declines of several freshwater species. The swim bladder nematode Anguillicola crassus was introduced into Europe in the 1980s and is considered a threat to the European eel (Anguilla anguilla). Infection affects stress resistance and swimming behaviour. European eels produce an immune response against the parasite during the late stages of infection and after repeated infections. We used RNA-seq to examine the molecular response to infection during the poorly understood early stage and identify expression of genes and associated processes that ar

Oxidative stress and genotoxic effects in gill and kidney of Anguilla anguilla L. exposed to chromium with or without pre-exposure to beta-naphthoflavone.

Fish in the aquatic environment can be subjected to a multipollution state and the occurrence of sequential exposures is an important aspect of eco-toxicological research. In this context, a preceding exposure can affect a toxic response to a subsequent exposure. Therefore, the current study was based on sequential exposures, viz. to a PAH-like compound (beta-naphthoflavone, BNF) followed by a heavy metal (chromium, Cr), focusing on the assessment of oxidative stress responses and their role in induction of genotoxicity. Oxidative stress responses in gill and kidney were investigated in Europe

An optimized whole-body corticosteroid hormones quantification method by LC-MS/MS for assessing stress levels in European glass eels (Anguilla anguilla).

The European eel (Anguilla anguilla) juvenile stage exhibits facultative estuarine migration. The causes of this behavior are yet unknown but it may have an impact on the population's fate by altering the sex ratio of the population. Recent studies have highlighted potential stress-related issues in glass eels settling in estuaries but studying stress response in small organisms requires sensitive, accurate and precise analytical methods. The aims of the present study are (i) to develop a whole-body Liquid Chromatography-Mass Spectrometry (LC-MS/MS) method for the simultaneous determination of
