Effect of undernutrition on the chemical composition and the activity of alkaline phosphatase in soluble and particulate fractions of the rat calvarium and femur. II: Effect of preweaning undernutrition in the suckling rat.

We studied the effects of undernutrition during the suckling period on the development of the calvarium and the femur of rat pups suckling dams fed a 20% or 5% protein diet. The results show that the maturation of bone is delayed by undernutrition. In both the calvarium and the femur, the particulate fraction alkaline phosphatase reflects the rate increments in calcium and phosphorus uptake in control animals. This association is affected in the undernourished pups, as both the alkaline phosphatase activity in particulate fraction and the calcium and the phosphorus uptake decreased. However, t

Effect of nutritional deprivation on mucosal viral infections.

Suckling BALB-c mice, subjected to nutritional deprivation in artificially expanded litters (18 to 20 pups), were compared to normally nourished pups (7-9 per litter) in a series of experiments designed to provide data on morphologic and functional alterations of the small intestine during malnutrition and infection. The effects of protein calorie malnutrition (PCM) on the viral replication pattern and severity of clinical disease were examined in suckling mice infected with murine rotavirus (MRV). The infection in nutritionally deprived animals was characterized by a significant decrease in t

Toxic effects of chromium (VI) by maternal ingestion on liver function of female rats and their suckling pups.

Potassium dichromate (K(2)Cr(2)O(7)) is an environmental contaminant widely recognized as a carcinogen, mutagen, and teratogen toward humans and animals. This study investigated the effects of K(2)Cr(2)O(7) on the hepatic function of pregnant and lactating rats and their suckling pups. Experiments were carried out on female Wistar rats given 700 ppm of K(2)Cr(2)O(7) in their drinking water from the 14th day of pregnancy until day 14 after delivery. Hepatotoxicity was objectified by the significant increase in liver malondialdehyde content and a significant accumulation of chromium in this soft

[Pathomorphological changes in colibacteriosis in pigs].

A total of 46 suckling piglets spontaneously affected with colibacteriosis and 20 pigs aged from 1 to 10 days, experimentally infected with two strains of Escherichia coli (0149 and 0138) were investigated morphologically. Both in the animals with spontaneous infection and in those with experimentally induced disease there were severe morphologic lesions not only in the small intestines, but also in the other viscera and the brain. Catarrhal to hemorrhagic enteritis was observed in 56.5 per cent of the spontaneous cases and 57.1 per cent of the test animals. Parenchymal dystrophy with hemorrha

Safety and efficacy of iron dextran as a feed additive for piglets.

The additive, 'Iron dextran 10%', is a liquid preparation containing iron dextran (25%, of which 10% is total iron), sodium chloride (1.5%), phenol (0.4%) and water (73.1%). Iron dextran 10% is considered safe for suckling piglets when given at an oral dose of 1 mL/kg body weight (bw) once in each of the first 2 weeks of life; this dose corresponds to 100 mg Fe/kg bw. The administration of iron dextran 10% to piglets deficient in vitamin E and/or selenium is considered a risk. The oral use of iron dextran 10% in suckling piglets does not pose any safety concerns to consumers, provided that the

Dexamethasone effects on group B streptococcal infection in newborn rats.

BACKGROUND: We previously published that human neutrophil-mediated bacterial killing of group B Streptococcus (GBS) in vitro was dependent on the timing and concentration of dexamethasone exposure. HYPOTHESIS: Dexamethasone treatment would affect neutrophil mediated killing of GBS in an animal model. METHODS: Wistar rat pups were randomly allocated to receive placebo or dexamethasone before, early or late after GBS infection. Suckling rats were infected with 104 or 105 colony-forming units of GBS or nothing. Pups were followed for survival, quantitative bacteremia, growth and neutrophil-medi

Jejunal inflammatory cytokines, barrier proteins and microbiome-metabolome responses to early supplementary feeding of Bamei suckling piglets.

BACKGROUND: Dietary intervention has been reported to improve intestinal health. The intestinal microbiota of newborn animals plays a fundamental role in the development of intestinal function and the innate immune system. However, little is currently known about dietary interventions in the gut microbiota and barrier function of livestock, especially suckling Bamei piglets. To this end, we studied the effect of early dietary supplementation on intestinal bacterial communities and intestinal barrier function in piglets. RESULTS: 10 purebred Bamei sows were randomly allocated into two groups.

Pregnant sows immunized with Cryptosporidium parvum significantly reduced infection in newborn piglets challenged with C. parvum but not with C. hominis.

BACKGROUND: The piglet is the only model to investigate the immunogenic relationship between Cryptosporidium hominis and C. parvum, the species responsible for diarrhea in humans. Despite being indistinguishable antigenically, and high genetic homology between them, they are only moderately cross protective after an active infection. METHODOLOGY/PRINCIPAL FINDINGS: Here we examined the degree of passive protection conferred to piglets suckling sows immunized during pregnancy with C. parvum. After birth suckling piglets were challenged orally with either C. parvum or C. hominis at age 5 days.

Cryptosporidium infection impairs growth and muscular protein synthesis in suckling rats.

This study aimed to explore the metabolic consequences of cryptosporidiosis in an acute experimental model both at the peak of infection and after parasite clearance. Four-day-old suckling rats were infected with 10(6) oocysts of Cryptosporidium parvum. At the peak of infection (day 8 PI), C. parvum resulted in a dramatic reduction both in nutrient intake (-50%) and body weight (16.3+/-5.2 vs 27.3+/-1.0 g, P<0.01) with a decrease in both lean body mass and adipose tissue. Muscular fractional and absolute synthesis rate were reduced (-15 and -55%, respectively). After parasite clearance (day 17

Maternal schistosomiasis: IL-2, IL-10 and regulatory T lymphocytes to unrelated antigen in adult offspring mice.

INTRODUCTION: We evaluated IL-10, IL-2 and regulatory T cells (Treg), in response to ovalbumin (OA), in offspring from schistosomotic mouse mothers. METHODS: We used animals born (BIM) or suckled (SIM) from infected mothers; and mice born/suckled from infected (BSIM) or non-infected mothers (CONTROL). After OA+adjuvant immunization, spleen cells were cultured, with or without OA, and doubly marked for cytometry. RESULTS: BIM showed fewer CD4+/IL-2+ and more B220+/IL-10+ cells, whereas the SIM group showed increased Treg frequency. BSIM had fewer B220+/IL-10+ and Treg cells. CONCLUSIONS: Sep

Bovine respiratory syncytial virus seroprevalence and risk factors in non-vaccinated dairy cattle herds in Brazil.

BACKGROUND: The cattle industry is one of the most important Brazilian agribusiness sectors and is a strong contributor to the national economy. Annually about 44.6 million calves are bred, which makes the optimal management of these animals extremely important. Several diseases can affect the initial stages of the bovine production chain, being the bovine respiratory syncytial virus (BRSV) one of the most relevant pathogens. This study aimed to characterize the epidemiology of BRSV infection in dairy cattle herds of São Paulo State, Brazil, using serological and risk factors analyses. For tha

Interactions between 11beta-hydroxysteroid dehydrogenase and COX-2 in kidney.

The syndrome of apparent mineralocorticoid excess (SAME) is an autosomal recessive form of salt-sensitive hypertension caused by deficiency of the kidney type 2 11beta-hydroxysteroid dehydrogenase (11betaHSD2). In this disorder, cortisol is not inactivated by 11betaHSD2, occupies mineralocorticoid receptors (MRs), and causes excessive sodium retention and hypertension. In renal medulla, prostaglandins derived from cyclooxygenase-2 (COX-2) stimulate sodium and water excretion, and renal medullary COX-2 expression increases after mineralocorticoid administration. We investigated whether medullar

Gestation and breastfeeding in schistosomotic mothers differently modulate the immune response of adult offspring to postnatal Schistosoma mansoni infection.

Schistosoma mansoni antigens in the early life alter homologous and heterologous immunity during postnatal infections. We evaluate the immunity to parasite antigens and ovalbumin (OA) in adult mice born/suckled by schistosomotic mothers. Newborns were divided into: born (BIM), suckled (SIM) or born/suckled (BSIM) in schistosomotic mothers, and animals from noninfected mothers (control). When adults, the mice were infected and compared the hepatic granuloma size and cellularity. Some animals were OA + adjuvant immunised. Safety of a feed additive consisting of Macleaya cordata (Willd.) R. Br. extract and leaves (Sangrovit® Extra) for suckling and weaned piglets and other growing Suidae (Phytobiotics Futterzusatzstoffe GmbH).

Following a request from the European Commission, EFSA was asked to deliver a scientific opinion on the safety of a feed additive consisting of Macleaya cordata (Willd.) R. Br. extract and leaves (Sangrovit® Extra) as a zootechnical feed additive for suckling and weaned piglets and other growing Suidae. In its previous assessment, the EFSA Panel on Additives and Products or Substances used in Animal Feed (FEEDAP) had no safety concerns for the target species when the additive is used at the recommended level of 0.750 mg sanguinarine/kg complete feed for suckling and weaned piglets and other gr

Effect of posterior pituitary denervation (PPD) on prolactin (PRL) and alpha-melanocyte-stimulating hormone (alpha-MSH) secretion of lactating rats.

Previous data have clearly suggested that the posterior pituitary (PP), consisting of neural lobe (NL) and intermediate lobe (IL), has a role in the control of anterior pituitary PRL secretion. However, basic aspects of this regulatory mechanism like (1), the role of an intact hypothalamic innervation of the PP as well as (2) the site of production of previously found PRL releasing substance(s) have not yet been characterized. Denervation of the PP (PPD) is an effective method for having a selective lesion of the innervation of PP, indeed, PPD results in a disappearance of neurosecretory mater

[Neuropathogenecity of Coxsackie B virus strains isolated from children with clinical manifestations of aseptic meningitis in different years].

The neuropathogenicity of Coxsackie B3, B1, and B5 virus strains was studied in experimentally infected suckling BALB/c mice and in contact animals from the same nest and litter infected due to the natural virus spread. The similar neurological disorders were found in both groups of animals. The data on pathological and morphological changes in the medulla oblongata as dystrophic and necrotic changes in the neurons with their lysis and deletion, brain tissue edema, and hemodynamic changes confirmed the neutrotropicity of the strains under study. Experimental pneumovirus infections. 2. Hydrocephalus of hamsters and mice due to infection with human respiratory syncytial virus (RS).

Respiratory syncytial virus (RS) injected into suckling hamsters and mice via the cerebral route induced hydrocephalus in 35% of the animals. The pathogenesis of the condition has been examined by light microscopy, immunofluorescence, and virus isolation. Infection of leptomeninges and ependyma was revealed by immunofluorescence of virus-specific antigens between Days 3 and 7 after inoculation. Histologically, eosinophilic inclusion bodies were visible within the cytoplasma of ependymal and meningeal cells. A patent aqueduct was observed in all hydrocephalic animals but one. These observations

Quantitative flow cytometric evaluation of maximal Cryptosporidium parvum oocyst infectivity in a neonate mouse model.

The importance of waterborne transmission of Cryptosporidium parvum to humans has been highlighted by recent outbreaks of cryptosporidiosis. The first step in a survey of contaminated water currently consists of counting C. parvum oocysts. Data suggest that an accurate risk evaluation should include a determination of viability and infectivity of counted oocysts in water. In this study, oocyst infectivity was addressed by using a suckling mouse model. Four-day-old NMRI (Naval Medical Research Institute) mice were inoculated per os with 1 to 1,000 oocysts in saline. Seven days later, the number
