[Allergic and pseudoallergic reactions to analgesics, antipyretics and non-steroidal anti-inflammatory drugs].

Antalgics, antipyretics and non-steroidal anti-inflammatory drugs (NSAIDs) are widely used, but suspected allergic reactions to these drugs are rare, especially in children. Most frequent reactions are cutaneous (urticaria, angioedema) and respiratory (rhinitis, asthma). Other reactions (anaphylaxis or anaphylactoid reactions, potentially harmful toxidermias) are rare. In a few patients, reactions may result from a specific (allergic) hypersensitivity (HS), with positive responses in prick and intradermal tests (anaphylaxis, immediate urticaria and/or angioedema) and in intradermal and patch t

Antipyretic Use in Noncritically Ill Patients With Fever: A Review.

Antipyretics are one of the most frequently used agents in medicine. Numerous pharmacological agents, such as acetaminophen, non-steroidal anti-inflammatory agents (NSAIDs), salicylates, and selective cyclooxygenase 2 (COX-2) inhibitors, and nonpharmacological treatment modalities, such as tepid sponging and cooling blankets, are available for temperature reduction. There is a scarcity of definitive clinical guidelines on the choice of various agents in noncritically ill febrile patients. Our review examined the various modalities available for antipyresis and compared their safety and efficac

[Paracetamol and other antipyretic analgesics: optimal doses in pediatrics].

Acetaminophen is a safe antipyretic therapy prescribed in children. The dosage usually applied in France ranges from 20 to 30 mg/kg/d, which is low and probably with little efficiency in many cases. From the data of the literature the optimal dosage seems to be 65 mg/kg/d using a 25 mg/kg loading dose, followed by a 12.5 mg/kg every 6 hours maintenance dose. The analgesic effect of a 15 mg/kg oral dose has been also demonstrated. Aspirin is equally effective, especially for rheumatic disorders, but its use is limited by adverse reactions. The other nonsteroidal anti-inflammatory drugs have a l

Analgesic, antipyretic and anti-inflammatory properties of Euphorbia hirta.

Lyophilised aqueous extract of Euphorbia hirta L. (Euphorbiaceae) has been evaluated for analgesic, antipyretic and anti-inflammatory properties in mice and rats, in order to complete its activity profile, after the confirmation of the existence of a central depressant activity particularly expressed by a strong sedative effect, associated with anxiolytic effects. This study leads us to the conclusion that this plant extract exerts central analgesic properties. Such a dose-dependent action was obtained against chemical (writhing test) and thermic (hot plate test) stimuli, respectively, from th

Effect of pyrogen and antipyretics on prostaglandin acitvity in cisternal c.s.f. of unanaesthetized cats.

1. Samples of cisternal cerebrospinal fluid (c.s.f.) were collected from unanaesthetized cats while rectal temperature was continuously recorded. From the same cat, samples were collected during normal body temperature, during pyrogen fever and when the fever was brought down by an I.P. injection of an antipyretic. Fever was produced by injection of the bacterial pyrogen of Shigella dysenteriae either into the third ventricle, cisterna magna or I.V. The samples of c.s.f. were assayed for PGE(1)-like activity on the rat stomach fundus strip preparation rendered insensitive to 5-HT.2. In samples

Analgesic, antipyretic and Ulcerogenic properties of an indigenous formulation--Kalpaamruthaa.

A modified indigenous Siddha formulation Kalpaamruthaa (KA), containing Semecarpus anacardium nut milk extract (SA), dried powder of Emblica officinalis (EO) fruit and honey was evaluated for its analgesic, antipyretic and Ulcerogenic properties. Both SA and KA, at a dose of 150 mg/kg b. wt were compared with the standard drug diclofenac sodium. KA exhibited an enhanced effect on all properties compared with that found with sole SA treatment, and is likely to be due to synergistic and additive interactions within the complex mixture of phytochemicals present in KA.

The use of antipyretic medications in the prevention of febrile convulsions in children.

Febrile convulsions are a relatively common outcome in paediatric febrile illness, although it is not known why some children suffer these. Antipyretic medications may form the basis for some treatment regimens, although they are not recommended in published guidelines. There is little evidence that the prophylactic use of antipyretics has any effect in reducing the incidence of febrile convulsions. Consequently, educational interventions aimed at reducing parental fear and helping them to care for their children during febrile illnesses may be more efficacious.

Hematologic effects of antipyretic analgesics. Drug-induced agranulocytosis.

Drug-induced agranulocytosis may be type I (involving the drug, antibodies and neutrophils), type II (associated with accumulated drug toxicity in hypersensitive persons), or type III (representing different etiologies induced by immune and toxic mechanisms). The pyrazolones (amidopyrine, dipyrone and butazones), phenothiazine derivatives, antithyroid drugs, and antibiotics are thought to be causative agents in agranulocytosis. The symptoms may involve sudden onset of high fever, sore throat with ulcerative angina, or stomatitis. Diagnosis of agranulocytosis is confirmed by severe granulocytop

[Analgetic and antipyretic activity of SL-573 (author's transl)].

Potency of analgetic activity of SL-573 was between that of indomethacin and aminopyrine in chemical stimulation tests. The analgetic activity of SL-573 was 3.2 times as potent as that of aminopyrine in the phenylquinone writhing test, 4.1 times as potent as aminopyrine in the acetic acid writhing test and 6.3 times as potent as aminopyrine in the Randall and Selitto test. Thus the analgetic activity of SL-573 appears to be comparable etic to that of codeine. SL-573, unlike narcotic analgesics, showed common properties to known antipyretic analgesics and anti-inflammatory agents in the followi

Alternating antipyretics for fever reduction in children: an unfounded practice passed down to parents from pediatricians.

Vitamin D toxicity also known as hypervitaminosis D was previously believed to be rare. But with an increase in vitamin D supplementation several cases have been reported in literature. Fat soluble vitamins like Vitamin D, due to their ability to accumulate in the body, have a higher potential for toxicity than water soluble vitamins. The main clinical consequence of vitamin D toxicity is hypercalcemia. In this report we describe an adult female patient who developed very high serum Vitamin D levels (746 ng/mL, RI: 20 to 50) as a result of medication error. Inspite of such high serum concentra

Central mediators involved in the febrile response: effects of antipyretic drugs.

Fever is a complex signal of inflammatory and infectious diseases. It is generally initiated when peripherally produced endogenous pyrogens reach areas that surround the hypothalamus. These peripheral endogenous pyrogens are cytokines that are produced by leukocytes and other cells, the most known of which are interleukin-1β, tumor necrosis factor-α, and interleukin-6. Because of the capacity of these molecules to induce their own synthesis and the synthesis of other cytokines, they can also be synthesized in the central nervous system. However, these pyrogens are not the final mediators of th
