Apolipoprotein AIMarburg: studies on two kindreds with a mutant of human apolipoprotein AI.

Three probands heterozygous for a mutant of apolipoprotein AI (apo AIMarburg, Utermann et al. 1982a) were detected by screening of 2282 unrelated individuals resulting an a frequency estimate of about 1/750 in the German population. All three probands with apo AIMarburg had hypertriglyceridemia (triglyceride above 250 mg/dl) and subnormal HDL-cholesterol (below 30 mg/dl), but no other lipoprotein abnormalities. The kindreds of two probands with AIMarburg were studied. The family data are consistent with an autosomal codominant inheritance of the trait. A total of 16 heterozygous blood relative

Genetics of apolipoprotein B and apolipoprotein AI and premature coronary artery disease.

Increased low-density lipoprotein (LDL) and decreased high-density lipoprotein cholesterol (HDL-C) predict premature coronary artery disease, as do elevated levels of apolipoprotein B or reduced levels of apolipoprotein AI. Probands were studied of families with common genetic forms of dyslipidaemia to determine if apo B or apo AI define genetic groups and if apo B or apo AI levels relate to premature coronary artery disease risk. Elevated apo B was characteristic of familial hypercholesterolaemia, familial combined hyperlipidaemia (FCHL), and was seen in individuals with elevated Lp(a). Norma

Radioimmunoassay for serum apolipoprotein D, an atypical apolipoprotein: validation and clinical application.

Apolipoprotein D (apoD) is an atypical apolipoprotein in terms of its structure, synthesis and probable function. It is the most abundant protein in breast cyst fluids of patients suffering from macrocystic disease of the breast, a disease associated with an increased risk of developing breast cancer. We have developed and optimized a competitive radioimmunoassay for the measurement of apoD in human serum. The assay uses a solid phase separation method and with a 4 h incubation period, it can be completed in a single working day. The within and between run coefficients of variation were < 8.5%

Association of the apolipoproteins with retinal arteriosclerosis in a health examination population.

BACKGROUND: Retinal arteriosclerosis is a non-invasive indicator for systemic microvascular damage and early atherosclerosis. While traditional lipid measures are used to establish cardiovascular risk predictors, Apolipoprotein B (ApoB), Apolipoprotein A1 (ApoA1), and their ratio might provide more insights into the atherogenic burden. However, their association with retinal arteriosclerosis in general health populations has not yet been explored. METHODS: This cross-sectional study included 4,938 adults from a health examination cohort. Retinal arteriosclerosis was evaluated by employing fun

Apolipoproteins Levels in Fatty Liver Disease: A Systematic Review and Meta-analysis.

BACKGROUND AND AIMS: Fatty liver disease (FLD) is a prevalent condition linked to metabolic disorders and can progress to severe liver diseases. Alterations in apolipoprotein (Apo) levels may provide valuable insights for diagnosing and managing FLD. This systematic review and meta-analysis evaluates these changes across different FLD phenotypes to evaluate their potential as diagnostic biomarkers. METHODS: We evaluated studies from PubMed, EMBASE, and Scopus using a predefined search string. Predefined inclusion and exclusion criteria were applied, and the risk of bias was assessed using the

Apolipoproteins in Chronic Kidney Disease and Kidney Transplant: A Long Unfinished Story.

Chronic kidney disease (CKD) is a growing global health burden, strongly associated with cardiovascular disease, the leading cause of mortality in this population. Dyslipidemia is a key metabolic abnormality in CKD, but traditional lipid measures such as total cholesterol, LDL cholesterol, HDL cholesterol, and triglycerides often fail to capture the complexity of lipid disturbances in CKD and after kidney transplantation. Apolipoproteins have emerged as more reliable markers of cardiovascular and renal risk. Elevated apolipoprotein B (ApoB), reduced apolipoprotein A1 (ApoA1), and a higher ApoB

Association of Biopsy-proven Nonalcoholic Fatty Liver Disease with Obesity and Apolipoproteins.

BACKGROUND AND AIMS: Insulin resistance is considered the most important key mechanism in the development of nonalcoholic fatty liver disease (NAFLD). Some studies have reported that hyperinsulinemia decreases the hepatic secretion of apolipoprotein (Apo) B. Chronic hyperinsulinemia in NAFLD may be responsible for the accumulation of triglycerides in hepatocytes. We aimed to investigate whether apolipoproteins are related to histological findings in patients with biopsy-proven NAFLD. We also aimed to evaluate the effects of obesity on apolipoproteins and the pathogenesis of NAFLD. METHODS: In

Association between Apolipoproteins AI and B and Ultrasound Indicators of Carotid Atherosclerosis.

BACKGROUND: Apolipoproteins A-I and B (apoA-I and apoB) may be better indicators of the risk of cardiovascular and cerebrovascular diseases than conventional risk factors (RFs). The onset of ischemic stroke (IS) may be preceded by the development of atherosclerotic changes in carotid arteries, which can be detected by ultrasound. Only a certain % of patients with IS have an (underlying) carotid etiology. OBJECTIVE: The aim of our study was to determine the association between ultrasound indicators of carotid atherosclerosis and the presence of apolipoproteins and other biomarkers in patients

Circadian Regulation of Apolipoproteins in the Brain: Implications in Lipid Metabolism and Disease.

The circadian rhythm is a 24 h internal clock within the body that regulates various factors, including sleep, body temperature, and hormone secretion. Circadian rhythm disruption is an important risk factor for many diseases including neurodegenerative illnesses. The central and peripheral oscillators' circadian clock network controls the circadian rhythm in mammals. The clock genes govern the central clock in the suprachiasmatic nucleus (SCN) of the brain. One function of the circadian clock is regulating lipid metabolism. However, investigations of the circadian regulation of lipid metaboli

Comparison of Conventional Lipoprotein Tests and Apolipoproteins in the Prediction of Cardiovascular Disease.

BACKGROUND: Total cholesterol and high-density lipoprotein cholesterol (HDL-C) measurements are central to cardiovascular disease (CVD) risk assessment, but there is continuing debate around the utility of other lipids for risk prediction. METHODS: Participants from UK Biobank without baseline CVD and not taking statins, with relevant lipid measurements (n=346 686), were included in the primary analysis. An incident fatal or nonfatal CVD event occurred in 6216 participants (1656 fatal) over a median of 8.9 years. Associations of nonfasting lipid measurements (total cholesterol, HDL-C, non-HDL

On the Aggregation of Apolipoprotein A-I.

In vivo, apolipoprotein A-I (ApoA-I) is commonly found together with lipids in so-called lipoprotein particles. The protein has also been associated with several diseases-such as atherosclerosis and amyloidosis-where insoluble aggregates containing ApoA-I are deposited in various organs or arteries. The deposited ApoA-I has been found in the form of amyloid fibrils, suggesting that amyloid formation may be involved in the development of these diseases. In the present study we investigated ApoA-I aggregation into amyloid fibrils and other aggregate morphologies. We studied the aggregation of wi

Apolipoprotein polymorphism in Hungary.

The disorders of plasma apolipoprotein metabolism caused by a number of genetic and environmental factors are associated with atherosclerosis. Because of the distinct structural, epidemiological and genetic properties of apolipoproteins we have analyzed some potentially important phenotypical markers of apolipoproteins(a)/Lp(a)/,E (apo E) and A-IV (apo-A-IV). In our Hungarian study population (n = 202) the frequency of Lp(a) concentrations (mean 10.5 +/- 13.5 mg/dl) and phenotypes (the dominance of high molecular weight isoforms) show similar distribution that of other Caucasian populations. Apolipoprotein E and amyloidogenesis.

Alzheimer's amyloid beta-protein (A beta) is a modified, pathogenic form of a constitutive host protein, soluble amyloid beta-protein (sA beta). Both are conformational isomers encoded by the gene for the beta-amyloid precursor protein (APP), located on chromosome 21. sA beta and A beta have identical sequence but are thought to differ in their secondary structure and physicochemical properties, hence they are conformational isomers. sA beta is easily degraded, while A beta is particularly resistant. A beta has a high beta-pleated sheet content, while sA beta is thought to be more random-coil

Apolipoprotein E in neurology.

Apolipoprotein E became relevant for neurologists in 1993 when the association of the apolipoprotein E-epsilon 4 allele with familial and sporadic late-onset Alzheimer disease was reported. Since that time, more than 100 confirmations and many research papers have appeared. A large neurobiological literature concerning the role of apolipoprotein E in the metabolism of the central nervous system is developing.
