Ancient pathogen DNA in archaeological samples detected with a Microbial Detection Array.

Ancient human remains of paleopathological interest typically contain highly degraded DNA in which pathogenic taxa are often minority components, making sequence-based metagenomic characterization costly. Microarrays may hold a potential solution to these challenges, offering a rapid, affordable, and highly informative snapshot of microbial diversity in complex samples without the lengthy analysis and/or high cost associated with high-throughput sequencing. Their versatility is well established for modern clinical specimens, but they have yet to be applied to ancient remains. Here we report ba

BPChAr-a Benzene Polycarboxylic Acid database to describe the molecular characteristics of laboratory-produced charcoal: Implications for soil science and archaeology.

The benzene polycarboxylic acid (BPCA) method is a technique to characterise the aromaticity and aromatic condensation of pyrogenic carbon (PyC) in charred residues. As a molecular marker for polycondensed aromatic moieties, the analysis of BPCAs in archaeological contexts has great potential as a means of detecting and characterising charred residues where past fire traces are not evident. Despite the increased frequency of applications and significant developments since the method's inception, no central database of BPCA results for modern charcoal pyrolysed under controlled laboratory condi

µCT scanning effects on aDNA and a multi-step workflow for archaeological petrous portions.

The petrous portion of the temporal bone (often informally referred to as the "petrous bone") is a key element in human evolutionary studies due to its exceptional preservation of biomolecules and morphological information. Intensive and often redundant sampling raises concerns about sustainability and long-term conservation, however. Digital recording of morphology (micro-computed and medical tomography) can preserve what destructive sampling destroys, enabling future analyses, yet there have been concerns regarding its effect on biomolecular preservation. Here, we present a systematic observ

Validating archaeological oyster morphometric studies and expanding their potential role in socioecological research.

Over the last decade, there has been a surge in archaeological studies of oyster shell morphometrics published in high-impact, high-profile scientific journals. Using large datasets of oyster size measurements yielded through archaeological investigations, these studies have aimed to contribute to inter- and trans-disciplinary bodies of work centered on issues of conservation, sustainability, ecosystem stability, and resource management. Indeed, teams of archaeologists, ecologists, geoscientists, and community partners have produced key insights into long-term trends in human-ecosystem dynamic

Silk proteins in archaeological soils: mass transfer, structural evolution, and an identification framework.

This study investigated the retention behavior of silk-derived proteins in archaeological soils and seeks to establish an analytical framework for the identification of silk residues preserved in burial environments. Silk fabric ageing and degradation were simulated using soil collected from the Tianzifen Site in Wulong, Chongqing. A multi-dimensional analytical framework combining multiple testing methods was used to characterize the transfer and preservation of silk proteins in soil. The results showed that silk degradation followed a clear staged trajectory. In the intermediate stage, fiber

Decoding past microbial life and antibiotic resistance in İnonü Cave's archaeological soil.

This study, which bridges the disciplines of archaeology and microbiology, examines the ancient bacterial communities and antibiotic-resistance genes in soil samples collected from İnönü Cave in Zonguldak, Turkiye. Our aim is to provide a comprehensive understanding of historical human activities and their influence on microbial communities. Soil samples were gathered from four distinct cultural levels from the Chalcolithic Age to the Early Iron Age. The microbial communities were characterized, and antibiotic-resistance genes were identified using high-throughput sequencing of 16S rRNA genes

The final plague outbreak in Scotland 1644-1649: Historical, archaeological, and genetic evidence.

This paper has several aims: to determine if Yersinia pestis was the causative agent in the last Scottish plague outbreak in the mid-17th century; map the geographic spread of the epidemic and isolate potential contributing factors to its spread and severity; and examine funerary behaviours in the context of a serious plague epidemic in early modern Scotland. Results confirm the presence of Y. pestis in individuals associated with a mid-17th century plague pit in Aberdeen. This is the first time this pathogen has been identified in an archaeological sample from Scotland. The geographic spread

Air pollution effect of SO2 and/or aliphatic hydrocarbons on marble statues in Archaeological Museums.

This study allowed the identification of the main physicochemical characteristics of deterioration of the materials used in the construction of Greek ancient statues in order to plan a correct methodology of restoration. The method of Reversed-Flow Inverse Gas Chromatography is appropriate to investigate the influence of air pollutants on authentic pieces from the Greek Archaeological Museum of Kavala, near Salonica. Six local physicochemical quantities which refer to the influence of one or two pollutants (synergistic effect) were determined for each system. These quantities answer the questi

Direct dating of archaeological pottery by compound-specific 14C analysis of preserved lipids.

A methodology is described demonstrating the utility of the compound-specific 14C technique as a direct means of dating archaeological pottery. The method uses automated preparative capillary gas chromatography employing wide-bore capillary columns to isolate individual compounds from lipid extracts of archaeological potsherds in high purity (>95%) and amounts (>200 microg) sufficient for radiocarbon dating using accelerator mass spectrometry (AMS). A protocol was developed and tested on n-alkanes and n-carboxylic acids possessing a broad range of 14C ages. Analytical blanks and controls allow

An elemental analysis of archaeological bone from Sicily as a test of predictability of diagenetic change.

Cortical human bone samples from three tightly dated components of a single Sicilian site were chemically analyzed employing the highly sensitive technique of inductively coupled plasma emission spectroscopy. Although the skeletons appeared to be excellently preserved, significant diagenesis was detected. Moreover, a majority of the elements tested showed no constant or linear variation over time, implying that diagenetic change tends not to be a predictable function of duration of interment. Variation among major long bones of a single skeleton was quite high, as was variation across the cort

Egyptian Canopic Jars at the Crossroad of Medicine and Archaeology: Overview of 100 Years of Research and Future Scientific Expectations.

Ancient Egyptian human remains have been of interest in the fields of both medical and Egyptological research for decades. However, canopic jar holders for internal organs (liver, lungs, stomach, intestines) of Egyptian mummies appear to be but a very occasional source of data for such investigations. The few medical approaches focusing on the content of these jars are summarized and listed according to pathogens and diseases to give a structured overview of this field of study. An extensive search of the literature has been conducted from different bibliographic databases with a total of n =

The "bear" essentials: actualistic research on Ursus arctos arctos in the Spanish Pyrenees and its implications for paleontology and archaeology.

Neotaphonomic studies of large carnivores are used to create models in order to explain the formation of terrestrial vertebrate fossil faunas. The research reported here adds to the growing body of knowledge on the taphonomic consequences of large carnivore behavior in temperate habitats and has important implications for paleontology and archaeology. Using photo- and videotrap data, we were able to describe the consumption of 17 ungulate carcasses by wild brown bears (Ursus arctos arctos) ranging the Spanish Pyrenees. Further, we analyzed the taphonomic impact of these feeding bouts on the bo
