The human hepatic asialoglycoprotein receptor is a target antigen for liver-infiltrating T cells in autoimmune chronic active hepatitis and primary biliary cirrhosis.

Autoantibodies to the human hepatic asialoglycoprotein receptor have been found in nearly 50% of the sera of patients with autoimmune chronic active hepatitis and in 15% of patients with primary biliary cirrhosis. In this study we demonstrate that the human hepatic asialoglycoprotein receptor is also a target antigen for T cell-mediated immune responses. Peripheral blood lymphocytes of 37% (7 of 19) of patients with autoimmune chronic active hepatitis and 33% (2 of 6) of patients with primary biliary cirrhosis showed a proliferative response to highly purified human hepatic asialoglycoprotein

Asialoglycoprotein receptor 1 positively regulates PTGS2-induced inflammatory response in polycystic ovary syndrome via ERK1/2 pathway.

Asialoglycoprotein receptor 1 regulates the inflammatory response in granulosa cells associated with polycystic ovary syndrome. In the granulosa cells of individuals with polycystic ovary syndrome, elevated levels of asialoglycoprotein receptor 1 may activate the extracellular signal-regulated kinase 1/2 (ERK1/2) signaling pathway, which in turn enhances the expression of prostaglandin-endoperoxide synthase 2. Abstract This study evaluated whether asialoglycoprotein receptor 1 (ASGR1) can regulate the expression of prostaglandin-endoperoxide synthase 2 (PTGS2) in granulosa cells (GCs) in polyc

Asialoglycoprotein receptor-1 mediated targeted drug delivery to liver cancer cells using stimuli responsive galactosylated electrospun nanofibers.

Synthesis of core-shell nanofiber (DOX@Gal-PVA/NSC) with galactose decorated poly(vinyl alcohol) as shell and doxorubicin loaded N-succinyl chitosan as core was achieved via coaxial electrospinning for site-specific targeting of liver cancer cells. Nanofibers of diameter 200 nm decorated with galactose were realised by in-situ photopolymerization during electrospinning. The galactose confers specific binding to asialoglycoprotein receptor-1 (ASGPR-1), overexpressed on hepatocellular carcinoma cells. The DOX loaded NSC core enable pH-responsive drug release, with higher rate of release to tumor

Asialoglycoprotein receptor-targeted perfluorooctylbromide as a targeted contrast agent for evaluating the severity of carbon tetrachloride-induced acute liver damage in rats.

Asialoglycoprotein receptor (ASGPR) is an endocytic C-type lectin receptor in hepatocytes. Acute and chronic liver diseases can result in the decreased expression and content of this receptor. The objective of this study was to determine whether ASGPR-targeted perfluorooctylbromide (PFOB) can enhance ultrasound imaging signals and evaluate the severity of carbon tetrachloride (CCl4)-induced acute liver damage in rats. The specificity of ASGPR-targeted PFOB for hepatocytes L-02 was investigated in vitro. In vivo, all rats were treated with either ASGPR-targeted PFOB or PFOB, and ultrasound imag

Synthesis and biological evaluation of technetium-99m labeled galactose derivatives as potential asialoglycoprotein receptor probes in a hepatic fibrosis mouse model.

Two galactose derivatives, a monovalent (99m)Tc-MAMA-MGal galactoside and a divalent (99m)Tc-MAMA-DGal galactoside, were synthesized and radiolabeled in high radiochemical purity (>98%). Dynamic microSPECT imaging and biodistribution study of two traces in normal and liver fibrosis mice showed that the (99m)Tc-MAMA-DGal revealed higher specific binding to asialoglycoprotein receptors in liver and then rapidly excreted via both hepatobiliary system and renal clearance. The results suggest that (99m)Tc-MAMA-DGal may be used as SPECT probes for noninvasive evaluation of asialoglycoprotein recepto

Porcine extrahepatic vascular endothelial asialoglycoprotein receptor 1 mediates xenogeneic platelet phagocytosis in vitro and in human-to-pig ex vivo xenoperfusion.

BACKGROUND: Asialoglycoprotein receptor-1 (ASGR1) mediates capture and phagocytosis of platelets in pig-to-primate liver xenotransplantation. However, thrombocytopenia is also observed in xenotransplantation or xenoperfusion of other porcine organs than liver. We therefore assessed ASGR1 expression as well as ASGR1-mediated xenogeneic platelet phagocytosis in vitro and ex vivo on porcine aortic, femoral arterial, and liver sinusoidal endothelial cells (PAEC/PFAEC/PLSEC). METHODS: Porcine forelimbs were perfused with whole, heparinized human or autologous pig blood. Platelets were counted at r

Liver-infiltrating T helper cells in autoimmune chronic active hepatitis stimulate the production of autoantibodies against the human asialoglycoprotein receptor in vitro.

Autoantibodies against the human asialoglycoprotein receptor (ASGPR) occur in the sera of patients with autoimmune liver disorders. Liver-infiltrating T cell clones that specifically recognize the ASGPR have been described in patients with autoimmune chronic active hepatitis (AI-CAH) and primary biliary cirrhosis (PBC). Recently, we have shown that peripheral blood mononuclear cells (PBMC) from patients with AI-CAH or PBC but not chronic viral hepatitis secreted anti-ASGPR antibodies in vitro. In this study we characterized the influence of liver-infiltrating T cells on the secretion of ASGPR-

Asialoglycoprotein receptor-1 promotes hyperandrogenism in polycystic ovary syndrome through direct interaction with FSHR, leading to cAMP-PKA-CREB pathway inhibition.

In brief: Asialoglycoprotein receptor 1 (ASGR1) impairs FSH receptor trafficking and reduces the cAMP-PKA-CREB signaling pathway, thereby suppressing the expression of the target gene CYP19A1. This impairs the conversion of testosterone to estradiol, ultimately contributing to the development of hyperandrogenemia in polycystic ovary syndrome. Abstract: Polycystic ovary syndrome (PCOS) is a prevalent multifactorial disorder characterized by hormone dysregulation and follicular development defects. Asialoglycoprotein receptor 1 (ASGR1), a C-type lectin predominantly expressed on the hepatic cell

Simplified quantification method for in vivo SPECT/CT imaging of asialoglycoprotein receptor with (99m)Tc-p(VLA-co-VNI) to assess and stage hepatic fibrosis in mice.

The goal of this study is to develop a noninvasive method of SPECT imaging to quantify and stage liver fibrosis with an Asialoglycoprotein receptor (ASGP-R) targeting tracer-(99m)Tc-p(VLA-co-VNI). ASGP-Rs are well known to specifically express in the mammalian liver. Here, we demonstrated ASGP-R expression decreased in carbon tetrachloride (CCl4)-induced mouse model. ASGP-R expression correlated with liver fibrosis progression. ASGP-R could be a useful marker in the stage of liver fibrosis. Liver uptake value (LUV) derived by SPECT imaging was used to assess liver fibrosis in the CCl4-induced

Fusarolides A-C, Three Pyran-Macrolide Hybrids from a Marine Derived Fungal Fusarium verticillioide G102 as Asialoglycoprotein receptor 1 Inhibitor and Phytopathogenic Fungicides.

Three undescribed 20-membered macrolides, fusarolides A-C (1-3), with relatively rare pyran-macrolide structures and complex chiral centers, were isolated from a marine derived fungal Fusarium verticillioide G102. Their chemical structures and stereoconfigurations were well elaborated. Compound 1 promoted cholesterol efflux from Huh-7 cells in a concentration-dependent manner by inhibiting Asialoglycoprotein receptor 1 (ASGR1), which was identified as a new therapeutic target in hypercholesterolemia in liver. Besides, compounds 1-3 exhibited obvious anti-phytopathogenic fungal activities.

Role of sialic acid for platelet life span: exposure of beta-galactose results in the rapid clearance of platelets from the circulation by asialoglycoprotein receptor-expressing liver macrophages and hepatocytes.

Although surface sialic acid is considered a key determinant for the survival of circulating blood cells and glycoproteins, its role in platelet circulation lifetime is not fully clarified. We show that thrombocytopenia in mice deficient in the St3gal4 sialyltransferase gene (St3Gal-IV(-/-) mice) is caused by the recognition of terminal galactose residues exposed on the platelet surface in the absence of sialylation. This results in accelerated platelet clearance by asialoglycoprotein receptor-expressing scavenger cells, a mechanism that was recently shown to induce thrombocytopenia during Str

Cationic lipid enhances in vitro receptor-mediated transfection.

The efficiency of cell-specific transfection by receptor-mediated uptake is improved by the use of cationic lipids. Asialoglycoprotein (AP) was conjugated to poly-L-lysine (PL) and complexed with the plasmid pCMVL that contains a luciferase reporter gene. The asialoglycoprotein-poly-L-lysine:pCMVL (AP-PL:pCMVL) complexes then were mixed with the cationic lipid dioctadecylamidoglycylspermine (DOGS). This complex was taken up by the hepatocyte-like cell line, Hep G2, via the asialoglycoprotein receptor. The expression of luciferase in cells transfected with the DOGS/AP-PL: pCMVL complexes were s

Influence of taurocholate on hepatic clearance and biliary excretion of asialo intestinal alkaline phosphatase in the rat in vivo and in isolated perfused rat liver.

We have used canine intestinal alkaline phosphatase, an asialoglycoprotein, as a tool to study the effect of taurocholate on hepatic asialoglycoprotein transport. After bolus injections of 20 U of alkaline phosphatase into anesthetized rats in vivo and isolated perfused rat livers, a biexponential disappearance pattern was observed; 6.2% of the dose and 0.8% of the dose was excreted in bile, respectively. Taurocholate infusion in vivo and in isolated perfused livers resulted in a virtually complete abolition of the plasma disappearance of alkaline phosphatase in the second slow phase of the cu

[Chemical galactosylation of acid alpha-glucosidase to provide directed transport of the enzyme into lysosomes of liver parenchymal cells].

Chemical galactosylation of human liver tissue lysosomal alpha-glucosidase was carried out. As a result of the modification some physicochemical properties of the enzyme were altered, while its stability and catalytic activity were maintained. An ability of the galactosylated alpha-glucosidase to interact with asialoglycoprotein receptor from mice liver tissue was studied in vitro. The reaction required Ca2+. A specific inhibitor of the receptor, N-acetyl galactosamine, as well as high concentrations of native glycoproteins and neoglycoproteins containing terminal galactose inhibited the recep

Modulation of the liver specific phenotype in the human hepatoblastoma line Hep G2.

The human hepatoblastoma line Hep G2 modulates gene expression in vitro in response to increasing culture density. Two stages of growth can be defined. At low density (less than 200,000 cells/cm2) the cultures have a doubling time of 24 h and exhibit several characteristics of fetal liver cells, including increased synthesis of alphafetoprotein, reduced synthesis of albumin, a predominance of the fetal isoenzymes of both aldolase and pyruvate kinase and a reduced level of the cell surface receptor for asialoglycoproteins. Confluent, high density cultures of Hep G2 (greater than 1 X 10(6) cells

T-lymphocyte sensitization to hepatocyte antigens in autoimmune chronic active hepatitis and primary biliary cirrhosis. Evidence for different underlying mechanisms and different antigenic determinants as targets.

Cultured with a liver-derived lipoprotein complex, T lymphocytes from 42 of 45 patients with autoimmune chronic active hepatitis generated migration inhibitory factor compared with 16 of 33 patients with primary biliary cirrhosis. Unlike T lymphocytes from patients with primary biliary cirrhosis, the T-cell reactivity of patients with chronic active hepatitis was always suppressed by T cells from normal subjects and, with two exceptions, by T cells from patients with primary biliary cirrhosis, even when these latter cells exhibited sensitization to this same antigen complex. Using a component

The role of the liver in catabolism of mouse and human IgA.

The fate of intravascular IgA which is produced in large quantities in humans and many animal species was investigated in vivo and in vitro with emphasis on the monomeric form of IgA. The site(s) of the catabolism of intravenously injected mouse monomeric IgA labeled with a residualizing tracer (dilactitol - 125I tyramine) was studied in mice. The greatest in vivo uptake of monomeric IgA was observed in the liver. In contrast to identically labeled IgG, liver accounted for more internal catabolism of monomeric IgA than all other tissues (spleen, muscle, skin, and kidney) combined. Although bot

Inhibition of liver metastasis in mice by blocking hepatocyte lectins with arabinogalactan infusions and D-galactose.

According to our hypothesis, organ-specific lectins (e.g., the D-galactose-specific hepatic binding protein) play an important role in the organ location of metastatic malignant cells. The rapid clearance and uptake by the liver of tritiated alpha 1-acid-(asialo)glycoprotein from the circulation of Balb/c mice was markedly delayed after preinjection of D-galactose or arabinogalactan. The preinjection (1 h) and regular application (for 3 days after tumor cell inoculation in Balb/c mice) of the receptor blocking agents D-galactose and arabinogalactan prevented the settling of sarcoma L-1 tumor i

Epstein-Barr virus as a trigger for autoimmune hepatitis in susceptible individuals.

During follow-up of healthy relatives of 13 patients with autoimmune hepatitis, seven cases of infectious mononucleosis due to Epstein-Barr virus (EBV) occurred. In two of these seven, before EBV infection, there was a defect in suppressor-inducer T lymphocytes specifically controlling immune responses to the asialoglycoprotein receptor, an antigen expressed on the hepatocyte surface. In these two, antibodies to this autoantigen persisted and increased after infectious mononucleosis, and autoimmune hepatitis developed within 4 months. In susceptible individuals, EBV is a trigger for autoimmune
