Apparent interspecific transmission of Aphanomyces astaci from invasive signal to virile crayfish in a sympatric wild population.

The crayfish plague pathogen (Aphanomyces astaci) causes mass mortalities of European crayfish when transmitted from its original North American crayfish hosts. Little is known, however, about interspecific transmission of the pathogen between different American crayfish species, although evidence from trade of ornamental crayfish suggests this may happen in captivity. We screened signal and virile crayfish for A. astaci at allopatric and sympatric sites in a UK river. Whilst the pathogen was detected in signal crayfish from both sites, infected virile crayfish were only found in sympatry. Gen

Recent acute crayfish mortality reveals Aphanomyces astaci presence in Bosnia and Herzegovina.

Although the introduction of the crayfish plague pathogen Aphanomyces astaci to Europe is responsible for substantial declines in native crayfish populations throughout the whole continent, its presence has never been officially confirmed in many European regions, including most of the Balkan Peninsula. We demonstrate that the recent crayfish mortality observed in Bosnia and Herzegovina (Mostarsko blato karst field, Neretva river drainage) was caused by A. astaci. The causative strain is known only from European crayfish, indicating that A. astaci poses a threat to native species in this regio

Distribution and epidemiology of genotypes of the crayfish plague agent Aphanomyces astaci from noble crayfish Astacus astacus in Finland.

The crayfish plague agent Aphanomyces astaci was isolated from 69 noble crayfish Astacus astacus samples in Finland between 1996 and 2006. All isolates were genotyped using randomly amplified polymorphic DNA-polymerase chain reaction (RAPD-PCR). Altogether, 43 isolates belonged to the genotype group of Astacus strains (As), which is assumed to represent the genotype originally introduced into Europe around 1860 and into Finland in 1893. There were 26 crayfish plague isolates belonging to the group of Pacifastacus strain I (Ps1), which appeared in Europe after the stocking of the North American

PCR detection of the crayfish plague pathogen in narrow-clawed crayfish inhabiting Lake Eğirdir in Turkey.

Many populations of the narrow-clawed crayfish Astacus leptodactylus in Turkey, including those inhabiting Lake Eğirdir, declined drastically in the mid-1980s due to introduction of crayfish plague Aphanomyces astaci. However, unlike many other localities, there has been some recovery in the A. leptodactylus population inhabiting this lake even though crayfish plague has been suspected to have persisted since then. In support of this, DNA from 5 of 34 healthy-looking crayfish sampled recently from the lake tested positive by both conventional and real-time PCR using species-specific primers ta

Infection status of freshwater crabs and crayfish with metacercariae of Paragonimus westermani in Korea.

The present study investigated the infection status of Paragonimus westermani metacercariae in freshwater crabs (n = 363) and crayfish (n = 31) from October 2007 to October 2008 using the crush method. All of the freshwater crabs, Eriocheir japonicus, were negative for P. westermani metacercariae while 10 (32.3%) of the 31 examined crayfish were positive. The 10 positive crayfish were caught in Haenam, Jeollanam-do, and there were 8-59 (mean 28.4) metacercariae per infected crayfish. These results suggest that P. westermani metacercariae are still transmitted by crayfish enzootically in southe

The effects of microcystin-LR and its location within an environmental pool on rusty crayfish (Faxonius rusticus) behavior and physiology.

Anthropogenic factors have negatively impacted the health of freshwater ecosystems by increasing the occurrence of harmful cyanobacterial blooms and their associated toxins. Water column mixing and reworking of substrate by organisms allow for cyanotoxin deposition into sediments and resuspension into the water column. Crayfish rework benthic substrates and are exposed to toxins in both benthic and pelagic phases. Since crayfish are keystone species and ecosystem engineers, understanding the spatial dynamics and effects of aquatic toxins on crayfish is necessary. In this study, crayfish were e

Thermal stress alters the allergen and proteome profile of the redclaw crayfish (Cheraxquadicarinatus).

Rising global temperatures and growing demand for sustainable protein are concurrent challenges driving the expansion of aquaculture, including farming of shellfish such as the Australian Redclaw Crayfish (Cherax quadricarinatus). Although environmental stressors alter animal physiology, their effects on food protein allergenicity remain unexplored, particularly in shellfish, a leading cause of food-induced anaphylaxis. This study investigated how rearing temperature affects protein and allergen expression in redclaw reared at either 17 °C, 28 °C, or 32 °C. Raw and cooked protein extracts were

High variation of virulence in Aphanomyces astaci strains lacks association with pathogenic traits and mtDNA haplogroups.

Introduced into Europe from North America 150 years ago alongside its native crayfish hosts, the invasive pathogen Aphanomyces astaci is considered one of the main causes of European crayfish population decline. For the past two centuries, this oomycete pathogen has been extensively studied, with the more recent efforts focused on containing and monitoring its spread across the continent. However, after the recent introduction of new strains, the newly-discovered diversity of A. astaci in North America and several years of coevolution with its European host, a new assessment of the traits link

Electric field-enabled aptasensing interfacial engineering to simultaneously enhance specific preconcentration and electrochemical detection of mercury and lead ions.

Aptamers with strong affinity to heavy metal ions (HMIs) allow fabrication of electrochemical sensors with high selectivity and sensitivity, while controllable regulation of aptamer-HMI recognition at the sensing interface, which is vital for better analytical performance, remains challenging. Here, an electric field-based strategy for engineering an aptasensing interface was proposed to realize the specific preconcentration and accurate detection of mercury (Hg2+) and lead (Pb2+) ions with a ratiometric electrochemical sensor. The working principle is to apply an electric field to drive HMIs

Tracing the origin of the crayfish plague pathogen, Aphanomyces astaci, to the Southeastern United States.

The oomycete Aphanomyces astaci is an emerging infectious pathogen affecting freshwater crayfish worldwide and is responsible for one of the most severe wildlife pandemics ever reported. The pathogen has caused mass mortalities of freshwater crayfish species in Europe and Asia, and threatens other susceptible species in Madagascar, Oceania and South America. The pathogen naturally coexists with some North American crayfish species that are its chronic carriers. Presumptions that A. astaci originated in North America are based on disease outbreaks that followed translocations of North American

Improved method for genotyping the causative agent of crayfish plague (Aphanomyces astaci) based on mitochondrial DNA.

Aphanomyces astaci causes crayfish plague, which is a devastating disease of European freshwater crayfish. The likely first introduction of A. astaci into Europe was in the mid-19th century in Italy, presumably with the introduction of North American crayfish. These crayfish can carry A. astaci in their cuticle as a benign infection. Aphanomyces astaci rapidly spread across Europe causing the decline of the highly susceptible indigenous crayfish species. Random amplified polymorphic DNA-PCR analysis of A. astaci pure cultures characterized five genotype groups (A, B, C, D and E). Current A. as

Caridean prawn (Crustacea, Decapoda) with description of a new species and an invasive crayfish from Yuelu mountain national park, Hunan, China.

Yuelu mountain national park (5A scenic area) is a rare urban mountain scenic area. The major impacts from human activity are growing, such as largely unplanned tourist activity, urbanization, and accelerated the others responsible for environmental changes. It is unavoidable to have great pressure and influence on the diversity of fauna on the scenic area. In order to better understand the diversity of the decapod fauna in the Yuelu mountain national park, an intensive field survey has been carried out. A total of five species was collected, three species of atyid shrimps, Neocaridina palmata

Monitoring the spore dynamics of Aphanomyces astaci in the ambient water of latent carrier crayfish.

The specialized crayfish parasite Aphanomyces astaci causes the devastating crayfish plague in European crayfish. Even though A. astaci sporulation has been thoroughly studied under pure culture conditions, little is known about the sporulation dynamic from its live host. Our purpose was to investigate the A. astaci spore dynamic in its native parasite-host relationship by monitoring the sporulation from carrier crayfish into the ambient water using agent specific qPCR. American signal crayfish (Pacifastacus leniusculus) with known positive carrier status were housed individually and communall

Antioxidation, immunity and hepatopancreatic histology of red swamp crayfish (Procambarus clarkii) subjected to low-temperature aerial exposure stress and re-immersion.

Desiccation is a stressful situation that decapods often experience during live transportation. This study investigated the effects of low-temperature aerial exposures (LTAEs) (dry exposure (DL) and moist exposure (ML) at 6 °C) and re-immersion on the antioxidative and immune responses and hepatopancreatic histopathology in P. clarkii. Compared to the control group (normally feeding at 24.0 °C water temperature), the crayfish under LTAEs showed overall severe hepatopancreatic oxidative damage, with significantly increased malondialdehyde (MDA) contents and significantly reduced total antioxida

Targeted and untargeted metabolomic analysis of Procambarus clarkii exposed to a "chemical cocktail" of heavy metals and diclofenac.

Water pollution poses an important problem, but limited information is available about the joined effects of xenobiotics of different chemical groups to evaluate the real biological response. Procambarus clarkii (P. clarkii) has been demonstrated to be a good bioindicator for assessing the quality of aquatic ecosystems. In this work, we studied the bioaccumulation of cadmium (Cd), arsenic (As) and diclofenac (DCF) in different tissues of P. clarkii during 21 days after the exposure to a "chemical cocktail" of As, Cd and DCF, and until 28 days considering a depuration period. In addition, a com

Crayfish plague affects juvenile survival and adult behaviour of invasive signal crayfish.

The spread of invasive, non-native species is a key threat to biodiversity. Parasites can play a significant role by influencing their invasive host's survival or behaviour, which can subsequently alter invasion dynamics. The North American signal crayfish (Pacifastacus leniusculus) is a known carrier of Aphanomyces astaci, an oomycete pathogen that is the causative agent of crayfish plague and fatal to European crayfish species, whereas North American species are considered to be largely resistant. There is some evidence, however, that North American species, can also succumb to crayfish plag

Crayfish plague Aphanomyces astaci detected in redclaw crayfish, Cherax quadricarinatus in Taiwan.

Between December 2013 and January 2014, five outbreaks of an unknown disease with moderate to high cumulative mortality were observed among the freshwater redclaw crayfish (Cherax quadricarinatus) populations at four crayfish farms in Miaoli and Changhua counties (northern Taiwan) and at one crayfish farm in Pingtung County (southern Taiwan). Polymerase chain reaction (PCR) analysis allowed the detection of Aphanomyces astaci DNA in dead crayfish. Histopathological examination revealed an infection of host tissue by fungal hyphae that presented as typical non-septate hyphae within the soft abd

Potent infection reservoir of crayfish plague now permanently established in Norway.

Noble crayfish Astacus astacus is threatened in Europe due to invasive crayfish carrying the crayfish plague agent Aphanomyces astaci. Norway is among the last countries in which the introduction of non-indigenous crayfish has been limited through strict legislation practices. However, North American signal crayfish Pacifastacus leniusculus were recently discovered in a water-course that has been repeatedly hit by the plague. We mapped the distribution and relative density (catch per unit effort) of signal crayfish within this lake, and performed agent-specific real-time PCR to estimate the pr

Confirmation of crayfish plague in Italy: detection of Aphanomyces astaci in white clawed crayfish.

In the summer of 2009, high levels of mortality among white clawed crayfish Austropotamobius pallipes were observed in 3 watercourses of central Italy. PCR and culture methods were used to detect the causative agent of the disease. Two strains of Aphanomyces spp. were isolated and identified by PCR and DNA sequencing as Aphanomyces astaci and A. repetans. This is the first crayfish plague outbreak in Italy to be confirmed by the isolation in culture of a pathogen from Austropotamobius pallipes.

In-vitro puncture experiment using alligator teeth tracks the formation of dental microwear and its association with hardness of the diet.

With the development of dental microwear texture analysis (DMTA), there has been an increasing application of DMTA for dietary estimation in extant and fossil reptiles, including dinosaurs. While numerous feeding experiments exist for herbivorous mammals, knowledge remains limited for carnivorous reptiles. This study aimed to qualitatively and quantitatively evaluate the formation of dental microwear through repeated puncture of different types of food using isolated teeth from the American alligator (Alligator mississippiensis) in an in-vitro experiment. Eleven isolated teeth were mounted on
