Intoxication of sheep by Astragalus arequipensis in northwestern Argentina.

Spontaneous intoxication by Astragalus arequipensis was diagnosed in a flock of 300 sheep in Jujuy province, northwestern Argentina, that grazed an area heavily invaded by this plant. The main clinical signs were intention tremors, ataxia, and progressive loss of condition. Autopsy of 2 affected animals revealed loss of body condition. The main microscopic changes were fine cytoplasmic vacuolation of cells in the cerebrum, cerebellum, thyroid and adrenal glands, kidney, liver, pancreas, urinary bladder, and lymph nodes, and swollen axons in the cerebellum. Ultrastructurally, the cytoplasmic va

Exploring the complex immunomodulatory effects and gut defense via oral administration of Astragali radix water extract to normal mice.

BACKGROUND: Astragali radix (AR) is one of the most widely used traditional Chinese herbal medicines. It exhibits diverse biological activities, including immunomodulatory and anti-inflammatory properties; however, some of its activities have only been demonstrated in vitro. OBJECTIVE: To examine the effects of orally administered AR extract on immune cells and the intestine under physiological conditions, which bridges the gap between previously observed in vitro outcomes and in vivo results. METHODS: AR extract was prepared by hot water extraction. Three separate animal experiments were co

Field and experimental studies in cattle and sheep poisoned by nitro-bearing Astragalus or their toxins.

Various species of nitro-containing Astragalus, and the toxic principle of these plants, 3-nitropropionic acid or 3-nitro-1-propanol, were fed to sheep and cattle at doses which produced chronic or acute intoxication. The signs of intoxication produced were similar to those observed in poisoning that occurs under field conditions. The signs of intoxication were emphysema and difficulty in locomotion. Primary microscopic lesions were observed in the lungs and CNS. The lungs had varying degrees of alveolar emphysema and there were varying degrees of wallerian degeneration of the spinal cord.

Molecular cloning and biotic elicitation response of phenylalanine ammonia-lyase gene of Astragalus chrysochlorus.

Phenylalanine ammonia lyase (PAL) is the first enzyme of the phenylpropanoid pathway, and it is necessary to upregulate flavonoid biosynthesis in most of the plant species. In this study, we have cloned PAL gene from endemic Astragalus chrysochlorus which is a producer of phenolic nicotiflorin (kaempferol-3-O-rutinoside). The cDNA encoding PAL was cloned from A. chrysochlorus using RT-PCR (reverse transcription-polymerase chain reaction) with conserved primer pairs. Amino acid sequence alignments showed that AcPAL (2160 bp, Accession number: KM189182) has more than 95% amino acid identity with

A Protective Role of Phenylalanine Ammonia-Lyase from Astragalus membranaceus against Saline-Alkali Stress.

Phenylalanine ammonia-lyase (PAL, E.C.4.3.1.5) catalyzes the benzene propane metabolism and is the most extensively studied enzyme of the phenylpropanoid pathway. However, the role of PAL genes in Astragalus membranaceus, a non-model plant showing high capability toward abiotic stress, is less studied. Here, we cloned AmPAL and found that it encodes a protein that resides in the cytoplasmic membrane. The mRNA of AmPAL was strongly induced by NaCl or NaHCO3 treatment, especially in the root. Overexpressing AmPAL in Nicotiana tabacum resulted in higher PAL enzyme activities, lower levels of malo

Dietary aqueous extract of Astragalus sinicus alleviates obesity by reshaping gut microbiota and metabolism.

BACKGROUND: Obesity induced by long-term high-fat diet (HFD) consumption is closely associated with metabolic disorders and gut microbiota dysbiosis. Although plant-derived extracts show anti-obesity potential, microbiota-dependent mechanisms and causal relevance remain incompletely understood. Astragalus sinicus L. is a traditional medicinal and dietary plant, yet the metabolic effects of its aqueous extract have not been systematically evaluated. This study investigated the anti-obesity effects of an aqueous extract of Astragalus sinicus (ASAE) and determined whether gut microbiota remodelin

Recent Advances in the Mechanisms and Applications of Astragalus Polysaccharides in Liver Cancer Treatment: An Overview.

Astragalus polysaccharides (APS), bioactive compounds derived from Astragalus membranaceus, have emerged as promising natural agents in the treatment of hepatocellular carcinoma, a leading cause of cancer-related mortality. Preclinical studies indicate that APS exerts significant anti-liver cancer effects through multiple biological actions, including the promotion of apoptosis, inhibition of proliferation, suppression of epithelial-mesenchymal transition, regulation of autophagy, and modulation of immune responses. These therapeutic effects are closely associated with the regulation of critic

Context-dependent immunomodulation by Astragalus polysaccharides: Structural determinants, structure-activity relationships, mechanisms, and applications: A review.

Astragalus polysaccharides (APS), derived mainly from Astragalus membranaceus (Fisch.) Bunge and Astragalus membranaceus var. mongholicus (Bunge) P. K. Hsiao, have attracted considerable attention as plant-derived immunomodulatory macromolecules. Accumulating evidence suggests that APS-mediated immunomodulation is context-dependent rather than unidirectional. Raw-material characteristics, processing methods, extraction procedures, fractionation, and purification can generate APS fractions with different structural characteristics, including molecular weight distribution, monosaccharide composi

Clinical and pathological comparison of Astragalus lentiginosus and Ipomoea carnea poisoning in goats.

The indolizidine alkaloid swainsonine, found in some Astragalus and Oxytropis (i.e., locoweed) species, is a potent cellular glycosidase inhibitor that often poisons livestock. Other toxic genera such as some Ipomoea species also contain swainsonine as well as calystegines which are similar polyhydroxy alkaloids. The toxicity of calystegines is poorly characterized; however, they are also potent glycoside inhibitors capable of intestinal and cellular glycoside dysfunction. The objective of this study was to directly compare A. lentiginosus and I. carnea poisoning in goats to better characteriz

Intoxication by Astragalus garbancillo var. garbancillo in llamas.

Lysosomal storage diseases are inherited and acquired disorders characterized by dysfunctional lysosomes. Intracytoplasmic accumulation of undegraded substrates leads to impaired cellular function and death. Several plant species are toxic to livestock because of the presence of indolizidine alkaloids, including swainsonine, which cause a storage disease. Swainsonine-induced nervous disease (i.e., locoism) of sheep and cattle is well recognized in several parts of the world, particularly in the western United States and in parts of Australia. Spontaneous intoxication by Astragalus garbancillo

Astragalus polysaccharides restore neutrophil functions and gut microbiota homeostasis in dairy cows with subclinical ketosis.

Subclinical ketosis (SCK) in periparturient cows is associated with gut microbiota dysbiosis and energy metabolism disorders. Although in vitro studies show that free fatty acids (FFA) and BHB impair polymorphonuclear neutrophil (PMN) functions-potentially causing secondary infections-limited in vivo evidence exists. Astragalus polysaccharides (APS) modulate metabolism, immunity, and gut microbiota, but their effects on PMN functions and gut microbiota in SCK cows remain unclear. This study aims to evaluate PMN dysfunction and gut microbiota dysbiosis in SCK cows, and clarify the protective ef

Dietary supplementation of Astragalus flavonoids regulates intestinal immunology and the gut microbiota to improve growth performance and intestinal health in weaned piglets.

Astragali Radix (AS) is a widely used herb in traditional Chinese medicine, with calycosin as its main isoflavonoid. Our previous study discovered that calycosin triggers host defense peptide (HDP) production in IPEC-J2 cells. The aim of this study is to investigate the alleviation effects of AS total flavone and AS calycosin on growth performance, intestinal immunity, and microflora in weaned piglets. Sixty-four piglets were assigned randomly to 4 treatment groups, (1) CON: the basal diet, (2) P-CON: the basal diet plus antibiotics (1 g/kg), (3) AS-TF: the basal diet plus AS total flavone at

Phytochemicals from Astragalus zederbaueri as Acetylcholinesterase Inhibitors for Alzheimer's Therapy.

Alzheimer's Disease (AD), the dominant form of dementia that evolves with age, involves several mechanisms by which it progresses. The cholinergic hypothesis proposes that the activity of Acetylcholinesterase (AChE) causes the decline of cholinergic neurotransmission, which leads to Alzheimer's Disease. Considerable studies are being conducted to determine the best AChE inhibitor. This study evaluated 40 phytocompounds from the Astragalus zederbaueri plant as potential AChE inhibitors for Alzheimer's disease. The analysis of the phytochemicals was conducted using the control drug donepezil (co
