Goose astrovirus infection affects uric acid production and excretion in goslings.

In 2018, a new goose astrovirus (GAstrV) was reported in China, which causes 2 to 20% deaths in 4- to 16-day-old goslings causing great damages to the livestock industry. Gout is the typical feature of GAstrV infection in goslings. However, the mechanism of gout formation remains unclear. In the present study, 2-day-old goslings were infected intramuscularly with GAstrV for 14 D. One quarter of the infected goslings died, and typical gout pathological changes were found in the dead infected goslings. Pathological changes were observed in the morphology of the kidney and liver, such as degenera

Loop-mediated isothermal amplification combined with a Pyrococcus furiosus argonaute system for the rapid detection of goose astrovirus.

Goose astrovirus (GAstV) is a newly emerged viral pathogen in goose, characterized by a high incidence and significant mortality rates. This etiology has repeatedly occurred in coastal areas of China, rapidly spreading to inland provinces in recent decade, and thus imposed huge economic losses on China's goose industry. Therefore, it is essential to establish a rapid, accurate and sensitive method for GAstV diagnosis. In this study, we employed reverse transcription loop-mediated isothermal amplification (RT-LAMP) assay combined with a Pyrococcus furiosus Argonaute (PfAgo) system, providing a

Duplex MIRA assays for simultaneous detection of novel goose astrovirus and goose circovirus.

BACKGROUND: Novel goose astrovirus (NGAstV) and goose circovirus (GoCV) are two major pathogens responsible for disease outbreaks in goslings, causing substantial economic losses to the goose farming industry. In this study, two duplex multienzyme isothermal rapid amplification (MIRA) assays were developed for the simultaneous detection of NGAstV and GoCV, and the specificity and sensitivity of these detection methods were evaluated. RESULTS: Both MIRA assays demonstrated high specificity for NGAstV and GoCV, with no cross-reactivity observed with six waterfowl pathogens, including duck enter

Development and protective efficacy of a live attenuated vaccine candidate against goose astrovirus.

Goose astrovirus (GAstV) is a significant pathogen affecting goslings by inducing visceral gout, yet no commercial vaccine is currently available. This study involved the serial passaging of the GAstV-GXNN strain in LMH cells to investigate alterations in viral replication, genomic stability, and pathogenicity, as well as to assess the potential of a vaccine candidate. The findings indicated that the viral titer increased progressively with each passage, reaching 107.35 TCID50/mL by the 120th passage (GAstV-GXNNP120). Whole-genome sequencing revealed the presence of 6, 19, 26, and 28 nucleotid

Antibody Response in Pullets Vaccinated with Inactivated Chicken Astrovirus for White Chick Syndrome.

White chick syndrome (WCS), a vertically transmitted disease caused by chicken astrovirus (CAstV), causes decreased hatchability of broiler breeder progeny and increased hatch of weak, white chicks. In the absence of a commercial vaccine, control measures for WCS have included serological surveillance of pullets, then movement of litter from farms with confirmed CAstV seroconversion to farms with seronegative birds. However, litter movement carries the risk of introducing other pathogens to farms, such as Salmonella. CAstVs have been isolated from clinical cases of WCS and genetically characte

Goose Nephritic Astrovirus Infection of Goslings Induces Lymphocyte Apoptosis, Reticular Fiber Destruction, and CD8 T-Cell Depletion in Spleen Tissue.

The emergence of a novel goose nephritic astrovirus (GNAstV) has caused economic losses to the Chinese goose industry. High viral load is found in the spleen of goslings infected with GNAstV, but pathological injuries to the spleen due to GNAstV are largely unknown. In this study, 50 two-day-old goslings were infected orally with GNAstV, and 50 goslings were treated with PBS as control. Spleens were collected at different times following infection to assess damage. GNAstV infection caused visceral gout and urate deposition in joints, and resulted in 16% mortality. GNAstV was found in the lymph

Transcriptome Sequencing of the Spleen Reveals Antiviral Response Genes in Chickens Infected with CAstV.

Astrovirus infections pose a significant problem in the poultry industry, leading to multiple adverse effects such as a decreased egg production, breeding disorders, poor weight gain, and even increased mortality. The commonly observed chicken astrovirus (CAstV) was recently reported to be responsible for the "white chicks syndrome" associated with an increased embryo/chick mortality. CAstV-mediated pathogenesis in chickens occurs due to complex interactions between the infectious pathogen and the immune system. Many aspects of CAstV-chicken interactions remain unclear, and there is no informa

First Molecular Evidence of the Presence of Avian Astroviruses in Turkey Flocks of Ecuador Through the Standardization of RT-qPCR Assays Based on SYBR Green.

Enteric diseases are a significant challenge for the poultry industry, causing substantial economic losses and affecting productivity. Turkey astrovirus (TAstV) types 1 and 2 and avian nephritis virus (ANV) are recognized as viral pathogens contributing with enteric diseases in turkeys, particularly in young poults. These viruses, part of the Astroviridae family, are small, round, non-enveloped, positive-sense RNA viruses with high prevalence in turkey flocks. Despite their importance, they had not been identified in Ecuador until now. This study presents the first detection and molecular char

Emerging fatal gout disease in Chinese goslings linked to acute kidney injury induced by novel goose astrovirus infection.

A novel goose astrovirus (GAstV) has broken out across China in recent years, causing widespread damage to the poultry industry. In goslings infected with GAstV, the leading cause of death is visceral gout. However, our understanding of the mechanism of gout formation in GAstV infection is largely inadequate. The aim of this study was to examine the pathogenicity of a GAstV strain and explore the molecular mechanisms of visceral gout caused by viral infection in goslings. The virulent GAstV strain HR2105/1 was effectively isolated from the visceral tissue of goslings in gout-affected areas. Th

Establishment and application of multiplex real-time qPCR detection for Tembusu virus and goose astrovirus genotypes 1 and 2.

The concurrent infection and epidemic of goose astrovirus type 1 (GoAstV-1), goose astrovirus type 2 (GoAstV-2), and Tembusu virus (TMUV) in geese have resulted in substantial economic losses to the goose industry. Therefore, establishing a rapid and accurate diagnostic method for these three viruses is imperative. In the present study, we designed specific primers targeting the Cap gene of GoAstV and the E gene of TMUV and developed a SYBR Green I-based multiplex real-time qPCR method for the simultaneous detection of GoAstV-1, GoAstV-2, and TMUV. The results revealed that the R2 values for t

Isolation of avian nephritis virus from chickens showing enteric disorders.

Runting-stunting syndrome (RSS) is one of the diseases associated with many detected viruses. In Brazil, there were reports of several enteric disease outbreaks in chickens in which avian nephritis virus (ANV) was detected; however, the role of ANV in the outbreaks and whether the virus was a causative agent of these cases of enteric diseases were not determined. The aim of this study was to isolate ANV in specific pathogen-free (SPF) chicken embryonated eggs (CEE) from the enteric contents of chickens showing signs of RSS. For this purpose, 22 samples of chicken enteric contents that were pos

Investigation into the aetiology of runting and stunting syndrome in chickens.

Currently, the aetiology of runting and stunting syndrome (RSS) in chickens is unknown. The impact of RSS on weight gain and microscopic lesions in immunological organs and the duodenum, was investigated in 1-day-old commercial broilers at 12 days following exposure to RSS-contaminated litter. Furthermore, the presence of the viral nucleic acids of three astroviruses and one parvovirus was analysed by in situ hybridization from days 1 through 5 post exposure. A 70% decrease in weight was observed in the RSS-exposed group at the end of the experiments when compared with the unexposed controls.

Maternal antibody-mediated protection against chicken astrovirus: efficacy of novel inactivated vaccine.

Chicken astrovirus (CAstV) infections cause significant economic losses in global poultry production through nephritis, enteritis, and growth retardation, yet commercial vaccines remain unavailable. This study evaluated a novel inactivated CAstV vaccine's capacity to induce protective maternal antibodies in specific-pathogen free breeder chickens and subsequent progeny protection. One hundred SPF breeder chickens received two dose vaccination (10 and 16 weeks) with inactivated vaccine containing Genogroup B strain IP2024-00664 (GenBank: PX146716.1, complete ORF-2 capsid protein gene, 2,247 bp)

Detection of antibodies against Turkey astrovirus in humans.

Astroviruses are a leading cause of gastroenteritis in mammals and birds worldwide. Although historically thought to be species-specific, increasing evidence suggests that astroviruses may cross species barriers. In this report, we used enzyme-linked immunosorbent assays to screen sera from three distinct human cohorts involved in influenza studies in Memphis, TN or Chapel Hill, NC, and Midwestern poultry abattoir workers for antibodies to turkey astrovirus type 2 (TAstV-2). Surprisingly, 26% of one cohort's population was TAstV-2 positive as compared to 0 and 8.9% in the other cohorts. This c

Research note: Simultaneous detection of GPV, H5 AIV, and GoAstV via TaqMan probe-based multiplex qPCR.

The endemic status of goose parvovirus (GPV), H5 subtype avian influenza virus (AIV), and goose astrovirus (GoAstV) infections continues to devastate the poultry industry in China. Despite this, there exists a notable gap in the application of molecular diagnostic techniques. This investigation described the development of a multiplex qualitative polymerase chain reaction (qPCR) assay capable of concurrently detecting GPV, H5 AIV, and GoAstV, with no cross-reactivity observed with other avian viral pathogens. The assay exhibited a detection threshold of 10 copies/μL for both GPV and GoAstV, an

A novel exonuclease-assisted isothermal nucleic acid amplification with ultrahigh specificity mediated by full-length Bst DNA polymerase.

Here, we introduced a novel exonuclease-assisted isothermal nucleic acid amplification (Exo-NAT) utilizing the full-length Bst DNA polymerase combined with a melting curve analysis. This method achieved an ultrahigh specificity with a good detection limit (102 copies) in both singleplex and multiplex detection, which was validated by detecting three diarrhea-inducing pathogens, rotavirus A, astrovirus, and adenovirus, in 42 clinical samples.

Co-detection and genomic characterization of avian rotavirus A, avian orthoreovirus, and chicken megrivirus-C using nontargeted metagenomic surveillance in Indian broiler chickens.

Nontargeted metagenomic surveillance of the poultry enteric virome reveals underrecognized threats to poultry health and productivity in intensive production systems. In South Asia, avian rotavirus A (AvRV-A) and avian orthoreovirus (ARV) are frequently detected in broilers by conventional diagnostics, whereas chicken megrivirus genotype C (ChMeV-C) is often identified through metagenomic surveillance. Often present in both clinical disease and coinfections, these viruses may impair gut function, immune responses, and growth performance, yet their genomic diversity and evolutionary dynamics in
