Lorazepam reinstates punishment-suppressed remifentanil self-administration in rats.

RATIONALE: We recently described a reinstatement procedure that models relapse to drug abuse in cases where abstinence results from aversive consequences of drug use. The potential value of this punishment-based model of relapse depends on its sensitivity to relapse-inducing events that are ineffective in the widely used extinction-based model. OBJECTIVES: It is known that certain drugs can have anti-punishment effects, but these drugs have not been tested in the punishment-based reinstatement procedure. Therefore, the effects of the benzodiazepine, lorazepam, were examined using punishment-b

Value control of punishment.

Punishment raises a basic question for human cognition: Do agents suppress actions because they represent the negative consequences those actions produce? Everyday deterrence, from parental reprimands to legal sanctions, assumes that people regulate behavior by learning what their actions cause and by evaluating those consequences negatively. However, in spite of this widely held assumption, experimental evidence that punished instrumental behavior is controlled by the current value of the punisher is currently lacking. To test this idea, we applied an outcome revaluation procedure to instrume

Neuropeptide Y tonically inhibits an NMDAR➔AC1➔TRPA1/TRPV1 mechanism of the affective dimension of chronic neuropathic pain.

Transection of the sural and common peroneal branches of the sciatic nerve produces cutaneous hypersensitivity at the tibial innervation territory of the mouse hindpaw that resolves within a few weeks. We report that interruption of endogenous neuropeptide Y (NPY) signaling during remission, with either conditional NPY knockdown in NPYtet/tet mice or intrathecal administration of the Y1 receptor antagonist BIBO3304, reinstated hypersensitivity. These data indicate that nerve injury establishes a long-lasting latent sensitization of spinal nociceptive neurons that is masked by spinal NPY-Y1 neu

Mechanisms of amnesia induced by impairment of long-term memory reconsolidation in edible snail.

Involvement of neurotransmitter receptors and translation and transcription processes in reconsolidation of conditioned food aversion memory was investigated in experiments on edible snails. Injections of neurotransmitter receptor antagonists and protein synthesis inhibitors before the reminder session were found to induce amnesia that was characterized by the possibility of memory recovery in repeated training and under the effect of mnemotropic agent D-cycloserine (early stage of amnesia) or by resistance to the mentioned actions (late stage). It has been shown that amnesia induction by memo

Frequency Dependent Electrical Stimulation of PFC and ACC for Acute Pain Treatment in Rats.

As pain consists of both sensory and affective components, its management by pharmaceutical agents remains difficult. Alternative forms of neuromodulation, such as electrical stimulation, have been studied in recent years as potential pain treatment options. Although electrical stimulation of the brain has shown promise, more research into stimulation frequency and targets is required to support its clinical applications. Here, we studied the effect that stimulation frequency has on pain modulation in the prefrontal cortex (PFC) and the anterior cingulate cortex (ACC) in acute pain models in r

Neural Plasticity in the Ventral Tegmental Area, Aversive Motivation during Drug Withdrawal and Hallucinogenic Therapy.

Aberrant glutamatergic signaling has been closely related to several pathologies of the central nervous system. Glutamatergic activity can induce an increase in neural plasticity mediated by brain-derived neurotrophic factor (BDNF) in the ventral tegmental area (VTA), a nodal point in the mesolimbic dopamine system. Recent studies have related BDNF dependent plasticity in the VTA with the modulation of aversive motivation to deal with noxious environmental stimuli. The disarray of these learning mechanisms would produce an abnormal augmentation in the representation of the emotional informatio

Dissociating retrieval-dependent contextual aversive memory processes in female rats: Are there cycle-dependent differences?

Upon retrieval, aversive associative memories may engage alternative processes depending on the conditioned stimulus exposure length. Generally, a short session maintains it through reconsolidation, and a long session inhibits it because of extinction learning. However, various experimental interventions have produced no memory changes when given after intermediate conditioned stimulus exposure events. The lack of effectiveness in the latter case has been explained by a stage of transition from reconsolidation to extinction, during which both phases are engaged but neither prevails. Alternativ

Gamma radiation-induced conditioned taste aversions in rats: a comparison of the protective effects of area postrema lesions with differing doses of radiation.

Lesions which destroy the area postrema (AP) and damage the adjacent nucleus of the solitary tract (NTS) attenuate or abolish conditioned taste aversions (CTA) induced by a variety of pharmacological agents as well as exposure to radiation. In the present experiment, 4 groups of male rats received lesions of AP and 4 groups were given sham lesions. One sham-lesioned and one AP-lesioned group were given a single pairing of 1-hr access to a novel 0.10% sodium saccharin solution followed immediately with exposure to 0, 100, 200, or 400 rad of gamma radiation, respectively. Four days later all gro

Disrupted population coding in the prefrontal cortex underlies pain aversion.

The prefrontal cortex (PFC) regulates a wide range of sensory experiences. Chronic pain is known to impair normal neural response, leading to enhanced aversion. However, it remains unknown how nociceptive responses in the cortex are processed at the population level and whether such processes are disrupted by chronic pain. Using in vivo endoscopic calcium imaging, we identify increased population activity in response to noxious stimuli and stable patterns of functional connectivity among neurons in the prelimbic (PL) PFC from freely behaving rats. Inflammatory pain disrupts functional connecti

The Role of New Agents and Supportive Care in a Multimodal Approach to Cancer Cachexia.

Given the multi-faceted nature of cancer cachexia, a combination of pharmacologic and supportive measures such as exercise and nutrition seems intuitive to most clinicians. Clinical trials have also suggested that a multimodal approach to cancer cachexia (CC) is feasible and potentially effective. However, past trials have been limited by medications that were partially effective or had the potential for serious adverse events such as thromboembolism. We review new agents that have demonstrated efficacy in phase II trials or are involved in multi-center phase III studies. The advent of several

Relative amnesic potency of five inhalational anesthetics follows the Meyer-Overton rule.

BACKGROUND: Doses of volatile anesthetics around 0.3 minimum alveolar concentration (MAC) inhibit learning. However, threshold amnesic doses and relative potencies between agents are not well established. The authors determined amnesic potency in rats for four common volatiles and nitrous oxide. METHODS: After institutional review board approval, adult Sprague-Dawley rats received inhibitory avoidance training during exposure to either air or various subanesthetic doses of desflurane, sevoflurane, isoflurane, halothane, or nitrous oxide (4-21 rats/dose). Animals were trained to remain in a st

Anti-aversive effects of cannabidiol on innate fear-induced behaviors evoked by an ethological model of panic attacks based on a prey vs the wild snake Epicrates cenchria crassus confrontation paradigm.

Several pharmacological targets have been proposed as modulators of panic-like reactions. However, interest should be given to other potential therapeutic neurochemical agents. Recent attention has been given to the potential anxiolytic properties of cannabidiol, because of its complex actions on the endocannabinoid system together with its effects on other neurotransmitter systems. The aim of this study was to investigate the effects of cannabidiol on innate fear-related behaviors evoked by a prey vs predator paradigm. Male Swiss mice were submitted to habituation in an arena containing a bur

Effects of Voluntary Ingestion of Synthetic Delta-9-Tetrahydrocannabinol on the Hedonic Value of Rewarding and Aversive Substances and CB1 Receptor Expression.

BACKGROUND: Human studies indicate a relationship between delta-9-tetrahydrocannabinol (THC) and alcohol use, yet research on their codependency remains limited. Rodent models have shown that cannabinoids can enhance the incentive salience and hedonic value of sucrose and alcohol. However, results seem to vary by the functions of THC dose and route of administration. MATERIALS AND METHODS: The present study addresses these discrepancies by using a translational model of THC consumption, using lower oral doses of synthetic THC (Dronabinol). Male Long-Evans (n = 24) rats consumed a vehicle or T

Phosphorylation of MAP kinase and CREB in mouse cortex and amygdala during taste aversion learning.

Mice were subjected to a taste aversion conditioning procedure in which they drank water, familiar saccharin, or novel saccharin, followed by injection of either NaCl or the emetic agent LiCl. Immunohistochemistry was used to localize phosphorylated MAP kinase (ppERK) and phosphoCREB (pCREB) in the brain shortly after conditioning. An examination of insular cortex and amygdala revealed specific phosphorylation of MAP kinase by novel saccharin and LiCl, and CREB by LiCl. Pairing of novel saccharin with LiCl was the only stimulus sufficient to increase ppERK and pCREB immunoreactivity in the lat

Pharmacotherapy in alcoholism.

This review categorizes five main uses of pharmacologic agents in the treatment of alcoholism: reversing the active pharmacologic effects of alcohol; controlling withdrawal symptoms; blocking the desire for alcohol use; treating psychiatric symptoms induced by alcohol and other drugs; and treating independent, but concurrent, psychopathologic conditions. No medication, including stimulants such as caffeine, has been found to actually reverse the action of alcohol. Because of their cross-tolerance and dependence with alcohol, benzodiazepines--especially intermediate acting preparations such as

Decision for self and other modulates risk attitude and electrophysiological processing: evidence from a behavioral and electrophysiological experiment.

Understanding how people make risky decisions for others compared to themselves is central to decision neuroscience. However, the cognitive and neural underpinnings of such self-other shifts in risk preference-and the mechanisms driving individual differences-remain unclear. To address this, we employed a mixed gambling task with feedback in which participants made risky decisions for themselves and for others while electroencephalography was recorded. Although individuals generally exhibited similar patterns across agents, decisions made for others were associated with a higher degree of risk

Approach-avoidance conflict recruits lateral frontoparietal and cinguloinsular networks in a predator-prey game setting.

Objects associated with both reward and threat produce approach-avoidance conflict (AAC). Although our day-to-day encounters with AAC objects are dynamic and interactive, the cognitive neuroscience literature on AAC is largely based on experiments that use static stimuli. Here, we used a dynamic, interactive, video-game environment to test neural substrates implicated in processing AAC in a more ecologically valid setting. While undergoing functional magnetic resonance imaging (fMRI), subjects (N = 31) played a predator-prey video game, guiding an avatar through a maze containing six types of

Buffering lidocaine heightens aversion to cornual nerve injections in dairy calves.

Best practice is to numb the horn buds before disbudding dairy calves, which can be achieved by injecting lidocaine, a local anesthetic, around the cornual nerve. In humans, the acute pain that occurs upon injection of lidocaine can be reduced by neutralizing the pH of the solution with an alkalizing agent, such as sodium bicarbonate. We evaluated whether buffering lidocaine would reduce calves' behavioral and physiological responses to cornual nerve blocks on the left and right sides of the head. Female Holstein calves were assigned to 1 of 3 treatments: cornual nerve blocks with 5.5 mL of un

Impact of Fluoxetine on Behavioral Invigoration of Appetitive and Aversively Motivated Responses: Interaction With Dopamine Depletion.

Impaired behavioral activation and effort-related motivational dysfunctions like fatigue and anergia are debilitating treatment-resistant symptoms of depression. Depressed people show a bias towards the selection of low effort activities. To determine if the broadly used antidepressant fluoxetine can improve behavioral activation and reverse dopamine (DA) depletion-induced anergia, male CD1 mice were evaluated for vigorous escape behaviors in an aversive context (forced swim test, FST), and also with an exercise preference choice task [running wheel (RW)-T-maze choice task]. In the FST, fluoxe

Folic acid prevents behavioral impairment and Na(+), K(+) -ATPase inhibition caused by neonatal hypoxia-ischemia.

Folic acid plays an important role in neuroplasticity and acts as a neuroprotective agent, as observed in experimental brain ischemia studies. The aim of this study was to investigate the effects of folic acid on locomotor activity, aversive memory and Na(+),K(+)-ATPase activity in the frontal cortex and striatum in animals subjected to neonatal hypoxia-ischemia (HI). Wistar rats of both sexes at postnatal day 7 underwent HI procedure and were treated with intraperitoneal injections of folic acid (0.011 μmol/g body weight) once a day, until the 30th postnatal day.
