The directed growth of retinal axons towards surgically transposed tecta in Xenopus; an examination of homing behaviour by retinal ganglion cell axons.

The growth of optic axons towards experimentally rotated tecta has been studied. In stage 24/25 embryos, a piece of the dorsal neural tube, containing the dorsal midbrain rudiment, was rotated through 180 degrees. At later stages of development, the pathways of growing optic axons were investigated by labelling with either horseradish peroxidase or fluorescent dye. It is shown that retinal ganglion cell axons followed well-defined pathways, in spite of the abnormal structure of the brain, and were able to locate displaced tecta. This directed outgrowth of retinal axons in the optic tracts appe

Spatial confinement shapes organelle architecture and remodeling in axons.

Axons are extended cellular compartments that mediate neuronal connectivity over extraordinary distances, placing unique demands on local organelle organization and trafficking. How these geometric constraints influence organelle architecture remains poorly understood. Here, we used cryo-electron tomography and quantitative morphometric analyses to define the three-dimensional ultrastructural organization of dorsal root ganglion axons in a near-native state. We identify distinct vesicle populations with tightly versus broadly constrained size distributions and reveal a continuum of endolysosom

Selective inhibition of small-diameter axons using infrared light.

Novel clinical treatments to target peripheral nerves are being developed which primarily use electrical current. Recently, infrared (IR) light was shown to inhibit peripheral nerves with high spatial and temporal specificity. Here, for the first time, we demonstrate that IR can selectively and reversibly inhibit small-diameter axons at lower radiant exposures than large-diameter axons. We provide a mathematical rationale, and then demonstrate it experimentally in individual axons of identified neurons in the marine mollusk Aplysia californica, and in axons within the vagus nerve of a mammal,

Optic nerve regeneration in mammals: Regenerated or spared axons?

Intraorbital optic nerve crush in rodents is widely used as a model to study axon regeneration in the adult mammalian central nervous system. Recent studies using appropriate genetic manipulations have revealed remarkable abilities of mature retinal ganglion cell (RGC) axons to regenerate after optic nerve injury, with some studies demonstrating that axons can then go on to re-innervate a number of central visual targets with partial functional restoration. However, one confounding factor inherent to optic nerve crush injury is the potential incompleteness of the initial lesion, leaving spared

Molecular disorganization of axons adjacent to human lacunar infarcts.

Cerebral microvascular disease predominantly affects brain white matter and deep grey matter, resulting in ischaemic damage that ranges from lacunar infarcts to white matter hyperintensities seen on magnetic resonance imaging. These lesions are common and result in both clinical stroke syndromes and accumulate over time, resulting in cognitive deficits and dementia. Magnetic resonance imaging studies suggest that these lesions progress over time, accumulate adjacent to prior lesions and have a penumbral region susceptible to further injury. The pathological correlates of this adjacent injury i

Segregation and pathfinding of callosal axons through EphA3 signaling.

The corpus callosum, composed of callosal axons, is the largest structure among commissural connections in eutherian animals. Axon pathfinding of callosal neurons has been shown to be guided by intermediate targets, such as midline glial structures. However, it has not yet been understood completely how axon-axon interactions, another major mechanism for axon pathfinding, are involved in the pathfinding of callosal neurons. Here, we show that callosal axons from the medial and lateral regions of the mouse cerebral cortex pass through the dorsal and ventral parts, respectively, of the corpus ca

Glutamate receptor-mediated ischemic injury of premyelinated central axons.

OBJECTIVE: Ischemic injury of axons is a feature of periventricular leukomalacia, a pathological correlate of cerebral palsy. Recent evidence suggests that axons are damaged before they receive the first layer of compact myelin. Here we examine the cellular mechanisms underlying ischemic-type injury of premyelinated central axons. METHODS: Two-thirds of axons in the postnatal day 10 (P10) rat optic nerve are small premyelinated axons (<0.4microm in diameter), and one-third have undergone radial expansion in preparation for glial contact and the onset of myelination. Compound action potential

Erythropoietin protects sensory axons against paclitaxel-induced distal degeneration.

Paclitaxel causes a sensory polyneuropathy with characteristic features of distal axonal degeneration. Although the exact mechanisms underlying distal axonal degeneration are unknown, paclitaxel-induced axonal degeneration has been shown to be associated with an increase in detyrosinated tubulin. Here we show that recombinant human erythropoietin prevents axonal degeneration in sensory neurons in vitro and this effect is associated with downregulation of detyrosinated tubulin. Furthermore, in an animal model of paclitaxel-induced distal sensory polyneuropathy, recombinant human erythropoietin

Morphometry of axons in optic nerves of Siamese's twins.

PURPOSE: To observe the development of optic nerve, we examined four optic nerves from Siameses Twins by absolute counts of axons. METHODS: Mean axon diameter, mean axon density, totally axonal population and optic nerve area were noted for each optic nerve. The mean axon diameter and the mean axon density were compared between paraxial (inner sectors) and cortical(outer sectors) areas of the nerves. RESULTS: More myelinated axons were seen in the inner sectors as compared to the outer sectors (average 11 axons/1000 microm2 in inner sectors and 34 axons/1000 microm2 in outer sectors (P = 0.0

Sympathetic axons pathfind successfully in the absence of target.

To determine whether sympathetic axons require the presence of a peripheral target to grow to the correct destination, we examined the developing footpad innervation in tabby mutant mice which lack sweat glands. Despite the absence of sweat glands, noradrenergic sympathetic axons are transiently present in the presumptive target area and avoid the more distal epidermal/dermal domain occupied by sensory axons. Since sympathetic axon pathfinding was not dependent upon the target tissue, we compared the subsequent development of sweat gland axons in tabby footpads with that in control footpads. I

Molecular and cellular properties of mammalian primary olfactory axons.

How are the axonal projections of olfactory and vomeronasal receptor neurons to the olfactory bulb formed during development? How are the primary olfactory axonal connections functionally organized? With progress in molecular biological techniques and histochemical methods, it became possible to study cellular strategies and molecular mechanisms which guide the primary olfactory axons of the main and accessory olfactory systems to the target glomeruli in the bulb. In addition, new methodologies have begun to elucidate various subsets of the primary olfactory axons with distinctive central conn

Myelination of two axons by a single Schwann cell.

A Schwann cell can form only one internode of myelin around an axon. However, we observed the formation by a single Schwann cell of myelin around two axons of different diameters in the sural nerve of a 45-year-old man with mononeuritis multiplex. Schwann cell processes spiraled in the same direction around each axon, forming mesaxons. The findings in this case appear to be an undescribed type of aberrant myelination.

Target size regulates calibre and myelination of sympathetic axons.

Axons in vertebrate peripheral nerves are ensheathed by Schwann cells. For some axons, this sheath consists of a single layer of glial cell cytoplasm and plasma membranes; for other axons, Schwann cells form multilayered myelin. Whether or not a Schwann cell makes myelin is determined by a signal from the axon, but the nature of this signal is not known. Here I show that sympathetic postganglionic axons, which are normally not myelinated, become myelinated when their calibre is increased as a result of increasing the size of the peripheral target they innervate. This result implies that axon c

Observation on the presence of Listeria monocytogenes in axons.

The brains from 16 sheep diagnosed as listerial encephalitis on the basis of clinical signs and histopathology were examined to determine the types of inflammatory cells present and for bacteria. Listeria monocytogenes were detected by immunocytochemistry in the brains of the 16 sheep and were also demonstrated in the trigeminal ganglion from the same side as the most severely affected side of the medulla. Bacteria were observed within myelinated axons in white matter tracts of the brain and in peripheral nerve axons of the trigeminal nerve. Their location in axoplasm was confirmed by electron

Nerve axons in carotid body hyperplasia. A quantitative study.

The nerve density, expressed as total length of nerve fibers per square millimeter of tissue section, was measured in both carotid bodies from five subjects free from cardiopulmonary disease and from eight subjects with carotid body hyperplasia. Six of the cases with enlargement of the carotid body had been exposed to chronic hypoxemia in life (due to panacinar pulmonary emphysema in five instances and the pickwickian syndrome in one case), and the other two cases had had systemic hypertension. The cases of carotid body hyperplasia showed an increased nerve density associated with the characte

Optic axons ignore denervated foreign territory in goldfish tectum.

Ultrastructural morphometry was used to test firstly, whether regenerating optic axons in goldfish tectum will form connections in adjacent denervated foreign territory and secondly, whether intact optic axons will collaterally sprout to innervate this territory. The stratum fibrosum marginale of the tectum was partially denervated by removing the torus longitudinalis and cutting the tectal commissure. Optic axons do not normally synapse in this layer. Thirty days after partial denervation and optic tract section the numbers of normal synapses in a micron 2 column through the stratum fibrosum

4 S RNA in regenerating optic axons of goldfish.

Previous experiments have demonstrated that 4 S RNA is transported axonally during the reconnection period of optic nerve regeneration in goldfish. The present experiments were performed to determine whether 4 S RNA is transported axonally during later, maturational stages of nerve regeneration and to examine some of the characteristics of 4 S RNA in regenerating axons. [3H]Uridine was injected into both eyes of fish 18 days (near the time of reconnection) after bilateral optic nerve crush and the fish were sacrificed at various times up to 180 days after injection. Noradrenergic axons hitch hiking along the human abducens nerve.

The abducens nerve (AN; cranial nerve VI) exits the brainstem at the inferior pontine sulcus, pierces the dura of the posterior cranial fossa, passes through the cavernous sinus in close contact to the internal carotid artery (ICA) and traverses the superior orbital fissure to reach the orbit to innervate the lateral rectus muscle. At its exit from the brainstem, the AN includes only axons from lower motor neurons in the abducens nucleus. However, as the AN crosses the ICA it receives a number of branches from the internal carotid sympathetic plexus. The arrangement, neurochemical profile and
