LYTIC CAPABILITY OF BACTERIOPHAGES (FAMILY MYOVIRIDAE) ON BURKHOLDERIA PSEUDOMALLEI.

Burkholderia pseudomallei, a gram-negative bacillus found in soil and water, is the causative agent of melioidosis. It can produce a biofilm, which increases resistance to antibacterial agents. Bacteriophages (phages) have been suggested as alternative antibacterial agents. In this study, the ability of six phages (family Myoviridae) to lyse B. pseudomallei isolates was examined using a microplate phage virulence assay. The six phages were more efficient in lysing soil than clinical B. pseudomallei isolates. Phage ST79 had the highest lytic capability, independent of inoculating phage quantity

Human antibodies against spores of the genus Bacillus: a model study for detection of and protection against anthrax and the bioterrorist threat.

A naive, human single-chain Fv (scFv) phage-display library was used in bio-panning against live, native spores of Bacillus subtilis IFO 3336 suspended in solution. A direct in vitro panning and enzyme-linked immunosorbent assay-based selection afforded a panel of nine scFv-phage clones of which two, 5B and 7E, were chosen for further study. These two clones differed in their relative specificity and affinity for spores of B. subtilis IFO 3336 vs. a panel of spores from 11 other Bacillus species/strains. A variety of enzyme-linked immunosorbent assay protocols indicated these scFv-phage clones

Molecular evolution of bacteriophages: evidence of selection against the recognition sites of host restriction enzymes.

Restriction enzymes produced by bacteria serve as a defense against invading bacteriophages, and so phages without other protection would be expected to undergo selection to eliminate recognition sites for these enzymes from their genomes. The observed frequencies of all restriction sites in the genomes of all completely sequenced DNA phages (T7, lambda, phi X174, G4, M13, f1, fd, and IKe) have been compared to expected frequencies derived from trinucleotide frequencies. Attention was focused on 6-base palindromes since they comprise the typical recognition sites for type II restriction enzyme

[Bacteriophages and bacteriocins of the genus Listeria].

Since the discovery of the first Listeria bacteriophage by Schultz in 1945, more than 219 phages were isolated for L. monocytogenes, L. invanovii, L. innocua, L. seeligeri and L. welshimeri. To date, no phage for L. murrayi and L. grayi is described in the literature. Unless two phages found by Jasińska in a water sample, all the other phages, when the origin is mentioned, were isolated from lysogenic strains, with or without induction. Electron microscopic examination of 55 Listeria phages revealed that two L. innocua phages belonged to the Myoviridae family (tail with contractile sheath) whe

In Vivo Studies on the Influence of Bacteriophage Preparations on the Autoimmune Inflammatory Process.

Phage preparations used for phage therapy may have not only direct antibacterial action but also immunomodulating effects mediated by phages themselves as well as by bacterial antigens. Therefore phage application in patients with immune disorders, and especially with autoimmune diseases, requires special attention. The aim of this study was to investigate the effect of phage lysates (staphylococcal phages A3/R, phi200, and MS-1 cocktail, enterococcal phage 15/P, Pseudomonas phage 119x, and E. coli T4 phage) as well as purified T4 phage on the course of murine collagen-induced arthritis (CIA),

Efficacy and durability of Bacillus anthracis bacteriophages used against spores.

Antibiotics and vaccines help fight anthrax disease, but there are no anthrax spore control methods suitable for use in environments where humans are present. The work reported in this article indicates that bacteriophages may help reduce risk from anthrax spores. Dose-response studies demonstrated that higher concentrations of mixed Bacillus anthracis bacteriophages (3.5 x 10(8) plaque-forming units per milliliter) inhibited subsequent growth of bacteria when sprayed on B. anthracis spores. Phages also were tested for durability under conditions designed to simulate environments possibly enco

[Treatment of late stages of anthracic infection].

Ways for increasing the anthrax treatment efficacy by combined use of antibiotics, toxin neutralizing, immunomodulating, symptomatic and pathogenetic agents are considered. The dynamics of the infection development was studied on rabbits by the microbiological, cytochemical, biochemical and cytomorphologic criteria. The efficacy of monoantibacterial and complex schemes of the etiopathogenetic therapy of the disease at the early and late stages (phases) of the infection generalization was estimated.

Polyphasic characterization of Bacillus species from anthrax outbreaks in animals from South Africa and Lesotho.

INTRODUCTION: Bacillus anthracis is the causative agent of anthrax, a disease endemic in regions of Northern Cape Province and Kruger National Park of South Africa. Accurate identification of virulent B. anthracis is essential but challenging due to its close relationship with other members of B. cereus group. This study characterized B. anthracis and Bacillus species that were recovered from animals and the environment where animals died of anthrax symptoms in southern Africa using a polyphasic approach. METHODOLOGY: For this purpose, 3 B. anthracis and 10 Bacillus isolates were subjected to

Polyvalent Phage CoNShP-3 as a Natural Antimicrobial Agent Showing Lytic and Antibiofilm Activities against Antibiotic-Resistant Coagulase-Negative Staphylococci Strains.

Synthetic antimicrobials have a negative impact on food quality and consumer health, which is why natural antimicrobials are urgently needed. Coagulase-negative staphylococci (CoNS) has gained considerable importance for food poisoning and infection in humans and animals, particularly in biofilms. As a result, this study was conducted to control the CoNS isolated from food samples in Egypt. CoNS isolates were selected on the basis of their antibiotic susceptibility profiles and their biofilm-associated behavior. In this context, a total of 29 different bacteriophages were isolated and, in part

Isolation and Characterization of a New Bacteriophage: Host Range Assessment and Prospects of Use in the Identification of Moraxella spp.

Bacteria of the genus Moraxella are significant pathogens in both human clinical practice and veterinary medicine. A novel bacteriophage, designated vB_MboM_ZALNahr, was isolated from environmental sources (wastewater from the Moscow Region) and morphologically classified as a myovirus lytically active against Moraxella spp. The phage exhibits a broad spectrum of lytic activity, affecting 76% of the tested Moraxella isolates of various origins. Genome sequencing revealed integrase genes, indicating the temperate nature of the phage and limiting its therapeutic potential. Phylogenetic analysis

Development of a high throughput assay for indirectly measuring phage growth using the OmniLog(TM) system.

The conventional and most accepted method of measuring the lytic activity of a phage against its bacterial host is the plaque assay. This method is laborious, time consuming and expensive, especially in high throughput analyses where multiple phage-bacterial interactions are required to be monitored simultaneously. It can also vary considerably with the experimenter and by the growth and plating conditions. Alternatively, the lytic activity can be measured indirectly by following the decrease in optical density of the bacterial cultures owing to lysis. Here we describe an automated, high throu

Evaluation of different methods to discriminate Bacillus anthracis from other bacteria of the Bacillus cereus group.

AIMS: To evaluate different methods that are useful for rapid and definitive discrimination of Bacillus anthracis from other bacteria of the Bacillus cereus group in environmental samples like letters claimed to contain anthrax spores. METHODS AND RESULTS: Characterized strains and bacteria from environmental samples were analysed by microbiological and molecular methods (PCR and restriction analysis). Environmental isolates often shared several microbiological features with B. anthracis, e.g. lack of beta-haemolysis and phospholipase C activity, and only the gamma phage assay was specific fo

Genome sequence and gene expression of Bacillus anthracis bacteriophage Fah.

Fah, a lytic bacteriophage of Bacillus anthracis, is used widely in the former Soviet Union to identify anthrax bacteria. Here, we present the analysis of a 37,974 bp sequence of the Fah genome and examine gene expression of the phage in a model host, Bacillus cereus. Half of the Fah genome contains genes coding for structural proteins and host lysis functions in an arrangement typical of Syphoviridae. The other half of the genome contains genes coding for enzymes of viral genome replication and for numerous predicted transcription factors that are likely to regulate viral gene expression. Pri

A heat-inducible Bacillus subtilis bacteriophage phi 105 expression system for the production of the protective antigen of Bacillus anthracis.

The protective antigen of Bacillus anthracis is the major protective immunogen in the current human vaccine. A heat-inducible protective antigen expression system was constructed based on a derivative of Bacillus subtilis phage phi 105. The recombinant protein produced by this system protected immunised animals against challenge with spores of B. anthracis. Gene instability and protease activity of the host strain contributed to the low level of recoverable protein in culture supernatant (approximately 2 mg l-1).

Antibacterial activity of single Klebsiella phage and phage cocktail against clinical carbapenem-resistant Klebsiella pneumoniae in vitro.

Klebsiella pneumoniae is a gram-negative, facultative anaerobic bacillus, that represents a significant global health concern due to its role as a major pathogen in multidrug-resistant infections. The growing prevalence of carbapenem-resistant K. pneumoniae infections among human necessitates the development of alternative and complementary therapies. Phages can effectively lyse drug-resistant pathogenic bacteria. In this study, stability tests confirmed that phage P560 was stable over a wide range of pH (3 to 10) and temperatures (4 to 50 °C) conditions. The growth characteristics showed that

Identification and biosynthesis of thymidine hypermodifications in the genomic DNA of widespread bacterial viruses.

Certain viruses of bacteria (bacteriophages) enzymatically hypermodify their DNA to protect their genetic material from host restriction endonuclease-mediated cleavage. Historically, it has been known that virion DNAs from the Delftia phage ΦW-14 and the Bacillus phage SP10 contain the hypermodified pyrimidines α-putrescinylthymidine and α-glutamylthymidine, respectively. These bases derive from the modification of 5-hydroxymethyl-2'-deoxyuridine (5-hmdU) in newly replicated phage DNA via a pyrophosphorylated intermediate. Like ΦW-14 and SP10, the Pseudomonas phage M6 and the Salmonella phage

[A short history of phage therapy].

From 1920 to 1940, phage therapy was extensively used to treat various infectious diseases. In 1915, Félix d'Herelle followed cases of bacillary dysentery in the Institut Pasteur hospital. He observed "clear spots" on bacteria culture. In 1917, d'Herelle presented a note to the "Académie des sciences de Paris" entitled: "on an invisible microbe, an antagonist of the dysentery bacillus". D'Herelle named this microbial agent bactériophage. In 1919, there was an epidemic of "fowl typhoid" in France. For d'Herelle, it was an opportunity to study phage behavior. He suggested that it was necessary t

Identification and characterization of clinical Bacillus spp. isolates phenotypically similar to Bacillus anthracis.

Bacillus anthracis, the etiological agent of anthrax, is a gram-positive, spore-forming rod, with colonies exhibiting a unique ground-glass appearance, and lacking hemolysis and motility. In addition to these phenotypes, several others traits are characteristic of B. anthracis such as susceptibility to gamma phage, the presence of two virulence plasmids (pX01 and pX02), and specific cell wall and capsular antigens that are commonly detected by direct fluorescent-antibody assays. We report on the identification and characterization of 14 Bacillus megaterium and four Bacillus sp. clinical isolat

Modulating phage susceptibility with membrane vesicles and bacteriophage cocktails for Acute Hepatopancreatic Necrosis Disease (AHPND) management in shrimp aquaculture.

Acute Hepatopancreatic Necrosis Disease (AHPND) is one of the most destructive bacterial diseases in shrimp aquaculture and is caused by virulent Vibrio parahaemolyticus strains carrying the pVA1 plasmid encoding the binary toxins PirA and PirB, resulting in severe economic losses and threatening sustainable production worldwide. Although lytic bacteriophages have emerged as promising alternatives to antibiotics because of their specificity and self-amplifying properties, their long-term effectiveness is constrained by the rapid evolution of bacterial resistance through multiple mechanisms, in
