Recent advances in the development of ultrasensitive micromechnical thermal detectors have led to the advent of novel subfemtojoule microcalorimetric spectoscopy (CalSpec). On the basis of principles of photothermal IR spectroscopy combined with efficient thermomechanical transduction, CalSpec provides acquisition of vibrational spectra of microscopic samples and absorbates. We use CalSpec as a method of identifying nanogram quantities of biological micro-organisms. Our studies focus on Bacillus subtilis and Bacillus cereus spores as simulants for Bacillus anthracis spores. Using CalSpec, we m

Anthrax is a worldwide zoonotic disease caused by the spore-forming bacterium Bacillus anthracis. Primarily a disease of herbivores, human infections often result from direct contact with contaminated animal products (cutaneous and inhalational anthrax) or through consumption of infected meat (gastrointestinal anthrax). The genetic near neighbor, Bacillus cereus biovar anthracis (Bcbva), causes an anthrax-like illness in the wildlife and livestock of west and central Africa due to the presence and expression of B. anthracis-specific virulence factors in this background. While Bcbva infections

Emerging strains of Bacillus cereus, traditionally considered a self-limiting foodborne pathogen, have been associated with anthrax-like disease in mammals, including humans. The strains have emerged by divergent evolution and, as exchange of genetic material in the Bacillus genus occurs naturally, it is possible that further isolates will be identified in the future. The strains vary in their genotypes and phenotypes, combining traits of both B. cereus and B. anthracis species. Cases of anthrax-like disease associated with these strains result in similar symptoms and mortality rates as those

Of the numerous infectious diseases afflicting humans, anthrax disease, caused by Bacillus anthracis, poses a major threat in its virulence and lack of effective treatment. The currently lacking standards of care, as well as the lengthy drug approval process, demonstrate the pressing demand for treatment for B. anthracis infections. The present study screened 1586 clinically approved drugs in an attempt to identify repurposable compounds against B. cereus, a relative strain that shares many physical and genetic characteristics with B. anthracis. Our study yielded five drugs that successfully i

A quantitative immunofluorescence assay based on fiber optic microscopy was used to measure the reaction of formalized spores of Bacillus anthracis and Bacillus cereus isolates with fluorescein conjugates prepared by hyperimmunization with B. anthracis Vollum spores. The spores of 11 of the 20 B. cereus strains reacted with the anti-anthrax conjugate to such an extent that they were indistinguishable from the spores of the several B. anthracis isolates tested. However, absorption of the conjugate with spores of B. cereus NCTC 8035 and B. cereus NCTC 10320 greatly reduced the cross-reaction wit

Bacillus anthracis, the aetiological agent of anthrax, has been taxonomically classified with the Bacillus cereus group, which comprises B. cereus, Bacillus thuringiensis, Bacillus mycoides, Bacillus pseudomycoides and Bacillus weihenstephanensis. Although the pathogenesis and ecological manifestations may be different, B. anthracis shares a high degree of DNA sequence similarity with its group member species. As a result, the discrimination of B. anthracis from its close relatives in the B. cereus group is still quite difficult. Suppression subtractive hybridization (SSH) was performed to sea

A 77-year-old man, initially diagnosed with pneumonia presented with fever and was administered continuous amino acid preparations and meropenem. Bacillus cereus was isolated from two separate sets of blood cultures, leading to a diagnosis of catheter-related bloodstream infection. Despite treatment with teicoplanin (TEIC) and additional antibiotics, B. cereus was continuously yielded from blood cultures for approximately four weeks. Ultimately, the patient was successfully treated with a combination of TEIC and clindamycin for six weeks, along with gentamicin for two weeks. This case report f

The toxin-producing bacterium Bacillus cereus is an important and neglected human pathogen and a common cause of food poisoning. Several toxins have been implicated in disease, including the pore-forming toxins hemolysin BL (HBL) and nonhemolytic enterotoxin (NHE). Recent work revealed that HBL binds to the mammalian surface receptors LITAF and CDIP1 and that both HBL and NHE induce potassium efflux and activate the NLRP3 inflammasome, leading to pyroptosis. These mammalian receptors, in part, contribute to inflammation and pathology. Other putative virulence factors of B. cereus include cytot

The ability to distinguish microbial pathogens from closely related but nonpathogenic strains is key to understanding the population biology of these organisms. In this regard, Bacillus anthracis, the bacterium that causes inhalational anthrax, is of interest because it is closely related and often difficult to distinguish from other members of the B. cereus group that can cause diverse diseases. We employed custom-designed resequencing arrays (RAs) based on the genome sequence of Bacillus anthracis to generate 422 kb of genomic sequence from a panel of 41 Bacillus cereus sensu lato strains. Bacillus cereus is a food-borne pathogen that causes a self-limiting gastroenteritis. We describe the case of a 72-year-old woman admitted to our hospital because of acute abdominal colic pain. Over a 2-day period, her clinical condition deteriorated rapidly, with the appearance of acute abdomen. Computed tomography investigation of the abdomen showed a liver abscess (diameter approximately 3 cm). At laparotomy, the abscess was found to be ruptured to the free peritoneal cavity. The final clinical diagnosis was acute peritonitis due to a ruptured liver abscess. Bacillus cereus was isolated fro

Seven emetic toxin-producing strains of Bacillus cereus were examined for toxin production in Skim Milk Medium at incubation temperatures ranging from 10 to 50 degrees C. Minimum and maximum growth temperatures were found to be 12 and 46 degrees C, respectively. At 12 and 15 degrees C, levels of toxin production were significantly higher (P < 0.01) than that observed at 30 degrees C, while no toxin was produced above 37 degrees C. Increased levels of sporulation were observed at increased temperatures, and no correlation was found between levels of sporulation and toxin production (R(2) = 0.08

PURPOSE: Bacillus cereus endophthalmitis occurring after penetrating ocular trauma has been almost always associated with a poor visual outcome. The purpose of our study was to review and report patients who had useful visual acuity outcomes.
METHODS: The study group consisted of five patients from a single medical center with penetrating ocular trauma and endophthalmitis caused by B. cereus. The study population was derived from a review of the microbiology records, clinical records, and operative reports of patients with culture-proven, post-traumatic endophthalmitis over a 15-year period.

Two cases of postoperative gas-gangrene-like infection due to Bacillus cereus are reported, drawing attention to the fact that Bacillus cereus, a common environmental bacterium, can occasionally give rise to severe post-operative infection. Characteristics of the organism related to the epidemiology and pathogenesis of such infections are discussed.

BACKGROUND: Cerebral infection with the opportunistic pathogen Bacillus cereus can lead to severe lesions, especially in premature newborns. Early diagnosis would be highly beneficial. The aim of this study was to describe the imaging findings associated with B. cereus infections.
MATERIALS AND METHODS: We included all newborns with a positive B. cereus blood culture and transfontanellar ultrasound between 2012 and 2022 at the Tours University Hospital. Imaging was assessed by both a junior and a senior radiologist to differentiate between hemorrhages, leukomalacia, and lesions attributed to

BACKGROUND: Bacillus cereus is a Gram-positive bacterium that can be found in various natural and human-made environments. It is often involved in gastrointestinal infections and food poisoning; yet, it can rarely cause serious non-gastrointestinal tract infections.
CASE PRESENTATION: Here we describe a case of B. cereus cutaneous infection of a wound on the hand of a young woman from a rural area in Iran. On admission, she had no systemic symptoms other than a cutaneous lesion. The identification of the causative agent was performed using sequencing of the 16S rRNA gene of the bacteria isola

Emetic Bacillus cereus strains produce a potent cereulide cytotoxin, which can cause acute and fatal cases of food poisoning. We isolated 18 emetic B. cereus strains from a food poisoning event, and from clinical and non-random food surveillance in China and phenotypic characteristics of haemolysis, starch hydrolysis, salicin fermentation, gelatin liquefaction, cytotoxicity, and susceptibility to antibiotics were assessed. All isolates were positive for haemolysis and gelatin liquefaction, and negative for starch hydrolysis and salicin fermentation. Their haemolytic potentials were intermediat

BACKGROUND: Bacillus cereus sensu lato (s. l.) is an ecologically diverse bacterial group of medical and agricultural significance. In this study, I use publicly available genomes and novel bioinformatic workflows to characterize the B. cereus s. l. pan-genome and perform the largest phylogenetic and population genetic analyses of this group to date in terms of the number of genes and taxa included. With these fundamental data in hand, I identify genes associated with particular phenotypic traits (i.e., "pan-GWAS" analysis), and quantify the degree to which taxa sharing common attributes are p
