Size determination of bacterial capsular oligosaccharides used to prepare conjugate vaccines.

We recently described the use of ion exchange chromatography for analysis and the industrial scale preparation of pools of oligosaccharides of intermediate chain length from polysaccharides of Haemophilus influenzae type b (Hib) and Neisseria meningitidis groups A and C. These negatively charged "sized" oligosaccharides are activated and conjugated to the carrier protein (CRM197) to prepare the corresponding glycoconjugate vaccines. Characterization and accurate determination of the degree of polymerization (DP) of the pool of oligosaccharides is essential for the consistent production of thes

Peptide vaccines: an innovative therapeutic approach against antibiotic-resistant bacterial infections.

Bacterial infections continue to pose a serious threat to global health, especially with the growing challenge of multidrug-resistant pathogens. While traditional vaccines have been pivotal in reducing disease burden, they come with limitations such as variable efficacy, safety concerns, and limited ability to address the diversity of bacterial strains. This review highlights the promise of peptide-based vaccines as an innovative approach to overcoming these hurdles. By targeting specific regions of bacterial proteins, peptide vaccines can elicit precise immune responses with improved safety a

Role of O-Acetylation in the Immunogenicity of Bacterial Polysaccharide Vaccines.

The incidence of infectious diseases caused by several bacterial pathogens such as Haemophilus influenzae type b, Streptococcus pneumoniae, and Neisseria meningitidis, has been dramatically reduced over the last 25 years through the use of glycoconjugate vaccines. The structures of the bacterial capsular polysaccharide (CPS) antigens, extracted and purified from microbial cultures and obtained with very high purity, show that many of them are decorated by O-acetyl groups. While these groups are often considered important for the structural identity of the polysaccharides, they play a major rol

The Unexpected Impact of Vaccines on Secondary Bacterial Infections Following Influenza.

Influenza virus infections remain a significant health burden worldwide, despite available vaccines. Factors that contribute to this include a lack of broad coverage by current vaccines and continual emergence of novel virus strains. Further complicating matters, when influenza viruses infect a host, severe infections can develop when bacterial pathogens invade. Secondary bacterial infections (SBIs) contribute to a significant proportion of influenza-related mortality, with Streptococcus pneumoniae, Staphylococcus aureus, Streptococcus pyogenes, and Haemophilus influenzae as major coinfecting

Antigenicity testing of inactivated oral enteric bacterial vaccines for human use.

This study deals with the laboratory testing of the antigenicity of inactivated oral enteric bacterial vaccines for human use, viz. typhoid, dysentery, Escherichia coli and cholera vaccines. For the assays the following methods are applied : 1) various types of active mouse protection tests 2) serum and coproantibody determinations in immunized persons and experimental animals, 3) protective ability of the immune sera in mice and embryonated eggs, 4) hemolytic and bacteriolytic plaque assays. It is pointed out that the most important question: how far the results of these tests are in agreemen

Vaccines for viral and bacterial pathogens causing acute gastroenteritis: Part I: Overview, vaccines for enteric viruses and Vibrio cholerae.

Efforts to develop vaccines for prevention of acute diarrhea have been going on for more than 40 y with partial success. The myriad of pathogens, more than 20, that have been identified as a cause of acute diarrhea throughout the years pose a significant challenge for selecting and further developing the most relevant vaccine candidates. Based on pathogen distribution as identified in epidemiological studies performed mostly in low-resource countries, rotavirus, Cryptosporidium, Shigella, diarrheogenic E. coli and V. cholerae are predominant, and thus the main targets for vaccine development a

The return of the mixed respiratory bacterial vaccine.

Background: Orally or sublingually administered lysates of mixed respiratory pathogenic bacteria have been used in Europe, Asia, and other parts of the world, but not the United States, since the mid 1950s, first to prevent recurrent respiratory tract infections, later to prevent wheezing and asthma exacerbations associated with respiratory infections, and, more recently, for the treatment of allergic rhinitis. The apparent success of this treatment contrasts with the negative experience of treating with injections of similar mixed respiratory bacterial vaccines (MRBV or BV) to prevent asthma

Recombinant bacterial lipoproteins as vaccine candidates.

Recombinant bacterial lipoproteins (RLP) with built-in immuno-stimulating properties for novel subunit vaccine development are reviewed. This platform technology offers the following advantages: easily converts antigens into highly immunogenic RLP using a fusion sequence containing lipobox; the lipid moiety of RLP is recognized as the danger signals in the immune system through the Toll-like receptor 2, so both innate and adaptive immune responses can be induced by RLP; serves as an efficient and cost-effective bioprocess for producing RLP in Escherichia coli and the feasibility and safety of

Vaccine Delivery with a Detoxified Bacterial Toxin.

It is still a challenge to develop needle-free mucosal vaccines. Despite progress in the development of the influenza vaccine, it must be reformulated annually because of antigenic changes in circulating influenza viral strains. Due to seasonal drift and shift of circulating strains, the influenza vaccine does not always match the circulating strains, and included adjuvants are not sufficient to induce a protective effect with long-lived memory cells. The adjuvants play a major role in the immune responses to a vaccine. Interestingly, the Bacillus anthracis detoxified anthrax edema toxin, whic

Bacterial enteric infections and vaccine development.

There is a great need for effective vaccines against the major bacterial enteropathogens. Bacterial enteric infections resulting in diarrhea, dysentery, or enteric fever constitute a huge public health problem, with more than a billion episodes of disease and several million deaths annually in developing countries. Diarrhea caused by a bacterial enteric infection is also the commonest illness experienced by international travellers. Studies of pathogenesis have established the importance of specific ligand-receptor interactions between the enteropathogens and the intestinal epithelium, resulti

Coated Bacterial Vaccine: A New Approach for Antigen Display on Bacterial Surface.

Bacterial vaccines using recombinant antigens displayed on the bacterial surface represent a major advancement in vaccine development. This approach leverages bacteria's natural ability to induce immune responses while improving vaccine targeting and efficacy. Existing platforms, such as live bacterial vaccines, inactivated bacterial vaccines, and bacterial-like particles (BLPs), face challenges like genetic modifications, antigen dilution, and degradation. To address these limitations, a novel system called coated bacterial vaccine (CBV) is proposed, combining the properties of inactivated ba

Pulmonary mRNA-LNP vaccines for rapid and durable protection against bacterial infection.

Pulmonary bacterial infections remain a major clinical challenge. Although vaccination reduces infection rates and mortality, the vulnerable post-vaccination immunity gap can still result in infection and vaccine failure. In addition, effective vaccines are unavailable for many clinically important bacterial pathogens. Here, we report a pulmonary mRNA-lipid nanoparticle (mRNA-LNP) vaccine incorporating an ionizable lipid engineered for localized high-level expression, which elicits both rapid and durable protection against bacterial lung infections in female mice, effectively bridging this cri
