An in vitro binding system is described to display large full-length proteins on bacteriophage T4 capsid surface at high density. The phage T4 icosahedral capsid features 155 copies of a nonessential highly antigenic outer capsid protein, Hoc, at the center of each major capsid protein hexon. Gene fusions were engineered to express the 83-kDa protective antigen (PA) from Bacillus anthracis fused to the N-terminus of Hoc and the 130-kDa PA-Hoc protein was expressed in Escherichia coli and purified. The purified PA-Hoc was assembled in vitro on hoc(-) phage particles. Binding was specific, stabl

Endonuclease V (denV) from bacteriophage T4 was examined for its ability to complement the DNA repair defect in xeroderma pigmentosum (XP) cells from complementation groups A, C, D, F and G. The denV gene was introduced into SV40-transformed normal and XP cells using a retroviral vector. Expression of denV resulted in partial correction of UV sensitivity and increased host cell reactivation (HCR) of a UV-damaged reporter gene for XP cells from groups A, C and D, but not those from group G. Expression of denV in XP-F cells resulted in enhanced HCR of a UV-damaged reporter but did not affect UV

Bacteriophage T4 capsid is a prolate icosahedron composed of the major capsid protein gp23*, the vertex protein gp24*, and the portal protein gp20. Assembled on its surface are 810 molecules of the non-essential small outer capsid protein, Soc (10 kDa), and 155 molecules of the highly antigenic outer capsid protein, Hoc (39 kDa). In this study Soc, a "triplex" protein that stabilizes T4 capsid, is targeted for molecular engineering of T4 particle surface. Using a defined in vitro assembly system, anthrax toxins, protective antigen, lethal factor and their domains, fused to Soc were efficiently

The use of bacteriophages to treat bacterial infections, known as phage therapy, has regained interest due to the rise of antibiotic-resistant bacteria. To make phage therapy more widely available, scalable purification methods that can adequately remove endotoxins, proteins and host cell DNA must be implemented. This is particularly important when considering intravenous (IV) administration, since the presence of these impurities is highly controlled by regulatory agencies. This work aimed at developing a purification workflow amenable to large-scale manufacturing, centred on the use anion-ex

Bioconjugation of polymers to proteins is a method to impart improved stability and pharmacokinetic properties to biologic systems. However, the precise effects of polymer architecture on the resulting bioconjugates are not well understood. Particularly, cyclic polymers are known to possess unique features such as a decreased hydrodynamic radius when compared to their linear counterparts of the same molecular weight, but have not yet been studied. Here, we report the first bioconjugation of a cyclic polymer, poly(ethylene glycol) (PEG), to a model protein, T4 lysozyme, containing a single engi

A rapid, efficient technique for the attachment of bacteriophages (phages) onto polyhydroxyalkanoate (PHA) surfaces has been developed and compared to three reported methods for phage immobilization. Polymer surfaces were modified to facilitate phage attachment using (1) plasma treatment alone, (2) plasma treatment followed by activation by 1-ethyl-3-(3-(dimethylamino)propyl)carbodiimide hydrochloride (EDC) and N-hydroxysulfosuccinimide (sulfo-NHS), (3) plasma-initiated acrylic acid grafting, or (4) plasma-initiated acrylic acid grafting with activation by EDC and sulfo-NHS. The impact of each

AIMS: Molecular methods have shown bacteriophage diversity in human milk, but this does not reflect viability, particularly within donor human milk which has been subjected to multiple freeze-thaw cycles and heat treatment. Our aim was to isolate bacteriophages from frozen human milk and explore the effects of sterilization and storage on bacteriophage survival.
METHODS AND RESULTS: Twenty frozen human milk samples were chosen for bacteriophages isolation. Analysis showed that these samples stored for 12 months at -20°C did not contain viable bacteriophages. We artificially inoculated 10 huma

Environmental factors play a critical role in viral persistence, yet the combined effects of temperature and surface chemistry on viral fitness are not fully understood. In this study, we examined bacteriophages T4 and φ X174 across copper, stainless steel, and polypropylene plastic surfaces at 4°C and 37°C to assess how these variables influence survival and growth. Both phages exhibited marked differences in survival across surfaces: Copper at 37°C caused rapid inactivation and cooler conditions promoted persistence. Growth varied independently of survival, indicating that viral stability a

E217 is a Pseudomonas phage used in an experimental cocktail to eradicate cystic fibrosis-associated Pseudomonas aeruginosa. Here, we describe the structure of the whole E217 virion before and after DNA ejection at 3.1 Å and 4.5 Å resolution, respectively, determined using cryogenic electron microscopy (cryo-EM). We identify and build de novo structures for 19 unique E217 gene products, resolve the tail genome-ejection machine in both extended and contracted states, and decipher the complete architecture of the baseplate formed by 66 polypeptide chains. We also determine that E217 recognizes t

Shigellosis, caused by Shigella bacterial spp., is one of the leading causes of diarrheal morbidity and mortality. An increasing prevalence of multidrug-resistant Shigella species has revived the importance of bacteriophages as an alternative therapy to antibiotics. In this study, a novel bacteriophage, Sfk20, has been isolated from water bodies of a diarrheal outbreak area in Kolkata (India) with lytic activity against many Shigella spp. Phage Sfk20 showed a latent period of 20 min and a large burst size of 123 pfu per infected cell in a one-step growth analysis. Enzymes, key catalysts in biochemical reactions, are prone to denaturation under harsh conditions, leading to reduced stability and higher costs. Enzyme immobilization, using carriers like magnetic nanoparticles, metal-organic frameworks, and viruses, is a common solution. T4 bacteriophage, a virulent E. coli phage containing 155 Hoc and 870 Soc proteins, offers a cost-effective and highly stable platform for enzyme immobilization. In this study, Soc-β-galactosidase (Soc-β-gal) was immobilized on the surface of T4 bacteriophage via affinity fixation and further encapsulated with a metal-polyph

Enzyme immobilization is pivotal for industrial biocatalysis yet often suffers from activity loss, structural distortion, operational instability, and high production costs. This study presents a magnetic bead-T4 capsid biocarrier (MBTCB) platform for efficient immobilization of an engineered tryptophan synthase Pf0A9 to enable sustainable synthesis of high-value tryptophan analogs. Soc-fused Pf0A9 was immobilized onto T4 phage capsids via high-affinity Soc-capsid binding under mild physiological conditions, preserving native enzyme conformation, and combining with magnetic bead conjugation fo

The coil-globule transition is a key phenomenon in polymer physics, where polymer chains shift between collapsed and extended states. This study investigates the relaxation dynamics of T4 DNA transitioning from an electrohydrodynamically compressed globule to a relaxed coil state in polyethylene glycol (PEG) solutions of varying molecular weights. Using an AC electric field to induce globule formation, we analyzed DNA expansion upon field cessation. Higher molecular weight PEG, particularly PEG 20000, markedly prolonged relaxation times and exhibited broader distributions compared to PEG 4000

Mammals have become "an environment" for enterobacterial phage life cycles. Therefore it could be expected that bacteriophages adapt to them. This adaptation must comprise bacteriophage proteins. Gp Hoc seems to have significance neither for phage particle structure nor for phage antibacterial activity. It is evidently not necessary for the "typical" antibacterial actions of bacteriophages. But the rules of evolution make it improbable that gp Hoc really has no function, and non-essential genes of T4-type phages are probably important for phages' adaptation to their particular lifestyle. A bacteriophage T4 DNA "synapsis model" proposes that the bacteriophage T4 terminase small subunit (TerS) apposes two pac site containing dsDNA homologs to gauge concatemer maturation adequate for packaging initiation. N-terminus, C-terminus, or both ends modified fusion Ter S proteins retain function. Replacements of the TerS gene in the T4 genome with fusion genes encoding larger (18-45 kDa) TerS-eGFP and TerS-mCherry fluorescent fusion proteins function without significant change in phenotype. Co-infection and co-expression by T4 phages encoding TerS-eGFP and TerS-mCherry shows in vivo FRET

Generating mechanistic pathways of rare events like ligand unbinding or protein folding is a central challenge in molecular dynamics simulation of complex molecular processes. Here, we present PathGennie, a direction-guided adaptive sampling framework that rapidly generates transition pathways without external biasing forces or thermal perturbation. The method launches swarms of ultrashort (a few femtoseconds long), unbiased trajectories and selectively propagates only those that exhibit progress toward a defined goal in a possibly high-dimensional collective variable space, thus circumventing

Porcine delta coronavirus (PDCoV) poses a significant threat to the swine industry. Thus, the development of innovative vaccine candidates is critical for PDCoV prevention. This study details the creation of a PDCoV nanoparticle vaccine utilizing bacteriophage (phage) T4 as a delivery platform. B cell and T cell epitopes of the PDCoV spike (S) protein were identified through bioinformatics and assembled into a tandem construct (termed Pep) using a linker. In silico molecular docking revealed stable interactions between Pep and TLR3. Immune stimulation predictions indicated that Pep could trigg

Bacteriophage T4 and other bacteriophages have a protein component known as a molecular needle which is used for the transmembrane reaction in the infection process. In this paper, the transmembrane reaction mechanisms of artificial protein needles (PNs) constructed by protein engineering of the component protein of bacteriophage T4 are elucidated by observation of single-molecules by high-speed atomic force microscopy (HS-AFM) and molecular dynamics (MD) simulations. The HS-AFM images indicate that the tip of the needle structure stabilizes the interaction of the needle with the membrane surf

Many studies report that copper can be used to control microbial growth, including that of viruses. We determined the rates of copper-mediated inactivation for a wide range of bacteriophages. We used two methods to test the effect of copper on bacteriophage survival. One method involved placing small volumes of bacteriophage lysate on copper and stainless steel coupons. Following exposure, metal coupons were rinsed with lysogeny broth, and the resulting fluid was serially diluted and plated on agar with the corresponding bacterial host. The second method involved adding copper sulfate (CuSO(4)
