Gut microbiota composition and tumor immune features in meningioma patients.

UNLABELLED: Meningiomas are prevalent intracranial tumors with poorly understood extraneural drivers. While the gut-brain axis influences neuro-oncogenesis, meningioma-specific gut microbiome alterations and their clinical implications remain uncharacterized. This study integrated 16S rRNA sequencing of fecal samples from 15 treatment-naïve WHO grade I meningioma patients (MPs) and 15 healthy controls (HCs) with immunohistochemical profiling of tumor immune infiltrates (MPO⁺ neutrophils, CD68⁺ macrophages, CD3⁺ T cells). Compared with HCs, MPs exhibited significantly reduced alpha diversity (S

Current research progress on Prevotella intermedia and associated diseases.

Prevotella intermedia is a Gram-negative anaerobic bacterium that is a common pathogen of periodontitis. Recent studies have revealed that P. intermedia is closely associated with a variety of diseases involving multiple systems. Under the action of its virulence factors such as cysteine protease and adhesins, P. intermedia has the ability to bind and invade various host cells including gingival fibroblasts. It can also copolymerize a variety of pathogenic bacteria, leading to interference with the host's immune inflammatory response and causing various diseases. In this article, we review the

Leptotrichia buccalis bacteremia in neutropenic children.

Two new cases of Leptotrichia buccalis bacteremia in seriously ill patients were described. The anaerobic, gram-negative microorganism L. buccalis was isolated from blood cultures of two children with severe leukopenia. Anaerobic organisms should be taken into account when a standard protective antibiotic chemotherapy is considered in the immunosuppressed host.

Heme acquisition mechanisms of Porphyromonas gingivalis - strategies used in a polymicrobial community in a heme-limited host environment.

Porphyromonas gingivalis, a main etiologic agent and key pathogen responsible for initiation and progression of chronic periodontitis requires heme as a source of iron and protoporphyrin IX for its survival and the ability to establish an infection. Porphyromonas gingivalis is able to accumulate a defensive cell-surface heme-containing pigment in the form of μ-oxo bisheme. The main sources of heme for P. gingivalis in vivo are hemoproteins present in saliva, gingival crevicular fluid, and erythrocytes. To acquire heme, P. gingivalis uses several mechanisms. Among them, the best characterized a

Individuals at risk for rheumatoid arthritis harbor differential intestinal bacteriophage communities with distinct metabolic potential.

Rheumatoid arthritis (RA) is an autoimmune disease characterized in seropositive individuals by the presence of anti-cyclic citrullinated protein (CCP) antibodies. RA is linked to the intestinal microbiota, yet the association of microbes with CCP serology and their contribution to RA is unclear. We describe intestinal phage communities of individuals at risk for developing RA, with or without anti-CCP antibodies, whose first-degree relatives have been diagnosed with RA. We show that at-risk individuals harbor intestinal phage compositions that diverge based on CCP serology, are dominated by S

Yeast cell wall product enhanced intestinal IgA response and changed cecum microflora species after oral vaccination in chickens.

The study was designed to explore the effect of a commercial yeast cell wall product (YP) on chicken intestinal IgA response and cecum microbiome after oral vaccination. Chickens were fed with YP during the experiments and orally immunized with live Newcastle disease virus (NDV) vaccine at 2 wk of age. Then, the animals were sacrificed, and samples were collected to measure the indicators of hemagglutination inhibition (HI), IgA response, IgA + cells, and cecum microbiome populations. The results showed that supplement of YP significantly enhanced serum NDV HI titer, intestinal NDV-specific se

Modulating effects of polysaccharides from the fruits of Lycium barbarum on the immune response and gut microbiota in cyclophosphamide-treated mice.

In the present study, the effects of Lycium barbarum polysaccharides (LBPS) on immunoregulation and gut microbiota dysbiosis in cyclophosphamide (CTX)-induced mice were investigated to elucidate whether the attenuation of immunosuppression is related to the modulation of the gut microbiota. The results showed that administration of LBPS could protect immune organs (enhancing immune organ indexes and alleviating immune organ damage), enhance the production of immune-related cytokines (IL-2, IL-6, IL-1β, TNF-α and IFN-γ) and prevent the hepatotoxicity in CTX-induced mice. Additionally, LBPS trea

Polytrauma independent of therapeutic intervention alters the gastrointestinal microbiome.

BACKGROUND: This study characterizes the gastrointestinal (GI) microbiome in a pre-clinical polytrauma hemorrhage model. METHODS: Rats (n = 6) were anesthetized, hemorrhaged 20% of their blood volume, and subjected to a femur fracture and crush injuries to the small intestine, liver, and limb skeletal muscle without resuscitation. Fecal samples were collected pre-injury and 2 h post-injury. Purified DNA from the samples underwent 16s rRNA sequencing for microbial quantification. Bacterial diversity analysis and taxonomic classification were performed. RESULTS: Following injury, the gut micro

Microbiome and resistome of the European bison (Bison bonasus).

After facing extinction in the early 20th century, populations of the two remnant genetic lines of European bison are now under continuous health monitoring. Faecal samples were taken from five Polish and one German herd of European bison over the course of several years. Through metagenomic sequencing, the bacterial and archaeal microbiome as well as the resistome of these samples could be characterized. Significant differences were mainly found between the bacterial microbiome of samples taken from droppings as opposed to rectal samples. Apart from this, the microbiome and resistome had low

Effects of Differences in Resistant Starch Content of Rice on Intestinal Microbial Composition.

The aim of this study was to evaluate the effects of resistant starch (RS) and fat levels on the gut microbiome in C57BL/6 mice. Three levels of RS from three varieties of rice were the major source of carbohydrates and fat levels were low (10%) and high (39%). We confirmed that RS decreased the Firmicutes to Bacteroidetes ratio, increased SCFA production by higher Bacteroidaceae and S24-7 abundance, and enriched predicted gene families of glycosidases and functional pathways associated with carbohydrate and glycan metabolism. We also found correlations between microbial taxa and tissue gene e

Lentinula edodes-derived exosome-like nanoparticles improve curcumin stability and biotransformation via gut microbiota interactions.

Exosome-like nanoparticles (ELNs) from edible sources are emerging as natural carriers for stabilizing bioactive compounds. This study investigated Lentinula edodes-derived ELNs (LeELNs) as a delivery system for curcumin. LeELNs efficiently encapsulated curcumin (69.25 %) into stable vesicles <200 nm and significantly improved its stability during simulated gastrointestinal digestion. In vivo, Le-Cur exhibited prolonged intestinal retention and elevated levels of hydrogenated metabolites in the small intestine, liver, and plasma compared with free curcumin. To elucidate this enhancement, PKH26

Co-exposure to different bacterial species' lipopolysaccharides with the NASH diet exacerbates NASH and liver fibrosis progression in mice.

BACKGROUND AND AIM: With the obesity epidemic, nonalcoholic fatty liver disease (NAFLD) has become a public health concern, but its progression mechanism remains unclear. Experimental models mimicking human NAFLD/steatohepatitis (NASH) are crucial. This study simulates gut microbiota imbalance effects on NASH and liver fibrosis. METHODS: We used different bacterial sources of lipopolysaccharide (LPS), including Escherichia coli (GEC) and Salmonella abortus equi (GSE), combined with a Gubra Amylin NASH (GAN) diet to induce NASH and liver fibrosis. RESULTS: The GSE group showed significantly h

Porphyromonas gingivalis as a keystone pathogen in systemic diseases.

Porphyromonas gingivalis (P. gingivalis) is a Gram-negative anaerobic bacterium recognized as a keystone pathogen in periodontitis. Beyond its established role in periodontal tissue destruction, accumulating evidence links P. gingivalis to several systemic conditions, including cardiovascular disease, Alzheimer's disease, rheumatoid arthritis, diabetes mellitus, and adverse pregnancy outcomes. This review synthesizes current evidence on how P. gingivalis may contribute to systemic disease beyond the oral cavity, with particular emphasis on its virulence factors, including gingipains and lipopo

Distribution of luxS and production of autoinducer-2 among gut Bacteroidales.

UNLABELLED: One of the best studied quorum sensing (QS) molecules, autoinducer-2 (AI-2), regulates processes in numerous bacteria. LuxS is an enzyme of the activated methyl cycle which, along with MtnN, converts S -adenosyl-homocysteine to homocysteine, releasing DPD (4,5-dihydroxy-2,3-pentanedione) which is spontaneously converted to AI-2. Many bacteria do not encode MtnN-LuxS and instead encode SahH, which directly converts S -adenosyl-homocysteine to homocysteine without AI-2 production. The genomes of some gut Bacteroidales were shown to contain luxS , however, these reports, as well as re

The Forgotten One: Lemierre's Syndrome Due to Gram-Negative Rods Prevotella Bacteremia.

BACKGROUND: Lemierre's syndrome (LS) is a rare syndrome caused by an acute oropharyngeal infection with metastatic spreading. It was described in 1939 as jugular vein septic thrombophlebitis associated with retropharyngeal infection. Different organisms can cause LS, such as Fusobacterium species, Peptostreptococcus, group B and C, Streptococcus, Staphylococcus, and Enterococcus species, but the most commonly isolated pathogen is Fusobacterium necrophorum, a common oral flora. Management depends on the initial presentation, type of pathogen isolated, and proper selection of antibiotics. CASE

Genomic insights into activated antimicrobial resistance of in situ hospital-wastewater biofilm.

Antimicrobial resistance (AMR), particularly among carbapenemase-producing organisms, poses a major global health threat. Although hospital wastewater is considered an AMR hotspot, its functional contribution to resistance dynamics remains poorly defined. We developed in situ biofilms in hospital wastewater and applied integrated metagenomic, metatranscriptomic, and culture-based analyses to characterize community structure and gene expression. Biofilms exhibited greater biomass and higher contamination with extended-spectrum β-lactamase-producing Escherichia coli than planktonic wastewater. B

Distinct Evolutionary Selection Patterns in Temperate and Filamentous Phages of Phocaeicola vulgatus in Health and IBD.

The gut virome is profoundly altered in inflammatory bowel disease (IBD), though phage evolution during inflammation remains poorly understood. We used viral tagging (VT) to recover phages of Phocaeicola vulgatus , a gut-associated pathobiont, from healthy and Crohn's disease (CD) viromes, revealing phages related to the tailed phage BV01, and the first filamentous phage (PvIno1) of any Bacteroidaceae host. BV01-like phages accumulated point mutations in tail genes during health, diversity which was lost in CD, while the configuration of a tail-fiber shufflon varied with disease state. In PvIn
