In vitro pharmacodynamic studies investigating the antimicrobial properties of five fluoroquinolones, (trovafloxacin, sparfloxacin, clinafloxacin, levofloxacin, and ciprofloxacin) against Bacteroides fragilis ATCC 23745 were conducted. The times required to reduce the viable counts by 3 log units were as follows: clinafloxacin, 2.9 h; levofloxacin, 4.6 h; trovafloxacin, 6 h; and sparfloxacin, 10 h. Exposure to ciprofloxacin did not achieve a 3-log decrease in viable counts. The susceptibility of B. fragilis was determined both prior to exposure and following 24 h of exposure to each of the fiv

Fibrin deposition in the peritoneal cavity during acute peritonitis appears to predispose the host to abscess formation by providing an environment for bacterial proliferation protected from host defenses. The purpose of the present study was to determine whether the potent abscess-inducing anaerobe Bacteroides fragilis could promote fibrin deposition by inducing mononuclear cells to express procoagulant activity (PCA). B. fragilis stimulated PCA in a dose-dependent fashion, achieving a maximum at 10(7) CFU/ml. Heat-killed B. fragilis induced comparable levels of PCA, while a nonspecific phago

Bacteroidota are prolific members of the human gut microbiota, influencing overall health through the degradation of various polysaccharides. To aid in this process, these bacteria deploy cell surface lipoproteins, proteins anchored to the membrane by a lipidated N-terminal cysteine. Despite their importance, lipoprotein synthesis and transport in Bacteroidota is not well defined, particularly how lipoproteins reach the outer membrane and cell surface. In Escherichia coli, lipoproteins are inserted into the outer membrane by LolB, the final step of the Lol pathway for localizing lipoproteins,

Bacterial pyomyositis is characterized by the formation of abscesses in skeletal muscle groups, frequently in the lower extremities. It is most prevalent in tropical climates and associated with Staphylococcus aureus. The presentation of pyomyositis in the neck is rare. We present a unique case of pyomyositis caused by a gut bacterium, Bacteroides fragilis. Additionally, the case highlights the difficulty in identifying the cause of spread of the bacteria from the gut to the affected musculature. A 77-year-old diabetic Caucasian male presented with progressive bilateral neck swelling and limit

Lipopolysaccharides (LPS) were extracted from three Bacteroides nodosus isolates by the aqueous phenol method and purified by ultracentrifugation. The structure of B nodosus LPS appears to be similar to enterobacterial LPS, with the polysaccharide component joined to the lipid A moiety through an acid-labile 2-keto-3-deoxyoctonic acid (KDO) linkage. B nodosus LPS contained glucose, galactose, hexosamine, KDO and heptose and no sugars apart from ribose, which was possibly a nucleic acid contaminant, were unique to any of the isolates. Electron microscopic examination indicated a similar morphol

The name Bacteroides is used to describe a group of Gram-negative bacilli which are non-sporing obligate anaerobes (Wilson and Miles, 1964). Their natural habitat is the large intestine, mouth, and vagina. Bacteroides septicaemia is becoming increasingly well recognized as a complication of gastrointestinal and gynaecological surgery. Patients with underlying malignancy or prior antibiotic therapy are predisposed to this condition (Bodner, Koenig and Goodman, 1970). In a survey by Okubadejo, Green and Payne (1973), twenty-nine strains of B. fragilis were tested against co-trimoxazole—all were

OBJECTIVES: Enterotoxigenic Bacteroides fragilis (ETBF) is a subspecies of B. fragilis that is considered to contribute to colorectal carcinogenesis through the activity of the B. fragilis toxin (BFT). However, colonic carriage of ETBF is also evident in a subset of asymptomatic individuals. In vitro studies using the HT29 cell line have linked BFT to the disruption of cell-cell adhesion and inflammation. This cell line, however, fails to consider the role of the mucus barrier in protecting the colonic epithelium in healthy individuals. This study aimed to investigate the role of a cell-associ

Acute pyogenic vertebral osteomyelitis is mainly attributed to haematogenous spread of aerobic bacteria, while anaerobic osteomyelitis results from contiguous spread of polymicrobial infections through breaks in the gut mucosal barrier and involves the vertebral bodies in only about 2%-5%. Herein, we report two cases of vertebral osteomyelitis due to Bacteroides fragilis. It is noteworthy that cases of vertebral osteomyelitis due to Bacteroides fragilis have been attributed to the extension of intra-abdominal or pelvic floor infections. However, in the two cases described, there was no history

During short-lived perturbations, such as inflammation, the gut microbiota exhibits resilience and reverts to its original configuration. Although microbial access to the micronutrient iron is decreased during colitis, pathogens can scavenge iron by using siderophores. How commensal bacteria acquire iron during gut inflammation is incompletely understood. Curiously, the human commensal Bacteroides thetaiotaomicron does not produce siderophores but grows under iron-limiting conditions using enterobacterial siderophores. Using RNA-seq, we identify B. thetaiotaomicron genes that were upregulated

Commensal symbionts may become pathogens upon escaping their habitat. In the gut, Bacteroides fragilis protects against colitis through induction of interleukin 10 (IL-10) by CD4(+) T cells. When intestinal integrity is disrupted, B. fragilis and colonic contents escape into the peritoneum, causing abscesses and bacteremia. Whether the virulence mechanisms employed by B. fragilis during infections differ from those employed for symbiosis during commensalism is unknown. We demonstrate T cell-independent IL-10 production in response to B. fragilis during its pathogenic interactions with the host

Bacteroides spp., particularly B. fragilis, are well-recognised bacterial pathogens. Production of the typical beta-lactamases of Bacteroides restricts the therapeutic use of beta-lactam agents mainly to the beta-lactamase inhibitor combinations and carbapenems. These compounds have the advantage of broad-spectrum activity and the ability to combat polymicrobial infections. Resistance of Bacteroides spp. to beta-lactam antibiotics appears to be increasing, largely because of an overall increase in beta-lactamase activity. There has been a rise in the prevalence of isolates showing high-level p

Porphyromonas endodontalis (formerly Bacteroides endodontalis) is a black-pigmented anaerobic Gram-negative rod which is associated with endodontal infections. It has been isolated from infected dental root canals and submucous abscesses of endodontal origin. The presence of P. endodontalis in infected dental root canals has been correlated with symptoms of an acute infection. It is occasionally found on oral mucous membranes and periodontal pockets. P. endodontalis has shown relatively low virulence in experimental monoinfections. In anaerobic mixed infections it can play an essential role. D

The effects of immunization in modulating the pathogenesis of Bacteroides (Porphyromonas) gingivalis infection in a murine model system were examined. BALB/c mice were immunized by intraperitoneal injection with B. gingivalis ATCC 53977 (one injection per week for 3 weeks), or with a lithium diiodosalicylate (LIS) extract (one injection per week for 3 weeks), or with lipopolysaccharide (LPS; one intravenous or intraperitoneal injection) from this same strain. Two weeks after the final immunization, the mice were challenged by subcutaneous injection of B. gingivalis ATCC 53977. Mice immunized w

The ability of fresh isolates of B. gingivalis to establish abscesses in the mouse model was studied by comparing them with established laboratory strains of B. gingivalis. Eight fresh isolates obtained from plaque associated with periodontal disease and grown under similar conditions as established strains were injected subcutaneously on the back of the mouse. All of these strains produced secondary lesions on the abdomen. Septicemia was associated with seven of the strains. Two commonly used laboratory strains, W50 and W83, also produced secondary lesions and septicemia. Five other laborator

Imipenem has excellent antimicrobial activity owing in part to beta-lactamase stability. We found that only 2 of over 350 Bacteroides fragilis group clinical isolates were resistant to imipenem, with an MIC of more than 16 micrograms/ml. These two isolates from the Tufts Anaerobe Laboratory (TAL) were resistant to all other beta-lactam agents tested. The organisms were able to inactivate imipenem in broth cultures and contained similar beta-lactamases that were able to hydrolyze carbapenems, cephamycins, cephalosporins, and penicillins. The molecular sizes of the beta-lactamases in TAL2480 and

The activity of imipenem against Pseudomonas aeruginosa and Bacteroides fragilis in vitro was studied. MICs of imipenem for P. aeruginosa are approximately 2 micrograms/m. Unlike most other beta-lactam antibiotics, imipenem does not show cross-resistance in the presence of intrinsic resistance. Imipenem is a strong inducer of Id beta-lactamase but is stable to hydrolysis by this enzyme. B. fragilis is more susceptible to imipenem than to other beta-lactam antibiotics. The activity of imipenem against B. fragilis is similar to that of metronidazole. The cephalosporinases of B. fragilis do not i
