Time-series gene expression patterns and their characteristics of Beauveria bassiana in the process of infecting pest insects.

Beauveria bassiana has been widely used as an important biological control fungus for agricultural and forest pests, and clarifying the interaction mechanism between B. bassiana and its host will help to better exert the efficacy of the mycoinsecticide. Here, we proposed a novel pattern analysis (PA) method for analyzing time-series data and applied it to a transcriptomic data set of B. bassiana infecting Galleria mellonella. We screened out 14 patterns including 868 genes, which had some characteristics that were not inferior to differentially expressed genes (DEGs). Compared with the previou

Beauveria caledonica as an Antagonist Controlling Fusarium oxysporum f. sp. cubense Tropical Race 4 in Bananas.

Beauveria caledonica offers great potential as a biocontrol agent for certain pests and diseases. However, its ability to colonize plants as an endophyte and its interactions with pathogenic microorganisms within the plant are still not fully understood. In this study, we investigated the potential of B. caledonica isolated from banana weevils to colonize banana cultivar "Baxijiao" (Musa spp. AAA) and control the Tropical Race 4 (TR4) strain of Fusarium Wilt of Banana (Fusarium oxysporum f. sp. cubense (Foc). A four-point dual-culture confrontation test revealed that B. caledonica effectively

¹⁵N isotope tracing reveals Beauveria bassiana colonization promotes nitrogen use efficiency of maize under elevated CO₂.

While the entomopathogenic fungus Beauveria bassiana is well known for its endophytic plant colonization and growth-promoting capabilities, its role in regulating plant nutrient dynamics, especially under climate change, remains poorly understood. This study investigated the interactive effects of B. bassiana colonization and elevated atmospheric carbon dioxide (CO₂) on nitrogen metabolism in maize. Open-top chambers were employed to simulate ambient and elevated (600 ppm) CO₂ conditions, combined with ¹ ⁵N isotope tracing to quantify nitrogen translocation, transformation and rhizosphere micr

Differential roles of three subclass III C6 transcription factors in the insect-pathogenic lifecycle of Beauveria bassiana.

Six subclass III C6 transcription factors (TFs) with unique Zn(II)2Cys6 binuclear cluster DNA-binding (C6 or GAL4) domain not locating at N-terminus are underexplored in Beauveria bassiana, a wide-spectrum mycoinsecticide. Here, we report that three C6 TFs with the GAL4 domain locating at C-terminus (TF1), between C-terminal and central regions (TF2) and between central and N-terminal regions (TF3) are largely differentiated in function. TF1 was proven to localize in both nucleus and cytoplasm and regulate host immune repression in the course of host hemocoel colonization instead of cuticular

Screening of the highly pathogenic Beauveria bassiana BEdy1 against Sogatella furcifera and exploration of its infection mode.

The white-backed planthopper (WBPH, Sogatella furcifera) is a notorious pest affecting rice production in many Asian countries. Beauveria bassiana, as the most extensively studied and applied insect pathogenic fungus, is a type of green and safe biological control fungus compared to chemical insecticides, and it does not pose the "3R" problem. In this study, the strain BEdy1, which had better pathogenicity to WBPH, was screened out from eight strains of B. bassiana. The daily growth rate, sporulation, and germination rate of BEdy1 strain were 3.74 mm/d, 1.37 × 108 spores/cm2, and 96.00%, respe

OTU7B Modulates the Mosquito Immune Response to Beauveria bassiana Infection via Deubiquitination of the Toll Adaptor TRAF4.

The Aedes aegypti mosquito transmits devastating flaviviruses, such as Zika, dengue, and yellow fever viruses. For more effective control of the vector, the pathogenicity of Beauveria bassiana, a fungus commonly used for biological control of pest insects, may be enhanced based on in-depth knowledge of molecular interactions between the pathogen and its host. Here, we identified a mechanism employed by B. bassiana, which efficiently blocks the Ae. aegypti antifungal immune response by a protease that contains an ovarian tumor (OTU) domain. RNA-sequencing analysis showed that the depletion of O

Metabolic effect of an exogenous gene on transgenic Beauveria bassiana using liquid chromatography-mass spectrometry-based metabolomics.

Genetic modification of Beauveria bassiana with the scorpion neurotoxin aaIT gene can distinctly increase its insecticidal activity, whereas the effect of this exogenous gene on the metabolism of B. bassiana is unknown until now. Thus, we investigate the global metabolic profiling of mycelia and conidia of transgenic and wild-type B. bassiana by liquid chromatography-mass spectrometry (LC-MS). Principal component analysis (PCA) and orthogonal projection to latent structure discriminant analysis (OPLS-DA) reveal clear discrimination of wild-type mycelia and conidia from transgenic mycelia and c

Mitochondrial and peroxisomal Lon proteases are functionally divergent in environmental adaptation and pathobiology of the entomopathogenic fungus Beauveria bassiana.

Lon protease belongs to the AAA‑ATPase family and plays comprehensive physiological roles via maintaining cellular protein homeostasis. This study seeks to explore the functions of Lon protease in biocontrol potential of the filamentous entomopathogenic fungus Beauveria bassiana. B. bassiana contained mitochondrial and peroxisomal Lon proteases (BbLon1 and BbLon2) which were functionally analyzed via gene disruption and complementation. BbLon1 contributed to fungal tolerance to oxidative, thermal, hyperosmotic, and cell wall-perturbing stresses as well as iron deficiency. Both BbLon1 and BbLon

Differential contributions of five ABC transporters to mutidrug resistance, antioxidion and virulence of Beauveria bassiana, an entomopathogenic fungus.

Multidrug resistance (MDR) confers agrochemical compatibility to fungal cells-based mycoinsecticdes but mechanisms involved in MDR remain poorly understood for entomopathogenic fungi, which have been widely applied as biocontrol agents against arthropod pests. Here we characterized the functions of five ATP-binding cassette (ABC) transporters, which were classified to the subfamilies ABC-B (Mdr1), ABC-C (Mrp1) and ABC-G (Pdr1, Pdr2 and Pdr5) and selected from 54 full-size ABC proteins of Beauveria bassiana based on their main domain architecture, membrane topology and transcriptional responses

Encapsulation of Beauveria bassiana conidia in carboxymethylcellulose beads: Advancing UV protection and thermal resilience in bioinsecticide applications.

The encapsulation of entomopathogenic fungi in polysaccharide matrices is a promising green strategy for protecting them from environmental stressors such as UV radiation and high temperatures (>35 °C). This study presents a novel formulation for encapsulating Beauveria bassiana conidia within a sodium carboxymethylcellulose matrix cross-linked with Al3+ or Fe3+. Beads produced via ionic gelation were subjected to air-drying or freeze-drying and characterized using SEM-EDX, FT-IR, TGA, and XRD. Freeze-dried beads were larger and exhibited a higher swelling capacity than their air-dried counter

Beauveria bassiana delivered through a cellulose-based hydrogel is effective against the red poultry mite, Dermanyssus gallinae.

Though the entomopathogenic fungus Beauveria bassiana has shown to be efficacious for managing Dermanyssus gallinae infestations, its delivery as liquid formulation poses concerns related to environmental stability and efficacy. To overcome such concerns, here we proposed a hydrogel matrix based on hydroxyethyl-cellulose (HEC) for delivering B. bassiana to control D. gallinae. Nymph and adult mites were exposed to a hydrogel matrix or filter papers containing B. bassiana (107 conidia/mL), with and without a pre-incubation period of 4 days at 25 °C (treated groups). As control groups, mites wer

Rapid analysis of insecticidal metabolites from the entomopathogenic fungus Beauveria bassiana 331R using UPLC-Q-Orbitrap MS.

Beauveria bassiana, a representative entomopathogenic fungus, is increasingly being utilized as an eco-friendly pest management alternative to chemical insecticides. This fungus produces a range of insecticidal secondary metabolites that act as antimicrobial and immunosuppressive agents. However, detailed qualitative and quantitative analysis related to these compounds remains scarce, we developed a method for the rapid analysis of these metabolites. Eight secondary metabolites (bassianin, bassianolide, beauvericin, beauveriolide I, enniatin A, A1, and B, and tenellin) were efficiently extract

Expressing Parasitoid Venom Protein VRF1 in an Entomopathogen Beauveria bassiana Enhances Virulence toward Cotton Bollworm Helicoverpa armigera.

Despite entomopathogenic fungi being used in various insect pest control, it is recognized that they could replace more chemical insecticides if they were more efficient. We have found that cotton bollworm Helicoverpa armigera responded to the infection of entomopathogenic fungus Beauveria bassiana by activating the Toll pathway. Koinobiont wasps also regulate host immunity and development to ensure the survival of their progeny. Previously, venom protein VRF1 was identified in Microplitis mediator. It enters H. armigera hemocytes, suppresses the expression of antimicrobial peptides (AMPs) by

Construction of a versatile cloning vector for Agrobacterium tumefaciens-mediated transformation of Beauveria bassiana using benomyl for selection.

Beauveria bassiana is a filamentous fungus that has considerable potential to combat insect pests. Therefore, an easy to use genetic transformation tool helps to investigate which fungal genes play a role in virulence and host specificity. However, B. bassiana is often very resistant to antibiotics used as selective agents for genetic transformation. We developed a binary cloning vector, pMBE1, for Agrobacterium tumefaciens-mediated transformation of B. bassiana. Selection of fungal transformants is based on the benomyl resistance gene that was introduced in this vector, as B. bassiana is very

Entomopathogenic Metarhizium and Beauveria species are associated with distinct geographic, management, and environmental factors, as well as plant communities.

Entomopathogenic fungi of the genera Metarhizium and Beauveria are important natural regulators of arthropod populations. To investigate their distribution and ecological drivers in real-world permanent grasslands, we did a survey analyzing fungal communities across 72 sites in Switzerland using ITS2 amplicon sequencing. We identified 10 Metarhizium and six Beauveria amplicon sequence variants (ASVs), with at least one ASV present at every site. Frequent ASVs included M. robertsii/M. pingshaense, M. brunneum, B. bassiana, and B. brongniartii. These ASVs showed distinct presence/absence and abu
