Cassava mosaic disease (CMD), caused by the cassava mosaic virus (CMV) and transmitted primarily by the whitefly Bemisia tabaci, reflects the most severe and widespread viral threat to cassava cultivation. The emergence of CMD with a nonlinear saturated incidence of Holling type II form is stochastically modeled and analyzed in this paper utilizing the continuous-time Markov chain (CTMC) modeling approach. The most significant distinction between deterministic and stochastic models is that the deterministic model predicts disease persistence when the basic reproduction number R 0 > 1 , where

Chili leaf curl virus (ChiLCV) is a Begomovirus chillicapsici that is one of the most devastating viruses impacted on the production of chili in the world, especially in South Asia. In the present study, we combined high-throughput computational genomics with experimental analysis of global diversity. A workflow was created using automated Python scripts to download, curate and process ChiLCV genomes from public database. About 410 complete ChiLCV genomes download from public databases. Using a phylogenetic approach, these isolates were subdivided into 34 strains, belonging to 10 major clades,

Viruses must rewire the cells they infect to promote their own multiplication, a task they accomplish through the deployment of multifunctional virus-encoded proteins. However, the limited coding capacity imposed by small viral genomes constrains their functional repertoire. Recently, it has been proposed that proteins encoded by plant-infecting geminiviruses can physically interact with one another, forming intra-viral protein complexes that may acquire novel activities and thereby expand viral functionality. Here, we test this concept by focusing on the C4 proteins of two okra-infecting gemi

BACKGROUND: Tomato yellow leaf curl virus (TYLCV), a member of the genus Begomovirus within the family Geminiviridae, is an important pathogen of tomato in many tropical, subtropical and temperate regions. TYLCV is exclusively transmitted by the whitefly Bemisia tabaci in a circulative manner. The viral coat protein (CP) has been assumed to play important roles in the entry of TYLCV into the insect midgut cells.
RESULTS: Testing the hypothesis that CP plays an important role in TYLCV acquisition by B. tabaci, a soluble form of the CP was expressed and purified. The purified recombinant CP mad

BACKGROUND: Geminiviruses encode small, rapidly evolving proteins that reprogram host functions to facilitate infection. Among these, C4/AC4 proteins frequently act as determinants of pathogenicity and symptom expression. Parsley yellow leaf curl virus (PYLCV; Pylecuvirus petroselini), a recently described monopartite geminivirus associated with yellowing and leaf curling in parsley, encodes a C4 protein whose biological activities and cellular distribution remain uncharacterized.
METHODS: Here, we combined comparative sequence analysis, confocal microscopy, site-directed mutagenesis, heterol

BACKGROUND: Bitter gourd (Momordica charantia), ridge gourd (Luffa acutangula), and pumpkin (Cucurbita pepo) are major vegetables of the Cucurbitaceae family cultivated extensively in India. However, their production is severely affected by begomoviruses, which inflict significant global damage to cucurbits. Among these, tomato leaf curl New Delhi virus (ToLCNDV), a whitefly-transmitted, persistent, circulative, and non-propagative begomovirus, seriously threatens cucurbit cultivation, often leading to substantial yield losses.

Chili pepper (Capsicum annuum) is a major horticultural commodity in Indonesia whose productivity is constrained by viral diseases, particularly pepper yellow leaf curl virus (PepYLCV). This study investigated the spatial distribution, symptomatology, incidence, severity, and genetic diversity of PepYLCV in seven primary chili‑producing districts of Lampung Province, namely North, Central, South, and West Lampung, Pesawaran, Pringsewu, and Tanggamus. Field surveys combined with molecular diagnostics demonstrated widespread PepYLCV infection, with disease incidence reaching 91% and 85% and dise

Tomato spotted wilt virus (TSWV) is an economically devastating pathogen that rapidly overcomes genetic resistance in major crops. Reverse genetic systems are crucial for investigating plant-virus interactions and resistance-breaking mechanisms, and developing these tools for segmented ambisense RNA viruses remains a crucial challenge. Current TSWV clones rely on extensively modified Asian isolates requiring co-delivery of multiple replication helpers and viral silencing suppressors. Streamlining these systems for regionally significant strains with minimal genetic alterations is essential. He

Geminiviruses severely threaten global crop production. Tomato yellow leaf curl virus (TYLCV) encodes multifunctional effector C4, which participates in diverse biological processes and interacts with host proteins to facilitate infection. Despite extensive studies on geminiviral C4, how TYLCV C4 suppresses tomato immunity remains poorly characterized. In this study, we identified the tomato 14-3-3 protein SlTFT2 as a specific interaction partner of the TYLCV C4 protein. We demonstrated that SlTFT2 functions as a positive regulator of antiviral defence, as its silencing enhanced systemic viral

BACKGROUND: Insect vectors are responsible for transmitting the majority of plant viruses. While the interplays between insect vectors and viruses have been extensively investigated, how other organisms associated with the shared host plants of the insect and virus impact virus transmission dynamics, and the corresponding viral countermeasures, remain largely unknown.
RESULTS: We explored the modulation of whitefly-mediated transmission of geminiviruses by two non-vector herbivores and the role of viral C2 proteins. The infestation of cotton bollworm induced the accumulation of jasmonic acid

BACKGROUND: Tobacco curly shoot virus (TbCSV, Begomovirus, Geminiviridae) is a viral agent that poses a serious threat to economically important crops like tomato and tobacco in Southwest China. The geminiviral V2 protein has been demonstrated to play multifunctional roles essential for viral infection and pathogenicity. This study comprehensively investigated the biological importance of five cysteines within the conserved 'WCCH' motif of the TbCSV V2 protein, and evaluated the effects of cysteine mutations on viral pathogenicity and the efficacy for cross-protection against wild-type severe

Pulses are vital to sustainable global food systems, supplying plant-based protein, minerals, and vitamins while improving soil fertility through biological nitrogen fixation. Cultivated on ~99.26 million ha with ~99.06 million tonnes annual production, their demand is rising, particularly in Asia and Africa. Yet production stability is increasingly threatened by climate variability, expanding vector niches, and predominantly rain-fed cultivation systems, where viral diseases impose major yield losses. While begomovirus-induced yellow mosaic disease remains the most destructive in Asia, the th

Papaya leaf curl Guangdong virus (PaLCuGDV), a member of the genus Begomovirus, poses a significant threat to passion fruit (Passiflora edulis) cultivation. A recent field survey in the Rongjiang area of Guizhou Province revealed a high natural infection rate of PaLCuGDV (90.1%; 209/232 plants). From a naturally infected passion fruit plant, the complete genome of a PaLCuGDV isolate was cloned and sequenced. The obtained genome, designated PaLCuGDV-GZ (GenBank OP901623.1), comprises 2732 nucleotides and shows > 90% similarity to other Chinese PaLCuGDV isolates based on BLAST analysis. To exper

Tomato yellow leaf curl virus (Family Geminiviridae, Genus Begomovirus) is a serious menace in the cultivation of tomato causing Tomato leaf curl disease (ToLCD). Recently, we presented the TYLCV isolates having additional genomic features (nucleotides insertions) characterized from the tomato fields of Kuwait adding up to the genetic diversity repertoire of these viruses. The widespread prevalence of disease in tropics across the continents, emergence of genetic variants and ever increasing complete genome sequences of virus isolates in public database warrant a global analysis to infer the g

Hemipteran insects transmit viruses when infesting plants, during which vectors activate salicylic acid (SA)-regulated antiviral defenses. How vector-borne plant viruses circumvent these antiviral defenses is largely unexplored. During co-infections of begomoviruses and betasatellites in plants, betasatellite-encoded βC1 proteins interfere with SA signaling and reduce the activation of antiviral resistance. βC1 inhibits SA-induced degradation of NbNPR3 (Nicotiana benthamiana nonexpressor of pathogenesis-related genes 3), a negative regulator of SA signaling. βC1 does not bind directly to NbNPR

BACKGROUND: The whitefly, Bemisia tabaci (Hemiptera: Aleyrodidae), is one of the most widely distributed agricultural pests. In recent years, B. tabaci Q has invaded China, and Q has displaced B in many areas now. In a number of regions of the world, invasion by B and/or Q has been followed by outbreaks of tomato yellow leaf curl virus (TYLCV). Our previous study showed TYLCV directly and indirectly modified the feeding behavior of B. tabaci in favor of Q rather than B.
METHODOLOGY/PRINCIPAL FINDINGS: In this study, we quantified the salicylic acid (SA) titers and relative gene expression of

Tomato leaf curl New Delhi virus (ToLCNDV) is a major begomovirus in tomato, where suppressor proteins AV2, AC2, and AC4 counteract host RNA silencing and intensify pathogenicity. Current management largely depends on vector control and resistance breeding, both limited by environmental concerns and rapid viral evolution, underscoring the need for targeted antiviral agents. Fifteen plant-derived phytochemicals, previously reported to exhibit antiviral activity against human viruses and representing major phytochemical classes such as flavonoids, terpenoids, alkaloids, and phenolics, were scree

Geminiviruses pose a severe threat to grain and vegetable crops worldwide, often resulting in significant economic losses. In cultivated tomato (Solanum lycopersicum), Ty resistance alleles have been introduced from wild tomato relatives, providing partial to strong resistance to geminivirus infections. The Ty-6 resistance locus from Solanum chilense was previously mapped to chromosome 10. It was recently shown to contain a mutant allele of the DNA POLYMERASE DELTA 1 (POLD1) gene that provides resistance to Tomato yellow leaf curl virus (TYLCV) infections. However, the resistance mechanism rem

Sri Lankan cassava mosaic virus (SLCMV) is a major cause for mosaic infections in cassava leaves, resulting in significant economic losses in southern India. SLCMV leads to growth retardation, leaf curl, and chlorosis in the host, with rapid transmission through whitefly insect vectors. Detecting SLCMV promptly is crucial, and the study introduces a novel and efficient colorimetric Loop-mediated isothermal amplification (LAMP) assay for successful detection in 60 min. Three primer sets were designed to target the conserved region of the SLCMV genome, specifically the coat protein gene, making

Tomato yellow leaf curl virus (TYLCV) is a significant threat to tomato cultivation globally, transmitted exclusively by the whitefly Bemisia tabaci. While previous research suggests that the TYLCV C2 protein plays a role in fostering mutualistic interactions between the virus and its insect vectors, the specific mechanisms remain unclear. In this study, we show that the C2 protein interferes with the salicylic acid (SA) defence pathway by disrupting TCP7-like transcription factor-mediated regulation of TGA2 expression. Whitefly infestation increases the expression of TCP7-like transcription f
