Bovine besnoitiosis is a chronic and debilitating disease, caused by the apicomplexan parasite Besnoitia besnoiti. Infection of cattle by B. besnoiti is governed by the tachyzoite stage, which is related to acute infection, and the bradyzoite stage gathered into macroscopic cysts located in subcutaneous tissue in the skin, mucosal membranes and sclera conjunctiva and related to persistence and chronic infection. However, the entire life cycle of this parasite and the molecular mechanisms underlying tachyzoite-to-bradyzoite conversion remain unknown. In this context, a different antigenic patte

Coccidia are obligate apicomplexan parasites that affect humans and animals. In fast replicating species, in vitro merogony takes only 24–48 h. In this context, successful parasite proliferation requires nutrients and other building blocks. Coccidian parasites are auxotrophic for cholesterol, so they need to obtain this molecule from host cells. In humans, ezetimibe has been applied successfully as hypolipidaemic compound, since it reduces intestinal cholesterol absorption via blockage of Niemann−Pick C-1 like-1 protein (NPC1L1), a transmembrane protein expressed in enterocytes. To date, few d

BACKGROUND: Equine besnoitiosis, caused by Besnoitia bennetti, and equine protozoal myeloencephalitis (EPM), caused by Sarcocystis neurona and Neospora hughesi are relevant equine diseases in the Americas that have been scarcely studied in Europe. Thus, a serosurvey of these cystogenic coccidia was carried out in Southern Spain. A cross-sectional study was performed and serum samples from horses (n = 553), donkeys (n = 85) and mules (n = 83) were included. An in-house enzyme-linked immunosorbent assay (ELISA) was employed to identify a Besnoitia spp. infection and positive results were confirm

Livestock industry is a pivotal sector for providing food, work places and monetary support for Egyptian people. Besnoitia besnoiti and Neospora caninum are protozoan parasites that are responsible for significant economic losses in ruminants, particularly in cattle. Besnoitia besnoiti can cause fertility problems and a general loss in productivity, while N. caninum is a major cause of abortion and neonatal abnormalities in infected animals. There is little information on the existence of these protozoa in Egypt, thus we conducted this study to reveal the current situation in cattle (n = 264),

The objective of this study was to establish a standardized protocol to monitor Besnoitia tarandi prevalence and intensity in barren-ground caribou (Rangifer tarandus) herds by: 1) calculating the relative sensitivity and specificity of the gross visual assessment of four anatomical sites compared with microscopic evaluation, and 2) determining which of four anatomical sampling sites was the most sensitive for detecting B. tarandi cysts by microscopy. Sampled tissues consisted of the conjunctiva of the left eye and skin sections from the rostrum, metatarsus, and thigh from 312 harvested caribo

Besnoitia besnoiti is an apicomplexan that causes serious economic loss in cattle in many countries and the disease is now spreading in Europe. At least 2 phases of bovine besnoitiosis are recognized clinically. An acute febrile phase characterized by anasarca and necrosis of skin is associated with multiplication of tachyzoites in vascular endothelium; this phase is short-lived and rarely diagnosed. Chronic besnoitiosis characterized by dermal lesions is associated with the presence of macroscopic tissue cysts and is easily diagnosed. Here we report the development of early B. besnoiti tissue

BACKGROUND AND AIM: Protozoan infections such as Neospora caninum and Besnoitia besnoiti are significant causes of infertility, abortion, and productivity losses in livestock. Despite their economic impact, epidemiological data from the Kingdom of Saudi Arabia (KSA) remain scarce. This study aimed to determine the seroprevalence of N. caninum and B. besnoiti in cattle and sheep across Asir, Jeddah, and Al-Qassim regions of KSA and to assess the influence of potential risk factors, including species, sex, age, breed, season, and management system.
MATERIALS AND METHODS: A cross-sectional study

Bovine besnoitiosis is an emerging disease in Europe, transmitted mechanically by the stable fly Stomoxys calcitrans. Infected cattle harbouring large quantities of parasitic cysts in their dermis are considered super-spreaders. The objective of this study was to demonstrate that Besnoitia besnoiti DNA can be detected in stable flies captured in the immediate vicinity of these hosts. In this case, we wanted to assess how far away from the super-spreaders we could capture infected Stomoxys flies. On two cattle farms affected by besnoitiosis in the south of France, super-spreaders were identifie

The study aimed to assess the prevalence of horses reactive to Sarcocystis neurona, Neospora caninum, and Besnoitia besnoiti in horses from the state of Rio Grande do Sul, Brazil. Sera reactivity was tested by the Immunofluorescent Antibody Test (IFAT) in 354 equids sampled at slaughter, using a cut-off of 1:80 for S. neurona and 1:50 for N. caninum and B. besnoiti. Detection of N. caninum was attempted by PCR and bioassay in gerbils (Meriones unguiculatus) using brain samples collected from seropositive horses. Sera from 158 horses (44.63 %) (158/354; CI: 39.54-49.84 %) reacted to S. neurona

Bumped kinase inhibitors (BKIs) have demonstrated safety and promising efficacy against various apicomplexan pathogens both in vitro and in vivo, but do not act parasiticidal in vitro. In the closely related cyst-forming coccidians Toxoplasma gondii, Neospora caninum and Besnoitia besnoiti, treatments with BKI-1708 induce the conversion of intracellular tachyzoites into atypical multinucleated complexes named "baryzoites". In this study, we comparatively assessed tachyzoites and baryzoites of all three species with respect to ultrastructure, differential antigen expression by immunofluorescenc

We here assessed the in vitro efficacy of the naptho-quinone buparvaquone (BPQ) against Besnoitia besnoiti tachyzoites in vitro. BPQ is currently licensed for the treatment of theileriosis in cattle in many countries, but not in the EU. In 4-day treatment assays, BPQ massively impaired tachyzoite proliferation with an IC50 of 10 ± 3 nm, and virtually complete inhibition was obtained in the presence of nm BPQ. Exposure to 1 µm BPQ leads to ultrastructural changes affecting initially the mitochondrial matrix and the cristae. After 96 h, most parasites were largely distorted, filled with cytoplas

Besnoitiosis is an emerging parasitic disease of equids. Italy is one of the few European countries where the circulation of Besnoitia spp. antibodies was demonstrated. In this study, a case of clinical besnoitiosis in two donkeys in northern Italy is reported. The two animals were clinically examined. Serum and blood samples were analyzed for the detection of Besnoitia spp. antibodies and for hematology, biochemistry, and enzyme activity, respectively. ITS-1 PCR and sequencing were carried out on DNA extracted from skin biopsies. Clinical examination revealed numerous scleral pearls in eyes o

Herein, we report the first incidence of systemic besnoitiosis in a male juvenile roe deer Capreolus capreolus. The animal was found dead in an area where bovine besnoitiosis is endemic and showed cachexia and multiple skin erosions in the metacarpal and metatarsal areas. Moreover, round and elevated white structures suggestive of Besnoitia spp. tissue cysts were also present. Twenty-eight tissue samples from different anatomical locations were collected for microscopic lesion and parasite detection through histopathology and PCR. Immunohistochemistry was performed to confirm Besnoitia-positiv

Recently, Besnoitia neotomofelis was described from a southern plains woodrat (Neotoma micropus) from southern Texas. During May 2010, 1 of 55 southern plains woodrats trapped in Uvalde County, Texas, was diagnosed with besnoitiosis. Grossly, the woodrat had bilateral swellings of the cheeks, and numerous Besnoitia sp.-like cysts were observed in the tongue, facial region, musculature of the limbs, and subcutis of the dorsum and flanks. Little to no inflammation was noted around cysts. The cysts were morphologically similar to B. neotomofelis based on light and transmission electron microcopy.

Six captive maras (Dolichotis patagonum) were found to have cysts consistent with Besnoitia in the lungs by light microscopy and electron microscopy. Three were juveniles that died with no premonitory signs during a 17-month period. The most prominent finding consisted of severe, subacute, diffuse interstitial pneumonia with syncytia and Besnoitia cysts. The severity of pneumonia correlated with the number of cysts and the presence of lytic cysts, often surrounded by chronic granulomatous inflammation. Disseminated infection was observed in one of these maras. The three other maras died of unr

An adult female rabbit was submitted for necropsy after sudden death. Pneumonia and nephritis were diagnosed by gross examination. On histological examination. Besnoitia cysts were seen in the pulmonary interalveolar tissue. The Besnoitia cysts were morphologically normal in appearance, measured 127 to 185 microns in diameter and provoked mild mononuclear inflammatory reaction. To the best of our knowledge there has been no previous report of natural besnoitiosis in rabbits.

Besnoitiosis is a parasitic disease of economic importance caused by cyst-forming protozoa from the genus Besnoitia. The disease affects the skin, subcutis, blood vessels, and mucous membranes of the animals. It is traditionally endemic in the tropical and sub-tropical regions of the world, and causes enormous economic loss associated with impaired productivity and reproduction, as well as skin lesions. Therefore, knowledge of the epidemiology of the disease, including the current Besnoitia species occurring in sub-Saharan Africa, the wide range of mammalian species hosts they use as intermedi

Besnoitiosis is a chronic and emerging parasitic disease caused by protozoa belonging to the genus Besnoitia. The disease is economically and ecologically threatening livestock and wildlife in Iran. Besnoitia caprae infecting goats can lead to cyst formation in host tissues and cause reproductive symptoms, resulting in economic losses due to abortions and/or reduced productive efficiency. Its significance is particularly pronounced in provinces such as Fars and Kermanو attributed to high goat population density, climatic conditions favoring transmission, and specific animal husbandry practices

Bovine besnoitiosis is a parasitic disease caused by the protozoan Besnoitia besnoiti, leading to infertility in bulls and abortions in cows. In Italy, it is considered an emerging disease, recently introduced by the importation of animals from Spain and France. In the last decade, many outbreaks have been reported and confirmed in native cattle, mostly in northern and central Italy. This study reports on an autochthonous outbreak of bovine besnoitiosis in two nearby farms located in northwestern Sicily. A 15-month-old Limousine bull born on the farm showed typical clinical signs of the chroni
