Using betaxolol for the prevention of paronychia induced by epidermal growth factor receptor inhibitors: a case-control cohort study.

BACKGROUND: High rates of posttreatment discomfort, infection, recurrence, and increased time to return to work have been noted after nail plate avulsion resulting from epidermal growth factor receptor tyrosine kinase inhibitor(EGFR-TKI)-induced paronychia, which may even interrupt the course of treatment for EGFR-TKI therapy. Thus, we conducted this study to determine how effectively a topical β-blocker, betaxolol, prevents EGFR-TKI-induced paronychia. METHODS: This case-control cohort study included a total of 131 non-small-cell lung cancer patients. The prevention group comprised 40 patien

Concentrations of betaxolol in ocular tissues of patients with glaucoma and normal monkeys after 1 month of topical ocular administration.

PURPOSE: To measure the concentration of betaxolol in tissues of humans with glaucoma and normal monkeys after topical administration. METHODS: Enucleated eyes (n = 7) of patients with glaucoma (age range, 27-79 years), without apparent anatomic disruption that would be likely to influence betaxolol absorption and intraocular distribution (exceptions: one pseudophakic, one aphakic) or other disease, were analyzed for betaxolol concentrations after self-administration of 0.25% betaxolol twice daily for 28 days or longer. The last instillation was made within 6 hours of surgery. Cynomolgus monk

Dose related efficacy of betaxolol in combination with nitroglycerin in patients with angina pectoris: a double blind cross-over study.

The effect of three different oral doses, 5 mg, 10 mg and 20 mg of betaxolol (BTX), a new long acting cardioselective beta-adrenergic blocking drug, on exercise tolerance was studied in a randomized double blind, cross-over study with latin square distribution in nine male patients with angina pectoris, under continuous therapy with oral nitroglycerin. BTX caused a significant reduction of heart rate (p less than 0.001) for all different dose levels during exercise. Systolic blood pressure was significantly decreased at peak exercise for BTX 10 (p less than 0.01) and BTX 20 (p less than 0.001)

Primary open-angle glaucoma patients characterized by ocular vasospasm demonstrate a different ocular vascular response to timolol versus betaxolol.

We evaluated the retrobulbar response to a selective versus nonselective beta blocker in a subgroup of primary open-angle glaucoma patients (POAG) characterized by ocular vasospasm. Eleven patients who exhibited ocular vasospasm (i.e. a significant increase in ophthalmic artery blood flow velocity or a significant decrease in ophthalmic artery resistance index during hypercapnia) underwent medication washout for 4 weeks and were enrolled in a double-masked cross-over study (betaxolol versus timolol). Patients were evaluated for blood flow velocity of the retrobulbar vessels using color Doppler

Development and validation of stability-indicating TLC-densitometric method for determination of betaxolol with LC-ESI/MS analysis of degradation product.

The purpose of this work was to develop a sensitive stability indicating TLC-densitometric method for the determination of betaxolol (Bx) in pharmaceutical preparations and to study the stability of Bx in acidic solutions. The method was developed on TLC aluminium plates precoated with silica gel F254using the mobile phase chloroform-methanol-ammonia 25% (18 : 4: 0.2, v/v/v) which gives compact spots for Bx (R(f) approximately equal to 0.64) and its degradation product (R(f) approximately equal to 0.39). Densitometric analysis was carried out in UV at 280 nm. The developed method is highly sen

Comparison of the effects of intravenous and oral betaxolol on antegrade and retrograde conduction in patients with atrioventricular nodal reentrant and atrioventricular reentrant tachycardia.

The electrophysiologic effects of intravenous (0.15 mg kg-1) and oral (20 mg day-1) betaxolol have been investigated in 11 patients with atrioventricular (A-V) nodal reentrant tachycardia and eight patients with orthodromic A-V reentrant tachycardia. Betaxolol significantly (P greater than 0.01) prolonged sinus cycle length, sinus node recovery time, intranodal conduction time, and the antegrade functional refractory period of the A-V node. When the effective refractory period of the A-V node could be determined it was increased by betaxolol, whereas no significant electrophysiologic effects w

Quality-by-design-based fabrication of betaxolol hydrochloride-loaded nano-ocular carrier via ternary phase mapping of solid/liquid lipids and BBD modeling for enhanced transcorneal flux and IOP-lowering effect.

The anti-glaucoma drug betaxolol hydrochloride (BH) exhibits poor ocular bioavailability due to limited corneal permeability, rapid precorneal loss, short retention time, and lacrimal drainage when administered conventionally. In the present study, a solid-liquid lipid-based nano-ocular carrier (NC) of BH was developed to modulate transcorneal permeability and improve intraocular efficacy. Initially, solid and liquid lipid excipients were screened on the basis of BH solubility, followed by ternary phase behavior analysis in normal saline to identify a one-phase region (OPR). It was processed a

[Nanocapsules of beta-blocking agents: a new drug carrier in ophthalmology. Application to medical treatment of glaucoma in rabbits].

In order to reduce the lacrimal elimination and to increase the intraocular penetration of betaxolol after ocular administration, we prepared a new drug carrier, polycaprolactone nanocapsules, containing betaxolol in the base form. The instillation of nanocapsules containing only 0.1% of betaxolol into glaucomatous rabbit eyes induced a greater decrease of IOP in intensity, time and stability compared to the reduction of IOP induced after instillation of the commercial eye drops (Betoptic) containing 5 times more betaxolol (0.56%). This new pharmaceutical dosage form produces very interesting
