Age and the aging process significantly alter the small bowel microbiome.

Gut microbial diversity decreases with aging, but existing studies have used stool samples, which do not represent the entire gut. We analyzed the duodenal microbiome in 251 subjects aged 18-35 (n = 32), 36-50 (n = 41), 51-65 (n = 96), and 66-80 (n = 82). Decreased duodenal microbial diversity in older subjects is associated with combinations of chronological age, number of concomitant diseases, and number of medications used, and also correlated with increasing coliform numbers (p < 0.0001). Relative abundance (RA) of phylum Proteobacteria increases in older subjects, with increased RA of fam

Long-lasting gut microbiome and fecal metabolome alterations after colorectal adenoma removal and their relationship to colorectal cancer.

Although the gut microbiome is implicated in colorectal cancer (CRC), microbiome and metabolome alterations along the adenoma-carcinoma sequence remain unclear. Here, we profile stool metagenomes obtained from 354 women 12.1 ± 4.8 years following adenoma resection and from their 1:1-matched controls, as well as stool metabolomes from 184 pairs. Metagenomic profiles are compared with those from 14 independent CRC case-control studies. Microbial composition differs between adenoma cases and controls and agrees with CRC-associated alterations (Pearson's rho = 0.26, p < 0.0001). Thirty-one microbe

Mechanism of high-fat diet exacerbating α-lactalbumin-induced allergic responses: Microbiome and metabolomic insights.

High-fat diet (HFD) is a key contributor in exacerbating food allergy, but there is little in-depth research on the mechanisms by which HFD affects allergic responses induced by α-lactalbumin (ALA). The mechanism of HFD exacerbating ALA-induced allergic responses (H-ALA) was studied via microbiome and metabolomics. HFD induced alterations in body weight and temperature in ALA-sensitized mice, while exacerbating allergic responses by increasing ALA-specific IgG/IgE levels and disrupting Th1/Th2 cytokine balance. In H-ALA, 16S rRNA gene sequencing revealed that Staphylococcus and Bilophila were

In vitro Study of Bifidobacterium lactis BL-99 With Fructooligosaccharide Synbiotics Effected on the Intestinal Microbiota.

Probiotics and prebiotics relieve constipation by altering the composition of the intestinal microbiota. However, their synergistic mechanism of action remains unclear. Herein, an in vitro fermentation model was constructed to examine the synergistic effects of Bifidobacterium lactis BL-99 and fructooligosaccharide (FOS) on the regulation of intestinal microbiota from a population with constipation. The utilization of FOS was promoted by BL-99, and the increase rate being 22.33%. Relative to the BL-99 and the FOS groups, the BL-99_FOS group showed a highly significant increase in acetic acid c

Black tea polysaccharides: purification, structural characterization, fermentation characteristics, and interaction with gut microbiota.

Although black tea polysaccharides have numerous health benefits, limited studies have investigated the structural and fermentation characteristics. Herein, the structural characteristics and interaction with the gut microbiota of a black tea polysaccharide (BTPS3) fraction were systematically evaluated. Results showed that BTPS3 was an acidic heteropolysaccharide with a molecular weight of 2.10 × 105 Da, primarily composed of arabinose, galactose, and galacturonic acid in a molar ratio of 31.23:24.06:16.69. Methylation and NMR analyses indicated that BTPS3 is a pectic polysaccharide, with gly

Cooked Adzuki Bean Reduces High-Fat Diet-Induced Body Weight Gain, Ameliorates Inflammation, and Modulates Intestinal Homeostasis in Mice.

Adzuki bean is widely consumed in East Asia. Although the positive effects of its biologically active ingredients on obesity have been confirmed, the role of whole cooked adzuki bean in preventing obesity and the relationship between the effects and gut microbiota remain unclear. Mice were fed either a low-fat diet (LFD) or high-fat diet (HFD) with or without 15% cooked adzuki bean for 12 weeks. Cooked adzuki bean significantly inhibited weight gain and hepatic steatosis, reduced high levels of serum triacylglycerol (TG), alanine aminotransferase (ALT), and aspartate aminotransferase (AST), an

Modulation of gut homeostasis by exopolysaccharides from Aspergillus cristatus (MK346334), a strain of fungus isolated from Fuzhuan brick tea, contributes to immunomodulatory activity in cyclophosphamide-treated mice.

In this study, the crude exopolysaccharides (CEPSs) from fungus Aspergillus cristatus (MK346334, NCBI) isolated from Fuzhuan brick tea and its main purified fraction (EPSs-2) were investigated. Using the RAW264.7 cell model, EPSs-2 exhibited an excellent immunomodulatory effect in vitro. Then, the regulating effects of EPSs on immune function and gut microbiota were evaluated using a cyclophosphamide (Cy)-induced mice model. It was found that both CEPSs and EPSs-2 improved the body weight loss, immune organ indexes as well as the levels of TNF-α, IL-1β, IFN-γ and IgA, exhibiting potent immunor

Arsenic Metabolism and Toxicity Influenced by Ferric Iron in Simulated Gastrointestinal Tract and the Roles of Gut Microbiota.

Iron (Fe) is a common trace element in drinking water. However, little is known about how environmental concentrations of Fe affect the metabolism and toxicity of arsenic (As) in drinking water. In this study, influence of Fe at drinking water-related concentrations (0.1, 0.3, and 3 mg Fe (total)/L) on As metabolism and toxicity, and the roles of gut microbiota during this process were investigated by using in vitro Simulator of the Human Intestinal Microbial Ecosystem (SHIME). Results showed that Fe had ability to decrease bioaccessible As by coflocculation in small intestine. 0.1 and 0.3 mg/

The spleen-strengthening and liver-draining herbal formula treatment of non-alcoholic fatty liver disease by regulation of intestinal flora in clinical trial.

OBJECTIVE: As a metabolic disease, one important feature of non-alcoholic fatty liver disease (NAFLD) is the disturbance of the intestinal flora. Spleen-strengthening and liver-draining formula (SLF) is a formula formed according to the theory of "One Qi Circulation" (Qing Dynasty, 1749) of Traditional Chinese Medicine (TCM), which has shown significant therapeutic effect in patients with NAFLD in a preliminary clinical observation. In this study, we aim to explore the mechanism of SLF against NAFLD, especially its effect on glucolipid metabolism, from the perspective of intestinal flora. MET

Profiling Gut Microbiome and Metabolites Aids Diagnosis and Prediction of Progression in Mild Cognitive Impairment.

Accumulating evidence suggests a link between the gut microbiota and metabolome and mild cognitive impairment (MCI); however, it remains largely unknown whether these parameters can aid MCI diagnosis and predict disease progression. We comprehensively characterized gut microbiota and metabolomic profiles in fecal samples from 35 individuals, including 16 spousal pairs in which one partner had MCI. Compared with controls, MCI cases exhibited significantly elevated levels of Izemoplasmatales spp. in the fecal microbiome and proline in the metabolome, along with decreased levels of Ruminococcus s

Intestinal Microbiomics and Liver Metabolomics Insights into the Improvement Effects of Lactiplantibacillus plantarum FJG2627 on Lipopolysaccharide-Induced Acute Liver Injury in Mice.

Endotoxin is one of the chemical substances that cause acute liver injury (ALI), which become a common health problem worldwide. Lactic acid bacteria (LAB) have emerged as a promising candidate for attenuating endotoxin-induced ALI. Lactiplantibacillus plantarum FJG2627 is a LAB isolated from traditional pickles with excellent probiotic effects. In this study, the protective functions of L. plantarum FJG2627 against ALI caused by lipopolysaccharides (LPS) in mice were investigated. Administration of L. plantarum FJG2627 significantly reduced serum ALT, AST, and pro-inflammatory factors (IL-1β

Intestinal microbial community assessment in patients with different forms of epilepsy and autoimmune encephalitis: An exploratory study.

Despite the availability of antiseizure medications (ASMs), approximately one-third of patients with epilepsy remain refractory to treatment. Evidence suggests that the gut microbiota modulates central nervous system function through neuroimmune and metabolic pathways. We characterized the fecal microbiome of patients with epilepsy and autoimmune encephalitis (AE) using 16S rRNA sequencing. Ninety-six participants from a tertiary hospital in Brazil were included: mesial temporal lobe epilepsy (MTLE; n = 38), genetic generalized epilepsy (GGE; n = 11), AE (n = 10), and healthy controls (n = 37)

Comparison of four characteristic carotenoids (lutein, β-carotene, lycopene, and astaxanthin) in alleviating DSS-induced colitis through gut microbiota regulation.

Carotenoids, particularly lutein, β-carotene, lycopene, and astaxanthin, possess established anti-inflammatory and antioxidant properties. Although these compounds are known to interact with the gut microbiota and ameliorate microbial dysbiosis, their structure-activity relationships in colitis alleviation remain poorly understood. Using a dextran sulfate sodium (DSS)-induced colitis model, we made a systematic comparison of these four structurally distinct carotenoids. All treatments markedly improved colitis-associated clinical symptoms, including weight loss, colon shortening, bloody stool,

Causal effects of gut microbiota on scoliosis: A bidirectional two-sample mendelian randomization study.

BACKGROUND: Recent studies have shown altered gut microbiome composition in patients with scoliosis. However, the causal effect of gut microbiota on scoliosis remains unknown. METHODS: A Mendelian randomization (MR) study was conducted to quantify the impact of 191 gut microbiome taxa's instrumental variables from the MibioGen Genome-wide association study (GWAS) on scoliosis risk using data from the FinnGen GWAS (1168 cases and 16,4682 controls). Inverse variance weighted (IVW) was the main method, and MR results were verified by sensitive analysis. RESULTS: Bilophila, Eubacterium (eligens

The signatures of liver metabolomics and gut microbiota in high-fat diet fed mice supplemented with rhododendrol.

As aromatic compounds found within red fruits and berries, raspberry ketones (RK) have the potential for nonalcoholic fatty liver disease (NAFLD) amelioration. However, the mechanism of RK on NAFLD is unclear, and their bioactive metabolite is unknown. As the major metabolites of RK that are mainly distributed in the liver, rhododendrol (RHO) is used in our current study to test whether RHO accounts for the beneficial effect of RK on NAFLD and the underlying mechanism. In a 16-week trial, RHO significantly decreased final body weight, improved serum lipid profile and ameliorated liver inflamma

The small bowel microbiome changes significantly with age and aspects of the ageing process.

Gut microbiome changes have been associated with human ageing and implicated in age-related diseases including Alzheimer's disease and Parkinson's disease. However, studies to date have used stool samples, which do not represent the entire gut. Although more challenging to access, the small intestine plays critical roles in host metabolism and immune function. In this paper (Leite et al. (2021), Cell Reports, doi: 10.1016/j.celrep.2021.109765), we demonstrate significant differences in the small intestinal microbiome in older subjects, using duodenal aspirates from 251 subjects aged 18-80 year

Antianaerobic antimicrobials: spectrum and susceptibility testing.

Susceptibility testing of anaerobic bacteria recovered from selected cases can influence the choice of antimicrobial therapy. The Clinical and Laboratory Standards Institute (CLSI) has standardized many laboratory procedures, including anaerobic susceptibility testing (AST), and has published documents for AST. The standardization of testing methods by the CLSI allows comparisons of resistance trends among various laboratories. Susceptibility testing should be performed on organisms recovered from sterile body sites, those that are isolated in pure culture, or those that are clinically importa
