PEGylated organophosphorus hydrolase combined with butyrylcholinesterase: dual-enzyme synergistic biocatalysis for acute organophosphate poisoning.

Organophosphates (OPs), widely used as insecticides, exhibit severe acute toxicity and pose a significant threat to public health. Current standard therapy, which mainly relies on atropine and oxime reactivators, alleviates symptoms but fails to effectively eliminate circulating toxin or excess acetylcholine (ACh), thus representing a non-causal intervention. This study proposes a novel, efficient synergistic dual-enzyme strategy for the treatment of acute OP poisoning in a Sprague-Dawley rat model. Intravenous co-administration of organophosphorus hydrolase (OPH) from Pseudomonas diminuta, si

Exploring the biocatalysis of psilocybin and other tryptamines: Enzymatic pathways, synthetic strategies, and industrial implications.

Tryptamines play diverse roles as neurotransmitters and psychoactive compounds found in various organisms. Psilocybin, a notable tryptamine, has garnered attention for its therapeutic potential in treating mental health disorders like depression and anxiety. Despite its promising applications, current extraction methods for psilocybin are labor-intensive and economically limiting. We suggest biocatalysis as a sustainable alternative, leveraging enzymes to synthesize psilocybin and other tryptamines efficiently. By elucidating psilocybin biosynthesis pathways, researchers aim to advance synthet

Hydroxyapatite as an Enzyme Carrier in Continuous-Flow Biocatalysis: An Improved Synthesis of γ‑Glutamyl Derivatives.

Biocatalysis has proved to be of great importance to pursue a more sustainable production of fine chemicals, and enzyme immobilization is a crucial tool to achieve this goal. In this work, hydroxyapatite, an eco-friendly material, was used as a support for the immobilization of a γ-glutamyltransferase from Escherichia coli, an enzyme that catalyzes the synthesis of bioactive γ-glutamyl derivatives, exploiting the noncovalent interactions between the support and the enzyme. After screening the immobilization conditions, the storage (up to 90 days) and the thermal stability (50 °C) of the obtain

Unlocking the catalytic potential of transaminase: A two-decade evolution toward green and scalable biocatalysis.

Over the past two decades, transaminases or aminotransferases have been involved in the chemoenzymatic synthesis of active pharmaceutical ingredients by highly efficient and straightforward catalysis of prochiral ketones to chiral amines. These enzymes are promising targets used in the pharmaceutical sector since they are relatively easy to clone and express from a variety of bacteria and other species, where they play key catalytic roles in amino acid metabolic pathways. Also, their high thermal and solvent stability makes them very suitable candidates for use in industry. These enzymes are s

Enantioselective Synthesis of Secondary Amines by Combining Oxidative Rearrangement and Biocatalysis in a One-Pot Process.

This contribution describes the development of chemoenzymatic one-pot processes, which combine an oxidative rearrangement and a biotransformation catalyzed by an imine reductase (IRED), for the synthesis of highly enantiomerically enriched secondary amines, such as an aryl-substituted pyrrolidine and a benzazepine. The benefits of this chemoenzymatic one-pot approach include high overall conversions (up to >99%), high enantiomeric excesses (up to >99% ee), and a straightforward synthetic approach toward secondary amines without the need to isolate the formed intermediate. For the initial chemi

Sustainable biocatalysis via lipase immobilization: pyrolyzed lignocellulosic bamboo supports in batch and continuous flow systems.

Bamboo-derived biochars obtained via fast and slow pyrolysis conditions were used as supports for the immobilization of Candida antarctica lipase B (CALB) through physical adsorption. The FP- and SP-based materials were then evaluated in esterification and hydrolysis reactions under both batch and continuous-flow conditions. In all cases, high immobilization yields were obtained, although the catalytic performance of FP-derived systems was consistently higher. In batch esterification of long-chain fatty acids, FP-CALB achieved high conversions, reaching up to 97% under optimized conditions (50

Thermostable Enzyme Variants in the Lower Mevalonate Pathway Improve Isoprenoid Production by Cell-Free Biocatalysis.

Cell-free biocatalysis is a rapidly evolving field with great potential for sustainably producing valuable chemicals. Some challenges in cell-free biocatalysis include reaction longevity, enzyme stability, and the cost of the biocatalysts. Here, the challenge of enzyme instability was addressed by employing thermophilic enzymes to improve the productivity of the lower mevalonate pathway, using limonene as an example isoprenoid product. The Classical mesophilic mevalonate pathway was compared to a newly assembled set of thermophilic enzymes comprising the Archaea I mevalonate pathway. The therm

Thermodynamic Microenvironment Engineering in Mesoporous Nanoreactors to Enhance Biocatalysis for AI-Empowered Ultrasensitive Pathogen Detection.

Harmonizing enzyme-support microenvironments to govern thermodynamic interaction landscapes presents a critical yet underexplored frontier in nanobiocatalysis for pathogen detection. Herein, we architecturally engineer mesoporous resorcinol formaldehyde nanospheres (mRFNSs, 9.95 nm pores) with tailored surface chemistry to elucidate how microenvironment modulation dictates enzyme immobilization energetics. Thermodynamic dissection demonstrates that betaine-tailored mRFNSs with optimal immobilization efficiency and activity dramatically reshape binding energetics, achieving record affinity thro

S-adenosyl-methionine-dependent methyltransferases: highly versatile enzymes in biocatalysis, biosynthesis and other biotechnological applications.

S-adenosyl methionine (SAM) is a universal biological cofactor that is found in all branches of life where it plays a critical role in the transfer of methyl groups to various biomolecules, including DNA, proteins and small-molecule secondary metabolites. The methylation process thus has important implications in various disease processes and applications in industrial chemical processing. This methyl transfer is catalysed by SAM-dependent methyltransferases (MTases), which are by far the largest groups of SAM-dependent enzymes. A significant amount is now known regarding the structural biolog

The Atmospheric Pressure Capillary Plasma Jet Is Well-Suited to Supply H2O2 for Plasma-Driven Biocatalysis.

Plasma-generated H2O2 can be used to fuel biocatalytic reactions that require H2O2 as a cosubstrate, such as the conversion of ethylbenzene to (R)-1-phenylethanol ((R)-1-PhOl) catalyzed by unspecific peroxygenase from Agrocybe aegerita (rAaeUPO). Immobilization is recently shown to protect biocatalysts from inactivation by highly reactive plasma-produced species; however, H2O2 supply by the employed plasma sources (μAPPJ and DBD) is limiting for rAaeUPO performance. This study evaluates a recently introduced capillary plasma jet for suitability to supply H2O2 in situ. H2O2 production is modula

Optimization of the Biocatalysis for D-DIBOA Synthesis Using a Quick and Sensitive New Spectrophotometric Quantification Method.

D-DIBOA (4-hydroxy-(2H)-1,4-benzoxazin-3-(4H)-one) is an allelopathic-derived compound with interesting herbicidal, fungicidal, and insecticide properties whose production has been successfully achieved by biocatalysis using a genetically engineered Escherichia coli strain. However, improvement and scaling-up of this process are hampered by the current methodology for D-DIBOA quantification, which is based on high-performance liquid chromatographic (HPLC), a time-consuming technique that requires expensive equipment and the use of environmentally unsafe solvents. In this work, we established a

Crosslinking-regulated nanoconfined biointerface enables highly stable and sensitive cooperative cascade biocatalysis for dual-mode biosensing.

Engineering catalytic microenvironments within hybrid materials is a critical strategy for enhancing enzyme stability and catalytic efficiency; however, precise regulation of intermediate transfer and interfacial catalytic coupling remains challenging. Here, we report a crosslinking-regulated, double-crystallized hierarchical hybrid nanoflowers system (D-Cu@c-GOx NFs) that establishes a nanoconfined biointerface for cooperative cascade biocatalysis. The system is constructed via in situ enzyme incorporation, followed by glutaraldehyde-mediated crosslinking and a second crystallization step, ge

Heterojunction-driven MXenzyme biocatalysis: a progressive strategy to remodel tumor immune microenvironments for improved therapeutic efficacy.

The hypoxic and immunosuppressive tumor microenvironment (TME) significantly impedes reactive oxygen species (ROS) generation and suppresses the release of tumor-associated antigens, thereby limiting the induction of immunogenic cell death (ICD) and compromising therapeutic outcomes. Reprogramming this immunosuppressive TME is therefore crucial for effective cancer treatment. In this study, we developed a heterojunction-engineered MXenzyme-based platform (NbMR@EDM) to address this challenge. By leveraging the exceptional photothermal conversion capability and multi-enzymatic activities of Nb2C

PEO/GG nanofiber-based lipase immobilization for efficient hydrolysis of selected vegetable oils and industrial biocatalysis.

The immobilization of lipase on electrospun nanofibers is a promising strategy to enhance enzyme stability, reusability, and catalytic efficiency. In this study, poly(ethylene oxide)/guar gum (PEO/GG) nanofibers with varying PEO content (4 %, 5 %, and 6 % w/v) and constant GG content (1 % w/v) were synthesized via electrospinning, referred to as PEO4/GG, PEO5/GG, and PEO6/GG. Among these, PEO4/GG was identified as the optimal support due to its higher porosity (28.75 ± 3.03 %) and surface area (354.16 ± 16.15 μm2), which enhanced enzyme loading and substrate diffusion. From discovery to application: Enabling technology-based optimizing carbonyl reductases biocatalysis for active pharmaceutical ingredient synthesis.

The catalytic conversion of chiral alcohols and corresponding carbonyl compounds by carbonyl reductases (alcohol dehydrogenases), which are NAD(P) or NAD(P)H-dependent oxidoreductases, has attracted considerable attention. However, existing carbonyl reductases are insufficient to meet the demands of diverse industrial applications; hence, new enzymes with functions that can expand the toolbox of biocatalysts are urgently required. Developing precisely controlled chiral biocatalysts is of great significance for the efficient development of a broad spectrum of active pharmaceutical ingredients v

Supramolecular assembly strategy of modified starch chains for achieving recyclable emulsion biocatalysis within a narrow pH range.

Stimuli-responsive Pickering emulsions are promising in biocatalysis for their ease of product separation and emulsifier recovery. However, pH responsiveness, though simple and cost-effective, faces challenges in precise control and narrow transition ranges, limiting its use in enzymatic catalysis. Herein we introduced amorphous octenyl succinic anhydride-modified debranched starch chains (Am-OSA-St) to control emulsion properties within a pH range suitable for enzymatic catalysis. By adjusting the OSA group density and molecular weight, Am-OSA-St allowed emulsions to transition reversibly bet

From oxygen detoxification to peroxide supply: Microbial NADH oxidases in redox physiology and H₂O₂-coupled biocatalysis.

NADH oxidases (NOXs) are flavin adenine dinucleotide (FAD)-dependent flavoproteins widely distributed in bacteria, archaea, and other microorganisms, where they couple NADH oxidation to oxygen reduction and contribute to redox homeostasis, oxygen adaptation, and peroxide metabolism. H₂O-forming NOX has traditionally been preferred for cofactor regeneration, whereas H₂O₂-forming NOX is increasingly recognized as a dual-function catalyst for NAD⁺ regeneration and in situ H₂O₂ production. This review summarizes the catalytic mechanisms, structural basis, and physiological functions of NOX, focusi

Pollen-Based Artificial Enzyme With Efficient Biocatalysis and Immune-Priming to Prevent Primary and Secondary Wound Infection.

Bacterial infection poses a significant threat to clinical treatment due to the emergence of drug resistance and the high risk of recurrence. Here, we report the de novo design of a spiky pollen-based vanadium artificial enzyme particle (VAE-Pollen) that integrates potent reactive oxygen species (ROS)-catalytic activity with immune priming to prevent both primary and secondary bacterial infections. Experimental and theoretical analyses confirm that bacteria are efficiently captured by the micro-structured surface of VAE-Pollen, and the introduction of oxygen vacancies modulates the electronic

The CYP152-family P450 enzyme CypC of Bacillus subtilis converts non-natural substrates in plasma-driven biocatalysis.

Plasma-driven biocatalysis utilizes in situ H2O2 production by atmospheric pressure plasmas to drive H2O2-dependent enzymatic reactions. Having previously established plasma-driven biocatalysis using recombinant unspecific peroxygenase from Agrocybe aegerita (rAaeUPO) to produce (R)-1-phenylethanol from ethylbenzene, we here employed CypC from Bacillus subtilis 168 (synonyms: YbdT, P450BSβ), an integral enzyme of surfactin and fengycin biosynthesis. CypC naturally hydroxylates medium and long-chain carboxylic acids. With short-chain carboxylic acids as decoy molecules, it also converts non-nat
