Plasma S100β is not a useful biomarker for tumor burden in neurofibromatosis.

OBJECTIVES: Neurofibromatosis 1 (NF1), NF2, and schwannomatosis are characterized by a predisposition to develop multiple neurofibromas and schwannomas. Currently, there is no blood test to estimate tumor burden in patients with these disorders. We explored whether S100β would act as a biomarker of tumor burden in NF since S100β is a classic immunohistochemical marker of astrocytes, oligodendrocytes and Schwann cells and a small study showed S100β concentrations correlate with the volume of vestibular schwannomas. DESIGN AND METHODS: We calculated whole-body tumor burden in subjects with NF1,

Wearable micromotor-based electrochemical microneedle sensors for monitoring tumor biomarkers in skin.

Wearable sensors show value in dynamic disease monitoring but face the challenge in efficiently detecting low-abundance biomarkers from trace skin interstitial fluid (ISF). Herein, we proposed an active microneedle sensor integrated with micromotors for highly sensitive, dynamic monitoring of trace tumor markers carcinoembryonic antigen (CEA) in ISF. The sensor employed a core-shell structured hydrogel microneedle array. The shell provided mechanical strength for minimally invasive skin penetration, while the water-soluble core dissolved upon contact with ISF, releasing self-propelled micromot

Circulating tumor cells as biomarkers in melanoma: techniques, challenges, and clinical applications.

Cutaneous melanoma is a malignant tumor and remains a clinical challenge due to unmet needs in risk stratification, real-time treatment monitoring, and early detection of progression or resistance. Early detection is crucial, as prognosis significantly worsens once metastasis occurs. Liquid biopsy has emerged as a powerful, minimally invasive tool for early diagnosis, real-time disease monitoring, and therapeutic guidance. Among its components, circulating tumor cells (CTCs) are rare yet clinically informative, providing insights into tumor heterogeneity, metastatic potential, and prognosis. G

Blood tumor load: combining biomarkers to increase the proportion of informative patients.

BACKGROUND: The load of circulating tumor cells (CTC) and tumor-derived extracellular vesicles (tdEV) strongly correlates with poor clinical outcomes and can be used to evaluate treatment response and the presence of treatment targets. However, the frequency of CTC is low, making an accurate assessment impossible in most patients. Here, we introduce blood tumor load (BTL), in which CTC and tdEV are combined into one value ranging from 0 (low) to 1 (high) to simplify result interpretation and increase the percentage of patients from which a reliable assessment can be made. PATIENTS AND METHODS

Tumor Mutational Burden as a Prognostic Biomarker in Follicular Lymphoma.

INTRODUCTION: Follicular lymphoma (FL) is the most common indolent non-Hodgkin lymphoma. Approximately 20% of patients experience early relapse within 24 months (POD24) of immunochemotherapy (ICT), a subgroup associated with poor prognosis and not well-identified by current prognostic indices. Given that tumor mutational burden (TMB) has been associated with outcome in various cancers, we investigated its potential as a prognostic biomarker in FL. METHODS: TMB was estimated by next-generation sequencing using a 409-gene panel in a cohort of 119 patients diagnosed with FL grades 1-3A and treat

The Role of Vimentin 3 in Ameloblastomas: A Novel Tumor Biomarker.

INTRODUCTION: Ameloblastoma (ABL), a common odontogenic tumor in the maxillofacial region, presents primarily as unicystic (U-ABL) and conventional (C-ABL) variants. Despite shared epithelial features, their distinct biological behaviors may stem from interactions between connective tissue and epithelial cells. Vimentin 3 (VIM3), a truncated variant of Vimentin-Full Length (VIMFL), exhibits unique biological properties. This study is the first to investigate VIM3 expression in ABLs. METHODS: Formalin-fixed paraffin-embedded (FFPE) samples of C-ABL (n = 30), U-ABL (n = 30), and dental follicle

Tumor DNA: an emerging biomarker in head and neck cancer.

Head and neck cancer (HNC) includes a diverse range of malignancies arising commonly from mucosal epithelia of the upper aerodigestive tract. Head and neck squamous cell carcinoma (HNSCC), the most common form of HNC, develops in the oral cavity, pharynx, and larynx and is associated with tobacco exposure, alcohol abuse, and infection with oncogenic viruses. Despite global advances in cancer care, HNSCC often presents with advanced disease and is associated with poor 5-year survival of ~50%. Genotyping tumor tissue to guide clinical decision-making is becoming commonplace in modern oncology, b

Circulating Tumor Cells as a Biomarker in Pancreatic Ductal Adenocarcinoma.

BACKGROUND/AIMS: Circulating tumor cells (CTCs) are valuable in both basic research and clinical application for cancer management. In the current study, we evaluated the diagnostic value of CTCs in pancreatic ductal adenocarcinoma (PDAC). METHODS: In total, 143 blood samples from 95 consecutively diagnosed PDAC patients and 48 healthy donors were collected. Combined data from immunostaining of CD45, DAPI and fluorescence in situ hybridization (FISH) with chromosome 8 centromere (CEP8) probe were used to identify CTCs. Cells with features of CD45-/DAPI+/CEP8>2 were detected as CTCs. RESULTS:

The role of TGR5 as an onco-immunological biomarker in tumor staging and prognosis by encompassing the tumor microenvironment.

The relationship between G protein-coupled bile acid receptor 1 (TGR5, GPBAR1) and, specifically, cancer has been studied in in vivo and in vitro experiments, but there is still a lack of pan-cancer analysis to understand the prognostic significance and functioning mechanism of TGR5 in different cancer-driving oncogenic processes. Here, we used Gene Expression Integration, Human Protein Atlas, and The Cancer Genome Atlas (TCGA) to perform a pan-cancer analysis of the role of TGR5 in all 33 tumors. In all TCGA tumors, the TGR5 gene expression has been assessed, and we found that the high TGR5 g

Tumor treating fields suppress tumor cell growth and induce immunogenic cell death biomarkers in biliary tract cancer cell lines.

Tumor Treating Fields (TTFields) are low-intensity, intermediate-frequency alternating electric fields that exert antimitotic effects on cancer cells. This study is the first to evaluate the in vitro efficacy of TTFields on biliary tract cancer (BTC) cell lines HCCC-9810 and RBE, investigating their sensitivity to TTFields across varying frequencies and electric field intensities (100-200 kHz, 1.3-2.1 V/cm). The results demonstrated that a frequency of 150 kHz induced the most pronounced cytotoxic effects, significantly impairing clonogenicity and migratory capacity while inducing multipolar s

APOs as promising prognostic biomarkers: correlation with tumor-infiltrating leukocytes in endometrial cancer.

BACKGROUND: Apolipoproteins (APOs) are essentially structural and functional components of lipoproteins, which are composed of 22 members and their effects on certain types of cancer have been studied. However, their roles in endometrial cancer (EC), which is one of the most common malignant tumors in gynecology were unclear and rarely investigated. METHODS: We investigated the expression levels of APOs genes in EC. Furthermore, we explored the roles of APOs in prognostic value, and immune infiltrates in EC patients by using different bioinformatics databases. In-vitro experiments were also c

TREM2⁺ tumor-associated macrophages in cancer: Context-dependent biology, biomarkers, and precision immunotherapy.

Tumor-associated macrophages (TAMs) are abundant and functionally important components of the tumor microenvironment (TME), yet the molecular programs that distinguish pathogenic macrophage states from tissue-adaptive populations remain incompletely defined. Triggering receptor expressed on myeloid cells 2 (TREM2) has emerged as a key marker of specialized immunoregulatory macrophage states across multiple solid tumors. Accumulating evidence links TREM2⁺ macrophages to metabolic adaptation, immune evasion, and impaired antitumor immunity, although their biological functions vary substantially

Dual-mode microfluidic immunostaining device for diagnostic biomarkers detection and tumor microenvironment evaluation.

The limited availability of tissue samples from rare tumors poses a major barrier to advances in precise diagnosis, prognostic evaluation, and therapeutic research-challenges exemplified by primary central nervous system diffuse large B cell lymphoma (PCNS-DLBCL). Here, we developed a dual-mode microfluidic immunostaining (Dumi) device, which integrates diagnostic and research workflows into a single, automated platform, reducing tissue section consumption by more than 90%. Using just one to two slides, it enables both regional detection of biomarkers for diagnostic subtyping (≤16) and constru

Biomarkers of mRNA vaccine efficacy derived from mechanistic modeling of tumor-immune interactions.

The success of mRNA vaccines against infectious diseases such as COVID-19 has opened new avenues for their application in oncology. In cancer immunotherapy, mRNA vaccines-typically encapsulated in lipid nanoparticles (LNPs) 100-200 nm in size-enable delivery of tumor-specific antigens to activate immune responses. Here, we investigated the efficacy of mRNA vaccines in cancer by modeling tumor-immune interactions and tumor microenvironment (TME) dynamics to identify predictive biomarkers. Using a mechanistic mathematical model, we simulated tumor growth, immune cell dynamics, and vaccine pharma

Coupling of response biomarkers between tumor and peripheral blood in patients undergoing chemoimmunotherapy.

Platinum-based chemotherapy in combination with anti-PD-L1 antibodies has shown promising results in mesothelioma. However, the immunological mechanisms underlying its efficacy are not well understood and there are no predictive biomarkers to guide treatment decisions. Here, we combine time course RNA sequencing (RNA-seq) of peripheral blood mononuclear cells with pre-treatment tumor transcriptome data from the single-arm, phase 2 DREAM trial (N = 54). Single-cell RNA-seq and T cell receptor sequencing (TCR-seq) reveal that CD8+ T effector memory (TEM) cells with stem-like properties are more
