Bacillus sp. Abq, belonging to Bacillus cereus sensu lato, was isolated from an aquifer in New Mexico, USA and phylogenetically classified. The isolate possesses the unusual property of precipitating Pb(II) by using cysteine, which is degraded intracellularly to hydrogen sulfide (H2S). H2S is then exported to the extracellular environment to react with Pb(II), yielding PbS (galena). Biochemical and growth tests showed that other sulfur sources tested (sulfate, thiosulfate, and methionine) were not reduced to hydrogen sulfide. Using equimolar concentration of cysteine, 1 mM of soluble Pb(II) wa

Destruction of the periodontium is normally associated with periodontal disease, although many other factors, such as trauma, aging, infections, orthodontic tooth movement and systemic and genetic diseases, can contribute to this process. Strategies (such as guided tissue regeneration) have been developed to guide and control regeneration using bioresorbable membranes and bone grafts. Although effective to a certain point, these strategies have the problem that they are not predictable and do not completely restore the architecture of the original periodontium. To achieve complete repair and r

INTRODUCTION: Mineral trioxide aggregate (MTA) and Portland cement have been shown to be bioactive because of their ability to produce biologically compatible carbonated apatite. This study analyzed the interaction of MTA and white Portland cement with dentin in vivo.
METHODS: Seventy-two human dentin tubes were filled with MTA Branco, MTA BIO, and white Portland cement + 20% bismuth oxide (PC1) or PC1 + 10% of calcium chloride (PC2) and implanted subcutaneously in 18 rats at 4 sites from the dorsal area. Empty dentin tubes, implanted in rats of a pilot study, were used as control. After 30,

Biomineralization, the biologically controlled formation of minerals by living organisms, is central to the development, maintenance, and repair of dental hard tissues. This perspective summarizes the current understanding of the molecular and cellular mechanisms governing enamel and dentin mineralization, with particular emphasis on the regulatory roles of amelogenins, enamel matrix proteins, dentin matrix proteins, and non-collagenous phosphoproteins. The clinical implications for dental caries, periodontal bone loss, and dentin hypersensitivity are critically evaluated in the context of bio

Bivalve planktonic development is a critical phase during which larvae must secrete the first calcium carbonate shell, the prodissoconch I (PD I). As PD I formation is in close contact with seawater, this process can be negatively affected by adverse seawater carbonate chemistry. It is hypothesized that bivalves can regulate shell formation under environmental stress through biologically controlled biomineralization involving a complex extracellular shell proteome. However, the plasticity of this regulatory mechanism during PD I development is unknown. We assessed the PD I shell proteome of th

Titanium (Ti) alloys have been applied to biomedical implants for a long time. Although Ti alloys are biocompatible, efforts have been continuously made to improve their bone conductivity and osteogenesis for enhancing their performance. Silk fibroin (SF) is a natural biomaterial with excellent biomedical and mechanical properties, and hydroxyapatite (HAP) nanocomposites derived from SF are promising for producing "artificial bone" owing to their biomedical applicability and strong mechanical functions. Therefore, we built an SF coating on the surface of Ti-6Al-4V alloy, and then the incubated

Nature has evolved several unique biomineralization strategies to direct the synthesis and growth of inorganic materials. These natural systems are complex, involving the interaction of multiple biomolecules to catalyze biomineralization and template growth. Herein we describe the first report to our knowledge of a single enzyme capable of both catalyzing mineralization in otherwise unreactive solution and of templating nanocrystal growth. A recombinant putative cystathionine γ-lyase (smCSE) mineralizes CdS from an aqueous cadmium acetate solution via reactive H2S generation from l-cysteine an

Biomineralization, the generation of hard tissues of living organisms, is a process strictly regulated by hormones, enzymes and a range of regulatory proteins of which several resisted structural characterization thus far. Without actual generalizations, there have been scattered observations in the literature for the structural disorder of these proteins. To address this issue in general, we have collected SwissProt proteins involved in the formation of bone and teeth in vertebrates, annotated for biomineralization. All these proteins show an extremely high level of predicted disorder (with a

Vesicles play a key role in biomineralization pathways by mediating ion concentration and transport while maintaining cellular homeostasis. Whereas calcium-loaded vesicles have been extensively studied in organisms such as coccolithophores and vertebrates, comparatively little is known about those involved in mollusc shell mineralization. Using electron microscopy and electron energy loss spectroscopy (EELS), we analyzed the ultrastructure and composition of three vesicle types predominant near the shell's growth surface in cryofixed juvenile Mytilus galloprovincialis: bilayered vesicles, ribo

Biomineralization refers to the process by which organisms form inorganic minerals, predominantly through the deposition of calcium phosphates. Calcium ions (Ca2+) serve not only as a fundamental component of the mineral phase but also as key signaling messengers that actively regulate the efficiency and progression of this process. The endoplasmic reticulum (ER) and mitochondria are two major organelles responsible for calcium ion storage and regulation within cells. Contact sites between mitochondria and ER, also called mitochondria-ER contacts (MERCs) or mitochondria-associated ER membranes

Enzyme-regulated biomineralization offers precise spatiotemporal control over tissue mineralization, overcoming key limitations of conventional regenerative therapies. This review systematically examines the underlying biological mechanisms, focusing on enzymatic regulation of phosphate metabolism, mineralization regulators, and matrix stabilization that orchestrate hierarchical mineral deposition. Organic matrices facilitate nanoconfinement-driven nucleation and spatially controlled mineralization through biochemical functionalization. These fundamental mechanisms have inspired the developmen

Microbially induced carbonate precipitation (MICP) offers a promising biological approach to sequester atmospheric CO2 as stable mineral carbonates, mitigating climate change impacts. This perspective highlights the complexity underpinning prokaryote-driven biomineralization processes, emphasizing the necessity for holistic evaluation beyond simple carbonate formation. Key metabolic pathways such as carbonic anhydrase-mediated CO2 hydration, ureolysis, photosynthesis, and sulfate reduction contribute variably to mineral precipitation and the carbon footprint. Furthermore, calcium carbonate pol

Crystallization of monosodium urate monohydrate (MSU) leads to painful gouty arthritis. Despite extensive research it is still unknown how this pathological biomineralization occurs, which hampers its prevention. Here we show how inflammatory MSU crystals form after a non-inflammatory amorphous precursor (AMSU) that nucleates heterogeneously on collagen fibrils from damaged articular cartilage of gout patients. This non-classical crystallization route imprints a nanogranular structure to biogenic acicular MSU crystals, which have smaller unit cell volume, lower microstrain, and higher crystall

BACKGROUND: Kidney stone formers (SFs) are at increased risk of stroke, myocardial infarction, and atherosclerosis of the carotid and coronary arteries. These cardiovascular and urologic pathologies can result from ectopic biomineral deposition. The objectives of this study are: (I) to evaluate risk factors for ectopic biomineralization, and (II) to characterize the overall burden of ectopic minerals in known SFs compared to non-stone formers (NSFs) matched for these risk factors.
METHODS: Presence and quantity of biominerals at eight anatomic locations (abdominal aorta, common iliac arteries

Recently, there has been growing attention and effort to search for new microbicidal drugs which present different mode of action from those already existing, as an alternative to the global threat of fungal and bacterial multi drug resistance (MDR). Here we propose biological synthesis of SnO₂ nanoparticles using mammalian cells as an economic and ecofriendly platform. This presents a novel biogenic method for SnO₂ synthesis using metal binding peptides extracted from MCF-7 human cancer cells, which induces the biomineralization of SnO₂ nanoparticles. A series of electron donor functional gro

There is a growing interest in cell therapies using mesenchymal stromal cells (MSCs) for repairing bone defects. MSCs have the ability to differentiate into osteoprogenitors and osteoblasts as well as to form calcified bone matrix. However, the molecular mechanisms governing mineralization during osteogenic differentiation remain unclear. Non-collagenous proteins in the extracellular matrix are believed to control different aspects of the mineralization. Since osteocalcin is the most abundant non-collagenous bone matrix protein, the purpose of this study is to investigate the roles of osteocal

Diatoms, shells, bones and teeth are exquisite examples of well-defined structures, arranged from nanometre to macroscopic length scale, produced by natural biomineralization using organic templates to control the growth of the inorganic phase. Although strategies mimicking Nature have partially succeeded in synthesizing human-designed bio-inorganic composite materials, our limited understanding of fundamental mechanisms has so far kept the level of hierarchical complexity found in biological organisms out of the chemists' reach. In this letter, we report on the synthesis of unprecedented doub

Osteogenesis is a crucial process in bone injury repair where osteoblasts mineralize collagen to form new bone at the fracture site. For effective bone regeneration in comminuted fractures, it is essential to develop an osteogenic bone adhesive that can be applied at the time of fixation to temporarily immobilize fragmented bone pieces. We present a flowable, injectable adhesive that enables on-demand bonding of irregular fracture surfaces and, after photocuring, provides enhanced mechanical integrity while supporting osteogenesis. To meet these clinical requirements, we engineered a gelatin m

This review focuses on two main activities that have been the subject of research in our group during the last 15 years that concerns i) tracking of the iron(II) pathways in the ferritin cage to reach the ferroxidase site or the nucleation sites as well as identifying the interactions between the protein matrix and the iron(III) products; ii) identifying the binding mode of gold(I) metal compounds to the cage surface for their targeted delivery towards cancer cells. The resulting metal interaction sites in the protein cage can be categorized based on their preference for binding to hard or sof

Calcium ions may adversely affect struvite biomineralization, but the exact role remains elusive and the way to eliminate its adverse effect needs to be explored. Herein, Shewanella oneidensis MR-1 was selected to induce struvite biomineralization. The increase of Ca/Mg ratios from 0 to 2:1 resulted in a decrease of struvite yield from 98.6 mg to 0 mg. Calcium ions inhibited struvite crystallization by forming amorphous calcium phosphate (ACP) at the early stage of biomineralization. The increase of disodium ethylenediaminetetraacetate (EDTA) concentrations from 1 mM to 3 mM led to a rise of s
