Multimodal bioprinting of pigmented skin with algorithm-tuned control.

This study addresses critical technical challenges in fabricating functional pigmented skin models via 3D bioprinting through the synergistic integration of droplet-based deposition and precision motion control. A hybrid bioprinting strategy was developed to create multilayer biomimetic architectures: the dermal layer was fabricated through extrusion of gelatin methacryloyl-polyacrylamide (GelMA-PAM) composites, while the epidermal layer incorporated precisely patterned melanocyte-laden GelMA-PAM arrays deposited via microvalve technology, subsequently solidified and populated with keratinocyt

Small Joint Organoids 3D Bioprinting: Construction Strategy and Application.

Osteoarthritis (OA) is a chronic disease that causes pain and disability in adults, affecting ≈300 million people worldwide. It is caused by damage to cartilage, including cellular inflammation and destruction of the extracellular matrix (ECM), leading to limited self-repairing ability due to the lack of blood vessels and nerves in the cartilage tissue. Organoid technology has emerged as a promising approach for cartilage repair, but constructing joint organoids with their complex structures and special mechanisms is still challenging. To overcome these boundaries, 3D bioprinting technology al

Application of regenerative medicine and 3d bioprinting in urology.

In the last two decades, a new purpose has collected great efforts from scientists in all branches of medicine. It is about the possibility to make the body regenerate ill tissues and organs by itself with de right artificial stimuli or the construction of new functional organs to replace the damaged ones. This process comprises various interdisciplinary approaches to healthcare, such as tissue engineering, molecular medicine, biotechnology, and three-dimensional printing. Urologists have been remarkably active in this field of medicine called Regenerative Medicine. The searching of the differ

In situ handheld three-dimensional bioprinting for cartilage regeneration.

Articular cartilage injuries experienced at an early age can lead to the development of osteoarthritis later in life. In situ three-dimensional (3D) printing is an exciting and innovative biofabrication technology that enables the surgeon to deliver tissue-engineering techniques at the time and location of need. We have created a hand-held 3D printing device (biopen) that allows the simultaneous coaxial extrusion of bioscaffold and cultured cells directly into the cartilage defect in vivo in a single-session surgery. This pilot study assessed the ability of the biopen to repair a full-thicknes

Coaxial Bioprinting in Regenerative Medicine: Advances and Emerging Applications.

Coaxial extrusion-based bioprinting (EBB) is an emerging technology that enables the fabrication of biomimetic tissues with precise structural and biological complexities. This three-dimensional bioprinting technique utilizes specialized concentric nozzles to facilitate the simultaneous extrusion of distinct biomaterials, enabling the fabrication of layered constructs that closely resemble native tissues. Unlike traditional extrusion-based methods, coaxial printing allows for independent control over core and shell materials. This enables multimaterial integration, and tailored microenvironmen

Challenges and opportunities in generating microvasculature using bioprinting techniques.

Successful tissue transplantation requires the formation of microvasculature within engineered tissues. However, the formation of perfusable microvascular networks and their integration with host tissues after transplantation remain major challenges. We overview current bioprinting approaches for engineering microvascular structures and discuss their advantages and limitations. We further highlight applications of the Kenzan method developed in our laboratory for scaffold-free microvascular tissue fabrication and discuss future perspectives for vascularized tissue engineering.

Smart Bioinks for 4D Bioprinting: Requirements, Design, and Applications.

3D bioprinting is known for its high precision and reproducibility in fabricating complex and customized biomedical constructs. However, its applications are limited by their static nature; i.e., unlike native tissues, they cannot change shape or functionality over time. To overcome this, 4D bioprinting has emerged as a groundbreaking strategy by incorporating time as the fourth dimension, enabling dynamic structures that adapt in response to stimuli, thereby more accurately replicating living tissues. The success of 4D bioprinting hinges on the development of advanced smart bioinks, as their

3D-Bioprinting of Stromal Vascular Fraction for Gastrointestinal Regeneration.

Intestinal disorders such as inflammatory bowel diseases (IBDs), Crohn's disease, malabsorption syndromes, and gastrointestinal fistulae (GIFs) are often characterized by chronic inflammation, epithelial barrier disruption, impaired stromal remodeling, and defective angiogenesis. These multifactorial alterations hinder tissue repair and contribute to poor clinical outcomes, with limited efficacy from current therapeutic options. Despite recent advances in surgical and endoscopic techniques, current treatment options remain limited and are frequently accompanied by high morbidity and costs. In

Preparation of Corneal Tissue Matrix Bioink for 3D Bioprinting.

There are many protocols available to decellularize tissues for the preparation of bioink for 3D bioprinting purposes. Almost all the methods comprise multiple chemicals and enzymes in different combinations. Here we describe the usage of sodium chloride that enables the decellularization of corneal tissues from human and animal sources, which is a simple, rapid, and detergent-free method, unlike conventional decellularization protocols. The method described here is for cornea tissue decellularization and its digestion and bioink preparation for 3D bioprinting applications. We demonstrate the

Photocurable biomaterials labeled with luminescent sensors dedicated to bioprinting.

In the present study, we focused on the development and characterization of formulations that function as biological inks. These inks were doped with coumarin derivatives to act as molecular luminescent sensors that allow the monitoring of the kinetics of in situ photopolymerization in 3D (DLP) printing and bioprinting using pneumatic extrusion techniques, making it possible to study the changes in the system in real time. The efficiency of the systems was tested on compositions containing monomers: poly(ethylene glycol) diacrylates and photoinitiators: 2,4,6-trimethylbenzoyldi-phenylphosphina

Ionically annealed zwitterionic microgels for bioprinting of cartilaginous constructs.

Foreign body response (FBR) is a pervasive problem for biomaterials used in tissue engineering. Zwitterionic hydrogels have emerged as an effective solution to this problem, due to their ultra-low fouling properties, which enable them to effectively inhibit FBRin vivo. However, no versatile zwitterionic bioink that allows for high resolution extrusion bioprinting of tissue implants has thus far been reported. In this work, we introduce a simple, novel method for producing zwitterionic microgel bioink, using alginate methacrylate (AlgMA) as crosslinker and mechanical fragmentation as a microgel

Preliminary Application Research of 3D Bioprinting in Craniofacial Reconstruction.

INTRODUCTION: In recent years, 3-dimensional (3D) printing has been widely used in regenerative medicine research and other fields because of its ability to customize macroscopic morphology and precisely control microstructure. Polymer scaffolds are 1 of the commonly used 3D bioprinting materials for defect repair and have recently been a research focus. Our article explored the bone-formation accelerating effect of 3D-printed porous scaffold Poly(glycerol sebacate) [PGS] in the critical bone defect of an enhancing rabbit mandibular model. Also, we overview and summarize the classification of

Bioprinting of hair follicle germs for hair regenerative medicine.

Hair regenerative medicine is a promising approach to treat hair loss. The replication of in vivo tissue configurations and microenvironments, such as hair follicle germs, has been studied to prepare tissue grafts for hair regenerative medicine. However, such approaches should be scalable, because a single patient with alopecia requires thousands of tissue grafts. In this paper, we propose an approach for the scalable and automated preparation of highly hair-inductive tissue grafts using a bioprinter. Two collagen droplets (2 µL each) containing mesenchymal and epithelial cells were placed adj

FRESH Bioprinting of Dynamic Hydrazone-Cross-Linked Synthetic Hydrogels.

Dynamic covalent chemistry is an attractive cross-linking strategy for hydrogel bioinks due to its ability to mimic the dynamic interactions that are natively present in the extracellular matrix. However, the inherent challenges in mixing the reactive precursor polymers during printing and the tendency of the soft printed hydrogels to collapse during free-form printing have limited the use of such chemistry in 3D bioprinting cell scaffolds. Herein, we demonstrate 3D printing of hydrazone-cross-linked poly(oligoethylene glycol methacrylate) (POEGMA) hydrogels using the freeform reversible embed

Bioinks and Bioprinting Strategies for Skeletal Muscle Tissue Engineering.

Skeletal muscles play important roles in critical body functions and their injury or disease can lead to limitation of mobility and loss of independence. Current treatments result in variable functional recovery, while reconstructive surgery, as the gold-standard approach, is limited due to donor shortage, donor-site morbidity, and limited functional recovery. Skeletal muscle tissue engineering (SMTE) has generated enthusiasm as an alternative solution for treatment of injured tissue and serves as a functional disease model. Recently, bioprinting has emerged as a promising tool for recapitulat
