Outbreaks of infectious bursal disease (IBD), a highly contagious immunosuppressive disease of young chickens, are still reported globally despite vaccination efforts. This study investigated the genetic characteristics of infectious bursal disease virus (IBDV) from 26 reported outbreaks in 2019 in Nigeria. Nucleotide sequences of VP2 hypervariable (hvVP2) region (n=26) and VP1 (n=23) of Nigerian IBDVs were determined. Our results revealed the detection of reassortant strains with segment A related to very virulent IBDV (vvIBDV) having virulence marker (222A, 242I, 256I, 294I and 299S), wherea

Infectious bursal disease (IBD) is an acute, highly contagious, economically important disease of young chickens caused by Avibirnavirus, the infectious bursal disease virus (IBDV). The causative virus is highly resilient in poultry environments and vaccination is the most effective measure for IBDV control. However, the susceptibility of highly attenuated IBDV vaccine strains to neutralization by maternally derived antibodies (MDA) and overwhelming virulence of partly attenuated strains have limited the application of conventional live IBDV vaccines in pre- and posthatch chicks. Nevertheless,

Infectious bursal disease (IBD) remains a potential worldwide threat to the poultry industry despite several vaccination approaches. Because maternally derived antibodies (MDA) constitute a critical problem for IBD vaccination, we examined the efficiency of the intracloacal vaccination approach in breaking through MDA. Experiment 1 determined the ability of the vaccinal strain to multiply in the bursa of Fabricius (BF) in chicks with a high level of MDA. Using real-time polymerase chain reaction, we quantified the strain in the bursae of vaccinated and non-vaccinated chicks. Experiment 2 was p

BACKGROUND: Lates calcarifer, known as seabass in Asia and barramundi in Australia, is a widely farmed species internationally and in Southeast Asia and any disease outbreak will have a great economic impact on the aquaculture industry. Through disease investigation of Asian seabass from a coastal fish farm in 2015 in Singapore, a novel birnavirus named Lates calcarifer Birnavirus (LCBV) was detected and we sought to isolate and characterize the virus through molecular and biochemical methods.
METHODS: In order to propagate the novel birnavirus LCBV, the virus was inoculated into the Bluegill

Major progress has been achieved since the first historical review of infectious bursal disease (H. N. Lasher and V. S. Davis, Avian Diseases, Vol. 41, pp. 11-19; 1997), much of it between 1977 and 2005. Significant findings in the 1980s were the presence of serotype 2 of infectious bursal disease virus (IBDV) and the diversity of antigenic and immunogenic types of IBDV. In the late 1980s, very virulent IBDV strains emerged and became widespread in many countries by the late 1990s. Soon after the discovery of the antigenic variants, specific commercial vaccines were developed and used successf

BACKGROUND: Since 2018, atrophy of the bursa has been found among vaccinated chickens with high antibody titres against infectious bursal disease virus (IBDV) in Fujian, China, suggesting poor vaccine efficacy against circulating IBDV strains.
METHODS: Novel IBDV strains were isolated, and vp2 and vp1 genes were sequenced and used to carry out phylogenetic analysis. Pathogenicity was investigated using 21-day-old specific pathogen-free (SPF) chickens. In addition, the effectiveness of current commercial vaccines used in China was evaluated against the isolated novel IBDV strains.
RESULTS: Si

Twenty chicks, 12 turkey poults and 10 ducklings, all 5 weeks old were infected with 2 x 10(3.5) chick LD(50) IBD virus to determine the course of the virus in the 3 poultry species. Uninfected control birds were kept separately. Two infected and 2 control birds/species were euthanized at time intervals between 3 and 168 hours post infection (pi). Sections of thymus, bursa of Fabricius, spleen, liver, kidney, proventriculus and ceacal tonsil were stained for the detection of IBD virus antigen using immunoperoxidase technique. IBD virus antigen positive cells stained reddish-brown and the amoun

Infectious pancreatic necrosis virus (IPNV) is a bisegmented double-stranded RNA virus belonging to the family Birnaviridae, genus Aquabirnavirus, which is a major viral pathogen of salmonid fish. The virus infects wild and cultured salmonids, causing high mortality in juvenile trout and salmon. A highly sensitive and specific real-time RT-PCR assay using the fluorogenic dye SYBR((R)) Green I was developed for the detection and quantitation of IPNV in rainbow trout (Oncorhynchus mykiss). Rainbow trout were infected experimentally with IPNV in the laboratory by injection or immersion and then p

The pathogenesis of highly virulent infectious bursal disease (IBD) virus (IBDV) infection was studied using 6-week-old intact and 5-week-old bursectomized chickens inoculated with highly virulent strain 90-11 or reference strain I. Chickens inoculated with 10(0.7) EID50 of strain 90-11 showed neither clinical signs nor lesions during the 4-day observation period. In contrast, birds inoculated with 10(2.7) or 10(4.7) EID50 developed severe clinical IBD, as well as gross and histologic lesions, typical of IBD, and produced IBDV antigen demonstrable by immunostaining in the bursa of Fabricius (B

Infectious pancreatic necrosis virus (IPNV) can cause devastating disease in Atlantic salmon (Salmon salar). IPNV has a broad host range and may threaten other aquaculture species. Understanding interspecies transmission of IPNV is crucial for protecting the aquaculture industry. With the expansion of fish farming (in Norway), it is important to assess whether a pathogen can transmit from one fish species to another and cause disease. We investigated whether IPNV-infected Atlantic cod can shed IPNV, leading to infection in other fish important to Norwegian aquaculture: halibut, salmon and lump

Infectious bursal disease (IBD), caused by the infectious bursal disease virus (IBDV), remains a major threat to global poultry production. The viral capsid protein VP2 is the primary protective antigen; however, conventional vaccines frequently provide only limited efficacy. In this study, we generated recombinant Eimeria acervulina strains expressing VP2 fused to the molecular adjuvant CTA1-DD (Cholera Toxin A1-D-domain dimer). Two transgenic strains were established: Ea-CVD (single-copy VP2) and Ea-VCVD (dual-copy VP2). Stable expression of the fusion protein was confirmed across multiple d

Infectious bursal disease virus (IBDV) causes an acute and highly contagious infectious disease characterized by severe immunosuppression, causing great economic losses to the poultry industry globally. Over the past 30 years, this disease has been well controlled through vaccination and strict biosafety measures. However, novel variant IBDV strains have emerged in recent years, posing a new threat to the poultry industry. Our previous epidemiological survey showed that few novel variant IBDV strains had been isolated from chickens immunized with the attenuated live vaccine W2512-, suggesting

Vaccination is one of effective means to prevent viral infectious diseases in poultry. However, the functions of circulating miRNAs in immune response remain unknown. In this study, miR-155, a key factor in the regulation of immune function, was selected to study its expression, potential function and mechanism in the 12-day-old chicken immune responses to three vaccines (avian influenza virus inactivated vaccine, Newcastle disease virus attenuated vaccine and infectious bursal disease virus attenuated vaccine), respectively. The experiment aimed to explore the relationships between the expres

In today's aquaculture of Atlantic salmon (Salmo salar L.), a majority of viral disease outbreaks occur after seawater transfer. A relevant question is how the parr-smolt transformation influences the efficacy of viral vaccines and the innate resistance against viral diseases. In this study, vaccinated and unvaccinated A. salmon parr were exposed to different photoperiodic regimens (1-, 3- or 6-week continuous light-WCL). Fish groups at different stages in the smoltification process were induced, as demonstrated by differences in morphological and physiological smolt parameters. At the time of

Studies have shown that infectious bursal disease virus (IBDV) infects lymphoid cells, mainly B cells and macrophages. This study was aimed to examine the involvement of chicken splenic-derived dendritic cells (ch-sDCs) in specific-pathogen-free chickens following inoculation with IBDV vaccine strain (D78) and a very virulent (vv) strain (UPM0081). Following IBDV infection, enriched activated ch-sDCs were collected by using the negative selection method and were examined based on morphology and immunophenotyping to confirm the isolation method for dendritic cells (DCs). The presence of IBDV on

Atlantic cod Gadus morhua L. juveniles weighing 40 g were challenged with infectious pancreatic necrosis virus (IPNV) by intraperitoneal (i.p.) or intramuscular (i.m.) injection or by bath. The amount of infectious virus was determined over 6 wk in head kidney, heart and pylorus tissues. No mortality or clinical signs were observed in either of the challenged groups. However, 6 wk after challenge virus was still present in the fish, which shows that IPNV can persist asymptomatically in cod. I.p. and i.m. injections were the most efficient routes of challenge giving the highest virus recovery.

The absence of an immediately deployable, broad-spectrum antiviral remains a critical vulnerability in global pandemic preparedness. Host-directed agents that activate innate immunity offer a pathogen-agnostic strategy, yet no such therapy is currently stockpiled or authorized for emergency use. Infectious Bursal Disease Virus (IBDV)-a non-replicating avian dsRNA vaccine virus with a 60-year safety record in poultry-induces robust interferon responses in mammals and has been administered orally in marmosets and more than 50 human patients with hepatitis A, B, C, SARS-CoV-2, and herpes zoster i

Infectious bursal disease (IBD) can result in significant immunosuppression and bursal damage in 3-6 week chickens. To investigate the role of important genes from chicken during IBD virus (IBDV) infection, Jinghong laying hens were used to clarify the pathways of immune response. The transcriptional profiles of cecal tonsil of chickens were performed by high-throughput sequencing at the first day and third day post IBDV infection (dpi) and the transcriptional levels of three immunity-related genes, namely IPMK, TAB3, and ZC3H12A, were confirmed in vitro by qPCR. The results showed that 1731 d

Recently, outbreaks of infectious pancreatic necrosis (IPN) in worldwide rainbow trout farms caused by IPN virus (IPNV) strains belonging to genogroup 1 and genogroup 5 are reported. In this study, formaldehyde-inactivated vaccines supplemented with or without GEL 02 PR adjuvant were developed by using both genogroups IPNV strains and were intraperitoneally injected into rainbow trout. At 30 days post-vaccination, the viral loads of IPNV challenged rainbow trout (Oncorhynchus mykiss) in the vaccine groups were significantly decreased compared with those in the PBS group (P < 0.05), about 2.4 l

Novel variant infectious bursal disease virus (nVarIBDV) infection causes mortality, morbidity, and immunosuppression in chickens worldwide. This study explored the pathogenicity, immunogenicity, and nVarIBDV load in commercial broiler chickens. Sixty 3-wk-old chicks were divided into nVarIBDV (28) and control (32) groups. The nVarIBDV group was inoculated with 1.0 ml (106.75 median egg infective dose [EID50]) of nVarIBDV through the oral and ocular routes at 0 days postinoculation (dpi), while the control group was uninoculated. The control group was sampled on 0 dpi, and both groups were sam
