Effect of di(n-butyl) phthalate on the blood-testis barrier during puberty onset.

Di(n-butyl) phthalate (DBP) is considered a substance of serious concern because of its reproductive toxicity and endocrine-disrupting properties. Exposure to DBP causes morphological and functional changes in the male reproductive system of birds and mammals. However, there are no detailed reports on the effects of DBP on the Sertoli cell and junctional complexes of the blood-testis barrier (BTB) in birds. The present study investigated dose-related ultrastructural changes in Sertoli cells and junctional complexes of the BTB in adult Japanese quail (Coturnix coturnix japonica) exposed to DBP

Arsenic interferes with spermatogenesis involving Rictor/mTORC2-mediated blood-testis barrier disruption in mice.

Ingestion of arsenic interferes with spermatogenesis and increases the risk of male infertility, but the underlying mechanism remines unclear. In this study, we investigated spermatogenic injury with a focus on blood-testis barrier (BTB) disruption by administrating 5 mg/L and 15 mg/L arsenic orally to adult male mice for 60 d. Our results showed that arsenic exposure reduced sperm quality, altered testicular architecture, and impaired Sertoli cell junctions at the BTB. Analysis of BTB junctional proteins revealed that arsenic intake downregulated Claudin-11 expression and increased protein le

Uropathogenic Escherichia coli Infection Compromises the Blood-Testis Barrier by Disturbing mTORC1-mTORC2 Balance.

The structural and functional destruction of the blood-testis barrier (BTB) following uropathogenic E. coli (UPEC) infection may be a critical component of the pathologic progress of orchitis. Recent findings indicate that the mammalian target of the rapamycin (mTOR)-signaling pathway is implicated in the regulation of BTB assembly and restructuring. To explore the mechanisms underlying BTB damage induced by UPEC infection, we analyzed BTB integrity and the involvement of the mTOR-signaling pathway using in vivo and in vitro UPEC-infection models. We initially confirmed that soluble virulent f

Adenylyl cyclase 3 deficiency results in dysfunction of blood-testis barrier during mouse spermiogenesis.

Human infertility has become a global medical and social health problem. Mice deficient in type 3 adenylyl cyclase (AC3), a key enzyme that synthesizes cyclic adenosine monophosphate (cAMP), develop male infertility, although the underlying molecular mechanisms remain unknown. We performed a label-free quantitative (LFQ) proteomics analyses to identify testicular differentially expressed proteins (DEPs) and their respective biological processes. Furthermore, histological examination demonstrated that AC3 deficiency in mice led to mild impairment of spermatogenesis, including the thinning of se

Naproxen and Ibuprofen Exposure Alter the Blood-Testis Barrier in a Novel In Vitro Model.

Semen parameters, including sperm counts, have rapidly declined in men across the globe over the last five decades. Although this decline remains unexplained, lifestyle factors may affect male fertility. Recently, several studies highlighted a potential link between non-steroidal anti-inflammatory drug (NSAID) usage, such as naproxen and ibuprofen, and declining male fertility. However, the mechanisms by which these common analgesics affect male fertility, including their effects on the blood-testis barrier (BTB), remain poorly characterized. Utilizing an in vitro rhesus macaque non-human prim

The effects of fine particulate matter on the blood-testis barrier and its potential mechanisms.

With the rapid expansion of industrial scale, an increasing number of fine particulate matter (PM2.5) has bringing health concerns. Although exposure to PM2.5 has been clearly associated with male reproductive toxicity, the exact mechanisms are still unclear. Recent studies demonstrated that exposure to PM2.5 can disturb spermatogenesis through destroying the blood-testis barrier (BTB), consisting of different junction types, containing tight junctions (TJs), gap junctions (GJs), ectoplasmic specialization (ES) and desmosomes. The BTB is one of the tightest blood-tissue barriers among mammals,

Black Phosphorus Nanosheets Penetrate the Blood-Testis Barrier and Induce Reproductive Toxicity in Male Mice.

Black phosphorus nanosheets (BPNSs), as an emerging subclass of two-dimensional (2D) nanomaterials, have exhibited extensive applicability in electronics, optoelectronics, and medical sciences. However, its expanding application in diverse fields has raised concerns about inadvertent environmental release, which might lead to potential ecological and health risks. Herein, mice were continuously exposed at doses of 0.1 mg/kg/day (possible acceptable daily intake dose, ADI) and 1 mg/kg/day (possible occupational exposure dose) to BPNSs for 28 days. Using hyperspectral imaging and Evans blue fluo

Icariin alleviates triptolide-induced testicular vacuolization via modulating germline ferroptosis and blood-testis barrier integrity.

INTRODUCTION: Environmental factors mediated testicular vacuolization injury is a prevalent occurrence, and its etiology and mitigation strategies have long remained inadequately elucidated. METHODS: A testicular injury model was established using Triptolide (TP), with Icariin (ICA) employed as a rescue compound. Function and mechanism investigations primarily employed testicular coefficient and sperm concentration, H&E staining, immunofluorescence staining, and western blot analysis. Single-cell RNA-sequencing (scRNA-seq) predominantly examined the impacts of TP and ICA on various cell popul

Inhibition of LncRNA H19 Attenuates Testicular Torsion-Induced Apoptosis and Preserves Blood-Testis Barrier Integrity.

Testicular torsion is a common emergency in adolescents, and can lead to severe ischemia reperfusion injury (IRI). LncRNA H19 has been shown to increase during ischemia, but its role in testicular IRI remains unknown. Focusing on this research gap, we utilized H19 biallelic mutant mice and Sertoli cell line (TM4) to construct in vivo and in vitro models of ischemia/reperfusion (I/R) and oxygen-glucose deprivation/reperfusion (OGD/R). Compared to WT I/R mice, H19-/- I/R mice showed milder tissue disorganization and cell loss, with a more intact blood-testis barrier (BTB). The cell viability dec

Inhibition of miR-143-3p Restores Blood-Testis Barrier Function and Ameliorates Sertoli Cell Senescence.

Due to the increasing trend of delayed childbirth, the age-related decline in male reproductive function has become a widely recognized issue. Sertoli cells (SCs) play a vital role in creating the necessary microenvironment for spermatogenesis in the testis. However, the mechanism underlying Sertoli cell aging is still unclear. In this study, senescent Sertoli cells showed a substantial upregulation of miR-143-3p expression. miR-143-3p was found to limit Sertoli cell proliferation, promote cellular senescence, and cause blood-testis barrier (BTB) dysfunction by targeting ubiquitin-conjugating

Ferroptosis is critical for phthalates driving the blood-testis barrier dysfunction via targeting transferrin receptor.

The global rate of human male infertility is rising at an alarming rate owing to environmental and lifestyle changes. Phthalates are the most hazardous chemical additives in plastics and have an apparently negative impact on the function of male reproductive system. Ferroptosis is a recently described form of iron-dependent cell death and has been linked to several diseases. Transferrin receptor (TfRC), a specific ferroptosis marker, is a universal iron importer for all cells using extracellular transferrin. We aim to investigate the potential involvement of ferroptosis during male reproductiv

Exogenous leptin affects sperm parameters and impairs blood testis barrier integrity in adult male mice.

BACKGROUND: Serum leptin levels are augmented in obese infertile men and in men with azoospermia. They also correlate inversely with sperm concentration, motility and normal forms. The mechanisms underlying the adverse effects of excess leptin on male reproductive function remain unclear. The present study aimed to evaluate the effects of exogenous leptin on sperm parameters in mice and to explore the underlying mechanisms. METHODS: We treated normal adult male mice with saline, 0.1, 0.5 or 3 mg/kg leptin daily for 2 weeks. After treatment, serum leptin levels, serum testosterone levels, sper

Sodium fluoride and sulfur dioxide affected male reproduction by disturbing blood-testis barrier in mice.

Fluoride and sulfur dioxide (SO2), two well-known environmental toxicants, have been implicated to have adverse effects on male reproductive health in humans and animals. The objective of this study to investigate if the BTB is one of the pathways that lead to reproductive toxicity of sodium fluoride and sulfur dioxide alone or in combination, in view of the key role of blood testis barrier (BTB) in testis. The results showed that a marked decrease in sperm quality, and altered morphology and ultrastructure of BTB in testis of mice exposure to fluoride (100 mg NaF/L in drinking water) or/and s

Cigarette smoke exposure disrupts the blood-testis barrier and negatively impacts reproductive capacity in mice.

Maintaining normal testicular structure and function is closely related to the blood-testis barrier (BTB). Meanwhile, cigarette smoke extract (CSE) affects testicular function. However, whether CSE mediates BTB damage and the underlying mechanism(s) are unclear. This study investigates the effects of CSE on the BTB in mice by focusing on changes in hormone levels. CSE was prepared and administered intranasally at different concentrations to 6-8-week-old male Balb/c mice for five weeks. After cigarette smoke components successfully entered and accumulated in the mice, the seminiferous tubules o

Glucagon-like peptide-1 analog, liraglutide, alters blood-testis barrier permeability in juvenile healthy rats.

Glucagon-like peptide-1 (GLP-1) analogue liraglutide has recently been approved for treating obese children. However, little is known about liraglutide's effect on testicular function during the juvenile developmental period, particularly regarding the blood-testis barrier (BTB) formation. This study analyses the impact of liraglutide treatment on BTB function in young rats and its consequences in adulthood. Rats received a daily dose of liraglutide from postnatal day (Pnd) 20 to 33, the period when a functional BTB is formed. At Pnd 34, liraglutide promoted a significant increase in permeable

Blood-testis barrier: a review on regulators in maintaining cell junction integrity between Sertoli cells.

The blood-testis barrier (BTB) is formed adjacent to the seminiferous basement membrane. It is a distinct ultrastructure, partitioning testicular seminiferous epithelium into apical (adluminal) and basal compartments. It plays a vital role in developing and maturing spermatocytes into spermatozoa via reorganizing its structure. This enables the transportation of preleptotene spermatocytes across the BTB, from basal to adluminal compartments in the seminiferous tubules. Several bioactive peptides and biomolecules secreted by testicular cells regulate the BTB function and support spermatogenesis

Adverse effects and potential mechanisms of polystyrene microplastics (PS-MPs) on the blood-testis barrier.

Microplastics (MPs) are defined as plastic particles or fragments with a diameter of less than 5 mm. These particles have been identified as causing male reproductive toxicity, although the precise mechanism behind this association is yet to be fully understood. Recent research has found that exposure to polystyrene microplastics (PS-MPs) can disrupt spermatogenesis by impacting the integrity of the blood-testis barrier (BTB), a formidable barrier within mammalian blood tissues. The BTB safeguards germ cells from harmful substances and infiltration by immune cells. However, the disruption of t
