Economic analysis of animal disease outbreaks--BSE and Bluetongue disease as examples.

Although there is a long tradition of research on animal disease control, economic evaluation of control measures is rather limited in veterinary medicine. This may, on the one hand, be due to the different types of costs and refunds and the different people and organizations bearing them, such as animal holders, county, region, state or European Union, but it may also be due to the fact that economic analyses are both complex and time consuming. Only recently attention has turned towards economic analysis in animal disease control. Examples include situations, when decisions between different

Bovine infection with bluetongue virus with special emphasis on European serotype 8.

Bluetongue virus (BTV) is an arthropod-borne virus infecting domestic and wild ruminants. Infection in cattle is commonly asymptomatic and characterised by a long viraemia. Associated with the emergence and the recrudescence of BTV serotype 8 (BTV-8) in Northern and Central Europe, remarkable differences have been noticed in the transmission and in the clinical expression of the disease, with cattle showing clinical illness and reproductive disorders such as abortion, stillbirth and fetal abnormalities. Several investigations have already indicated the putative ability of the European BTV-8 st

The preparation of purified bluetongue virus group antigen for use as a diagnostic reagent.

Soluble group antigen isolated from bluetongue virus infected BHK cells has been shown to be a single protein with a molecular weight of 38,000 and to comigrate on polyacrylamide gels with protein 7 isolated from virus particles. Purification by exclusion chromatography and isoelectric focussing yields a protein that appears to be antigenicaly identical to the native group antigen found in virus harvests; it is non infectious for sheep and is thought to be largely free from host cell and other virus specified proteins. It is suggested that this antigen could be used in diagnostic procedures wi

Inhibition of bluetongue and Colorado tick fever orbiviruses by selected antiviral substances.

The effects of four ribonucleic acid virus inhibitors were evaluated in cell cultures and in mice to determine inhibitory effects against bluetongue virus and Colorado tick fever virus (CTFV). Test compounds included 1-beta-D-ribofuranosyl-1,2,4-triazole-3-carboxamide (ribavirin), 3-deazaguanine, 3-deazauridine, and 9-(S)-(2,3-dihydroxypropyl)adenine. Ribavirin-2',3',5'-triacetate (ribavirin triacetate) was evaluated in vivo against CTFV. Inhibition of cytopathic effect and plaque reduction were used to evaluate antiviral activity. In cytopathic effect inhibition studies, bluetongue virus was

Increased Virulence of Culicoides Midge Cell-Derived Bluetongue Virus in IFNAR Mice.

Bluetongue (BT) is a Culicoides midge-borne hemorrhagic disease affecting cervids and ruminant livestock species, resulting in significant economic losses from animal production and trade restrictions. Experimental animal infections using the α/β interferon receptor knockout IFNAR mouse model and susceptible target species are critical for understanding viral pathogenesis, virulence, and evaluating vaccines. However, conducting experimental vector-borne transmission studies with the vector itself are logistically difficult and experimentally problematic. Therefore, experimental infections are

Bluetongue virus serotype 26: infection kinetics and pathogenesis in Dorset Poll sheep.

Bluetongue virus serotype 26 (BTV-26) has recently been isolated from sheep in Kuwait. The aim of this study was to assess the pathogenicity and infection kinetics of BTV-26 in Dorset Poll sheep. Six sheep were experimentally infected with BTV-26 and samples taken throughout the study were used to determine the kinetics of infection using a pan specific BTV real time RT-PCR assay and two group specific ELISAs. Five of the six sheep showed mild clinical signs characteristic of bluetongue including conjunctivitis, reddening of the mouth mucosal membranes, slight oedema of the face and nasal disc

Bluetongue in China: Current Status of Viruses, Vectors, Detection Methods, and Vaccines.

Bluetongue (BT) is a vector-borne viral disease caused by the bluetongue virus (BTV), which can affect a variety of wild and domestic ruminants. Due to its significant impact on ruminant health and national economies, BT is classified as a notifiable multispecies disease by the World Organization for Animal Health (WOAH). In China, BT is listed as a Class II multispecies animal disease. This article provides a comprehensive review of the distribution of BTV and its primary insect vector, Culicoides, in China. Since BTV was first reported in China in 1979, BTV antibody-positive samples have bee

Bluetongue and epizootic haemorrhagic disease in wildlife with emphasis on the South American scenario.

Bluetongue and epizootic hemorrhagic disease affect domestic ruminants and cervids. However, other species may act as pathogen carriers in the transition of bluetongue virus (BTV) and epizootic hemorrhagic disease virus (EHDV). The wild species affected by these diseases manifest a variable range of clinical signs and lesions, and while some species appear to be extremely susceptible, showing high levels of mortality, some are resistant to these pathogens, acting as potential reservoirs of these orbiviruses.  The purpose of the following review is to describe the clinical and pathological mani

Mechanistic insights into Bluetongue virus immunodynamics: a Bayesian within-host modelling approach.

Bluetongue virus (BTV) is a vector-transmitted orbivirus that infects ruminant livestock and wildlife, causing substantial economic losses owing to its impact on animal health and trade. Despite extensive study, the mechanisms underlying infection and immune dynamics remain unclear. Here, we develop and calibrate a mechanistic within-host model of BTV infection in experimentally infected sheep to quantify the contributions of innate and adaptive immune responses to viral control. The model integrates longitudinal data on viral RNA dynamics reverse transcriptase quantitative polymerase chain re

A Field-Applicable Method for Bluetongue Virus Detection Using Filter Paper Cards.

The diagnosis of Bluetongue Virus (BTV) in Argentina is primarily conducted via RT-qPCR and faces challenges due to the limited number of diagnostic laboratories, many of which are located in regions where BTV is exotic and far from primary sample collection areas. As a result, blood samples must be transported over considerable distances for analysis. This poses significant logistical challenges due to the specific transport conditions that are required. Consequently, the development of methodologies that optimize sample collection, preservation, and safe transport is of vital importance. The

Serological Cross-Reactions between Expressed VP2 Proteins from Different Bluetongue Virus Serotypes.

Bluetongue (BT) is a severe and economically important disease of ruminants that is widely distributed around the world, caused by the bluetongue virus (BTV). More than 28 different BTV serotypes have been identified in serum neutralisation tests (SNT), which, along with geographic variants (topotypes) within each serotype, reflect differences in BTV outer-capsid protein VP2. VP2 is the primary target for neutralising antibodies, although the basis for cross-reactions and serological variations between and within BTV serotypes is poorly understood. Recombinant BTV VP2 proteins (rVP2) were expr

Clinical observations from a study comparing two different treatments for bluetongue in sheep.

In September 2023, the official national reference laboratory confirmed the presence of BTV serotype 3 in The Netherlands. In the following weeks, it appeared that the infection caused very severe clinical signs in sheep, but it was challenging for the involved veterinary clinicians to select appropriate labeled drugs to treat them. The objective of this study was therefore to document the clinical presentation and survival outcomes of sheep affected by bluetongue and to evaluate the effects of a treatment protocol with and without antihistamines. From a group of 83 sheep with acute and clear

Plant-produced Bluetongue chimaeric VLP vaccine candidates elicit serotype-specific immunity in sheep.

Bluetongue (BT) is a hemorrhagic non-contagious, biting midge-transmitted disease of wild and domestic ruminants that is caused by bluetongue virus (BTV). Annual vaccination plays a pivotal role in BT disease control in endemic regions. Due to safety concerns of the current BTV multivalent live attenuated vaccine (LAV), a safe efficacious new generation subunit vaccine such as a plant-produced BT virus-like particle (VLP) vaccine is imperative. Previously, homogenous BTV serotype 8 (BTV-8) VLPs were successfully produced in Nicotiana benthamiana plants and provided protective immunity in sheep

Global Seroprevalence of bluetongue virus in camels: a first systematic review and meta-analysis.

Bluetongue (BT), caused by the bluetongue virus (BTV), is a non-contagious, vector-borne disease that primarily affects domestic and wild ruminants worldwide. Although recent evidence indicates BTV infection in camels, the epidemiological role of camels in the transmission and spread of the disease remains poorly understood. While some meta-analyses have evaluated the seroprevalence of BTV in traditional hosts, a comprehensive global meta-analysis specifically focusing on camels is lacking. This systematic review and meta-analysis aimed to estimate the global seroprevalence of BTV in camels an

Mitigation of the Risk of Importing Akabane and Exotic Bluetongue Virus in Ruminant-Sourced Sera.

Akabane virus is an arbovirus of the Bunyaviridae family that is endemic in certain tropical and subtropical regions, including parts of Asia, Africa, the Middle East, and Australia. Since Australian-sourced fetal bovine serum (FBS) is a preferred reagent for use in regulated cell culture applications (including but not limited to manufacturing processes for human and veterinary biologics), the importation of FBS from Australia represents a risk to United States livestock in the form of possible introduction of Akabane virus. This risk is currently mitigated, per United States Department of Ag

A duplex real-time RT-PCR for the detection of bluetongue virus in bovine semen.

The control measures prescribed by the World Organization for Animal Health (OIE) for international trade in extended semen implicate repeated free testing of the donor's blood for bluetongue virus (BTV). The aim of this study was to validate a real-time RT-PCR for the direct testing of semen for artificial insemination (AI). The amplification of the BTV target was combined with an internal control target in duplex format. Optimal RNA recovery and efficient removal of PCR inhibitors was established using Trizol-based RNA extraction. The total assay was highly repeatable, the preliminary analys

A comparison of laboratory and 'wild' strains of bluetongue virus--is there any difference and does it matter?

Bluetongue (BT) viruses (BTV) have been propagated in laboratory culture systems for more than 50 years, especially for the production of vaccines. An important outcome of the adaptation of these viruses to laboratory culture is attenuation of their virulence. As a consequence of this modification, it has been possible to produce vaccines that have reduced the clinical impact of BT in a number of countries. Unfortunately, the adaptation of these viruses has also introduced undesirable properties. In particular, modified live BT vaccine viruses have a high capacity to cross the ovine placenta a

Isolation of a capsid protein of bluetongue virus that induces a protective immune response in sheep.

A method to purify the neutralization specific antigen of bluetongue virus P2 in large amounts has been developed. The purified protein is free from virus-specified or cellular contaminants and its immunological specificity has been preserved. The purification is based on the observation that protein P2 can be dissociated from the virion by treatment with monovalent or divalent salts. The salt concentration required to solubilize the outer capsid proteins is pH dependent and in general decreases with a decrease in pH. P2 purified by extraction from polyacrylamide gels does not induce immune-pr
