Hyperbaric Oxygen Promotes Proximal Bone Regeneration and Organized Collagen Composition during Digit Regeneration.

Oxygen is critical for optimal bone regeneration. While axolotls and salamanders have retained the ability to regenerate whole limbs, mammalian regeneration is restricted to the distal tip of the digit (P3) in mice, primates, and humans. Our previous study revealed the oxygen microenvironment during regeneration is dynamic and temporally influential in building and degrading bone. Given that regeneration is dependent on a dynamic and changing oxygen environment, a better understanding of the effects of oxygen during wounding, scarring, and regeneration, and better ways to artificially generate

Chemical-Biological Fixation for Enhanced Bone Augmentation in Guided Bone Regeneration.

Stable fixation of barrier membranes is essential for achieving effective bone augmentation during guided bone regeneration (GBR). Conventional fixation techniques, such as membrane nails and sutures, provide suboptimal results and may lead to the displacement of bone substitute materials, eventually compromising regenerative outcomes. To address this, a thermosensitive injectable adhesive, M2N2ACa, was developed to solidify upon exposure to a 36°C wet environment. This material was found to facilitate robust adhesion between barrier membranes and moist bone surfaces at 36°C, achieving an adhe

Advances on T cell immunity in bone remodeling and bone regeneration.

Bone remodeling and bone regeneration are essential for preserving skeletal integrity and maintaining mineral homeostasis. T cells, as key members of adaptive immunity, play a pivotal role in bone remodeling and bone regeneration by producing a range of cytokines and growth factors. In the physiological state, T cells are involved in the maintenance of bone homeostasis through interactions with mesenchymal stem cells, osteoblasts, and osteoclasts. In pathological states, T cells participate in the pathological process of different types of osteoporosis through interaction with estrogen, glucoc

Spontaneous bone regeneration in surgically induced bone defects in young rabbits.

Defects of the femoral shaft (15%, 20%, 25%, and 30% of the femoral length) with intact periosteum were made in young rabbits to investigate differences in union time. The mean union time was 7.3, 7.1, 7.4, and 7.0 weeks, respectively and there was no significant difference ( >0.05) between the groups. The mean healing indices were 6.7, 4.6, 3.9, and 3.1 weeks/cm, respectively and these were significantly different ( <0.05) between the 15% bone defect group and the other bone defect groups. These results suggested that union time was not affected by the amount of bone defect. Femoral lengtheni

3D Printed Patient-Specific Resorbable Bone Scaffolds for Alveolar Bone Regeneration.

BACKGROUND: Alveolar bone loss following trauma, periodontal disease, congenital anomalies, or tooth extraction poses a major challenge for oral rehabilitation, especially in implant dentistry. Traditional grafting techniques using autografts, allografts, or xenografts provide limited predictability due to issues of resorption, donor morbidity, and immune incompatibility. Advances in three-dimensional (3D) printing now enable preparation of patient-specific biodegradable scaffolds designed using 3D imaging, like cone beam computed tomography (CBCT) and computer-aided design (CAD), allowing pre

Zn-Sr-sintered true bone ceramics enhance bone repair and regeneration.

Bone defects are one of the toughest challenges faced by orthopedic surgeons worldwide, especially at critical sizes, which are caused by severe trauma, malignancy, or congenital disease. The ideal bone tissue-engineered scaffold for bone regeneration is the one that has good osteoconductivity, osteoinductivity, pore structure, and antibacterial properties. Metal ions have been recognized in recent years to be essential regulators of bone metabolism, and they are widely used for bone tissue engineering. In particular, zinc ions are of interest because of their ideal biocompatibility, osteogene

Bone regeneration strategies with bone marrow stromal cells in orthopaedic surgery.

Bone is the most transplanted tissue human with 1 million procedures every year in Europe. Surgical interventions for bone repair are required for varied reasons such as trauma resulting non-union fractures, or diseases including osteoporosis or osteonecrosis. Autologous bone grafting is the gold standard in bone regeneration but it requires a second surgery with associated pain and complications, and is also limited by harvested bone quantity. Synthetic bone substitutes lack the osteoinductive properties to heal large bone defects. Cell therapies based on bone marrow or ex vivo expanded mesen

The Role of Extracellular Vesicles in Bone Regeneration and Associated Bone Diseases.

Extracellular vesicles (EVs) are nanoscale particles with a lipid bilayer membrane structure secreted by various cell types. Nearly all human cells secrete EVs, primarily mediating intercellular communication. In recent years, scientists have discovered that EVs can carry multiple biological cargos, such as DNA, non-coding RNAs (ncRNAs), proteins, cytokines, and lipids, and mediate intercellular signal transduction. Bone is a connective tissue with a nerve supply and high vascularization. The repair process after injury is highly complex, involving interactions among multiple cell types and bi

Aptamer-functionalized exosomes from bone marrow stromal cells target bone to promote bone regeneration.

In elderly people particularly in postmenopausal women, inadequate bone formation by osteoblasts originating from bone marrow mesenchymal stem cells (BMSCs) for compensation of bone resorption by osteoclasts is a major reason for osteoporosis. Enhancing osteoblastic differentiation of BMSCs is a feasible therapeutic strategy for osteoporosis. Here, bone marrow stromal cell (ST)-derived exosomes (STExos) are found to remarkably enhance osteoblastic differentiation of BMSCs in vitro. However, intravenous injection of STExos is inefficient in ameliorating osteoporotic phenotypes in an ovariectomy

Spontaneous regeneration of bone following mandibular ramus bone harvesting: a CBCT analysis.

The aim of this study was to evaluate healing at the donor site following autogenous block graft harvesting from the mandibular ramus. In this retrospective study, cone beam computed tomography images taken at different time-points were examined, and the volumes of preoperative and postoperative regions of interest were calculated in the software. Images were classified into four groups: 0-3 months, 4-6 months, 7-12 months, and >12 months post-surgery. To characterize the healing process over time, statistical analyses were conducted for both the 70% and 80% healing thresholds. Nearly half (n

Engineered Microalgae Promoting Angiogenesis for Diabetic Bone Regeneration.

Diabetic bone defects, characterized by hyperglycemia-induced accumulation of advanced glycation end products (AGEs), oxidative stress, and an inflammatory microenvironment, face impaired angiogenesis, and compromised bone regeneration. This study demonstrates that palladium (Pd)-engineered Spirulina platensis (SPP) effectively modulates the pathological diabetic bone microenvironment. Compared with native SP, SPP exhibits significantly enhanced capabilities: it reduces intracellular reactive oxygen species (ROS) and AGEs accumulation in endothelial cells, promotes pro-inflammatory M1 macropha

Alveolar Bone Regeneration: Smart Biomaterials and Physical Stimulation.

Large-area bone loss from disease, trauma, or congenital defects requires surgical procedures and bone grafting. Alveolar bone loss from severe periodontal disease and non-unions often demands immediate grafting. Treating large alveolar bone defects using grafts and substitutes is challenging due to the complex oral environment, infection risks, and unstable graft properties, which may compromise strength and bioactivity. Successful grafts must promote vasculature development and osteogenesis while maintaining mechanical stability at the graft site. Current bone graft substitutes are inadequat

Piezoelectric Smart Biomaterials for Craniomaxillofacial Bone-Regeneration Application.

This Perspective explores the transformative potential of piezoelectric biomaterials in addressing one of the most persistent challenges in craniomaxillofacial (CMF) bone regeneration: the reliable healing of large, irregular, and functionally demanding skeletal defects. Traditional approaches─autologous grafts, allografts, and inert synthetic scaffolds─are limited by donor-site morbidity, immunogenicity, mechanical insufficiency, and inadequate bioactivity. In contrast, piezoelectric materials offer a dynamic alternative, generating endogenous-like electrical cues in response to mechanical st

Polyphenols and Functionalized Hydrogels for Osteoporotic Bone Regeneration.

Osteoporosis induces severe oxidative stress and disrupts bone metabolism, complicating the treatment of bone defects. Current therapies often have side effects and require lengthy bone regeneration periods. Hydrogels, known for their flexible mechanical properties and degradability, are promising carriers for drugs and bioactive factors in bone tissue engineering. However, they lack the ability to regulate the local pathological environment of osteoporosis and expedite bone repair. Polyphenols, with antioxidative, anti-inflammatory, and bone metabolism-regulating properties, have emerged as a

Bone Regeneration and Oxidative Stress: An Updated Overview.

Bone tissue engineering is a complex domain that requires further investigation and benefits from data obtained over past decades. The models are increasing in complexity as they reveal new data from co-culturing and microfluidics applications. The in vitro models now focus on the 3D medium co-culturing of osteoblasts, osteoclasts, and osteocytes utilizing collagen for separation; this type of research allows for controlled medium and in-depth data analysis. Oxidative stress takes a toll on the domain, being beneficial as well as destructive. Reactive oxygen species (ROS) are molecules that in
