Smut infection of perennial hosts: the genome and the transcriptome of the Brassicaceae smut fungus Thecaphora thlaspeos reveal functionally conserved and novel effectors.

Biotrophic fungal plant pathogens can balance their virulence and form intricate relationships with their hosts. Sometimes, this leads to systemic host colonization over long time scales without macroscopic symptoms. However, how plant-pathogenic endophytes manage to establish their sustained systemic infection remains largely unknown. Here, we present a genomic and transcriptomic analysis of Thecaphora thlaspeos. This relative of the well studied grass smut Ustilago maydis is the only smut fungus adapted to Brassicaceae hosts. Its ability to overwinter with perennial hosts and its systemic pl

Disrupted FLOWERING LOCUS C alleles from weedy radish enhance the florigenic potential of rootstocks for graft-mediated floral induction in Brassicaceae.

Graft-mediated floral induction is a classical yet promising breeding technique that accelerates seed production and shortens generation time in crops. However, its application remains limited to a few species because floral induction is often unstable. Even among florally induced rootstocks, grafted scions can differ in their flowering response depending on rootstock genotype, but the genetic basis of this variation remains unclear. Here, using a Brassicaceae grafting model, we investigated two early-bolting radish (Raphanus sativus) accessions differing in their ability to induce flowering.

Resolving taxonomic complexity in the genus Boechera (Brassicaceae) using the Boechera Microsatellite Website: a case study of the rare triploid B. bodiensis.

BACKGROUND AND AIMS: The genus Boechera (rock cress) comprises ∼75 sexual diploid taxa and >355 genetically distinct hybrid lineages, many of which reproduce asexually through apomixis. This complex reproductive landscape poses substantial challenges for taxonomy, similar to those encountered in genera such as Taraxacum, Hieracium, Poa and Rubus. The Boechera Microsatellite Website (BMW) offers an extensive database and analytical tools that are proving instrumental in resolving these difficulties. Here, we demonstrate the utility of the BMW through analysis of Boechera bodiensis, a rare and p

Genes encoding light-harvesting chlorophyll a/b-binding proteins in papaya (Carica papaya L.) and insight into lineage-specific evolution in Brassicaceae.

Light-harvesting chlorophyll a/b-binding (Lhc) proteins comprise a plant-specific superfamily involved in photosynthesis and stress responses. Despite their importance, little is known in papaya (Carica papaya), an economically important tree fruit crop as well as a species close to the model plant arabidopsis (Arabidopsis thaliana). This study reports a first genome-wide analysis of Lhc superfamily genes in papaya, and a total of 28 members that represent four defined families or 26 orthologous groups were identified from the papaya genome. The superfamily number is comparable to 28 or 27 rep

Gas exchange, growth, and defense responses of invasive Alliaria petiolata (Brassicaceae) and native Geum vernum (Rosaceae) to elevated atmospheric CO2 and warm spring temperatures.

PREMISE OF STUDY: Global increases in atmospheric CO2 and temperature may interact in complex ways to influence plant physiology and growth, particularly for species that grow in cool, early spring conditions in temperate forests. Plant species may also vary in their responses to environmental changes; fast-growing invasives may be more responsive to rising CO2 than natives and may increase production of allelopathic compounds under these conditions, altering species' competitive interactions. METHODS: We examined growth and physiological responses of Alliaria petiolata, an allelopathic, inva

The Pieris climate accords: how climate change shapes molecular interactions between the butterfly Pieris rapae (Lepidoptera: Pieridae) and its host plant Arabidopsis thaliana (Brassicales: Brassicaceae).

Global temperatures are projected to rise by approximately 1.5 °C between 2030 and 2050, driven by increasing atmospheric carbon dioxide (CO2) levels. Such rapid climate change is predicted to drive correspondingly rapid change in animals and plants, particularly those that humans depend on for food and security. Here, we investigated how increased temperature and CO2 may affect herbivore-host plant interactions using controlled experiments with a widespread agricultural pest, the cabbage white butterfly Pieris rapae, and a model host plant, Arabidopsis thaliana. Potential effects of Alliaceae and Brassicaceae edible plants on blood glucose levels in patients with type 2 diabetes: A systematic review and meta-analysis of clinical trials.

Type 2 diabetes (T2D) is one of the main leading causes of mortality worldwide, and its global prevalence will increase within the next years. Many pre-clinical studies demonstrated the potential effects of edible plants from Alliaceae and Brassicaceae on the regulation of blood glucose levels. Of note, secondary metabolites from Alliaceae and Brassicaceae share the ability to slowly release hydrogen sulfide (H2S), which is emerging as a crucial modulator of the "glucose-insulin system". However, the results of clinical studies evaluating the effects of such edible plants on glycaemic control

Sub-proteome S-nitrosylation analysis in Brassica juncea hints at the regulation of Brassicaceae specific as well as other vital metabolic pathway(s) by nitric oxide and suggests post-translational modifications cross-talk.

Abiotic stress affects the normal physiology of the plants and results in crop loss. Brassica juncea is an oil yielding crop affected by abiotic stress. In future, over 30% yield loss by abiotic stress is predicted in India. Understanding the mechanism of plant response to stress would help in developing stress tolerant crops. Nitric oxide (NO) is now viewed as a remarkably important signaling molecule, involved in regulating stress responses. S-Nitrosylation is a NO based post-translational modification (PTM), linked with the regulation of many physiologically relevant targets. In the last de

Assessing the potential of turnip rape and radish cover crops as trap crops for Psylliodes chrysocephala in commercial field production conditions.

BACKGROUND: Cabbage stem flea beetle (CSFB; Psylliodes chrysocephala) is a major pest of winter oilseed rape (WOSR; Brassica napus), during autumn. Previous studies have shown some species of Brassicaceae, for example, turnip rape (Brassica rapa) and radish (Raphanus sativus var longipinnatus), to be attractive for this pest. Farmers' crop rotations regularly include the use of cover crops before spring sowing to avoid leaving the soil bare and unprotected. As these cover crops are generally destroyed in winter, incorporating these Brassicaceae into cover crops mixes could: (i) attract CSFB an

Simultaneous quantification of 14 glucosinolates in rapeseeds by ultra high performance liquid chromatography - Tandem mass spectrometry.

Glucosinolates' species and content serve as crucial indicators for Brassicaceae plants and their products. However, endogenous enzymes in Brassicaceae can hydrolyze glucosinolates, influencing their quantitative detection. Our study utilized rapeseed, a vital Brassicaceae oil crop, to establish a novel UHPLC-MS/MS method for glucosinolate determination, incorporating a pretreatment approach with enzyme inhibition. This enabled intact glucosinolate determination at room temperature. The straightforward process involved homogenizing samples with saturated sodium chloride and methanol, followed

Postharvest senescence of green vegetables and the role of glutathione metabolism: current state of knowledge and future research opportunities.

The rapid progression of senescence contributes to the postharvest perishability of green vegetables including Brassicaceae commodities. Tissue yellowing due to chlorophyll degradation is a hallmark feature of senescing green vegetables and is associated with the accumulation of reactive oxygen species. Moreover, oxidative stress metabolism in vegetables is exacerbated by postharvest temperature abuse and the senescence-related phytohormone ethylene. Glutathione is a primary antioxidant that detoxifies reactive oxygen species in plants. Glutathione status in postharvest vegetables is partially

Lineage-specific evolution of Methylthioalkylmalate synthases (MAMs) involved in glucosinolates biosynthesis.

Methylthioalkylmalate synthases (MAMs) encoded by MAM genes are central to the diversification of the glucosinolates, which are important secondary metabolites in Brassicaceae species. However, the evolutionary pathway of MAM genes is poorly understood. We analyzed the phylogenetic and synteny relationships of MAM genes from 13 sequenced Brassicaceae species. Based on these analyses, we propose that the syntenic loci of MAM genes, which underwent frequent tandem duplications, divided into two independent lineage-specific evolution routes and were driven by positive selection after the divergen

MYB Superfamily in Brassica napus: Evidence for Hormone-Mediated Expression Profiles, Large Expansion, and Functions in Root Hair Development.

MYB proteins are involved in diverse important biological processes in plants. Herein, we obtained the MYB superfamily from the allotetraploid Brassica napus, which contains 227 MYB-related (BnMYBR/Bn1R-MYB), 429 R2R3-MYB (Bn2R-MYB), 22 R1R2R3-MYB (Bn3R-MYB), and two R1R2R2R1/2-MYB (Bn4R-MYB) genes. Phylogenetic analysis classified the Bn2R-MYBs into 43 subfamilies, and the BnMYBRs into five subfamilies. Sequence characteristics and exon/intron structures within each subfamily of the Bn2R-MYBs and BnMYBRs were highly conserved. The whole superfamily was unevenly distributed on 19 chromosomes a

Cyanide in the chemical arsenal of garlic mustard, Alliaria petiolata.

Cyanide production has been reported from over 2500 plant species, including some members of the Brassicaceae. We report that the important invasive plant, Alliaria petiolata, produces levels of cyanide in its tissues that can reach 100 ppm fresh weight (FW), a level considered toxic to many vertebrates. In a comparative study, levels of cyanide in leaves of young first-year plants were 25 times higher than in leaves of young Arabidopsis thaliana plants and over 150 times higher than in leaves of young Brassica kaber, B. rapa, and B. napus. In first-year plants, cyanide levels were highest in

Evolution of fruit development genes in flowering plants.

The genetic mechanisms regulating dry fruit development and opercular dehiscence have been identified in Arabidopsis thaliana. In the bicarpellate silique, valve elongation and differentiation is controlled by FRUITFULL (FUL) that antagonizes SHATTERPROOF1-2 (SHP1/SHP2) and INDEHISCENT (IND) at the dehiscence zone where they control normal lignification. SHP1/2 are also repressed by REPLUMLESS (RPL), responsible for replum formation. Similarly, FUL indirectly controls two other factors ALCATRAZ (ALC) and SPATULA (SPT) that function in the proper formation of the separation layer. FUL and SHP1/

Eruca sativa Mill. seed extract promotes anti-obesity and hypoglycemic effects in mice fed with a high-fat diet.

Obesity is currently considered a major source of morbidity, with dramatic complications on health status and life expectancy. Several studies demonstrated the positive effects of Brassicaceae vegetables on obesity and related diseases, partially attributing these beneficial properties to glucosinolates and their derivatives isothiocyanates. Recently, isothiocyanates have been described as a hydrogen sulfide (H2 S)-releasing moiety, suggesting that H2 S may be at least in part responsible for the beneficial effects of Brassicaceae. In this work, the metabolic effects of an extract obtained fro

Evolution of nitrilases in glucosinolate-containing plants.

Nitrilases, enzymes that catalyze the hydrolysis of organic cyanides, are ubiquitous in the plant kingdom. The typical plant nitrilase is a nitrilase 4 homolog which is involved in the cyanide detoxification pathway. In this pathway, nitrilase 4 converts beta-cyanoalanine, the intermediate product of cyanide detoxification, into asparagine, aspartic acid and ammonia. In the Brassicaceae, a new family of nitrilases has evolved, the nitrilase 1 homologs. These enzymes are not able to use beta-cyanoalanine as a substrate. Instead, they display rather broad substrate specificities and are able to

Revisiting SA biosynthesis: new post-PAL route in plant immunity.

Salicylic acid (SA), the active component of aspirin, is a crucial defense phytohormone that coordinates both local and systemic acquired resistance. For decades, SA biosynthesis was attributed to two pathways: (i) the isochorismate synthase pathway, which is dominant in arabidopsis (Arabidopsis thaliana) and other Brassicaceae, and (ii) the phenylalanine ammonia-lyase (PAL) pathway, common in most other plants. However, the PAL pathway remained biochemically incomplete due to the lack of a molecularly identified benzoic acid 2-hydroxylase. Recent findings fill this gap by identifying a conser
