Anticancer effect of bromelain with and without cisplatin or 5-FU on malignant peritoneal mesothelioma cells.

Malignant peritoneal mesothelioma (MPM) is a rare neoplasm of the peritoneum, causally related to asbestos exposure. Nonspecific symptoms with a late diagnosis results in poor survival (<1 year). Treatment with cytoreductive surgery and hyperthermic intraperitoneal chemotherapy has improved survival in some patients (median 3-5 years). Hence, new therapies are urgently needed. MUC1 is a glycosylation-dependent protein that confers tumours with invasiveness, metastasis and chemoresistance. Bromelain (cysteine proteinase) hydrolyses glycosidic bonds. Therefore, we investigated the antitumour eff

Effect of wine inhibitors on free pineapple stem bromelain activity in a model wine system.

The influence of potential inhibitors, naturally present in wine, on the activity of stem bromelain was investigated in order to evaluate the applicability of this enzyme for protein stabilization in white wine. Bromelain proteolytic activity was tested against a synthetic substrate (Bz-Phe-Val-Arg-pNA) in a model wine system after adding ethanol, sulfur dioxide (SO(2)), skin, seed, and gallic and ellagic tannins at the average range of their concentration in wine. All the inhibitors of stem bromelain activity tested turned out to be reversible. Ethanol was a competitive inhibitor with a rathe

Investigation of the effects of a Kiperin Bromelain & Papain complex in normal colon, mammary, and skin cells.

Bromelain is a proteolytic enzyme complex naturally found in the stem, fruit, and juice of pineapple, while papain is a proteolytic enzyme derived from the latex of the papaya plant. These bioactive components have attracted increasing scientific interest due to their reported roles in cell proliferation and wound healing, suppression of oxidative stress through free radical scavenging, and apoptosis-related pathways. Although bromelain and papain have been associated with various health-related effects, their effects on healthy cells remain largely unexplored. In this study, different concent

Protective effect of bromelain on some metabolic enzyme activities in tyloxapol-induced hyperlipidemic rats.

Elevation of one or more plasma lipids, such as phospholipids, cholesterol esters, cholesterol, and triglycerides, is known as hyperlipidemia. In humans and experimental animals, bromelain, the primary active ingredient isolated from pineapple stems, has several positive effects, including anti-tumor growth, anticoagulation, and anti-inflammation. Hence, the purpose of this study was to determine the possible protective impact of bromelain on some metabolic enzymes (paraoxonase-1, glutathione S-transferase, glutathione reductase, sorbitol dehydrogenase [SDH], aldose reductase [AR], butyrylchol

Extraction and crosslinking of bromelain aggregates for improved stability and reusability from pineapple processing waste.

The present study is first of its kind that focuses upon the extraction of bromelain from pineapple core waste and stabilising it as insoluble cross-linked aggregates. The influence of process variables such as the choice of precipitant, type of cross-linker, concentration of cross-linker and the reaction time for cross-linking step was investigated upon the activity recovery of bromelain cross-linked aggregates. The optimization of this biocatalyst preparation specifically recovered 87% of the enzymatic activity available in pineapple core waste by ammonium sulphate (60%, w/v) precipitation f

Development of a contaminated ischemic porcine wound model and the evaluation of bromelain based enzymatic debridement.

OBJECTIVES: There are no well accepted animal models of chronic wounds, limiting advances in understanding and treatment of chronic ulcers. We developed a porcine wound model which combines multiple factors involved in chronic wounds to create a contaminated necrotic eschar and evaluated the debriding efficacy of a novel bromelain based enzymatic debriding agent (EscharEx). METHODS: Contaminated ischemic wounds were created on the flanks of domestic pigs by 'sandwiching' the skin between 2 'O' rings (1 placed on the surface of the skin and the other underneath the skin) for 24h prior to derma

Effects of Bromelain in Maternal Deprivation and Social Isolation Models of Schizophrenia in Male BALB/c Mice.

Bromelain is a mixture of enzymes found in several parts of the pineapple plant, including the stem, fruit, leaves, and peel. Its use was recommended to slow down the progression of neurological disorders such as Parkinson's disease. Although studies have examined the beneficial effects of bromelain (Br) in neurological disorders, its detailed benefits in schizophrenia are still poorly understood. We, thus, modeled schizophrenia using two developmental methods. In the first experiment, postweaning social isolation (SI) method where male mice were weaned at postnatal day (PND) 21 and isolated i

Sequential Bromelain-Based Enzymatic Debridement in a Near-Total (97%) TBSA Burn: A Case Report.

Introduction: Early debridement of burn eschar is a cornerstone of care for patients with extensive burns, yet early large-area surgical excision may be limited by blood loss, physiological instability, and substantial logistical demands. Evidence on bromelain-based enzymatic debridement involving treatment areas greater than 15% total body surface area (TBSA) remains scarce. Case Report: A 38-year-old man sustained partial- and full-thickness burns involving 97% TBSA, together with inhalation injury. Sequential enzymatic debridement with NexoBrid was performed at the bedside in five 4-h sessi

Effect of bromelain on ischemia-reperfusion injury in the torsion model created in polycystic and normal ovarian tissues.

PURPOSE: Due to its increased volume, polycystic ovarian tissue is more prone to torsion than normal ovarian tissue. In treating ovarian torsion, detorsion is applied to ensure oxygenation of hypoxic tissues. However, the resulting oxygen radicals cause tissue damage. Bromelain is a substance obtained from pineapple, and studies in the literature show it is used as an antioxidant. This study aimed to evaluate the damage caused by ischemia-reperfusion (I/R) in the torsion-detorsion model created in normal and polycystic ovarian tissue and investigate the role of bromelain in this damage. Bromelain enhances nitric oxide bioavailability: Bradykinin's link to TRPV1/Ca2+ /AMPK/autophagy signaling.

Bromelain, a protease enzyme extracted from the pineapple stem, is suggested to protect against atherosclerosis, non-alcohol fatty liver diseases, and coagulation dysfunction. However, the mechanism underlying the vascular protection of bromelain in the cardiovascular system is not fully understood. In this study, we explored the role of the kininogen-bradykinin system in bromelain-mediated nitric oxide (NO) bioavailability in endothelial cells (ECs). NO bioavailability was examined by Griess's assay, western blot analysis was used to assess protein expression, the level of urea and arginine w

Effective Use of Bromelain and Acetylcysteine (BromAc®) for Treatment of Perigastric Pseudomyxoma Peritonei: A Case Report.

BACKGROUND/AIM: Pseudomyxoma peritonei (PMP) is a rare clinical condition of progressive peritoneal mucin accumulation. PMP has a reasonable survivability but with a notable risk of tumour recurrence. Standard treatment, including for tumour relapse, aims for a cure with complete cytoreductive surgery with hyperthermic intraperitoneal chemotherapy. In the case of tumour recurrence, surgery becomes progressively complex, and some patients are not suitable for surgery either due to patient preference or morbidity and mortality risk. BromAc® is an emerging, novel mucolytic combination therapy com

Influence of UV radiation on molecular structure and catalytic activity of free and immobilized bromelain, ficin and papain.

Our research has shown that the degree of photosensitivity of the cysteine proteases can be arranged in the following order: bromelain → ficin → papain. After the UV irradiation with 151 J·m-2 intensity of a bromelain solution, the enzyme activity has increased. No decrease in the catalytic capacity and the change in the size of the molecule was recorded in the 151-6040 J·m-2 range of irradiation intensities. A decrease in the catalytic capacity of ficin and the increase of its globule size occurred after exposure to a radiation of 3020 J·m-2 intensity. The decrease in papain activity was obse

Umami characterization and molecular simulation of novel peptides from Pleurotus citrinopileatus Singer bromelain hydrolysate.

Pleurotus citrinopileatus Singer is the promising precursor of umami peptides. This study aimed to isolate P. citrinopileatus Singer-derived umami peptides, and then explore their taste characteristics and umami mechanism. Four novel umami peptides (LLVTVEDELRV, LDDWR, ELFEPT, and ENLLEK) were separated and identified from bromelain hydrolysate, and then predicted their umami potentials through umami fragment frequencies and T1R1/T1R3 receptor docking. Their umami characteristics were subsequently confirmed in sensory evaluation and electronic tongue verification. Particularly, LLVTVEDELRV (0.
