Sequencing and phylogenetic characterization of Brucella canis isolates, Ohio, 2016.

Brucella canis is one of zoonotic pathogens causing infection in human. In this study, we isolated and sequenced 38 B. canis strains from 11 cases. Core genome multilocus sequence typing analysis classified all B. canis isolates into two genogroups, GI and GII. All 38 isolates cluster together forming a 2016 Ohio cluster, in which they form five subclusters reflecting their geographical differences. Unlike GI, the isolates of the GII are from diverse geographical locations including Asia, America, Africa, and Europe and form Asia and South America clusters. Overall, our findings could be usefu

The vaccine candidate BLSOmp31 protects mice against Brucella canis infection.

Canine brucellosis represents a major reproductive problem worldwide and it is considered a zoonotic disease. New approaches are therefore urgently needed to develop an effective and safe immunization strategy against Brucella canis. In the present study, BALB/c mice were subcutaneously immunized with the recombinant chimera rBLSOmp31 formulated in different adjuvants. The different strategies induced a vigorous immunoglobulin G (IgG) response, with high titers of IgG1 as well as IgG2. Besides, spleen cells from rBLSOmp31-immunized mice produced gamma interferon and IL-4, suggesting the induct

"MLVA_5: A reduced marker scheme for phylogeographic analysis of Brucella canis".

Canine brucellosis, caused by Brucella canis, is a globally significant zoonosis. This study characterized the genetic diversity of B. canis in Argentina and South America using the MLVA_16 scheme, comparing 110 Latin American strains with 84 Asian strains (from Korea and China). The analysis revealed high overall diversity (Hunter-Gaston Index: 0.97) and marked geographical structuring. Using LASSO logistic regression, we identified a minimal panel of five markers (MLVA-5: BRUCE19, BRUCE18, BRUCE11, BRUCE07, BRUCE16) that optimally preserves the phylogeographic signal on a global scale. First detection of Brucella canis infections in a breeding kennel in Austria.

Brucella canis occurs almost worldwide and is a potential danger to the health of dogs and humans. The pathogen was detected in the placenta and fetuses of a Standard Poodle by direct culture and immunohistochemistry. Further, Brucellae were also isolated from the blood samples of two asymptomatic female Medium Poodles. The isolates were identified as B. canis by conventional microbiological methods and a novel Bruce-ladder multiplex PCR. Genotyping was performed by multiple locus variable number tandem repeats analysis (MLVA).

Clinical resolution of Brucella canis-induced ocular inflammation in a dog.

A 2-year-old spayed female mixed-breed dog was referred with an 8-month history of recurrent bilateral anterior uveitis and hyphema secondary to infection with Brucella canis. Treatment with doxycycline PO and atropine and prednisone acetate topically prior to referral had failed to resolve the ocular inflammation. Successful control of ocular inflammation was achieved after initiation of treatment with gentamicin SC and ciprofloxacin and doxycycline PO. The finite indirect fluorescent antibody (IFA) titer was reduced by half, and results of an agar gel immunodiffusion (AGID) test became negat

[A serologic study of human Brucella canis infections in the bursa region].

Serum samples of 123 patients of various ages and both sexes from Bursa area were tested by Mercaptoethanol Tube Agglutination Test on a random basis for Br. canis antibodies. Two patients, one male and one female, showed high titers (1:400) in mercaptoethanol tube agglutination test. A titer of 1:200 or higher was considered significant and indicative of active infection. Br. canis infection in man in Turkey is reported for the first time.

First Isolation of Brucella canis from a breeding kennel in Italy.

Brucella canis has been isolated for the first time in Italy in a commercial breeding kennel. It was diagnosed after a deep investigation related to the onset of reproductive disorders. Animals were tested with direct and indirect techniques. The agent was first detected in two Chihuahua aborted foetuses by direct culture. Further, it was also isolated from blood samples of dogs hosted in the kennel, which also showed reaction to conventional serological tests (microplate serum agglutination test). The isolates were identified as B. canis by standard microbiological methods and a Bruce‑ladder

[Comparative study of the antigenic composition of Brucella canis strains by immunoblotting].

Brucella cultures isolated from sick dogs have not been properly studied in Russia up to the present time. In 1994 a culture has been isolated from aborted fetus of a dog. Investigations by the traditional methods referred it to Brucella genus, species canis (strain K-01). Reference strain B. canis RM6/66 and B. canis K-01 were identical by the profiles of protein antigens in immunoblotting with a set of antibrucellosis sera. B. canis, B. suis, B. abortus, and B. melitensis. However, immunoblotting with sera to B. canis showed the similarity of B. canis cultures with the reference strain B. su

Early detection of Brucella canis via quantitative polymerase chain reaction analysis.

Canine brucellosis is a reportable zoonotic disease that can lead to canine reproductive losses and human infection through contact with infected urine or other genitourinary secretions. Although many locations require testing and euthanasia of positive dogs, current diagnosis is limited by the time required for seroconversion, for example, presence of B. canis-specific antibodies. The goal of this study was to determine the diagnostic ability of Brucella canis-specific quantitative polymerase chain reaction (qPCR) assay to detect B. canis in field samples prior to serological positivity for f

Seroepidemiological investigation of Brucella canis antibodies in different human population groups.

A study was conducted to determine the prevalence of Brucella canis antibodies in specified groups based on their exposure to dogs. The method used was a microtiter technique, and the presence of antibodies at a 1:12 or greater dilution of serum was considered a positive test. Eleven (5.7%) of the newborn infants had evidence of maternal antibodies, and 67.8% of the persons with an average exposure to dogs had B. canis antibodies, with a 62.1% prevalence in males and a 72.4% prevalence in females (P less than or equal to 0.001). Veterinarians had a much higher rate of infection (72.6%) than ma

Identification of Brucella by ribosomal-spacer-region PCR and differentiation of Brucella canis from other Brucella spp. pathogenic for humans by carbohydrate profiles.

Molecular and chemical characteristics often provide complementary information in the differentiation of closely related organisms. The genus Brucella consists of a highly conserved group of organisms. Identification of the four species pathogenic in humans (Brucella melitensis, Brucella abortus, Brucella suis, and Brucella canis) is problematic for many clinical laboratories that depend primarily on serology and phenotypic characteristics to differentiate species. PCR amplification of the 16S-23S ribosomal DNA interspace region was evaluated for species-specific polymorphism. B. abortus, B. m

Canine brucellosis due to Brucella canis: description of the disease and control measures.

Brucellosis is a contagious disease caused by bacteria of the genus Brucella, which can affect different animal species. Dogs may occasionally be infected with B. abortus, B. melitensis or B. suis, or by the endemic form of the disease, caused by B. canis. Among the brucellosis‑affecting domestic animals, that of the dog is certainly the least frequent, but also the least studied. Canine brucellosis due to B. canis represents the dog‑specific brucellosis, both because it is the main susceptible animal species, and because it constitutes its fundamental reservoir of infection. The disease can a
