Fungal biotransformation of chlorogenic and caffeic acids by Fusarium graminearum: New insights in the contribution of phenolic acids to resistance to deoxynivalenol accumulation in cereals.

Fusarium Head Blight and Gibberella Ear Rot, mainly caused by the fungi Fusarium graminearum and Fusarium culmorum, are two of the most devastating diseases of small-grain cereals and maize. In addition to yield loss, these diseases frequently result in contamination of kernels with toxic type B trichothecenes. The potential involvement of chlorogenic acid in cereal resistance to Fusarium Head Blight and Gibberella Ear Rot and to trichothecene accumulation was the focus of this study. The effects of chlorogenic acid and one of its hydrolyzed products, caffeic acid, on fungal growth and type B

Molecular cloning and characterization of caffeic acid O-methyltransferase from Acorus calamus.

Volatile phenylpropenes, including α- and β-asarone, are characteristic aromas of sweet flag (Acorus calamus) and have been used as ingredients in pharmaceutical applications. However, studies on the biosynthetic enzymes involved in the production of volatile phenylpropenes in A. calamus remain limited. In this study, we analyzed volatile phenylpropenes, including α- and β-asarone, in the A. calamus plants. Using RNA-sequencing analysis, we identified a gene encoding S-adenosyl-L-methionine-dependent O-methyltransferase (AcCOMT), which converts caffeic acid to ferulic acid via the methylation

The flavanoide caffeic acid phenethyl ester blocks 6-hydroxydopamine-induced neurotoxicity.

Parkinson's disease (PD) is a neurodegenerative disorder characterized by progressive loss of dopaminergic (DA) neurons of the substantia nigra pars compacta. 6-Hydroxydopamine (6-OHDA) is specific to dopaminergic neurons in intrastriatal rodent models. It induces neuronal death either via uncoupling mitochondrial oxidative phosphorylation resulting in energy deprivation or alternatively, is associated with its ability to produce hydrogen peroxide, hydroxyl and superoxide radicals. Caffeic acid phenethyl ester (CAPE), an antioxidant flavanoid, has antiviral, anti-inflammatory, antioxidant, and

Copper-Induced Interactions of Caffeic Acid and Sinapic Acid to Generate New Compounds in Artificial Biological Fluid Conditions.

Active ingredients may be ingested through foods, and they can cause several interactions in the human body. Although drug-drug or drug-food interactions are evaluated before the approval of medicines, several functional food interactions are not well-documented because of the wide range of possible combinations of interactions. In this study, we examined the chemical reactions between hydroxycinnamic acids (HCAs), a group of polyphenols, and metal ions in artificial gastric juice or artificial intestinal fluid. Caffeic acid (CaA) and sinapic acid (SA) reacted with copper ions under artificial

Preparation and characterization of caffeic acid-grafted electrospun poly(L-lactic acid) fiber mats for biomedical applications.

Caffeic acid (CA) was chemically immobilized onto the surfaces of the individual electrospun poly(l-lactic acid) (PLLA) fibers to enhance the hydrophilicity and impart the antioxidant activity to the obtained fibrous membranes. This was done in two sequential steps. First, amino groups were covalently introduced onto the surfaces through the reaction with 1,6-hexamethylenediamine (HMD). In the second step, the amino moieties reacted with CA, which had been preactivated sequentially with 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC) and N-hydroxysuccinimide (NHS). The success of the react

Periplasmic Engineering Enhances Terminal Hydroxylation for Efficient Caffeic Acid Biosynthesis in Escherichia coli.

Caffeic acid (CA) is a valuable phenylpropanoid with applications in food, pharmaceutical, and chemical industries. Microbial production of CA is often limited by the terminal hydroxylation catalyzed by 4-hydroxyphenylacetate 3-monooxygenase (HpaBC). Here, we constructed a de novo CA biosynthetic pathway in Escherichia coli and enhanced production through systematic metabolic and spatial engineering. Optimization of l-tyrosine supply and HpaBC expression increased CA production to 61.0 mg/L. However, further enhancement of glucose uptake and precursor supply was insufficient, indicating that h
