Differential effects of cooking in open cups and in sealed cellophane bags on the residual cyanide content of moi-moi, a processed legume product.

Bean pastes from the black eyed bean (Vigna unguiculata), pigeon pea (Cajanus cajan) and Ife brown (V. unguiculata) were processed into a popular West African snack moi-moi, cooked in open aluminium cups and in sealed, thin, transparent cellophane bags. The residual total, free and bound cyanide levels of moi-moi from both processes were determined using a combination of enzymatic and colorimetric procedures. Results showed that in all cases, cooking of moi-moi in open aluminium cups caused retention of significantly higher (P<0.05) residual free cyanide than cooking in sealed cellophane bags.

Enhanced Accumulation of Health-Promoting Cajaninstilbene Acid in Pigeon Pea Hairy Root Cultures Cocultured with an Endophytic Fungus during Early Stages of Colonization.

Endophytic fungi can effectively regulate the biosynthesis of health-beneficial metabolites in plants. However, few studies have revealed how the accumulation of host metabolites varies during interactions with endophytic fungi. Here, pigeon pea hairy root cultures (PPHRCs) were cocultured with an endophytic fungus Penicillium rubens to explore the impact on the biosynthesis and accumulation of cajaninstilbene acid (CSA). The results showed that CSA accumulation in PPHRCs increased significantly (15.29-fold) during the early stages of P. rubens colonization (fungal attachment and invasion phas

Cocultivation of pigeon pea hairy root cultures and Aspergillus for the enhanced production of cajaninstilbene acid.

Pigeon pea hairy root cultures (PPHRCs) have been proven to be a promising alternative for the production of health-beneficial phenolic compounds, such as the most important health-promoting compound, i.e., cajaninstilbene acid (CSA). In this study, PPHRCs were cocultured with live Aspergillus fungi for further improving phenolic productivity via biological elicitation. Aspergillus oryzae CGMCC 3.951 (AO 3.951) was found to be the optimal fungus that could achieve the maximum increment of CSA (10.73-fold increase) in 42-day-old PPHRCs under the inoculum size of mycelia 0.50% and cocultivation

Sporopollenin-chitosan microspheres loaded with an endophytic fungus Talaromyces neorugulosus R-209 for promoting development and controlling root rot in pigeon pea.

Plant-beneficial microbes can be effective as biological agents for promoting development and controlling diseases in plants. However, direct inoculation of non-encapsulated plant-beneficial microbes into the soils can affect their vitality and efficacy. A novel bio-based encapsulant, sporopollenin-chitosan microspheres (SCMs), was developed to load an endophytic fungus Talaromyces neorugulosus R-209 with antagonistic activities against the root rot pathogen (Rhizoctonia solani AG4) and plant growth-promoting functions. The results showed that T. neorugulosus R-209 encapsulated in SCMs (TnR-20

Design and Synthesis of Cajanine Analogues against Hepatitis C Virus through Down-Regulating Host Chondroitin Sulfate N-Acetylgalactosaminyltransferase 1.

There still remains a need to develop new anti-HCV agents with distinct mechanism of action (MOA) due to the occurrence of resistance to direct-acting antiviral agents (DAAs). Cajanine, a stilbenic component isolated from Cajanus cajan L., was identified as a potent HCV inhibitor by phenotypic screening in this work (EC50 = 3.17 ± 0.75 μM). The intensive structure optimization provided significant insights into the structure-activity relationships. Furthermore, the MOA study revealed that cajanine inhibited HCV replications via down-regulating a cellular protein chondroitin sulfate N-acetylgal

Harnessing the power of native biocontrol agents against wilt disease of Pigeonpea incited by Fusarium udum.

Fusarium wilt, caused by (Fusarium udum Butler), is a significant threat to pigeonpea crops worldwide, leading to substantial yield losses. Traditional approaches like fungicides and resistant cultivars are not practical due to the persistent and evolving nature of the pathogen. Therefore, native biocontrol agents are considered to be more sustainable solution, as they adapt well to local soil and climatic conditions. In this study, five isolates of F. udum infecting pigeonpea were isolated from various cultivars and characterized morphologically and molecularly. The isolate from the ICP 8858

Pangenome-wide insights into eukaryotic translation initiation factors (eIF) genes uncover targets for sterility mosaic disease resistance engineering in pigeonpea.

Sterility mosaic disease (SMD) poses a significant threat to pigeonpea production. Although resistant cultivars are widely used, the rapid evolution of viral pathogens often renders host resistance ineffective. Targeting host susceptibility factors-particularly eukaryotic translation initiation factors (eIFs) that many plant viruses exploit for replication-offers a promising alternative. Although eIF proteins are well characterized in several model crops, their roles in pigeonpea remain largely unexplored. In this study, we conducted a comprehensive analysis of the eIF gene family in pigeonpea

Immobilized pigeon pea urease on chitosan-PEG biocomposite for concurrent urea sensing in milk and blood samples.

The ubiquitous hydrolytic enzyme urease converts urea into ammonia and carbon dioxide, making it an essential indicator for dairy product quality and kidney health. We developed a biocomposite sensor to meet this diagnostic necessity. This innovation strategically incorporates pigeon pea seed urease into chitosan-PEG. We verified our predicted model with a 91.30 % immobilization efficiency via our thorough process. The immobilized urease showed strong catalytic activity, with a Vmax of 3194 μmol/min/mg and a Km of 0.373 mM. Optimal activity was observed at 90 °C and pH 8.0. Additionally, it ex

In vitro antihelmintic effect of fifteen tropical plant extracts on excysted flukes of Fasciola hepatica.

BACKGROUND: Fasciolosis due to Fasciola hepatica is the most important hepatic disease in veterinary medicine. Its relevance is important because of the major economical losses to the cattle industry such as: reduction in milk, meat and wool production; miscarriages, anemia, liver condemnation and occasionally deaths, are estimated in billons of dollars. The emergence of fluke resistance due to over or under dosing of fasciolides as well as environmental damage produced by the chemicals eliminated in field have stimulated the need for alternative methods to control Fasciola hepatica. The aim o

Melatonin enhances salt tolerance by promoting CcCAD10-mediated lignin biosynthesis in pigeon pea.

Melatonin plays a crucial role in enhancing plant resistance to salt stress by regulating biosynthesis of specialized metabolites. Phenylpropanoids, especially lignin, contribute to all aspects of plant responses toward biotic and abiotic stresses. However, the crosstalk between melatonin and lignin is largely unknown in pigeon pea under salt stress. In this study, the cinnamyl alcohol dehydrogenase CcCAD10 was identified to be involved in melatonin treatment and salt stress. The content of lignin was significantly increased in CcCAD10 over-expression (OE) lines, the enhanced antioxidant enzym
